  • MyoD, expressed in skeletal muscle lineages of vertebrate embryo, is one of muscle-specific basic helix-loop-helix (bHLH) transcription factors, which plays a key role in the determination and differentiation of all skeletal muscle lineages. In this study, a cDNA of grass carp MyoD was cloned and characterized from total RNA of grass carp embryos by RT-PCR. The full-length cDNA of grass carp MyoD is 1597 bp. The cDNA sequence analysis reveals an open reading frame of 825 bp coding for a protein of 275 amino acids, which includes a bHLH domain composed of basic domain (1-84th amino acids) and HLH domain (98-142th amino acids), without signal peptide. Then the MyoD cDNA of grass carp was cloned to yeast expression vector pPICZ$\alpha$A and transformed into P. pastoris GS115 strain, the recombinant MyoD protein with a molecular weight of about 31KD was obtained after inducing for 2d with 0.5% methanol in pH 8.0 BMGY medium, and the maximum yield was about 250 mg/L in shaking-flask fermentation. The results were expected to benefit for further studies on the crystal structure and physiological function of fish MyoD.

  • The purpose of this study is to investigate the actual conditions, to find problems, and to present an improvement plan of interior landscape in the subway stationis. Moreoiver, it will presume the possibilties to develop the subway and ways to utilie underground space effectively. The results are summarized as follows A light intensity was recorded 0-100 Lux as the lowest light and 500 Lux below as the highest in 61 subway stations. An average temperature of 12 $^{\circ}C$and an air humidity of 38% were recorded over a 4 month period from January to April, but includes a drastic variaton between -8.5$^{\circ}C$ and 21.5$^{\circ}C$. Soil acidity of pH 7 below and soil moisture of 1-2 wet degree were apeared in subway stations mostly. Plant forms consisted of artificial flower & flower. Plant species were recorded at a total of 54 species and appeared in the order of Araucaria heteropylla, Ophiophogon jaburan, Aspidistra elatior cv. 'Asahi' and Hedera helix. Plant height was, for the most part, below 0.5m. Plant species that was fined of conditions were Palm, Camellia japonica, Araucaria heterophylla as a high plant, dracaena fragrans, dracaena deremensis cv. Wakneckii as a middle plant, and Ophiopogon jaburan, Hedra helix, ytomium falcatum, Aspidistra elatior cv. Asahi as a low plant. It used to water materials such as small pool, small cascade, water cycles and natural materials such as natural rock, small rock, sand, bark and animal materials such as squirrels, birds, goldfish as an object for plants in the subway stations. From these actual conditions, First of all, It must make up physical environments such as light, temperature humidity, soil for plant growth, and is important to chooce suitable indoor plants and draw up systematic management in the subway environments. Also, it change plants frequently and uses variable objects for subway stationi individuatism, Moreover, indoor plants with strong environmental adaptation abities such as shade tolerance, drought tolerance and cold tolerance need to develope variable species possibly. If these improvements occur, utilization and amenity of subway stationis will increase, according to the use of interior landscape.

  • Background: Intradural-intramedullary intervertebral disc extrusion (IIVDE) is a rare condition of intervertebral disc disease. However, the diagnosis of IIVDE is challenging because the prognosis and imaging characteristics are poorly characterized. Objectives: We aimed to describe the clinical and imaging characteristics of tentatively diagnosed IIVDE in dogs to assess the prognostic utility of neurological grade and magnetic resonance imaging (MRI) findings. Methods: Twenty dogs were included in this retrospective cohort study. Results: Nonchondrodystrophic breeds (n = 16) were more predisposed than chondrodystrophic breeds. Most dogs showed acute onset of clinical signs. Neurological examination at admission showed predominant non-ambulatory paraparesis (n = 9); paresis (n = 16) was confirmed more frequently than paralysis (n = 4). Follow-up neurological examination results were only available for 11 dogs, ten of whom showed neurological improvement and 8 showed successful outcomes at 1 month. The characteristic MRI findings include thoracic vertebra (T)2 hyperintense, T1 hypointense, intramedullary linear tracts with reduced disc volume, and cleft of the annulus fibrosus. None of the MRI measurements were significantly correlated with neurological grade at admission. Neurological grade did not differ according to the presence of parenchymal hemorrhage, parenchymal contrast enhancement, and meningeal contrast enhancement. Neurological grades at admission showed a statistical correlation with those observed at the 1-month follow-up (r = 0.814, p = 0.02). Conclusions: IIVDE is a rare form of disc extrusion commonly experienced after physical activity or trauma and most frequently affects the cranial-cervical and thoracolumbar regions of nonchondrodystrophic dog breeds. Neurological score at admission emerged as a more useful prognostic indicator than MRI findings in dogs with suspected IIVDE.

  • This study was conducted to select efficient foliage plants for improving indoor environment conditions through the investigation of physiological responses including photosynthetic rate according to temperature, light intensity, and $CO_2$ level. Eight popular foliage plants used in this study were $Hedera$ $helix$ L., $Cissus$ $rhombifolia$ Vahl, $Ficus$ $benjamina$ L. 'Hawaii', $Syngonium$ $podophyllum$ Schott 'Albo-Virens', $Dieffenbachia$ $sp.$ 'Marrianne', $Pachira$ $aquatica$ Aubl., $Spathiphyllum$ $wallisii$ Regel, and $Scindapsus$ $aureus$ Engler. Photosynthetic rate and transpiration rate of the plants subjected to various light intensities (0, 25, 50, 75, 100, 150, 300, and $600{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$ PPFD), $CO_2$ levels (0, 50, 100, 200, 400, 700, and $1,000{\mu}molCO_2{\cdot}mol^{-1}$), and two different temperatures (16 and $22^{\circ}C$) were measured. In addition, various parameters in relation to photosynthesis were calculated from the measured data. As a result, the patterns of photosynthesis varied among 8 foliage plants according to light intensity, $CO_2$ level, and temperature. Most foliage plants except $Dieffenbachia$ had high levels of apparent quantum yield, which represents the photosynthetic rate under low light intensity (PPFD $0-100{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$). $Hedera$ $helix$, $Ficus$ $benjamina$, $Pachira$ $aquatica$, and $Spathiphyllum$ $wallisii$ exposed to high light intensity (PPFD $200-600{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$) showed high levels of photosynthesis. $Cissus$ $rhombifolia$ and $Syngonium$ $podophyllum$ were low in $CO_2$ fixation efficiency compared to the other 6 foliage indoor plants. $Hedera$ $helix$ and $Spathiphyllum$ $wallisii$ showed high photosynthetic rate under high $CO_2$ level and vigorous photosynthesis was also observed in $Ficus$ $benjamina$ and $Pachira$ $aquatica$ grown under $22^{\circ}C$. Considering characteristics of indoor environment such as low light, high $CO_2$ level, and low relative humidity, therefore, $Hedera$ $helix$, $Spathiphyllum$ $wallisii$, $Ficus$ $benjamina$, and $Pachira$ $aquatica$ were efficient indoor foliage plants to improve indoor environmental conditions.

  • A 21-residue peptide corresponding to amino acids 84-104 of $p16^{INK4A}$, the tumor suppressor, has been synthesized and its structure was studied by Circular Dichroism, $^1H$ NMR spectroscopy and molecular modeling. A p16-derived peptide (84-104 amino acids) forming stable complex with CDK4 and CDK6 inhibits the ability of CDK4/6 to phosphorylate pRb in vitro, and blocks cell-cycle progression through G1/S phase as shown in the function of the full-length p16. Its NMR spectral data including NOEs, $^3J_{NH-H{\alpha}}$ coupling constants, $C_{\alpha}H$ chemical shift, the average amplitude of amide chemical shift oscillation and temperature coefficients indicate that the secondary structure of a p16-derived peptide is similar to that of the same region of full-length p16, which consists of helix-turn-helix structure. The 3-D distance geometry structure based on NOE-hased distance and torsion angle restraints is characterized by ${\gamma}$-turn conformation between residues $Gly^{89}-Leu^{91}$(${\varphi}_{i+1}=-79.8^{\circ}$, ${\varphi}_{i+1}=60.2^{\circ}$) as evidenced in a single crystal structure for the corresponding region of p18 or p19, but is undefined at both the N and C termini. This compact and rigid ${\gamma}$-turn region is considered to stabilize the structure of p16-derived peptide and serve as a site recognizing cyelin dependent kinase, and this well-defined ${\gamma}$-turn structure could be utilized for the design of anti-cancer drug candidates.

  • Enzymatic activities and fluorescence spectroscopic properties of the double mutant proteins P96L/F139W, P96L/F258W and a triple mutant protein P96L/F139W/F258W of tryptophan synthase $\alpha$ subunit from Escherichia coli was examined to study tertiary and local structure changes around the tryptophan residues. The enzymatic activities of P96l./F139W and P96L/F258W were similar, but P96L/F139W/F258W had lower activity, as compared to wild type. The fluorescence intensities of double mutant, P96L/F139W and P96L/F258W, were decreased but that of a triple mutant, P96L/F139W/F258W, was increased when compared to wild type. The sum of the maximum fluorescence intensity (fluorescence intensity at the λ$_{max}$) for the double mutant proteins was not equal to the intensity seen in the triple mutant protein. The enzymatic activity and fluorescence data indicate that the replacement of Pro$^{96}$ longrightarrowLeu might affect on the stability of helix 8 and the loop located between strand 4 and helix4. The result suggests that the tertiary structure of triple mutant (P96L/F139W/F258W), being different from wild type, might have more compact residual structure at the vicinity of 139 and 258.8.

  • Five fusion proteins between Z domains derived from Staphylococcal Protein A and Green Fluorescent Protein or Human Proinsulin were produced on the periplasm of Escherichia coli. The effects of the molecular weight and amino acid composition of the translocated peptide, culture medium composition, and growth phase of the bacterial culture were analyzed regarding the expression and periplasmic secretion of the recombinant proteins. It was found that secretion was not affected by the size of the translocated peptide (17-42 kDa) and that the highest periplasmic production values were obtained on the exponential phase of growth. Moreover, the highest periplasmic values were obtained in minimal medium, showing the relevance of the culture medium composition on secretion. In silico prediction analysis suggested that with respect to the five proteins used in this study, those that are prone to form ${\alpha}$-helix structures are more translocated to the periplasm.

  • Since its defeat in the Second World War, Japan has been fostering good ties with Western countries, especially the United States. However, some East Asian leaders emboldened by their countries' economic success have proclaimed that the future belongs to Asia and have put forward the "Asian values" argument. It is interesting to note that some elements of the "Asian values" argument resemble ultranationalist discourse that was dominant in Japan before the war. The Japanese Government had a great opportunity to reappraise its role in international politics and take stock of its economic and diplomatic relations with East Asian countries after the end of the Cold War. To meet future challenges, Japan should fully overcome anti-Western sentiment and participate in establishing a truly democratic East Asian regionalism based on the "universal values" of human rights, democracy and freedom. This may prove to be one of the biggest challenges for Japan's East Asia policy in the new century.

  • ABA type block copolymers composed of r benzyle L-glutamate as the A component and poly(ethylene glycol) as the B components were obtained. Platelet adhesion on their sunfaces was investigated by a column elusion method to examine the effects of microdomain and secondary structure. The number of platelets adhered from whole blood and plasma rich platelet was smaller for the block copolymer systems than for the homopolymers. In the block copolymer system, the number of platelets adrered on their surfaces increased with increasing the content of PEG, that is, with decreasing of a-helix of block copolymers. A thick thrombus formation on the PBLG homopolymer was observed than block copolymer by scanning electron micrographs. The platelets adhesion increased with increasing the critical surface tension of the block copolymer.

  • The blockchain is still new and unfamiliar. But blockchain appears to shake an entire technology innovation system. Blockchain is rapidly drawing attention in that it will be able to fundamentally revolutionize industry ecosystem. While cryptocurrency transactions and market capitalization have been popular in mass media, several platform operators in non-cryptocurrency areas such as jewelry, social networks, and entertainment, are also moving to introduce blockchain technology in full swing. In this brief note, we intend to present integrated theoretical strands to summarize various prospects for blockchain technology. Further, we want to provide a reflection as to whether this new technology gives opportunities, challenges, or risks to future society. Particularly, we point out one of its alternative and promising adoption that gives way to new forms of decentralized and autonomous organizations (DAOs).
