The Biochemical Oxygen Demand(BOD) indicates that microbes are proliferating or that oxygen is being spent by breathing action when examining water under the same aerobic condition. In this research of the mesurement of BOD are the poisonous elements of heavy metal ions such as Cu-ion, Cr-ion, Pb-ion and Zn-ion. They exert an unfavorable influence in the analysis of BOD and research was performed to provide certain data of minimum negative influence by the poisonous matters. The results of the research confirm that heavy metal ion(Cu, Cr, Pb, Zn) do direct an influence upon the normal growth of aerobic microbes in actual tests of chemical analysis of portable water or sewage. The most critical concentration for a negative effect on lowering oxygen quantity and disturbing the aerobic mocrobes normal growth was found to be 0.01 mg/l. Therefore, test results are not valid if the heavy metal concentration is to or greater than 0.0mg/l, To improve comprehension through out the research the author uses the following abbreviations: 1. The Cu-ion is to be excluded before experimental analysis if it is over 0.01mg/l inorder to obtain a real value for the BOD. 2. The Cr-ion is to be excluded before experimental analysis if it is over 0.01mg/l in order to obtain a real value for the BOD. 3. The Pb-ion is to be excluded before experimental analysis if it is over 0.01mg/l in order to obtain a real value for the BOD. 4. The Zn-ion is to be excluded before experimental analysis if it is over 0.01mg/l in order to obtain a real value for the BOD.
The information related to the heavy metal pollution in the environment was obtained from studies on the effects of pH, phosphate and soil properties on the adsorption of metal ions (Cd, Cu, Ni, and Zn) by soils. Three soil materials; soil 1 with low CEC (8.2 me/100g) and low organic carbon content (0.34%); soil 2 with high CEC (36.4 me/100g) and low organic carbon content (1.8%) and soil 3 with high CEC (49.9 me/100g) and high organic carbon content (14.7%) were used. Soils were adjusted to several pH's and equilibrated with metal ion mixtures of 4 different concentrations, each having equal equivalents of each metal ion (0.63, 1.88, 3.12 and 4.38 micromoles per one gram soil with and without 10 micromoles of phosphate per one gram soil). Reported here are the results of the equilibrium study on soil I. The rest of the results on soil 2 and soil 3 will be repoted subsequeutly. Generally higher metal ion concentration solution resulted in higher final metal ion concentrations in the equilibrated solution and phosphate had minimal effect except it tended to enhance removal of cadmium and zinc from equilibrated solutions while it tended to decrease the removal of copper and nickel. In soil 1, percentages of added metal ions removed at pH 5.10 were; Cu 97, Ni 69, Cd 63, and Zn 55, while increasing pH to 6.40, they were increased to Cu 90.9, Zn 99, Ni 96, and Cd 92 per As initial metal ion concentration increased, final metal ion concentrations in the equilibrated solution showed a relationship with pH of the system as they fit to the equation $p[M^{++}]=a$ pH+b where $p[M^{++}]=-log$[metal ion concentration in Mol/liter]. The magnitude of pH and soil effects were reflected in slope (a) of the equation, and were different among metal ions and soils. Slopes (a) for metal ions in the aqueous system are all 2. In soil 1 they were; Zn 1.23, Cu 0.99, Ni 0.69 and Cd 0.59 at highest concentration. The adsorption of Cd, Ni, and Zn in soil 1 could be represented by the Iangmuir isotherm. However, construction of the Iangmuir isotherm required the correction for pH differences.
공장 폐수를 비롯한 중금속 오염 환경으로부터 효율적인 중금속 제거를 위해 생물학적 오염제거에 관한 연구가 많이 이루어지고 있다. 본 실험에서는 항생제 내성 유전자를 많이 가지고 있는 것으로 알려진 Serratia marcescens를 대상으로 이 균주가 중금속들에 대해 어떤 영향을 보이는지 조사하였다. 그 결과 lead, iron, magnesium, manganese 등의 처리군에서 24시간내에 1,000ppm 이상의 고농도 처리군에서 최소억제성장농도(MIC)를 나타냈으며, cadmium에서는 600ppm 처리군에서, 구리는 800ppm의 처리군에서 MIC를 보였으며, 아연 처리군에서는 800ppm에서 MIC를 나타냈다. 또한 48시간 배양에 따른 MIC 비교 결과, 중금속의 고농도 처리군에서 매우 긴 적응기를 갖는 것으로 확인하였다. 15가지의 항생체를 대상으로 저항성을 조사한 결과, ampicillin, tetracycline, cefamandole, cephalothin에서 저항성을 보엿으며 다른 S. marcescens 균주들에 비해 chloramphenicol에 대한 특이한 민감성을 보였다. 카드뮴과 납을 대상으로 중금속의 세포내 흡수를 조사한 결과 16.59%와 35.38%의 중금속이 세포내로 흡수되었음을 확인하였다.
Bacterial light-oxygen-voltage-sensing photoreceptor-derived flavin mononucleotide (FMN)-based fluorescent proteins act as a promising distinct class of fluorescent proteins utilized for various biomedical and biotechnological applications. The key property of its independency towards oxygen for its chromophore maturation has greatly helped this protein to outperform the other fluorescent proteins such as GFP and DsRed for anaerobic applications. Here, we describe the feasibility of FMN-containing fluorescent protein FbFP as a metal-sensing probe by measuring the fluorescence emission changes of a protein with respect to the concentration of metal ions. In the present study, we demonstrated the mercury-sensing ability of FbFP protein and the possible amino acids responsible for metal binding. A ratiometric approach was employed here in order to exploit the fluorescence changes observed at two different emission maxima with respect to Hg2+ at micromolar concentration. The engineered variant FbFPC56I showed high sensitivity towards Hg2+ and followed a good linear relationship from 0.1 to 3 μM of Hg2+. Thus, further engineering with a rational approach would enable the FbFP to be developed as a novel and highly selective and sensitive biosensor for other toxic heavy metal ions as well.
Aim of the present study was the development of a bioassay which enables the detection of toxic effects of heavy metal ions to a bacterium, Escherichia coli. Inhibition effects of the metals on growth rates of the bacterium were studied in the absence or presence of fulvic acid. This method does not clearly differentiate among metals, but does detect overall AGB inhibition rate (toxicity) for 5 different heavy metals. The toxicity of the metals in the absence of fulvic acid in the same testing conditions was significantly increased in following order: Hg < Pb, Zn < Cd < Cu, whereas the inhibition rate (toxicity) in the presence of FA was shown to be increased In following order: Cd < Pb, Hg < Cu < Zn. The results of the present study indicate that this simple and fast biomonitoring assay with direct exposure of E coli. might be a valuable supplement to analytical methods of contaminated media.
Functional properties of yam mucilage were investigated by physicochemical analysis. Yam mucilage was extracted from the root of yam and then separated into two fractions by its selective aggregation with isopropanol concentration. Each mucilge fraction showed the excellent binding properties with heavy metal ions Co, Cr and Cu. Cr showed the higher affinity with mucilage than Co and Cu at pH 6.3. In ACE inhibition, IC50 values of mucilage fraction 1 and 2 showed 8.99$\mu\textrm{g}$/${mu}ell$ and 7.1$\mu\textrm{g}$/${mu}ell$, respectively.
The stabilizing behavior of heavy metals in the silicate glass containing Electric Arc Furnace dust (EAF dust) were studied by the Toxic Characterization Leaching Procedure (TCLP) test, and the change of crystalline phase and glass network structure were investigated as a function of EAF dust content added. The glass containing EAF dust of $30\;wt\%$ an oxygen/network former ratio(R) of $2\~3$ allowing a fairly stable network structure thus showed much lower heavy metal leaching concentration than that for containing EAF dust above $50\;wt\%$ at TCLP test. For the glass containing EAF dust $50\~60\;wt\%$, however, the R was over 3, which weakened the glass network structure and increased the heavy metals leachate. Adding the EAF dust to a glass decreased the degree of Si-O-Si symmetry and increased the number of non-bridging oxygen, which decreased the chemical durability of glasses. When the dust content in a glass was over $70\;wt\%$, the Zn and Fe ions reacted to form the spinel crystal rather than to bind to network structure of glass and leaching concentration of those ions from the specimen decreased, so the spinel phase could be attributed to lowering a heavy metal leaching.
Emulsion Liquid Membrane (ELM) is a prominent technique for the separation of heavy metal ions from wastewater due to the fast extraction and is a single-stage operation of stripping-extraction. The selection of the components (Surfactant and Carrier) of ELM is a very significant step for its preparation. In the ELM technique, the primary water- in-oil (W/O) emulsion is emulsified in water to produce water-in-oil-in-water (W/O/W) emulsion. The water in oil emulsion was prepared by mixing the membrane phase and internal phase. To prepare the membrane phase, the extractant D2EHPA (di-2-ethylhexylphosphoric acid) was used as a mobile carrier, Span-80 as a surfactant, and Paraffin as a diluent. Moreover, the internal (receiving) phase was prepared by dissolving sulphuric acid in water. Di-(2- ethylhexyl) phosphoric acid such as surfactant concentration, carrier concentration, sulphuric acid concentration in the receiving (internal) phase, agitation time (emulsion phase and feed phase), the volume ratio of the membrane phase to the receiving phase, the volume ratio of the external feed phase to the primary water-in-oil emulsion and pH of feed were studied on the percentage extraction of metal ions at 20℃. The results show that it is possible to remove 78% for As(V), 98% for Cd(II), and 99% for Pb(II). The electrochemical adsorption of the Ag ions from aqueous solution on pelletized activated carbon monolith was investigated over wide range of operation time. The adsorption capacities of pelletized activated carbon monolith are associated with their internal porosity and are related properties such as surface area, pore size distribution. The chemical industry generates wastewater that contains toxic matters like heavy metals in small concentrations so that their economic recovery is not feasible. But, the method using activated carbon monolith can be used to withdrawal of heavy metals in waste water. After the electrochemical treatment, the quantitative properties in Ag ion solutions are also examined by pH concentration and studied elemental analysis by ICP-Atomic Emission Spectrometer and Energy Disperse X-ray (EDX) spectra. It is consider that the pH is very important factor at the reason of water pollutant with increasing acidity in industrial field. The result of quantitative analysis using Inductively Coupled Plasma-Atomic Emission Spectrometer of metal after electrochemical reaction in Ag ions solution depending on time are shown that the amount of Ag ions deposited was decreased with growth of Ag particles on the carbon surfaces as increasing electrochemically treated time. And, surface morphologies are investigated by scanning electron microscopy (SEM) to explain the changes in adsorption properties.
뱀장어 간세포 배양을 이용하여 cytochrome P4501A (CYP1A) 유전자 발현에 대한 중금속들의 영향에 대해 조사하였다. 첫째, CYP1A mRNA 발현에 대한 benzo[$\alpha$]pyrene (B[a]P)의 농도별 조사에서, B[$\alpha$]P $10^{-8}\sim10^{-5}$ M의 농도로 배양액에 첨가하여 배양한 후 세포를 수거하여 RT-PCR 방법으로 CYP1A mRNA 발현량을 조사하였다. CYP1A mRNA 발현은 B[$\alpha$농도 의존적으로 유도되었으며, $10^{-7}$ M 농도부터 통계적으로 유의차를 나타냈다 (p<0.05). 둘째로 뱀장어 간세포에 B[a]P ($10^{-5}$ M)와 카드뮴 ($10^{-6}$, $10^{-5}$ M)을 각각 또는 함께 첨가하여 CYP1A mRNA 발현을 조사하였다. 카드뮴을 첨가한 그룹은 vehicle에 비해 CYP1A 발현의 억제가 관찰되었으며, 고농도($10^{-5}$M)의 카드뮴에서 더 많이 억제되었다. 또한 B[$\alpha$]P와 카드뮴을 함께 처리한 그룹에서도 CYP1A 유전자 발현은 B[$\alpha$]P 단독 처리에 비해 현저한 억제가 관찰되었다. 셋째로, in vivo에서 B[a]P를 주사한 뱀장어의 배양 간세포를 이용하여 CYP1A 유전자 발현에 대한 중금속들의 영향을 조사하였는데, 뱀장어에 10mg/kg의 B[$\alpha$]P를 주사한 후, 48시간 후에 간을 채취하여 간세포 배양을 하였다. 카드뮴, 구리, 납 및 아연 ($10^{-6}$, $10^{-5}$ M) 등을 각각 배양액에 첨가하여 2일간 배양한 후 CYP1A mRNA 발현량을 조사하였다. 이미 B[$\alpha$]P 처리에 의해 CYP1A 발현이 유도된 대조구 (vehicle)에 비해 중금속들이 첨가된 모든 그룹에서 CYP1A 유전자 발현 억제가 관찰되었다. 본 연구는 여러 어종에서 CYP1A 유전자 발현에 대한 중금속들의 영향 및 중금속 독성을 연구하는데 매우 중요한 기초 자료로 활용되어질 것이다.
