Genome sequence of Caballeronia sordidicola strain PAMC 26592 isolated from an arctic lichen species (북극 지의류에서 분리한 Caballeronia sordidicola균주 PAMC 26592의 유전체 서열 분석)

  • Caballeronia sordidicola strain PAMC 26592 was isolated from Umbilicaria sp., a lichen material collected from Svalbard Archipelago in the Arctic Ocean. We report the draft genome sequence of the strain PAMC 26592, a metabolic generalist. As we have observed in previous genomic studies in the genus Caballeronia draft genomic sequences of PAMC 26592 had an assortment of genes of ecological importance and of bio-technical potentials, which include diverse metabolic genes for carbohydrates, aromatic compounds, amino acids, and vitamins, and genes for nitrogen / sulfur metabolisms, stress responses, membrane transporters, antibiotic resistance, and heavy metal resistance.

Analysis of the Emission Potential of Hazardous Pollutants Produced from disposal of the School Solid Wastes by Small-Scale Incinerator (학교 생활 쓰레기의 성분 분석과 소형소각로 운전에 따른 유해성 오염물의 배출 잠재성 분석 연구)

  • This study analyzed solid wastes generated from a school. The emission potential of hazardous pollutants generated from incineration of the school solid wastes (SSWs) was analyzed. Components of the SSWs were identified and the SSWs were classified into combustible and non-combustible wasts. The combustible wastes consisted of papers of 56.5^ plastics of 30.2% woods of 7.1% and fibers of 6.1% based on weight of the wastes. The moisture content and the ash content of the combustible wastes were 18~20% and 11~13% respectively. The combustible wastes of the SSWs were incinerated by using a small-scale incinerator. Fly and bottom ashes and volatile organic compounds (VOCs) were collected from the incineration. Also the metal leaching experiments on the fly and bottom ashes were performed, In analysis of metals leached from the ashes the total amounts of metals leached in the acid solution (pH=3) were much greater than those in the neutral solution (pH=5.8~6.2) For the same amounts of the fly and bottom ashes the total amounts of metals leached from the fly ashes were much greater than those from the bottom ashes. The VOCs produced from incineration of the SSWs consisted of aromatics of 42.1% aliphatic alkenes of 26.3% oxidized forms of 17.3% and aliphatic alkanes of 14.3% In addition the considerable amounts of hazardous air pollutants (e.g benzene chloro-benzene and chloro-alkanes) and compounds (e. g, aliphatic alkenes) with high potential of ozone or photochemical smog formation were identified from the incineration experiment of the SSWs.

Condensation and coagulation of metallic species with fly ash particles in a waste incinerator (폐기물 소각시 생성되는 유해 중금속물질과 연소실내 비산재와의 응축, 응집 현상에 대한 연구)

  • A numerical analysis on condensation and coagulation of the metallic species with fly ash particles pre-existing in an incinerator was performed. Waste was simplified as a mixture of methane, chlorine, and small amounts of Pb and Sn. Vapor-phase amounts of Pb- and Sn -compounds were first calculated assuming a thermodynamic equilibrium state. Then theories on vapor-to-particle conversion, vapor condensation onto the fly ash particles, and particle-particle interaction were examined and incorporated into equations of aerosol dynamics and vapor continuity. It was assumed that the particles followed a log-normal size distribution and thus a moment model was developed in order to predict the particle concentration and the particle size distribution simultaneously. Distributions of metallic vapor concentration (or vapor pressure) were also obtained. Temperature drop rate of combustion gas, fly ash concentration and its size were selected as parameters influencing the discharged amount of metallic species. In general, the coagulation between the newly formed metal particles and the fly ash particles was much greater than that between the metal particles themselves or between the fly ash particles themselves. It was also found that the amount of metallic species discharged into the atmosphere was increased due to coagulation. While most of PbO vapors produced from the combustion were eliminated due to combined effect of condensation and coagulation, the highly volatile species, PbCl$_{2}$ and SnCl$_{4}$ vapors tended to discharge into the atmosphere without experiencing either the condensation or the coagulation. For Sn vapors the tendency was between that of PbO vapors and that of PbCl$_{2}$ or SnCl$_{4}$. To restrain the discharged amount of hazardous metallic species, the coagulation should be restrained, the number concentration and the size of pre-existing fly ash particles should be increased, and the temperature drop rate of combustion gas should be kept low.

  • The silver selenide electrode has been prepared and its properties as an indicating electrode for silver ion have been investigated. Epoxy resin was used as a filler of silver selenide electrode. Silver metal plate was directly connected with the membrane of the electrode and the silver paste was used as its binder. The sintered electrode was more sensitive and stable than the pressed electrode, and the silver selenide electrode more sensitive than the silver sulfide electrode to silver ion. The linear relationship between the electrode potential and logarithmic concentration of silver ion has been observed down to 10-6 M for the electrode. Several heavy metal ions except mercuric ion did not interfere this linearity, but halide, cyanide, and thiocyanate ions did intensively interfere owing to the formation of silver compounds and complexes. This electrode has been applied to the potentiometric titration for determining halide ion. It is concluded that interferences from ,$CN^-, SCN^-, S^-, I^-, Br^-, Cl^- and Hg^{2+}$ ions are detrimental to the practical use of the electrodes for measuring pAg.

Assessing Heavy Metals for Estrogenicity Using a Combination of In vitro and In vivo Assays (In vitro 및 In vivo Assay를 통한 중금속의 에스트로겐성 평가)

  • The estrogenicities of six heavy metal compounds, which contaminate frequently in foods, were assayed using a combination of in vitro and in vivo assays. The assays were 1) estrogen receptor dependent transcriptional expression assay, 2) E-screen assay and, 3) the uterotropic assay in mice. The chemicals studied were 17$\beta$ -estradiol, diethylstilbestrol (DES), arsenic oxide, bis(tri-n-butyltin), cadmium chloride, chromium chloride, lead acetate, and mercuric chloride. Using the estrogen receptor dependent transcriptional expression assay, the following estrogenicity ranking was measured: bis(tri-n-butyltin) > cadmium chloride > chromium chloride >> mercuric chloride >lead acetate = arsenic oxide. Using E-screen test, the following estrogenicity ranking was measured: bis(tri-n-butyltin) > cadmium chloride > chromium chloride >> mercuric chloride > lead acetate = arsenic oxide. Results from the uterotropic assay showed that bis(tri-n-butyltin), cadmium chloride, chromium chloride caused an increase in uterine wet weight, while lead acetate, mercuric chloride, and arsenic oxide failed to do so. Bis(tri-n-butyltin), cadmium chloride and chromium chloride showed the highest estrogenicity in three assay systems. Recent studies suggesting that bis(tri-n-butyltin), cadmium chloride have estrogenicities are compatible with the present finding. Furthermore, our study is suggesting that chromium chloride may be estrogenic. The results demonstrate that this three level-assay combination (transcriptional activation, cell proliferation, and an in vivo effect in an estrogen-responsive tissue) could serve as a useful method to assess the estrogenicity of heavy metals.

Emulsion Liquid Membrane Transport of Heavy Metal Sons by Macrocyclic Carriers (거대고리 운반체에 의한 중금속이온의 에멀죤 액체막 수송)

  • New two macrocyclic compounds using as carriers of liquid emulsion menbrame, have been synthesized. These reuslts provide evidance for the usefulness of the theory in designing the systems. The efficiency of selective transport for heavy metal ions have been discussed from the membrane systems that make use of $SCN^-$,<>,$I^-$,CN- and $Cl^-$ ion as co-anions in source phase and make use of $S_2O_3^{2-}$ and $P_2O_7^{4-}$ ion as receiving phase, respectively. The transport rate of M(II) was highest when a maximum amount of the M(II) in the source phase was present as$Cd(SCN)_2$$(P[SCN^-]= 0.40M)$, $Hg(SCN)_2([SCN^-]=0.40M)$ and Pd(CN)$([CN^-]= 0.40M)$. The Cd(II) and Pb(II) over each competitive cations were well transprted with 0.3M-S2032- and 0.3M-P2O74-, respectively in the receiving phase. Results of this study indicate that two criteria must be met in order to have effective macrocycle-mediated transport in these emulsion system. First one must effective extraction of the $M^{n+}$ into the toluene systems. The effectiveness of this extraction is the greatest if locK for $M^{n+}$macrocycle interaction is large and if the macrocycle is very insoluble in the aqueous phase. Second, the ratio of the locK values (or Mn+-receiving phase ($S_2O_3^{2-}$- or $P_2O_7^{4-}$) to $M^{n+}$-macrocycle (($L_1$이나 $L_2$) interaction must be large enough to ensure quantitative stripping of Mn+(($Cd^{2+}$,$Pb^{2+}$)at the toluene receiving Phase interface. $L_1$(3.5-benzo-10,13,18,21-tetraoxa-1,7,diazabicyclo(8,5,5) eicosan) forms a stable ($Cd^{2+}$ and >,$Pb^{2+}$ complexes and $L_1$ is very insoluble in water and its $Cd^{2+}$ and >,$Pb^{2+}$ complex is considerably less stable than $Cd^{2+}$-(S2O3)22- and $Pd^{2+}-P_2O_7^{4-}$ complexes. On the other hand, the stability of the $Hg^{2+}$)+-$L_1$( complex exceed that of the $Hg^{2+}$- (S2O3)22- and Hg2+-P2O74-, and the distribution coefficient of $L_2$(5,8,15,18,23,26-hexaoxa-1,12- diazabicyclo-(10,8,8) octacosane) is much smaller than that of $L_1$. Therefore, the partitioning of Lr is favored by the aqueous receiving Phase, and little heavy metal ions transport is seen despite the large logK for $Hg^{2+}$+-$L_1$ and $Mn^+$($Cd^{2+}$+, $Pb^{2+}$+ and $Hg^{2+}$)-$L_2$ interactions. Characteristics of Fermented Wood Chips and Pig Manure (목질칩을 이용한 분뇨 발효 시 목질칩과 돈분뇨의 성분 변화)

  • After manufacturing fermentation system for degrading pig manure using environmentally friendly technique, performance of the system and characteristics of wood chips and pig manure fermented in the system were analyzed. Results from this study shows that proper fermentation temperature($55{\sim}60^{\circ}C$) reached 3days after the system started and degradation rate, which expresses fermentation performance of system, was $180{\iota}$/day. Even as progressing the fermentation of wood chips and pig manure mixture, the amount of extractives drawn out by alkali, and alcohol-benzene and lignin content was not varied. However, ash content in wood was increased. The inorganic compounds in pig manure seem to be transferred into wood chip. On the other hand holocellulose contents in wood were decreased a little. Holocellulose seems to be consumed as the second carbon source in fermentation process. Results through analysis of inorganic- and heavy metal elements contents in wood chips and pig manure fermented in long term process shows that inorganic elements($Ca^{2+},\;Mg^{2+},\;K^+,\;Na^+$ etc.) contents were increased with fermentation time and heavy metal elements(Cd, As, Cu etc.) which cause environmental pollution were not detected. Number of microorganisms including bacteria, actinomycetes, and fungi, the number of C.F.U(Colony Forming Unit) was increased while temperature in fermentation system was abruptly increased.

Evaluation of the Sediments Contamination in the Lake Sihwa (시화호 퇴적토의 오염도 평가 및 효과적 관리방안)

  • An investigation on the polluted sediments in the Lake Sihwa and the benthos that inhabited on the sediments was conducted. Cost effective remediation alternatives were derived form the results of the investigation. The sediment samples taken from four sampling points out of thirteen showed relatively high heavy metal (particularly copper) concentrations which exceeded the Effects Range Low (ERL) of the National Oceanic and Atmospheric Administration, USA. The four sampling points were located in front of industrial complexes. Although the heavy metals appeared to have affected the growth of the benthos, the concentration of it did not exceed the criteria of dredging that were developed by Netherlands or the State of Washington, USA. However, contamination by organic matters and sulfur compounds was severe, which exceeded the criteria of dredging that were established in Japan. The sediments taken from the four sampling points which were contaminated with heavy metals showed higher organic matter content in general. The organic matters in the sediments depleted oxygen in summer, which appeared to be fatal to the benthos. A comprehensive analysis on the sediments, benthos, and other environmental impact from the contaminated sediments drew a conclusion that the benthonic environment of the Lake Sihwa needed a stepwise remediation, giving a particular emphasis on the clean up of the sediments upstram of the Lake which could cause odor problems to the nearby residential area.

Studies about the bioactive component analysis and an oral glucose tolerance test of Add-Omit-Saenghyeoryunbu-eum(AO-SHU) for confirmation of diabetes therapy (가감생혈윤부음(加減生血潤膚飮)의 당뇨병 치료효과 확인을 위한 생리활성성분 분석과 경구포도당부하 연구)

  • Objectives : Instrumental chemical analysis was utilized to investigate the effect of Add-Omit-Saenghyeoryunbu-eum(AO-SHU) on diabetic treatment. One of the most exciting, yet also controversial, arguments is the safety and biological mechanisms of the natural medicine on human body. Therefore, the aim of this study is to provide a better understanding on bioactive chemical components, hazards of heavy metal contamination and biological mechanism of the diabetic medicine composed of 12 different natural herbs. Methods : To study bioactive compound and metallic component in the diabetic medicine in detail, LC-MS/MS (Liquid Chromatography-Mass/Mass), GC (Gas Chromatography) and ICP (Inductively Coupled Plasma) were utilized to characterize the extract of the diabetic medicine and the result was compared with 18 marker substances selected from literature survey. In addition, in vitro assay experiments including GPR 119 activity and human DGAT-1 inhibition, and OGTT (Oral Glucose Tolerance Test) were performed to verify the effectiveness of this medicine on diabetic treatment. Results : Out of 18 marker substances, 9 bioactive compounds were identified from LC-MS/MS analysis which include Citruline, Catalpol, Berberine, Ginsenoside Rb1, Ginsenoside Rg1, Oleanolic acid, β-Sitosterol, Mangiferin, and Schizandrin. ICP study on 245 residual pesticides revealed that 239 species were not detected but 6 species, Dimethomorph, Trifloxystrobin, Pyraclostrobin, Isoprocarb, Carbaryl and Flubendiamide, while the amounts are trace levels, below permitted concentrations. The biological activity was observed in vitro assay and Oral Glucose Tolerance Test(OGTT), which are consistent with a preliminary clinical test result, a drop in blood sugar level after taking this herbal medicine. Conclusions : Instrumental chemical analysis using LC-MS/MS, GC, and ICP was conducted successfully to identify bioactive compounds in AO-SHU for the treatment of diabetes, finding 9 bioactive compounds. Furthermore, in vitro assay experiments and OGTT show that AO-SHU has its biological activities, which imply that it can be a candidate for the future diabetes remedy.

Bacterial Productivity and Degradability of Organic Compounds in the Coastal Area of East Sea (동해 연안역의 박테리아 생산력과 유기물질의 분해능)

  • Bacterial population, signal compounds of pollution, bacterial productivity, heterotrophic activity and the activities of extracellular enzymes were analyzed four times from July 1994 to April 1995 at Daejin, Galnam, Pohang, and Ulsan in the coastal area of Bast Sea in order to investigate the state of eutrophication. The population of heavy metal resistant bacteria in Pohang was the highest among all stations. The bacterial populations which could utilize C -P compounds (phosphonate) showed the highest at Pohang that was expected for the industrial and agricultural effluents to inpour. It could be useful as an index of expectation about the influx of industrial sewages and agricultural effluents. The activities of extracellular enzymes for the degradation of organic matter were the highest in the Ulsan Chuyongam, but low in Daejin. These results showed positive relationship with the population of heterotrophic bacteria and bacterial productivity The ratio of poly-P and DNA in Pohang was the highest, and that in Ulsan, Galnam, and Daejin consecutively, corresponding with the distribution of heterotrophic bacteria. In case of the heterotrophic activities, each of the stations gives difference of seasonal variation in heterotrophic activities. Bacterial productivity seemed to be dependent on seasons; the highest in summer and the lowest in fall and winter. These seasonal variations of bacterial productivity seems to be similar with those of heterotrophic bacteria. It is suggested that the state of eutrophication could be evaluated by the criteria of the population sizes and the productivities of microorganisms, the amount of poly-P, heterotrophic activity, the activities of extracellular enzymes, etc.
