Preventive effects of sea cucumber (Apostichopus japonicus) ethanol extract on palmitate-induced vascular injury in vivo

  • Cardiovascular diseases (CVDs) have posed serious public health problems, accounting for nearly 30% of mortality worldwide and their incidence is still increasing. Therefore, new treatment resources are necessary to prevent or manage the ever-increasing population of patients with CVDs. Sea cucumber is well known for its medical and health benefit effects, but it is not well known what/how effect it has on vascular disease. In the present study, we examined the protect effect of sea cucumber, Apostichopus japonicus 80% ethanol extract (AJE) on zebrafish embryo with the stimulation of free fatty acid, palmitate (PA). In vivo study showed that AJE can attenuate PA-induced toxicity through relieving the rapid heartbeat, increasing the survival rate and reducing the malformation in both wild type and Tg (fli1a:eGFP) transgenic zebrafish lines. Additionally, compare with PA treated embryos, the yolk sac area, body length, axial vascular segment (AVS) and intersegmental vessel (ISV) of the co-treatment group of AJE and PA were comparable to the control group. Moreover, AJE lowered the expression of inducible nitric oxide synthase (iNOS), nitric oxide (NO) and inflammation-related genes induced by PA, and inhibited PA-induced vascular development disorders. Our data preliminarily verify that AJE could be a candidate resource for the prevention or therapy of CVDs.

  • Arthropods have an open circulatory system with a simple tubular heart, so it has been estimated that the contractile pumping structure of the cardiac muscle will be less efficient than that of vertebrates. Nevertheless, certain arthropods are known to have far superior properties and characteristics than vertebrates, so we investigated the fine structural features of intercalated discs and cardiac junctions of cardiac muscle cells in the black widow spider Latrodectus mactans. Characteristically, the spider cardiac muscle has typical striated features and represents a functional syncytium that supports multiple connections to adjacent cells by intercalated discs. Histologically, the boundary lamina of each sarcolemma connects to the basement membrane to form an elastic sheath, and the extracellular matrix allows the cells to be anchored to other tissues. Since the intercalated disc is also part of sarcolemma, it contains gap junctions for depolarization and desmosomes that keep the fibers together during cardiac muscle contraction. Furthermore, fascia adherens and macula adherens (desmosomes) were also identified as cell junctions in both sarcolemma and intercalated discs. To enable the coordinated heartbeat of the cardiac muscle, the muscle fibers have neuronal innervations by multiple axons from the motor ganglion.

청각장애 산모의 태아 초음파 심장박동 촉각 변환 시스템 개발 (Development of Touch-Conversion Device of Heartbeat for Deaf Parents in Fetal Ultrasonography)

  • 아기의 심장 박동 소리를 듣지 못하는 청각 장애 부모를 위한 태아 초음파 심장박동 촉각 변환 시스템을 개발하고자 하였다. 본 시스템은 아기의 심장박동 소리를 USB 마이크를 이용하여 소릿값을 입력받아 이에 대응하는 값을 진동 모터의 진동 출력 세기와 LED 밝기의 세기로 출력시키도록 하였다. 아기의 심장박동 느낌을 진동으로 표현해 내기 위해 진동의 샘플링 주기를 구체적인 수치로 조절하였고 산모에 따라 진동의 세기와 주변 잡음 제거를 할 수 있도록 조절하는 가변저항 2개를 연결하였다. 그 결과 태아 초음파 심장박동 촉각 변환 시스템을 성공적으로 개발하였으며 장치 체험자들로부터 총 평균 4.63점으로 높은 만족도를 얻었다. 개발 제품은 청각장애 산모들의 모성애를 충족시키고 아기의 심장박동을 느낌으로써 아기의 존재를 느낄 수 있을 것이며, 산부인과의 초음파 영상진단의 확장 신기술로 활용할 수 있을 것으로 기대된다.

  • 이 연구는 제주도 연안에 분포되어 있는 정치망에서 음향 집어기에 의한 어류의 유집에 대한 기초 자료를 제공할 목적으로, 정치망의 주요 어획 대상 어류인 독가시치를 선택하여, 가청 수중음과 전기 자극을 이용하여 음향 조건학습시켰으며, 주파수와 음압을 변화시켜가면서 심전도를 도출하여 심박 간격의 변화로부터 청각 능력을 알아내는 방법을 이용하여, 독가시치의 청각 문턱치 및 청각 임계비를 측정 조사 한 결과 75, 79, 83의 3단계 백색잡음을 방성하였을 때 독가시치의 청각 문턱치는 백색잡음이 없을 때보다 높게 나타나 마스킹 효과가 나타났고, 측정주파수 200Hz, 300Hz에서 보다 뚜렷하게 나타났다. 독가시치의 청각 임계비는 측정주파수 80, 100, 200, 300, 500, 800Hz에서 대략 28, 23, 31, 34, 40, 52dB이고 마스킹 현상은 음압이 약 70dB 이상의 잡음레벨에서 나타나기 시작하였고 측정주파수 100 - 200Hz에서 신호음을 인식하기 위해서는 음압이 대략 98dB이상, 배경잡음레벨보다 약 23dB이상의 높은 음압이 요구되었다.

DACUM을 이용한 고위험 산모·신생아 통합센터 간호사의 직무분석 (Job Analysis of Maternal Fetal Intensive Care Unit Nurses Using DACUM Technique)

  • Purpose: This study was performed to establish the role and to analyze the job of MFICU (Maternal Fetal Intensive Care Unit) nurses using DACUM (Developing a curriculum). Methods: A DACUM workshop was held to define MFICU nurses' role and identify their duties and tasks. A DACUM committee was consisted of 7 nurses, 2 nursing professors and 1 medical doctor and as a result, a survey was developed which contained duties and tasks of MFICU nurse. Pre-test was carried out for the validity, finally collected the data from 97 nurses who worked at 7 MFICU and 10 delivery room. Results: A total of 60 duties, 115 tasks and 822 elements of tasks were defined on the DACUM chart and survey. The importance, frequency and difficulty of the tasks were presented the determinant coefficient (DC), the highest DC duty was 'Manage maternal ventilator' (15.09) and the lowest DC was 'Provide nursing care for leisure to gestation extension mother' (6.52). Twenty-eight tasks were differentiated between MFICU and delivery nurses significantly. And the most important, frequently, difficulty task perceived by MFICU nurse was 'Check fetal heartbeat with electronic fetal heart monitor'. Conclusion: The organized educational program and policy was needed to develop for MFICU nurses.

Saccharomyces cerevisiae의 생물시계와 초단기 대사진동 (Biological Clock and Ultradian Metabolic Oscillation in Saccharomyces cerevisiae)

  • 생물시계(Biological clock)는 생명체에서 나타나는 반복되는 자율적인 리듬을 말하며, 단일세포는 물론 다세포 생명체의 기본적인 대사와 이에 따른 표현형과 행동을 직접적으로 조절하고 있다. 이러한 생물시계는 동면 리듬, 수면 리듬, 심장박동 리듬 및 짝짓기 노래 리듬 등 매우 다양하며, 24시간 이상의 주기를 infradian rhythm, 24시간 주기를 circadian rhythm, 24시간 이내의 짧은 주기를 ultradian rhythm으로 구분한다. 효모 Saccharomyces cerevisiae는 최소 5종 이상의 반복되는 자율적인 리듬이 알려져 있으며, 이중 일부는 생체시계로 인식되고 있다. 본 리뷰에서는 Saccharomyces cerevisiae의 glycolytic oscillation (T= 1~30분), cell cycle-dependent oscillation (T= 2~16 시간), ultradian metabolic oscillation (T= 15~50분), yeast colony oscillation (T= 수 시간) 및 circadian oscillation (T= 24시간)에 대한 연구 결과를 제시하고, 특히 ultradian metabolic oscillation의 특징, 집단 동조인자(population synchronizer), 동조인자의 조절 기작 및 효모 생물시계의 대사공학 분야의 이용성을 제시하여 효모를 이용한 동적 대사조절 및 생물시계 연구가 가능함을 제시하였다.

초음파검사에 의한 소의 번식장애 감별진단 및 치료법 개발 IV, 발정확인 및 조기 임신진단 (Development of Differential Diagnosis and Treatment Method of Reproductive Disorders Using Ultrasonography in Cows IV. Confirmation of Estrus Detection and Early Pregnancy Diagnosis)

  • Plasma progesterone (P$_4$) concentrations were measured for confirming the estrus observation and for the early pregnancy diagnosis in 130 cows of small farmers. Ultrasonographic examinations were performed from day 30 after artificial insemination to establish the characteristic ultrasonographic appearances of gestational structures in each pregnant stages. Of the 130 cows inseminated, 111 cows (85.4%) were an ovulatory estrus, 12 cows (9.2%) were an unovulatory estrus, and 7 cows (5.4%) were the error of estrus detection, respectively. The accuracy for early pregnancy diagnosis in 111 ovulatory estrus cows achieved when the discriminatory concentration at day 21 after artificial insemination was placed at 3.0 ng-/ml in plasma, was 86.7 % for positive diagnosis and 100% for negative diagnosis, respectively. Pregnancy diagnosis by ultrasonography were performed to evaluate gestational structures from day 30 after artificial insemination in 83 cows. Pregnant cows were 72 of 83 cows. The characteristic ultrasonography of gestational structures in each gestational stages was as follows. The embryo proper was observed within anechoic fetal fluid between 28 and 40 days after insemination, and amnion and embryonic heartbeat was also detected in this period. Between days 41 and 50, embryo proper was detected as an discriminated from head and body, and forelimb buds and hindlimb buds were also observed in this period. Between days 51 and 60, an embryo proper was clearly discriminated from head and body, and fetal movement, forelimb buds and hindlimb buds were observed in this period. Between days 61 and 70, fetus was completely developed, and fetal skeleton, organs and cotyledon were observed. After day 71, each organs of fetus were rapidly developed and a fetus was partially observed in screen because fetus was too big and larger, These results indicate that plasma P$_4$ determination at days 0,6 and 21 after artificial insemination can be utilized for confirming the estrus observation and for early pregnancy diagnosis. Also, ultrasonography was reliable method for early pregnancy diagnosis at day 30 after artificial insemination.

광전용적맥파 센서를 이용한 맥파전달시간의 측정 (Measurement of Cardiac Pulse Transit Time using Photoplethysmography Sensor)

  • In this study, we implemented the pulse transit time (PTT) system to examine usefulness of the monitoring method of distensibility and elasticity using photoplethysmography sensor in vivo. PTT is defined as the time interval between the peak of QRS complex in ECG signal and the maximum slope point of photoplethysmography. these two signals were converted to digital data by means of AID converter, then PTT was evaluated by heartbeat using PC. Results of analysis were displayed as a graph using spline interpolation method. The variance of PTT was measured repetitiously to verify efficiency of PTT system in resting state and hyperemic state. Repeated measurement of PTT was not same value but showed that coefficients of correlation were related with each other as 0.8302 (P<0.01) in resting state. And also repeated measurement of PTT showed significant correlation as 0.868 (P<0.01) in the hyperemic state. These result showed that PTT is reflect on transient pressure variance in the artery and is very useful method for the evaluation of prognosis of the hypertension and arteriosclerosis.

계층구조적 분류모델을 이용한 심전도에서의 비정상 비트 검출 (Detection of Abnormal Heartbeat using Hierarchical Qassification in ECG)

  • The more people use ambulatory electrocardiogram(ECG) for arrhythmia detection, the more researchers report the automatic classification algorithms. Most of the previous studies don't consider the un-balanced data distribution. Even in patients, there are much more normal beats than abnormal beats among the data from 24 hours. To solve this problem, the hierarchical classification using 21 features was adopted for arrhythmia abnormal beat detection. The features include R-R intervals and data to describe the morphology of the wave. To validate the algorithm, 44 non-pacemaker recordings from physionet were used. The hierarchical classification model with 2 stages on domain knowledge was constructed. Using our suggested method, we could improve the performance in abnormal beat classification from the conventional multi-class classification method. In conclusion, the domain knowledge based hierarchical classification is useful to the ECG beat classification with unbalanced data distribution.

유-헬스 앱 개발을 위한 센서 추상화: 정확도 향상을 위한 필터링 및 요약 (Sensor Abstraction for U-health Application Development: Filtering and Summarization for Accuracy Enhancement)

  • 최근 혈압, 체온 및 혈당 같은 개인 건강 정보를 알려주는 센서-기반의 유-헬스 앱에 대한 연구가 활발히 진행되고 있다. 그러나 센서들을 통해 얻어진 정보는 그 정확성에 문제가 있을 수 있으므로 가공되지 않은 상태로 사용하기 어려운 경우가 많다. 본 논문은 스마트폰과 연동하는 생체 센서들을 통해 얻어진 측정값들의 정확성을 향상시키기 위한 센서 추상화 계층을 제안한다. 이 계층은 연결된 센서의 종류를 인식하고 읽어온 센서 값들을 ISO/IEEE 11073 신체 건강 표준에 따라 변환하며, 필요한 경우 측정값들 중에서 이상치(outlier)를 제거하는 필터링(filtering) 기법과 구해진 값들을 보다 적합한 형태로 변환해주는 요약(summarization) 기법을 적용한다. 제안된 센서 추상화 계층의 평가를 위해 안드로이드 기반의 개발보드를 사용한다. 체온 센서와 심박 센서를 통해 얻어진 값들에 대해 필터링 및 요약 기법을 적용한 경우의 결과가 그렇지 않은 경우에 비해 향상된 정확성을 보인다.
