Effects of vitamin E and selenium on the ovariectomized rat (Vitamin E와 selenium이 난소가 제거된 흰쥐에 미치는 영향)

  • The present study was devised to determine the effects of body weight, organ weight, hematological values and biochemical parameters by vitamin E and selenium (Selevit) in the ovariectomized (OVX) rats. The animals were divided into 4 groups. Intact group (n=10) received no treatment and operation. Sham group (n=10) received only sham operation and no treatment. OVX group received operation and no treatment. OVX+Selevit received operation and Selevit. The body weights of the all group increased, and that of OVX+Selevit group was the lowest rates. There were significant differences (P<0.01, P<0.001) of body weights between OVX+Selevit group and all other groups. Also, organ weights such as heart, liver, spleen and kidney were measured. The heart and liver weight were significantly lower (P<0.01, P<0.001) in the OVX+Selevit group than in the Intact and Sham group. Also, the kidney weight was significantly lower (P<0.05, P<0.01) in the OVX+Selevit group than in the all other group. On the other hand, there was no significantly difference in the organ weight of spleen between the OVX+Selevit group and the other groups. The number of WBC was significantly higher (P<0.05) in the OVX+Selevit group than in the all other groups. The hematological values of RBC, MCV, MCH and MCHC were no significant differences in any groups. The biochemical parameters of serum total protein and alkaline phosphatase increased significantly in the OVX+Selevit group as compared to that in the OVX group. But, there were no significant differences in AST and ALT in any groups. We conclude that vitamin E and selenium were significantly decreased the body weights in the ovariectomized rats. Our findings suggest that vitamin E and selenium may influence the process of lipid packaging and absorption in the ovariectomized rats.

Acute Toxicity and General Pharmacological Action of QGC EXT

  • It has been shown that QGC isolated and purified from Rumecis folium found protective effects of gastritis and esophagitis which EXT is an ethanol extract of it. We examined acute toxicity and the general pharmacological action of QGC EXT to search for any side effects of it in rats, mice, guinea pigs, and cats. In a single dose toxicity study, QGC EXT didn't show toxicological effects in rats and mice, and the $LD_{50}$ was over 5 g/kg in both animals, and there were also no changes in weight, feed and water intake during these toxicological experimental periods. We examined the general pharmacological action on central controlled behavior responses, and peripheral organs including blood pressure, heart rate, respiration and gastrointestinal system, We found that there were no significant changes in body temperature, locomotors activity, stereotyped behaviors, sleeping time, and convulsion. In other studies, writhing reaction, normal body temperature, there did not appear to be any changes. The large intestine movement and electrical field stimulation-induced contraction was not changes by its EXT. In addition, the influences on blood pressure, heart rates, and respiration by QGC EXT were not found. These results indicate that QGC EXT may be very safe as a new drug, since its $LD_{50}$ was very high over 5 g/kg and any side effects were not found.

Change of energy consumption according to loading on the ankle of normal adults during gait (정상 성인의 발목에 부가된 하중에 의한 보행 중 에너지 소모도 변화)

  • The purpose of this study was to evaluate the change of the energy consumption when loading to leg of the 60persons who don't have past history of cardiopulmonary and neuromuscular disease, To evaluate the change or energy consumption, heart rate was measured in sitting position for 5minute, during walking for 3minute at for 4.8km on treadmill, and during resting state after walking with 1Kg loading to right ankle, and the other 1Kg loading was added to left ankle and then heart rates were measured in the The results were as follow; 1. PCI value without loading to Ankle were significantly increased compared to 1Kg, and 2Kg. (p<0.05) 2. Female Subjects showed mon increased PCI value in without leading and 2Kg loading compared to male subjects. ( p<0.05) 3. When 1Kg ana 2Ka loading to ankle significantly differences were showed between them. (p<0.05) 4. In the case of 1Kg and 2Kg loading, the difference among age groups was observed and the significant difference among PCI, PCI 1kg, PCI 2kg was showed in the only group that is less than 30 years old. 5. In every PCI condition the difference among height groups was observed and the significant difference among PCI conditions was showed in the only group that is less than 165cm. 6. The difference among weight groups in each PCI condition was not observed, but the significant differences among PCI conditions was showed in every group except the group that h from 60kg to 69kg. These results showed that energy consumption was increased according to loading on the ankle during Sate so weight of orthosis or prosthesis met be considered when choosing them and during gait training with these ones.

The Physiological Responses of Material-improved Working Clothes for Construction Site Worker (건설현장 작업복의 소재 기능성 향상에 따른 생리반응)

  • In this paper, experiments to develop working clothes and evaluated, including the current and material-improved working clothes in relation to physiological functionality measurement. Experiments were conducted on subjects after wearing working clothes in an climate chamber, and the thermo-physiological response, such as human body temperature, micro-climate within the clothes, blood pressure, heart rate were measured. In this manner, the physiological functionality of improved working clothes was compared with that of current working clothes and evaluated. The summary of obtained results is as follows: For physiological functionality evaluation through material-improved working clothes, P working clothes showed significantly lower rectal temperature than C working clothes. For mean skin temperature, P's skin temperature was significantly higher than C's in the second half of the experiment. P working clothes's temperature around the thighs in Micro climate was significantly lower than that of the C working clothes. Also, humidity within the clothes showed similar trends. During the exercise period, C working clothes showed higher blood pressure than P, but P showed higher heart rates than C. Also, the oxygen uptake amount was higher in C than P during the exercise period, it explains that the energy consumption amount of P working clothes was smaller than that of C working clothes. Of the subjective evaluation, for temperature sensation, workers wearing P working clothes felt cooler. For humidity, C working clothes showed more humidity. For comfort, P working clothes were better, and for sense of fatigue, workers felt less tired wearing P working clothes. From results above, we can see that physiological functionality improved in the material-improved working clothes in the working clothes for construction site workers. The improvement of working clothes through functionality improvements not only will provide personal pleasantness to constriction site workers, but will also generate efficiency and productivity improvements at construction sites. All in all, the continuous study of functionality improvements in working clothes taking into consideration the human body's physiological responses is required.

Acute Toxicity Assessment in Zebrafish Danio rerio of Arsenic-rich Extracts from Three Species of Seaweeds (제브라피쉬(Danio rerio)를 이용한 비소 고함류 3종 해조류 추출물의 급성 독성평가)

  • Seaweeds are composed of a variety of bioactive substances, including polysaccharides, pigments, minerals, peptides, and polyphenols. Among these substances, the arsenic content of seaweeds has been a significant cause for concern. The present study evaluated the toxicity of arsenic from three species of seaweed using a zebrafish Danio rerio model. The arsenic-rich extracts were obtained from Ecklonia cava (ECAE), Undaria pinnatifida (UPAE) and Hizikia fusiformis (HFAE) using a solvent of 50% methanol and 1% $HNO_3$. We investigated the toxicity of the arsenic-rich extracts in zebrafish embryos through survival rate, heart rate, yolk sac edema size, cell death, reactive oxygen species (ROS) production and real-time polymerase chain reaction (PCR). The hepatotoxicity of arsenic-rich extracts was assessed in the liver of adult zebrafish through real-time PCR and histopathology. The survival rates of embryos and adult zebrafish showed no significant changes at any concentration. At 100 ppm, embryos did not exhibit significant differences in heart rate, yolk sac edema size, cell death or ROS production. In addition, apoptosis-related genes in larvae and liver tissue were unaffected by treatment with arsenic-rich extracts. These data will help clarify that developmental changes, hepatic oxidative stress, and apoptosis are not associated with toxicity from arsenic-rich seaweed extracts in a zebrafish model.

  • In this study, we modeled moving-actuator type Total Artificial Heart (TAH) with cardiovascular system as a form of electric circuit. The bronchial circulation, important for the imbalance between the left cardiac output and the right one, was considered and added to the model. In the model, the relations of hemodynamic variables, just as blood pressures, volumes, or flow rates of each part of body, can be expressed as simultaneous first order ordinary differential equations. To solve the equations by the numerical analysis, Runge-Kutta forth order approximation method was adopted. The simulation software (SimTAH), implemented in C++ as a window-based application program, was developed to display the hemodynamic variables and to receive control inputs from users. SimTAH was evaluated by comparison of the simulation results with the results of mock-circulation tests, in vitro.

Differential effects of the valenced content and the interaction with pacing on information processing while watching video clips (영상물 시청에 발현된 감성 유인가의 차별적 영향과 편집속도와의 상호작용)

  • This study investigates differential impacts of the positive and negative content and the interaction with pacing, as a structural feature, on information processing while watching televised video clips with moderately intensive emotional tone. College participants watched six positive messages and six negative video clips lasting approximately 60 seconds. Heart rate was used to index attention and skin conductance was used to measure arousal. After all of the stimuli were shown, the participants performed the free recall questionnaire. The result demonstrates, first, positivity superiority on attention in which participants' heart rates were slower during positive content compared to during negative content. Secondly, negativity superiority was shown on free recall memory as participants remembered positive content better than did negative content. The result also manifests the interaction of emotional valence and pacing as the effects of pacing were less for the negatively emotional content compared to those for the positively emotional content. It is suggested that future studies should examine further about the differential and independent functions of positive and negative contents on information processing and the potential interaction with formal features.

A Comparison of the Effects of Concomitant Analgesics with Midazolam for Sedative Dental Therapy

  • Purpose: Intravenous sedation with midazolam is common in contemporary dentistry. That is effective for anxious patients, but additional analgesic agent needs to be used, because midazolam alone doesn't have an analgesic effect. This study was performed to select an analgesic agent between an opioid agent, and nonsteroidal anti-inflammatory drugs as adjunctives in intravenous sedation with midazolam. Methods: The subjects were 60 patients who visited the Department of Oral and Maxillofacial Surgery, Sacred Heart Hospital, Hallym University, between August 2009 and February 2010. Conscious sedation was performed on 20 patients of 3 groups (control group, ketorolac group, and fentanyl group), who were divided randomly. The analgesic agent was administrated preoperatively. For sedation, vital signs were recorded. After sedation and operation, subjective questionnaires of the patient and operator were implemented. Results: All of the $SPO_2$, blood pressure, and heart rates stayed within the normal range for sedation. The sedation depth and analgesic effect of the ketorolac group and fentanyl group were similar. In the case of sedation depth, 12 patients in the ketorolac group and 14 patients in the fentanyl group had no memory of surgery. In the case of analgesic effect, the visual analogue scale of pain scored 2~3 in 13 patients in the ketorolac group, and 0~2 in 12 patients in the fentanyl group. The satisfaction of patients and doctors was also similar. Conclusion: Considering the management and complication of an opioid agent, non-steroidal anti-inflammatory drugs is more effective than an opioid agent.

Study on the Inhibitory Mechanism of Deterioration Rate of Contractility in Papillary Muscle from Ginseng Treated Rat Heart. (인삼을 경구투여한 흰쥐 심장근의 수축력 퇴화율 억제에 미치는 작용 기전 연구)

  • It was previously reported that the deterioration rate of contractile force of the isolated heart from ginseng extract treated rat was slower than that from control. Present study was carried out to investigate the mechanism of the action of ginseng on the contractile force of the papillary muscle in terms of calcium metabolism. Rats weighing 200-300g were administered orally with ginseng ethanol extract (100mg/kg/day) for more than 10 days. The isolated papillary muscles from rat hearts were suspended in bath containing Krebs-Henseleit solution. When equilibration of contractile force of papillary muscle was reached, the rates of deterioration of contractile forces of papillary muscle from ginseng component treated rats were determined by washing with Ca-free Krebs-Henseleit solution and compared with that of normal hearts. At the beginning of washing, the rate of deterioration of contractile force of the papillary muscle was slower significantly in ginseng treated rats than in control rats, suggesting that calcium may be somehow involved in sustaining the contractility in ginseng treated hearts. Anoxia of papillary muscle with nitrogen gas to muscles inhibited the contractility, but differences between control and ginseng treated groups in the rate of deterioration were not observed. Influence of diltiazem, calcium blocker, on the contractility of papillary muscle from ginseng treated and control hearts was studied. Contractility of papillary muscle from control and ginseng treated hearts was inhibited by diltiazem in dose dependent manner but the inhibition of the ginseng treated muscles was much weak. The effect was significantly different. From the results, it seemed that slowing in deterioration rate of papillary muscle from ginseng treated hearts might be related to calcium which was mobilized from plasma membrane of internal organelle by ginseng.

Interaction effects of pen environment and sex on behavior, skin lesions and physiology of Windsnyer pigs

  • Objective: The study was carried to determine the interaction effects of pen enrichment and sex on behavioral activities, skin lesions and physiology of Windsnyer pigs. Methods: Forty-eight growing Windsnyer pigs of both sex, with an average initial body weight of 21.6 (${\pm}9.01$) kg were used. Four pigs were randomly assigned to either enriched or barren pens at a stocking density of $0.35m^2/pig$. Enriched pens contained 2 L bottles filled with stones and suspended at head level on ropes stretching across the pens. In addition, two plastic balls (90 mm in diameter) and 500 mL bottles (235 mm long) were placed on the floor of each enriched pen. Results: Pigs in barren environments had higher heart rates (p<0.001) than those in enriched pens. There was an interaction of pen environment and sex on rectal temperature (p<0.001). Females in enriched pens had higher rectal temperatures (p<0.05) than females in barren pens. There was no interaction of pen environment and sex on time spent eating and drinking (p>0.05). Time spent bullying was influenced (p<0.05) by pen environment and sex. Female pigs in barren environment spent more time on bullying than females in enriched pens. There was an interaction of pen environment and sex on time spent lying down and walking (p<0.05). Female pigs in enriched pens spent more time lying down than females in barren pens. Males in barren pens spent more time walking than males in enriched pens while no effect of pen environment was observed in females. There was an interaction of pen environment and sex on the number of skin lesions in the head, neck and shoulder region and other parts of the body (p<0.05). Conclusion: It was concluded that pen enrichment reduced the number of skin lesions and anti-social behaviors, especially for female pigs. There is a need, therefore of housing indigenous pigs under confinement.
