부착성 HeLa 세포의 탈리 유도에 의한 다공성 미립담체의 담체간 전이 배양 (Bead-to-Bead Cell Transfer by Induction of Detachment of Anchorage Dependent HeLa Cells Grown on Macroporous Microcarriers)

  • Cellulose로 만들어진 다공성 미립담체를 이용하여 HeLa cell을 working volume 100mL의 spinner flask에서 배양하였으며, 세포가 완전히 자란 미립담제를 계대배양하기 위하여 담체간 세포 전이 배양 방법을 시도하였다. 부유세포의 농도는 다공성 미립담체-HeLa 시스템의 경우에 담체간 세포 전이 배양에 영향을 미치는 중요한 인자로 작용하였으며, 낮은 칼슘농도의 배지인 RPMI-1640과 빠른 교반 속도를 이용하여 활성을 유지한 많은 세포가 떨어지도록 유도하였으며, 담체간 세포 전이 배양을 효과적으로 3회 이상 실시할 수 있었다. 이렇게 배양한 세포에 재 조합 Vaccinia virus를 감염하여 그 수율을 비교한 결과 T-flask에서 떼어낸 세포로 접종한 미립담체 배양과 거의 비슷한 재조합 단백질($\beta$-galactosidase) 수율을 나타내었다. Trypsin 처리 방법에 의한 미립담체 계대 배양도 경우에 따라서는 유용한 미립담체 계대 배양 방법이 될 수 있지만 실제 생산 규모에의 적용에는 공정이 복잡해지고 정확한 제어가 필요하다는 등의 문제가 있다. 따라서, 추가적인 비용이나 공정이 필요 없는 간편한 방법인 담체간 세포 전이 배양은 동물세포 배양을 이용한 유용 단백질 및 바이러스 생산 공정의 규모 증대에 매우 유용한 수단이다.

아드리아마이신에 의한 HeLa 세포의 자살 과정 중 p53 유전자의 돌연변이 빈발 부위에서의 핵산 변화의 부재 (Lack of p53 Gene Nucleotide Change in Mutation Hot Spots During HeLa Cell Apoptosis by Adriamycin)

  • 세포의 자살 현상은 항암제를 이용한 치료법에 있어 중요하다. p53 은 세포 자살을 유발하여 암세포를 죽이는 핵심적 요소임이 밝혀졌다. 그러나 최근의 연구는 p53 과 무관한 세포 자살 경로가 있음을 보였다 (Gaftenhaus 외, 1996). 본 저자들은 유전독성적 항암제인 아드리아마이신에 의하여 유도된 세포 자살 과정에서 p53 유전자의 돌연변이가 일어나는지를 관찰하였다. 그러므로 본 연구는 아드리아마이신에 의하여 유도된 세포 자살 과정에서 HeLa 세포 집단에서의 p53 유전자의 돌연변이 상태를 조사하였다. DNA 분절 현상으로 관찰한 바, 본 실험 조건하에서는 아드리아마이신을 1 uM 농도로 12 시간 처리하였을 때 세포 자살 현상이 일어났다. p53 유전자의 돌연 변이 상태를 관찰하고자 돌연변이 빈발 부위가 존재한다고 알려진 exon 5, 7 및 8 부위를 종합 효소연쇄반응으로 증폭변화는 관찰되지 않았다. 그러므로 본 연구는 p53 유전자 손상없이 아드리아마이신에 의하여 세포 자살이 유도됨을 밝혔다.

Inotodiol Inhabits Proliferation and Induces Apoptosis through Modulating Expression of cyclinE, p27, bcl-2, and bax in Human Cervical Cancer HeLa Cells

  • Inonotus obliquus is a medicinal mushroom that has been used as an effective agent to treat various diseases such as diabetes, tuberculosis and cancer. Inotodiol, an included triterpenoid shows significant anti-tumor effect. However, the mechanisms have not been well documented. In this study, we aimed to explore the effect of inotodiol on proliferation and apoptosis in human cervical cancer HeLa cells and investigated the underlying molecular mechanisms. HeLa cells were treated with different concentrations of inotodiol. The MTT assay was used to evaluate cell proliferating ability, flow cytometry (FCM) was employed for cell cycle analysis and cell apoptosis, while expression of cyclinE, p27, bcl-2 and bax was detected by immunocytochemistry. Proliferation of HeLa cells was inhibited by inotodiolin a dose-dependent manner at 24h (r=0.9999, p<0.01). A sub-$G_1$ peak (apoptotic cells) of HeLa cells was detected after treatment and the apoptosis rate with the concentration and longer incubation time (r=1.0, p<0.01), while the percentage of cells in S phase and $G_2$/M phase decreased significantly. Immunocytochemistry assay showed that the expression of cyclin E and bcl-2 in the treated cells significantly decreased, while the expression of p27 and bax obviously increased, compared with the control group (p<0.05). The results of our research indicate that inotodiol isolated from Inonotus obliquus inhibited the proliferation of HeLa cells and induced apoptosis in vitro. The mechanisms may be related to promoting apoptosis through increasing the expression of bax and cutting bcl-2 and affecting the cell cycle by down-regulation the expression of cyclin E and up-regulation of p27. The results further indicate the potential value of inotodiol for treatment of human cervical cancer.

TRAIL 유도 세포사멸에 있어서 IFN-γ의한 증가 기전 연구: IRF-1과의 관련성 (IRF-1-mediated IFN-γ enhancement of TRAIL-induced apoptosis)

  • Tumor necrosis factor (TNF)-related apoptosis-inducing ligand (TRAIL) is a member of the TNF family and potent inducer of apoptosis. TRAIL has been shown to effectively limit tumor growth in vivo without detectable cytotoxic side effects. Interferon (IFN)-${\gamma}$ often modulates the anti-cancer activities of TNF family members including TRAIL. We previously reported that IFN-${\gamma}$ enhanced TRAIL-induced Apoptosis in HeLa cells without the unknown mechanism. In this study, we investigated whether IRF-1 involves in IFN-${\gamma}$-enhanced TRAIL-induced apoptosis. We exposed HeLa cells to IFN-${\gamma}$ for 12 hours and then treated with recombinant TRAIL protein. No apoptosis was induced in cells pretreated with IFN-${\gamma}$, and TRAIL only induced 30% apoptosis after 3 hours treatment. In HeLa cells pretreated with IFN-${\gamma}$, TRAIL induced cell death to more than 75% at 3 hours, showed that IFN-${\gamma}$-pretreatment enhanced HeLa cell death to TRAIL-induced apoptosis. To investigate the functional role of IRF-1 in IFN-${\gamma}$-enhanced TRAIL-induced apoptosis, IRF-1 was overexpressed by using an adenoviral vector AdIRF-1. IRF-1 overexpression increased apoptotic cell death and significantly enhanced apoptotic cell death induced by TRAIL when infected cells were treated with TRAIL. Our findings show that IFN-${\gamma}$ enhances TRAIL-induced apoptosis by IRF-1 in HeLa cells.

몰약(沒藥)이 자궁경부암세포(子宮經部癌細胞)(HeLa Cell)의 Apoptosis에 미치는 영향(影響) (Myrrha-induced Apoptosis in Human Cervical Carcinoma HeLa Cells)

  • Purpose : To address the ability of Myrrha (MY) to induce cell death, we investigated the effect of MY on apoptosis. In human cervical carcinoma HeLa cells, apoptosis occurred following MY exposure in a dose-dependent manner. Methods : We have tested several kinds of anti-oxidants to investigate the MY-induced apoptotic mechanism. Among the anti-oxidants, N-acetyl cysteine(NAC) or reduced glutathione (GSH) protects MY-induced apoptosis. NAC is an aminothiol and synthetic precursor of intracellular cysteine and GSH. To confirm the role of GSH in MY-induced apoptosis, methionine and cystathionine-glutathione extrusion inhibitors were treated in the presence of MY. Results : NAC, GSH, methionine or cystathionine led to protective effect against MY-induced apoptosis in HeLa cells. The GSH and GSH-associated reagents regulate MY-induced cytochrome c release and the resultant caspase-3 activation. Furthermore, the two specific inhibitors of carrier-mediated GSH extrusion, methionine and cystathionine demonstrate GSH extrusion occurs via a specific mechanism. While decreasing GSH extrusion and protecting against MY-induced apoptosis, methionine and cystathionine failed to exert anti-apoptotic activity in cells previously deprived of GSH. Conclusion : the target of the protection is indeed GSH extrusion. This shows that the protective effect is achieved by forcing GSH to stay within the cells during apoptogenic treatment. All this evidence indicates the extrusion of GSH precedes andis responsible for the apoptosis, probably by altering the intracellular redox state, thus giving a rationale for the development of redox-dependent apoptosis in MY-treated human cervical carcinoma HeLa cells.

Mitofusin-2 Promotes the Epithelial-Mesenchymal Transition-Induced Cervical Cancer Progression

  • High expression of mitofusin-2 (MFN2), a mitochondrial fusion protein, has been frequently associated with poor prognosis of patients with cervical cancer. Here, we aimed to identify the function of MFN2 in cervical cancer to understand its influence on disease prognosis. To this end, from cervical adenocarcinoma, we performed an MTT assay and quantitative RT-PCR (qRT-PCR) analysis to assess the effects of MFN2 on the proliferation and of HeLa cells. Then, colony-formation ability and tumorigenesis were evaluated using a tumor xenograft mouse model. The migration ability related to MFN2 was also measured using a wound healing assay. Consequently, epithelial-mesenchymal transition (EMT) of MFN2-knockdowned HeLa cells originating from adenocarcinoma. markers related to MFN2 were assessed by qRT-PCR. Clinical data were analyzed using cBioPortal and The Cancer Genome Atlas. We found that MFN2 knockdown reduced the proliferation, colony formation ability, migration, and in vivo tumorigenesis of HeLa cells. Primarily, migration of MFN2-knockdowned HeLa cells decreased through the suppression of EMT. Thus, we concluded that MFN2 facilitates cancer progression and in vivo tumorigenesis in HeLa cells. These findings suggest that MFN2 could be a novel target to regulate the EMT program and tumorigenic potential in HeLa cells and might serve as a therapeutic target for cervical cancer. Taken together, this study is expected to contribute to the treatment of patients with cervical cancer.

Inhibition of Cell Cycle Progression and Induction of Apoptosis in HeLa Cells by HY558-1, a Novel CDK Inhibitor Isolated from Penicillium minioluteum F558

  • In the course of screening for a novel inhibitor of CDC2, HY558-1 was isolated from a culture broth of Penicillium minioluteum F558. Moreover, it was found that HY558-1 had an effect on both the cell cycle regulation and apoptosis of human cervical adenocarcinoma HeLa cells. A flow cytometric analysis of HeLa cells revealed appreciable cell cycle arrest at the G1 and G2/M phases following treatment with HY558-1. Furthermore, DNA fragmentation due to apoptosis was observed in HeLa cells treated with HY558-1. To obtain further information on the cell cycle arrest and apoptotic induction induced by HY558-1, the expression of certain cell cycle and apoptosis-associated proteins was examined using a Western blot analysis. The results revealed that HY558-1 inhibited the phosphorylation of pRb and decreased the expression levels of CDK2, CDC2, and cyclin A in the cell cycle progression. It was also shown that the level of $p21^{WAF1/CIP1}$ was increased in HeLa cells treated with 0.52 mM of HY558-1. Accordingly, HY558-1 was found to inhibit the proliferation of HeLa cells through the induction of G1 phase arrest by inhibiting pRb phosphorylation via an upregulation of $p21^{WAF1/CIP1}$, and G2/M phase arrest by directly inhibiting CDC2 and cyclin A. Moreover, HeLa cells treated with 0.52 mM of HY558-1 exhibited apoptotic induction associated with the cleavage of Bid and release of cytochrome c from mitochondria into the cytosol. Subsequent investigation of the activation of caspase-3 and cleavage of poly (ADP-ribose) polymerase (PARP) suggested that the mitochondrial pathway was primarily involved in the HY558-1-induced apoptosis in HeLa cells.

Betaine Effects on Morphology, Proliferation, and p53-induced Apoptosis of HeLa Cervical Carcinoma Cells in Vitro

  • Objectives: To investigate the effects of betaine on HeLa cell growth and apoptosis and molecular mechanisms. Materials and Methods: Concentrations of 0.1, 1.0, 5.0, 20.0, 100.0 mg/ml of betaine were used to evaluate the anticancer efficacy for HeLa cells respectively, and MCF-10A was also detected as a normal diploid cell control. Results: We found that proliferation of HeLa cells was inhibited significantly upon exposure to increasing betaine levels with the MTT test (p<0.05). The percentage of S phase cells in the low dose groups (<5mg/ml) were distinctly higher than in high dose groups, and the rates of Sub-G1 phase were the opposite (p<0.01); A high concentration of betaine (>5.0mg/ml) significantly promoted the apoptosis of HeLa cells (p<0.01). SOD activities of the low dose groups were slightly higher than the control group (p<0.05) and there were obvious synchronicity and correlation among the expression of promoting apoptosis genes Bax, P53, Caspase 3 and apoptosis suppression gene Bcl-2. In response to an apoptosis-inducing stimulus, p53 and cyclin D1 could be activated with blockage of the cell cycle at G1/S or S/G2 checkpoints. Conclusions: Our data showed that betaine could promote HeLa cells proliferation in vitro at low concentrations. In contrast, high concentrations could significantly inhibit cell growth and migration, and induce apoptosis of HeLa cells through caspase 3 signaling and further promoted necrosis. This might imply that betaine exhibits tumoricidal effects and acts as a biological response modifier in cancer treatment by inducing apoptosis and cell cycle arrest in a dose and time-dependent manner.

넓패 추출물이 HeLa 자궁암세포의 세포사멸에 미치는 영향 (Ishige sinicola Extracts Induce Apoptosis via Activation of a Caspase Cascade in Human HeLa Cells)

  • 본 연구에서는 넓패 메탄올 추출물의 농도별 처리가 인체 자궁암 세포 HeLa의 세포사멸에 미치는 영향을 확인하기 위하여 세포독성 측정, Hoechst 33258 staining, flow cytometry 분석을 통하여 세포사멸을 확인하였다. 넓패 메탄올 추출물 처리 시 HeLa 세포에서 농도 의존적으로 세포의 증식을 억제하였으며, 또한 넓패 메탄올 추출물은 농도 의존적으로 핵을 응축하고 apoptotic bodies을 생성하였다. 유세포 분석을 통하여 apoptosis를 측정한 결과, 넓패 메탄올 추출물의 농도가 증가함에 따라 유의적으로 apoptotic 세포가 증가하였다. Western blot을 통해 PARP 단백질의 절단 현상을 분석한 결과, 넓패 메탄올 추출물의 처리 농도와 시간에 따라 PARP 단백질의 절단 현상이 증가하였다. 또한 넓패 메탄올 추출물은 caspase-8, caspase-9 및 caspase-3 활성을 농도와 시간에 따라 증가시켰으며, caspase 저해제인 z-VAD-fmk로 처리 시 넓패 메탄올 추출물에 의한 세포사멸이 유의적으로 감소되어 넓패 메탄올 추출물에 의한 HeLa 세포의 apoptosis 유도에 caspase가 중요한 역할을 하고 있음을 확인하였다. 따라서 넓패 메탄올 추출물은 HeLa 자궁암 세포의 apoptosis를 유도하는 것으로 나타나 넓패의 항암효과 가능성을 제시하였다.

선택적 COX-2 억제제 NS 398과 EGF 수용체 차단제 AG 1478의 복합투여가 HeLa 세포주의 방사선 감수성에 미치는 영향 (The Modulation of Radiosensitivity by Combined Treatment of Selective COX-2 Inhibitor, NS 398 and EGF Receptor Blocker AG 1478 in HeLa Cell Line)

  • 목적 : 분자 표적의 선택적 억제가 방사선 세포 살상 효과를 증진시키는 것으로 알려져 있으므로 선택적 COX-2 억제제와 EGF 수용체 차단제를 HeLa 세포주에 처리한 후 방사선 효과의 상승작용을 알아보고자 하였다. 대상 및 방법 : 자궁경부암 세포주인 HeLa세포에서 EGF 수용체 차단제 AG 1478, 선택적 COX-2 억제제 NS 398과 방사선을 복합 투여하여 세포성장 억제 분석(cell graph inhibition assay)과 세포사멸 분석(apoptosis assay)을 시행하였고, 방사선 감수성 변화를 살펴보기 위해 세포생존 분석(clonogenic survival assay)을 시행하였다. 방사선 감수성 인자로는 2 Gy에서의 세포생존분획($SF_2$)과 linear-quadratic model을 이용한 dose enhancement ratio (DER)를 사용하였다. 방사선 감수성에 대한 작용기전 분석을 위해 flow cytometry로 세포주기 분석(cell cycle analysls)을 시행하였고, western blot 분석을 통하여 bcl-2와 bax 단백질의 발현 변화를 살펴보았다. 결과 : HeLa세포에 NS 398과 AG 1478을 방사선과 함께 복합 투여한 실험 군에서 세포사멸 정도가 가장 높게 나타났다($8.49\%$ vs. $22.70\%$). 세포주기 분석 결과, 방사선과 복합 약물 처리군에서 $G_0/G_l$ 세포주기 정체와 5 세포 분획 소실이 나타났으며 이러한 변화는 72시간 이후까지 지속되었다 세포생존 분석 결과로는 방사선과 AG 1478군에서 $SF_{2}0.68{\pm}0.07$, DER 1.12를 보인 반면, 방사선과 복합약물처리군에서는 $SF_{2}0.12{\pm}0.01,\;DER\;3.00$으로 나타났다. Western blot분석에서는 방사선과 복합약물처리군에서 bcl-2와 bax 단백질 발현이 모두 감소하는 양상을 보였다. 결론 : 신호전달 체계를 억제하는 분자 표적 약제인 선택적 COX-2 억제제와 EGF 수용체 차단제를 방사선과 복합투여함으로써 HeLa세포의 방사선 감수성이 증가됨을 확인하였다.
