Effect of deep-sea mineral water on growth performance, water intake, blood characteristics and serum immunoglobulins in the growing-finishing pigs

  • Brine mineral water (BMW) is groundwater near the deep sea, and the mineral component of the BMW is different from the water of the deep sea because the components of BMW are derived from the unique geographical features surrounding it. Recently, BMW has attracted attention due to the unique health-related minerals it possesses; however, the influence of BMW on physiological function has not yet been determined in domestic animals. Therefore, this experiment investigated the influence of BMW on the growth performance, water intake, blood properties, and immunoglobulin (Ig) levels of serum in growing-finishing pigs. A total of 64 pig barrows (Landrace × Yorkshire × Duroc) with an average initial weight of 40.56 ± 0.17 kg were used in the experiment, and 0%, 2%, 3%, and 5% samples of BMW diluted with freshwater were provided to experimental animals during the 56 days. We found that the gain/feed ratio in the 3% BMW group was significantly higher than that in the 5% BMW group of growing-finishing pigs (p < 0.05). The water intake was significantly increased in the 5% BMW group compared with the other groups (p < 0.05) of growing-finishing pigs. Additionally, the concentrations of red blood cells (RBCs), hemoglobin (HGB), and hematocrit (HCT) were significantly higher in the 3% BMW group than in the control group. The level of high-density lipoprotein cholesterol was higher in the 3% BMW group than in the 5% BMW group (p < 0.05). Furthermore, IgG and IgM levels in the serum were significantly higher in the 2% and 3% BMW groups than in the control group (p < 0.05). These results suggest that a dilution of 3% BMW in the intake water could improve the levels of RBCs and serum Igs in growing-finishing pigs.

Iron Deficiency and Early, Low-dose Iron Supplementation in Breast-fed Infants (모유 수유아의 철 결핍과 조기 저용량 철분보충요법의 효과에 대한 연구)

  • Purpose: The purpose of this study was to determine the efficacy of early low-dose iron supplementation in term breast-fed infants. Methods: Eighty-seven healthy term infants were divided into 3 groups: A, formula-fed; B, breast-fed only; S, breast-fed with iron supplementation (5 mg/day from 2 months of age). We measured ferritin, iron, total iron binding capacity (TIBC), transferrin saturation rate (TFSAT), hemoglobin (Hb), hematocrit (Hct), mean corpuscular volume (MCV), mean corpuscular hemoglobin (MCH), and red cell distribution width (RDW) at birth, 6 months of age, and 12 months of age. Results: 1) At 6 months of age, ferritin, iron, TFSAT, and Hb in Group B were the lowest among the 3 groups, whereas TIBC and RDW were the highest. The incidences of iron deficiency (ID) and iron deficiency anemia (IDA) in Group B were 33% and 30%, respectively, significantly higher than those seen in Groups A (5% and 8%, respectively) and S (7% and 5%, respectively). 2) At 12 months of age, ferritin, TFSAT, Hb, MCV, and MCH in Group B were the lowest among the 3 groups, whereas TIBC and RDW were the highest. Iron and Hct did not differ among the 3 groups. The incidences of ID and IDA in Group B were 64% and 50%, respectively, again significantly higher than those seen in Groups A (4% and 3%, respectively) and S (9% and 7%, respectively). Conclusion: The prevalences of ID and IDA were higher in breast-fed infants than in formula-fed infants, even at 6 months of age. Early and low-dose iron supplementation in breast-fed infants improved iron status and lowered the incidence of iron deficiency anemia in early infancy.

The Relation of BMI, Smoking and Drinking with Biomedical Parameters in 20s and 30s Men (20~30대 성인 남성에서 체질량지수, 흡연량 및 음주량과 혈액검사치의 관련성)

  • The purpose of this study was to explore the relation of BMI, smoking and drinking with biomedical parameters in 20s and 30s men. This was a retrospective exploratory study and the subjects were 603 men who participated in health check-up for clinical trial at a tertiary hospital between October 2006 and June 2011. RBC, hemoglobin(Hb), hematocrit(Hct), triglyceride(TG), uric acid(UA), ALT in obese group were significantly higher than those of normal weight group. WBC and Hb in smokers were significantly higher than those of nonsmokers, protein in smokers was significantly lower than that of nonsmokers. Platelet in moderate smokers who smoke 10~19 cigarettes a day was significantly higher than that of nonsmokers and light smokers who smoke below 10 cigarettes a day, albumin in moderate smokers was significantly lower than that of nonsmokers and light smokers. TG in moderate smokers was significantly higher than that of nonsmokers, and blood glucose(BG) in moderate smokers was significantly lower than that of nonsmokers. AST and ALT in drinkers who drink over 10g a day were significantly higher than those of nondrinkers. In regression, BMI was significant factors of increasing RBC, Hb, Hct, total cholesterol(TC), TG, BG, UA, AST and ALT. Smoking amount was significant factors of increasing WBC, Hb, Hct and TG, and was significant factors of decreasing BG and Ca. Smoking amount was significant factors of increasing TC, AST and ALT, and was significant factors of decreasing WBC. In conclusion, there was significant relations of BMI, smoking, and drinking with biomedical parameters such as Hb, TC, TG, and ALT in young-aged men. Therefore, practice of healthy lifestyle activities to include preventing obesity and heavy drinking, and quitting smoking needs to the 20s and 30s men for preventing lifestyle disease.

  • 배경: 혈전 생성은 심혈관계 삽입물이나 순환 보조장치의 발전에 있어서 중요한 문제이므로 이러한 장치들의 혈전성에 대한 평가는 필수적이다. 따라서 이러한 장치들의 혈전성 연구를 시행하기 위한 효과적인 생체 외 실험 방식이 개발되어 원심성 순환 펌프의 발전에 큰 도움이 되고 있으며, 최근 엄밀하게 혈액과 공기의 접촉을 차단하는 방식이 믿을 수 있는 생체 외 실험 방식으로 강조되고 있다. 본 논문에서는 모의 순환 회로를 이용하여 기존에 행해진 연구 방법 상의 공기 접촉에 따른 핼액 응고 기전의 활성화 여부를 알아보기 위하여 공기 접촉 여부에 따른 혈액 인자의 변화를 관찰하여 확인하고, 또한 회로 내에서 생성된 혈전을 생체 내에서 형성된 혈전과 비교 분석하여 이 모의 순환 회로를 이용한 생체 외 검사가 새로 고안된 심혈관계 장치들의 혈전성을 평가하는데 있어서 동물실험을 대신할 수 있다는 기존의 주장을 검증하고자 하였다. 연구재료 및 방법: 바이오펌프를 이용한 같은 모의 순환 회로 한 쌍을 준비하고 동일 개체에서 얻은 헤파린을 첨가한(1u/$m\ell$) 신선한 혈액을 공기와 접촉시키지 않도록 한 A-회로와 공기와 접촉시킨 B-회로에 충전시켜 실험을 동시에 진행하여 결과를 얻었으며 총 12번 시행하였다. 회로에 충전한 혈액의 activated clotting time(ACT)은 정상의 3~5배였으며 ACT가 정상 범위의 1.5배가되면 실험을 끝냈고, Hematocrit(HCT), 혈소판, 동맥혈가스분석(ABGA), factor Ⅷ, factor \ulcorner, 섬유소원, thromboxane B2(TXB2), free hemoglobin(fHb) 등을 측정하였다. 실험이 끝난 후 A-회로와 B-회로에서 얻은 각 검사를 평가하고 분석하였으며 생성된 혈전을 생체 내에서 생성된 혈전과 조직 검사로 비교하였다. 본 연구의 자료분석은 SPSS 통계 프로그램을 이용하였고 유의수준 0.05를 기준으로 유의도를 판단하였다. 결과: 정상 ACT는 186.9$\pm$20.5초(평균$\pm$표준편차)이었고, 헤파린을 넣은 초기 ACT는 982.2$\pm$165.5초이었으며, 실험시간은 보통 60분에서 180분 사이였다. HCT, fHb, 혈소판, TXB2, factor Ⅷ, factor \ulcorner, 섬유원소의 초기값은 35.5$\pm$3.2%, 12.4$\pm$6.5 mg/dL, 354$\pm$56($\times$103)/$\mu$L, 72.6$\pm$15.1 mg/dL, 29.3$\pm$1.2%, 137.9$\pm$42.1 mg/dL, 그리고 17.7$\pm$9.7%이었다. 공기 접촉에 따른 차이를 보았을 때 ACT(p=0.398), HCT(p=0.988), fHb(p=0.898), 혈소판 (p=0.904), TXB2(p=0.985), factor Ⅷ(p=0.872), factor \ulcorner(p=0.489), 섬유원소(p=0.973)으로 전 예에서 통계적 유의성이 없었다. 실험 후 양 군에서 생성된 혈전의 현미경 소견은 B-회로의 혈전에 여러 군데 공기 방울이 관찰되는 것 이외에 A-회로에서 생성된 혈전과 차이가 없었고, 생체에서 생성된 혈전과 비교했을 때 그 양상이 같았다. A-회로와 B-회로에서 생성된 혈전의 총 량은 8.4$\pm$3.7 mL와 9.4$\pm$3.1 mL로 통계상 유의성은 없었다. (p=0.624). 결론: 모의 순환 회로를 이용한 혈전성 평가를 위한 생체 외 실험에 있어서 공기와의 접촉 여부는 혈액 응고 기전의 활성화나 혈전 생성에 별다른 영향을 미치지 않는다. 모의 순환 회로에서 형성된 혈전과 문헌에 있는 인체 내에서 형성된 혈전을 비교한 결과 그 형태가 일치하므로 이 모의 순환회로를 이용한 실험이 동물실험을 대신할 수 있다고 판단된다.

  • To establish the hematological reference values in the healthy adults visited our hospitals, following examination were done on 2823 persons by Coulter Counter Model S-plus II ; white blood cell count: (WBC), red blood cell count(RBC), hemoglobin(Hb), hematocrit(Hct), mean corpuscular volume (MCV), mean corpuscular hemoglobin(MCH), mean corpuscular hemoglobin concentration(MCHC), red cell distribution width(RDW), platelet, plateletcrit, mean platelet volume(MPV) and platelet distribution width(PDW). Effect of Acute Ethanol Intoxication on the Pulmonary Compliance and Surfactant in Rats (급성(急性) Ethyl 알콜 중독(中毒) 흰쥐의 폐용압률(肺容壓率)과 폐포활성물질(肺胞活性物質)의 변화(變化))

  • Relatively little has been done on the metabolic changes of the lung produced by the excessive alcohol ingestion to the point of the acute alcohol intoxication. In the present study, an effort was made to clarify the possible changes of the pulmonary surfactant system by the acute alcohol ingestion. The dynamic pulmonary compliance and the levels of protein and inorganic phosphorus (Pi) of both lung lavage and extract were chosen as the parameters of the pulmonary surfactant activities. The albino rats of both sexes were used, and 1.5 ml of 50% ethanol per 100 g body weight was given by oral intubation, and the experiment was performed at 1, 3, 6, 12, and 24 hours after the alcohol ingestion. The rat was sacrificed by cutting the carotid arteries, and blood sample for the determination of hematocrit(Hct) and the blood alcohol concentration was obtained. Both lungs were completely removed without dammage to the lung tissue, and the pulmonary compliance was measured by the changes of pressure-volume(P-V) curves by inflating or deflating the lung with air. Immediately after the P-V curves were recorded, the lung lavage was obtained by washing the lobes with 15ml of isotonic saline 3 times with a syringe. Next, total lungs were homogenized and filtered to obtain the lung extract. The protein and Pi levels were measured using the lung lavage and extract as the samples, and the lung/body weight ratio(L/B ratio) was also calculated. The results thus obtained were compared with the normal values and summarized as follows. The blood alcohol concentration reached the highest level of $0.71{\pm}0.02\;g\;%$ at 1 hr and gradually decreased until 24 hrs$(0.36{\pm}0.02\;g%)$ after the alcohol ingestion, but all the experimental groups showed significant increase comparing with the normal. The highest Hct value was obtained at 1hr$(64.86{\pm}2.45%)$ and significantly elevated value was continued throughout the experiment. The L/B ratio was significantly lowered from 3hrs until 24hrs after the alcohol ingestion but from 6 th hr on, a generally elevated value was observed with a significant value at 12 hrs and gradual recovery to the normal value at 24 hrs after the alcohol ingestion. The pulmonary compliance at inflation and deflation did not change appreciablly from the normal until 3 hrs after the alcohol ingestion but from 6 th hr on, a generally elevated value was observed with a significant value at 12 hrs and gradual recovery to the normal value at 24 hrs after the alcohol ingestion. The protein level of the lung lavage stowed a significantly increased value of $12.36{\pm}0.35\;mg/gm(3rd hr)$, $12.70{\pm}0.74\;mg/gm(12 th hr)$, and $12.65{\pm}0.88\;mg/gm(24 th hr)$, respectively, comparing with the normal value of $10.65{\pm}0.62\;mg/gm$, and the Pi level also showed a similar tendency of significant increase at 12th hr $(7.65{\pm}0.63\;{\mu}mol/gm)$ and 24 th hr$(6.70{\pm}0.36\;{\mu}mol/gm)$ comparing with the normal value of $5.32{\pm}0.20\;{\mu}mol/gm$. The protein level of the lung extract in the alcohol group was generally similar to the normal value with a slight decrease at 1st and 3 rd hr, tut the Pi level of the lung extract was generally increased in the alcohol group, and a significant increase was observed at 6 th hr$(17.77{\pm}1.54\;{\mu}mol/gm)$, 12 th hr$(13.92{\pm}0.78\;{\mu}mol/gm)$ and 24 th hr$(14.57{\pm}0.53\;{\mu}mol/gm)$ of the alcohol ingestion comparing with the normal value of $10.34{\pm}0.37\;{\mu}mol/gm$. From the above, it may be concluded that the acute alcohol intoxication produces the metabolic changes of the lungs by the increased surfactant activities and elevated pulmonary compliance.

Assessment of DNA Damage using an Alkaline Single Cell Gel Electrophoresis (SCGE) Comet Assay and Toxic Effects in Chickens by T-2 Toxin Treatment (T-2 toxin을 투여한 닭에서 Comet assay 방법을 이용한 DNA 손상 평가와 독성)

  • This study was designed to evaluate the possible DNA damaging effects of T-2 toxin using an alkaline single cell gel electrophoresis (SCGE) comet assay and also to investigate toxic effects in chickens. A total of 20 chickens were used in these experiments. Graded concentrations of dietary T-2 toxin (0, 4, 8, and $16{\mu}g/g$ of diet) were given to groups of 5 broiler chickens. In comet assay, The DNA damage was analysed by the tail extent moment (TEM) and tail length (TL), which were used as markers of DNA strand breaks in SCGE. A significant dose-dependent increase in the extent of DNA migration as well as in the percentage of cells with tails was observed after treatment with T-2 toxin (P<0.05). Treatment with the low T-2 toxin ($4{\mu}/g$ of diet) induced a relatively low level of DNA damage in comparison with the high T-2 toxin ($16{\mu}/g$ of diet) group. The growth rate was significantly reduced by concentrations of 8, and $16{\mu}/g$ of diet (P < 0.05). The feed conversion ratio were significantly affected by any concentrations (P < 0.05). The relative weight of the spleen, and lung was decreased by the growth inhibitory concentrations. The bursa of Fabricius, thymus, and kid- ney were decreased in relative weight by concentrations of $16{\mu}/g$ of diet. The relative weight of the liver and heart were unaffected. The hemoglobin (Hb), hematocrit (HCT), and mean corpuscular hemoglobin (MCH) were decreased at concentration of $16{\mu}/g$ of diet. As compared with control chickens, there was no marked change in serum components except uric acid in T-2 treated chickens. All lymphoid tissues retained atrophic and lymphoid cell depletion throughout the three weeks trial.

  • This study was performed to evaluate the effect of hemoglobin (Hb) on bone mineral density (BMD) in university students by performing a quantitative analysis. The subjects included healthy university students aged 20 to 30 years. Although osteoporosis has traditionally been considered as a disease of aging women, it is becoming an increasingly concerning male health problem. Diagnosis of osteoporosis is calculated with a quantitative assessment of BMD. Laboratory blood and urine tests are mainly used with low BMD or fragility fractures to identify any possible causes of bone metabolism disorders. In this study, there was no difference in BMD according to gender. The average red blood cell (RBC), Hb, and Hematocrit (HCT) were significantly higher in males (p<0.01). The correlation between lumbar spine, skeletal muscle mass (SMM), and basal metabolic rate (BMR) was statistically significant (p<0.01). Hb showed a 51.7% statistical influence on BMD by multiple regression analysis. These findings are useful to understand the relationship between BMD and Hb; lower Hb level is associated with lower BMD. The Hb level was the strongest predictor of abnormal BMD. In conclusion, this study showed that a low Hb value was significantly correlated with low bone mass, suggesting that a low Hb value is a risk factor for changes in bone turnover that leads to a decrease bone density.

Effects of Lentinus edodes Extract on the Loperamide-induced Constipation in Rats (표고버섯 추출액이 loperamide로 유도된 변비에 미치는 영향)

  • The preventive effect of Lentinus edodes extract (LE) against constipation was studied in rats. Rats were pretreated with LE contained in drinking water at the concentration of 10%, 20% and 40% over 30 days. Constipation was induced by subcutaneous injection of loperamide (4 mg/kg/day) 3 days prior to sacrifice. Treatment of loperamide resulted in decreases in the number and wet weight of fecal pellets, and increase in the number of fecal pellet in the distal colon and cecocolon weight. In contrast, the number and wet weight of fecal pellets were increased, and the number of fecal pellet in the distal colon and the cecocolon weight were decreased in LE-pretreated groups compared to the loperamide-treated group. Blood parameters such as white blood cell, red blood cell, hemoglobin, hematocrit, platelet, mean corpuscular volume, mean corpuscular hemoglobin and mean corpuscular hemoglobin concentration, and serum alanine transaminase, aspartate aminotransferase and alkaline phosphatase activities, and blood urea nitrogen and creatinine values were not significantly different between the groups. In addition, LE (0.5 mg/mL) increased spontaneous contractile activity, which was reduced by atropine or loperamide in isolated rat ileum. Theses results suggest that the improvement of constipation symptoms in LE-pretreated rats resulted from a stimulatory effect of LE on intestine contractile activity.

A Study on the Distribution of Tongue Colors and the Correlation of Tongue Colors with the Clinical Laboratory Parameters (설색의 분포 및 설색과 임상병리검사 결과의 연관성에 대한 연구)

  • Objectives : The aim of this study was to evaluate the distribution of tongue color and the correlation of the tongue colors with clinical laboratory parameters. Methods : Statistical analysis on the frequency of each tongue color and the results of the clinical laboratory parameters in each tongue color group. Results : Tongue colors that appeared most frequently were 702c, 695c, 493c, 694c, 701c, 500c, 7432c, 7431c, 7634c, 7419c in that order. Following them, 494c and 507c were rarer colors, and 501c, 673c, 687c, 693c, 696c, 703c, 7418c were equally rarest colors. Among each tongue color group, a significant difference was found in the result of complete blood cell count (CBC). The level of red blood cell (RBC) of 7432c group was significantly higher than that of the other groups except 694c group. The level of hemoglobin (Hb), hematocrit (Hct) of 7432c group were significantly higher than those of all the other groups. The level of RBC of 7431c group was lowest and significantly different from 694c, 7432c group. In results of the other tests were not significantly different. However the level of total cholesterol was higher in 695c, 694c, 500c group, and the level of triglyceride was higher in 500c, 7431c, 694c group. Conclusions : The tongue color is distributed throughout following colors by order of frequency: 702c, 695c, 493c, 694c, 701c, 500c, 7432c, 7431c, 7634c, 7419c, 708c, 494c, 507c, 501c, 673c, 687c, 693c, 696c, 703c, 7418c. Some parameters of CBC has a significant correlation with some tongue colors. Other results including lipid profile, renal function test, and liver function test had no significant difference according to tongue color. However, the colors which have low chroma or take blue violet color tented to be higher in the levels of total cholesterol and triglyceride.
