Studies on hematologic values and types of blood protein in Jindo dogs II. Types of blood protein of Jindo dogs (진도견(珍島犬)의 혈액상(血液像)과 혈액단백질형(血液蛋白質型)에 관(關)한 연구(硏究) II. 진도견(珍島犬)의 혈액단백질형(血液蛋白質型))

  • The phenotypes of hemoglobin, albumin and transferrin of 3U2 Jindo dogs in Jindo area were studied by starch gel electrophoresis for hemoglobin and albumin, and by polyacrylamide gradient gel electrophoresis for transferrin. The results obtained were as follows: 1. In the hemoglobin phenotypes, three phenotypes, HbAA, HbAB and HbBB, which were controlled by two allelic genes, $Hb^A$ and $Hb^B$, were observed and their frequencies of appearance were 1.65%, 10.60% and 87.75% respectively. The distribution of gene frequency was calculated as 0.0695 in $Hb^A$ and 0.9305 in $Hb^B$. 2. In the albumin phenotypes, three phenotypes, Alb FF, Alb FS and Alb SS, which were controlled by two allelic genes, $AIb^F$ and $AIb^S$ were observed and their frequencies of appearance were 12.59%, 25.56% and 61.85% respectively. The distribution of gene frequency was calculated as 0.2537 in $AIb^F$ and 0.7463 in $AIb^S$. 3. Analysis of transferrin phenotypes showed 6 different types which were controlled by three allelic genes, $Tf^B$, $Tf^C$ and $Tf^D$ and their frequencies of appearance were 54.04% in TfBB, 17.54% in TfBC, 9.82% in TfBD, 8.07% in TfCC, 7.37% in TfCD and 3.16% in TfDD. The distribution of gene frequency was calculated as 0.6772 in $Tf^B$, 0.2053 in $Tf^C$ and 0.1175 in $Tf^D$.

Genetic Polymorphisms of Blood Proteins and Enzymes in Korean Native Cattle (재래한우의 혈액단백질 및 효소의 유전적 다형현상)

  • This study was conducted to investigate the genetic constitution of blood proteins and enzymes in 238 Korean Native cattle reared at Korean Native Cattle Breeding Center, National Livestock Cooperative Federation. The genetic polymorphisms of post-transferrin-2(pTf-2), transferrin(Tf), post-albumin(pAlb), albumin(Alb), ceruloplamin(Cp), amylase-I(Am-I) and hemoglobin(Hb) were analyzed by using PAGE(polyacrylamide gel electrophoresis) and STAGE(starch gel electrophoresis). The genotypes and gene frequencies were estimated at these loci for each blood proteins and enzymes. The results obtained from this study were summarized as follows : 1. The pTf-2 locus were identified to be genetically controlled by codominant alleles designated pTf-2 F and S, and the distribution of genotypes were 46.22, 46.64 and 7.14% for pTf-2 FF, FS and SS types, and the gene frequencies of the pTf-2 F and S allele were 0,695 and 0.305, respectiveley. 2. The Tf locus were found to be controlled by Tf A, D1, D2 and E alleles, and the distributioin of genotypes were 0.84, 13.87, 13.03, 10.92, 22.27, 12.61, 2.94, 15.51, 6.72 and 1.68% for Tf AA, AD1, AD2, AE, D1D1, D1D2, D1E, D2E and EE types, and the gene frequencies of Tf A, D1, D2 and E were 0.197, 0.430, 0.191 and 0.081, respectively. 3. The pAlb locus were observed to be controlled by two alleles, pAlb F and S, and the distribution of genotypes were 42.86, 33.19 and 23.95% for pAlb FF, FS and SS types, and the gene frequencies were 0.595 and 0.405 for Tf F and S allele, respectively. Also the gene frequencies of Alb was 1.000 of Alb A allele. 4. The Cp locus were identified to be controlled by Cp F and S allele, and the distribution of genotypes were 23.11, 34.87 and 42.02% for Cp FF, FS and SS types, and the gene frequencies were 0.405 and 0.595 for Cp F and S allele, respectively. 5. The Am-I locus were observed to be genetically controlled by Am-I B and C allele, and the distribution of genotypes were 51.26, 16.81 and 31.92% for Am-I BB, BC and CC types, and the gene frequencies of Am-I B and C alleles were 0.597 and 0.403, respectively. 6. The Hb locus were found to be controlled by Hb A and B alleles, and the distribution of genotypes were 93.19, 16.39 and 0.42% for Hb AA, AB and BB types, and the gene frequencies of Hb A and B alleles were 0.914 and 0.086, respectively.

Enhanced Efficacy of Human Brain-Derived Neural Stem Cells by Transplantation of Cell Aggregates in a Rat Model of Parkinson's Disease

  • Objective : Neural tissue transplantation has been a promising strategy for the treatment of Parkinson's disease (PD). However, transplantation has the disadvantages of low-cell survival and/or development of dyskinesia. Transplantation of cell aggregates has the potential to overcome these problems, because the cells can extend their axons into the host brain and establish synaptic connections with host neurons. In this present study, aggregates of human brain-derived neural stem cells (HB-NSC) were transplanted into a PD animal model and compared to previous report on transplantation of single-cell suspensions. Methods : Rats received an injection of 6-OHDA into the right medial forebrain bundle to generate the PD model and followed by injections of PBS only, or HB-NSC aggregates in PBS into the ipsilateral striatum. Behavioral tests, multitracer (2-deoxy-2-[$^{18}F$]-fluoro-D-glucose ([$^{18}F$]-FDG) and [$^{18}F$]-N-(3-fluoropropyl)-2-carbomethoxy-3-(4-iodophenyl)nortropane ([$^{18}F$]-FP-CIT) microPET scans, as well as immunohistochemical (IHC) and immunofluorescent (IF) staining were conducted to evaluate the results. Results : The stepping test showed significant improvement of contralateral forelimb control in the HB-NSC group from 6-10 weeks compared to the control group (p<0.05). [$^{18}F$]-FP-CIT microPET at 10 weeks posttransplantation demonstrated a significant increase in uptake in the HB-NSC group compared to pretransplantation (p<0.05). In IHC and IF staining, tyrosine hydroxylase and human ${\beta}2$ microglobulin (a human cell marker) positive cells were visualized at the transplant site. Conclusion : These results suggest that the HB-NSC aggregates can survive in the striatum and exert therapeutic effects in a PD model by secreting dopamine.

Genetic Variants of the Blood Proteins and Enzymes in Beef Cattle (육우의 혈액단백질 및 효소의 유전적 변이체)

  • This study was conducted to examine the genetic variants of the blood proteins and enzymes in beef cattle breeds, Hereford, Angus and Sharolais reared at the Daekwanryuong Branch of the National Livestock Research Institute. Genetic polymorphisms of transferrin(Tf), post-transferrin2(pTf-2), albumin(Alb), post-albumin (pAlb), ceruloplasmin(Cp), amylase-I(Am-I) and hemoglobin(Hb) in blood were analyzed by the methods of PAGE(polyacrylamide gel electrophoresis) and STAGE(starch gel electrophoresis). The results obtained from this study were summarized as follows: 1. Tf and pTf-2 locus assumed to be controlled by codominant alleles, A. $D_1$, $D_2$ and E allele for Tf, F and S allele for pTf-2. In genotype frequencies, 25% and 90% for Tf $D_1D_2$ and pTf-2 SS in Hereford, 25% and 100% for Tf $AD_1$ and pTf-2 FF in Angus, 50% for Tf $D_1D_1$ and pTf-2 FS in Sharolais were found to have the highest frequency, respectively. In gene frequencies, 0.400 and 0.900 for Tf E and pTf-2 S allele in Hereford, 0.678 and 0.607 for Tf $D_1$ and pTf-2S in Sharolais were appeared to have the highest frequency. 2. Alb and pAlb locus assumed to be controlled by codominant alleles, only A allele for Alb, F and S allele for pAlb. In genotype frequencies, 70% for pAlb SS in Hereford, 90% for pAlb FF in Angus and 57.15% for pAlb SS in Sharolais were found to have the highest frequency. In gene frequencies, 0.825 and 0.750 for pAlb S in Hereford and Charolais, 0.900 for pAlb F in Angus were found to have the highest frequency. 3. Cp and Am-I locus appeared to be controlled by two alleles, F and S allele for Cp, B and C allele for Am-I. In genotype frequencies, 100% and 65% for Cp FF and Am-I BB in Hereford, 45% and 85% for Cp FF, and Am-I CC in Angus, 50% and 64.29% for Cp FF and Am-I BC in Sharolais were found to have the highest frequency. Gene frequencies were 1,000, 0.600 and 0.750 for Cp F in Herehord, Angus and Sharolais, 0.800, 0.875 and 0.680 for Am-I B, C and C allele in Hereford, Angus and sharolais, respectively. 4. Hb locus assumed to be controlled by codominant alleles, only A allele in Hereford and Angus, A and B allele in Sharolais. Genotype frequencies were 57.14% and 42.86% for Hb AA and AB in Sharolais, and gene frequencies were 0.785 and 0.215 for Hb A and B in Sharolais.

MEASUREMENTS OF SHRINKAGE STRESS AND REDUCTION OF INTER-CUSPAL DISTANCE IN MAXILLARY PREMOLARS RESULTING FROM POLYMERIZATION OF COMPOSITES AND COMPOMERS (광중합형 구치부 수복재료의 중합 수축력과 교두 변위의 상관관계)

  • The purpose of present study was to evaluate the polymerization shrinkage stress and cuspal deflection in maxillary premolars resulting from polymerization shrinkage of composites and compomers. Composites and compomers which were used in this study were as follows: Dyract AP, Z100, Surefil. Pyramid, Synergy Compact, Heliomolar, Heliomolar HB, and Compoglass F. For measuring of polymerization shrinkage stress, Stress measuring machine (R&B, Daejon, Korea) was used. One-way ANOVA analysis with Duncan's multiple comparison test were used to determine significant differences between the materials. For measuring of cuspal deflection of tooth, MOD cavities were prepared in 10 extracted maxillary premolars. And reduction of intercuspal distance was measured by strain measuring machine (R&B, Daejon, Korea) One-way ANOVA analysis with Turkey test were used to determine significant differences between the materials. Polymerization shrinkage stress is $\mathbb{\ulcorner}$Heliomolar, Z100, Pyramid < Synergy Compact Compoglass F < Dyract AP < Heliomolr HB, surefil$\mathbb{\lrcorner}$ (P < 0.05). And cuspal delfelction is $\mathbb{\ulcorner}$Z100, Heliomolar, Heliomolar HB, Synergy Compact Surefil. < Compoglass F < Pyramid, Dyract AP$\mathbb{\lrcorner}$ (P < 0.05). Measurements of ploymerization shrinkage stress and those of cuspal deflection of the teeth was different. There is no correlation between polymerization shrinkage stress and cuspal deflection of the teeth(p > 0.05).

Study on the Prevalence of Iron-deficient Anemia in Korean Preschool Children (학령 전 아동의 철 결핍성 빈혈 영양 상태 판정에 관한 연구)

  • The objective of this study was to identify the nutritional risk factors by blood analysis, in 1,083 preschool children of age 3 to 6 years. The frequency of anemic children was 7.3% with Hb<11.1 g/dL, 29.9% with ferritin<20 ng/mL, and 16.7% with transferrin Fe saturation(%)<15%. The prevalence of anemia in these children were 12.8% for those with MCV<79 fL, and it was 71% for those with TIBC> $400{\mu}g/dL$. Serum ferritin concentration was 20 ng/mL in the normal children. Thirty two percent of the children had anemia with Hb<12 g/dL, which is below the normal range of Hb. Exactly 15.4% of the children had serum Fe concentration of $60{\mu}g/dL$. The transferrin Fe saturation of the children (16.3%) was >15%. The serum ferritin concentration showed low correlations with Hb, Fe, transferrin Fe saturation, and MCV. The transferrin Fe saturation, higher Hb concentration, MCV, and Hct values were increased significantly. Consequently, iron-deficiency anaemia was thus defined as having Hb concentration <12 g/dL accompanied by ferritin concentration <20 ng/mL or Hct <33%.

The Effect of the Experience of Diabetes Education on Knowledge, Self-Care Behavior and Glycosylated Hemoglobin in Type 2 Diabetic Patients (당뇨교육경험이 제2형 당뇨환자의 지식, 자가간호행위, 당화혈색소에 미치는 영향)

  • Purpose: This study was to identify the effect of the experience of diabetes education on knowledge, self-care behavior and glycosylated hemoglobin (HbA1C). Further, this study was held to examine about patient's preferred methods of education and re-education frequency. Methods: 166 type 2 diabetes patients from two hospitals in Incheon participated in this study. Data were analyzed by using descriptive analysis, t-test, ANOVA, Scheffe's test and multiple regression analysis. Results: 72.3% patients needed re-education and the average interval of re-education was 8.53 months. Patients preferred education methods were lectures, practical training, and studying from pamphlet. Depending on the frequency of diabetes education, there were significant differences in the level of diabetes knowledge (F=10.88, p<.001) and self-care behaviors (F=4.59, p=.012), but there was not significant difference with HbA1C (F=1.53, p=.220). As to how much the diabetes education helped managing diabetes, there was a significant difference in the level of self-care behaviors (t=2.01, p=.049), but there were not significant differences in level of knowledge (t=1.10, p=.275) and HbA1C (t=-.33, p=.746). The experience of diabetes education was a significant factor which influenced patient's knowledge (t=3.93, p<.001) and self-care behaviors (t=2.21, p<.001). But HbA1C was not influenced by the experience of diabetes education (t=-1.68, p=.096). Conclusion: It is necessary to provide diabetes education with appropriate interval and methods and subjects that reflect the needs of patient through the study results.

Effect of Vitamin C Supplementation on Blood Sugar and Antioxidative Status in Types II Diabetes Mellitus Patients (비타민 C 보충이 제 2형 당뇨병 환자의 혈당 및 항산화상태에 미치는 효과)

  • Purpose: This study was to determine the effect of oral vitamin C supplements on blood sugar and antioxidative status in Types II diabetes mellitus patients. Method: Data for the study were collected from June 24 to August 31, 2001. Participants(31) took 1g/day vitamin C for 4 weeks, after a 1 - week taking no Vitamin C, followed by Vitamin C 3g/day for 4 weeks. A baseline blood sample was obtained following a 12hour overnight fast and at the end of each 4week Vitamin C administration. Blood samples were taken for plasma vitamin C concentration, fasting blood sugar, HbA1c, superoxide scavenging activity and hydrogen peroxide scavenging activity. The data were analyzed by SPSS for repeated measures ANOVA. Result: Plasma vitamin C concentration was significantly increased over dose(F=3.316, p=.043). Fasting blood sugar and HbA1c was significantly decreased over dose(F=13.192, p=.000; F=11.995, p=.000). Superoxide scavenging activity and hydrogen peroxide scavenging activity was significantly increased over dose(F=486.138, p=.000; F=177.704, p= .000). Conclusion: The results suggest that megadose vitamin C supplementation may have a beneficial effect in diabetes mellitus patients on both glycemic control and antioxidant status. Thus dietary measures to increase plasma vitamin C may be an important health strategy for reducing the compliance of diabetic patients

Effect of General obesity and Abdominal obesity on Hemoglobin in Korean Adult (한국성인의 비만과 복부비만이 헤모글로빈에 미치는 영향)

  • The purpose of this study was to investigate the effect of body mass index(BMI) and waist circumference(WC) on hemoglobin(Hb) in Korean Adult. This study analyzed the data of 2015, The Sixth KNHANES. Data were analyzed using descriptive statistics, t-test, ANOVA and Scheffe's test, Pearson correlation coefficient and multivariate logistic regression analysis(SPSS 25.0). The major findings, The subject's Hb In BMI, obese was the highest(F=97.862, p<0.001). The Hb In WC, $WC{\geq}90cm$ was higher for male(t=4.909, p<0.001), and $WC{\geq}85cm$ was higher for female(t=5.088, p<0.001). Hb has a significant positive correlated with BMI(r=0.188, p<0.001), WC(r=0.298, p<0.001). In male, Hyperhemoglobinemia was 2.30 times(p<0.001) higher in obese and 1.81 times(p<0.001) higher in $WC{\geq}90cm$. In female, Hyperhemoglobinemia was 1.99 times(p=0.002) higher in overweight, 5.66 times(p<0.001) higher in obese, and 4.27 times(p<0.001) higher in $WC{\geq}85cm$. In conclusion, adult obesity prevention and management programs should include exercises, diets, and education, and exercise interventions specifically for eliminating abdominal obesity.

The Effects of Resistance Exercise Program for Elders with Type 2 Diabetes on the Self Care, Stress and HbA1c (저항운동 프로그램이 제 2형 당뇨병 노인의 자가간호, 스트레스 및 당화혈색소에 미치는 효과)

  • Purpose: The purpose of this study was to examine the effects of resistance exercise program for elders with type 2 Diabetes on the self care, stress and HbA1c. Methods: Thirty three elders with type 2 Diabetes(18 experimental and 15 control subjects) were selected conveniently among the aged who had been enrolled in a community senior center. The subjects in experimental group participated in a resistance exercise program for 8 weeks. Data were analyzed using the SPSS/Win 12.0. Results: The resistance exercise program showed a statistical difference in self care(t=-4.39, p=.000) and stress(t=2.22, p=.034). However, there was not a statistical difference in HbA1c (F=.556, p=.557), but experimental group had decreased continuously in HbA1c after the program. Conclusion: The resistance exercise program improved self care and reduced the stress in the elders with type 2 Diabetes. Therefore, resistance exercise program can be applied as an effective nursing intervention to promote self care, decrease the stress and to prevent complication for the elders with type 2 Diabetes.
