The effect of three different water temperatures in our research facility on Huanren brown frog (Rana huanrensis) egg's hatching rate, hatching periods, and larvae's growth (인위적으로 조성한 세 가지 수온이 계곡산개구리(Rana huanrensis) 알의 부화율, 부화기간 및 유생의 생장에 미치는 영향 연구)

  • This study was executed to know the effect of three differently controlled temperature conditions on Huanren brown frog (Rana huanrensis )'s growth in 2013. We've collected nine Huanren brown frog egg's sacs on Mt. Surak ($37^{\circ}40^{\prime}55.86^{{\prime}{\prime}}N$, $127^{\circ}05^{\prime}19.99^{{\prime}{\prime}}E$) in Seoul. We put those nine egg sacs in the controlled growth chambers under low temperature (LT, $5{\pm}2^{\circ}C$), medium temperature (MT, $10{\pm}2^{\circ}C$), and high temperature (HT, $13{\pm}2^{\circ}C$) conditions with three egg sacs, respectively. We measured the eggs' hatching rate, their hatching periods, and the size of the hatched individuals. The hatching rate was higher in MT (95.6%) and the rates of the other treatments were relatively lower but very similar such as LT (82.2%) and HT (82.6%). The three hatching periods were 10 days at HT, 14 days at MT and 23 days at LT. The body sizes of the hatched individuals were biggest at MT ($7.62{\pm}0.11mm$), smallest at LT ($6.82{\pm}0.10mm$) and medium at HT ($7.19{\pm}0.15mm$) (P-value ${\leq}0.0001$). From our results, we found that the various water temperatures could be very effective to Huanren brown frog eggs' hatch and growth including their body sizes. We suggest if we study more about the growth of Huanren brown adult frogs under similar temperature conditions for a long term period, it must be very helpful for conservation study about metamorphosis rate and size of adult frog as well as we could understand about the amphibians who are adapting to the climate change.

Biological Characteristic of Obolodiplosis robiniae and Control Effects of Some Insecticides (아까시잎혹파리(Obolodiplosis robiniae)의 생물학적 특성 및 약제 살충 효과)

  • Biological Characteristic of Obolodiplosis robiniae and insecticidal activity of some insecticides against larvae of O. robiniae were investigated. Egg was in oval shape, and its color was light orange and became red when close to hatch. Length of the major axis and the minor axis of egg was 0.4 mm and 0.1 mm, respectively. Larval color was milky and size was 2.6 mm. Pupa was deep brown and its size was about 3.2 mm. Wing and abdomen of adult was black and reddish, respectively. Size of female adult was about 3.3 mm, and larger than male adult. Number of eggs in the ovary was $192.3{\pm}50.7$. First emergence was from late April to late May, and second from late May to late June. Third emergence was from late June to late July. Newly emerged adult escaped from soil, and second and third emerged adult escaped directly from gall of Robinia pseudoacacia leaf. Egg parasitoid was identified as Platygaster robiniae and parasite rate was 51.6%. Among test insecticides, imidacloprid 10% WP and thiacoprid 10% FL showed very strong insecticidal activity against larvae of O. robiniae at 48h later after treatment.

  • The hatchability of the artificially induced hybrid between two groupers (family Serranidae), red spotted grouper (Epinephelus akaara) and brown-marbled grouper (E. fuscoguttatus) that lives in different habit environment was investigated. There was no difference in the required time of each developmental stages after fertilization between hybrid (red spotted grouper ♀ ${\times}$ brown-marbled grouper ♂) and purebred (red spotted grouper ♀${\times}$♂) and required 25.6 hours to hatch at incubated in $25^{\circ}C$, but a noticeable unequal cleavage in cell size was observed in hybrid eggs unlikely to purebred. The hatching rate of fertilized eggs of hybrid was generally low across the four incubate temperatures (22, 25, 28, $31^{\circ}C$) with highest 9.8% in $25^{\circ}C$. This study demonstrated the possibility of artificial hybridization between two groupers, red spotted grouper and brown-marbled grouper, thus preparing the groundwork on developmental characteristics, deformities of hatched larvae and early survival ability for further studies on aquaculture.

Effects of sewage sludge on the cocoon production and its hatchability of tiger worm(Eisenia fetida) (하수슬러지가 줄지렁이(Eisenia fetida)의 산란 및 부화율에 미치는 영향)

  • When the clitellate tiger worms(Eisenia fetida) were fed with paper mill sludge, sewage sludge of agricultural-urban area and sewage sludge of agricultural area, number of cocoons produced by individual clitellate earthworm for 104 days were 11.8, 8.6 and 3.5, respectively. However, clitellate earthworms that had been fed with sewage sludge of agricultural-urban area for 52 days or fed with sewage sludge of agricultural area for 95 days could not produce cocoons, whereas the earthworms that were fed with paper mill sludge kept producing cocoons through the investigating period. The hatchability of cocoons produced by the clitellate tiger worms(Eisenia fetida) fed with sewage sludge of agricultural-urban region, sewage sludge of agricultural region and paper mill sludge were 42.8%, 38.3% and 39.2%, respectively. And hatching periods of cocoons were 33.8~36.0 days, which were not affected by the kinds of sludges. However, the cocoons produced by clitellate earthworms which had been fed with sewage sludge for about 1.5 months could not hatch at all. Death rates of second generation hatched from cocoons produced by tiger worms(Eisenia fetida) which had been fed with the sewage sludge from agricultural-urban region and the sewage sludge from agricultural region reached to 100% and 90% at 60 days after hatching, respectively, whereas death rate of second generation fed with paper mill sludge reached to 16%.

Variance Component Quantitative Trait Locus Analysis for Body Weight Traits in Purebred Korean Native Chicken

  • Quantitative trait locus (QTL) is a particular region of the genome containing one or more genes associated with economically important quantitative traits. This study was conducted to identify QTL regions for body weight and growth traits in purebred Korean native chicken (KNC). F1 samples (n = 595) were genotyped using 127 microsatellite markers and 8 single nucleotide polymorphisms that covered 2,616.1 centi Morgan (cM) of map length for 26 autosomal linkage groups. Body weight traits were measured every 2 weeks from hatch to 20 weeks of age. Weight of half carcass was also collected together with growth rate. A multipoint variance component linkage approach was used to identify QTLs for the body weight traits. Two significant QTLs for growth were identified on chicken chromosome 3 (GGA3) for growth 16 to18 weeks (logarithm of the odds [LOD] = 3.24, Nominal p value = 0.0001) and GGA4 for growth 6 to 8 weeks (LOD = 2.88, Nominal p value = 0.0003). Additionally, one significant QTL and three suggestive QTLs were detected for body weight traits in KNC; significant QTL for body weight at 4 weeks (LOD = 2.52, nominal p value = 0.0007) and suggestive QTL for 8 weeks (LOD = 1.96, Nominal p value = 0.0027) were detected on GGA4; QTLs were also detected for two different body weight traits: body weight at 16 weeks on GGA3 and body weight at 18 weeks on GGA19. Additionally, two suggestive QTLs for carcass weight were detected at 0 and 70 cM on GGA19. In conclusion, the current study identified several significant and suggestive QTLs that affect growth related traits in a unique resource pedigree in purebred KNC. This information will contribute to improving the body weight traits in native chicken breeds, especially for the Asian native chicken breeds.

First Sexual Maturity, Spawning Frequency and Deposition of the Egg Capsules of the Female Purple Shell Rapana venosa in the Slag Deposit Area, Gwangyang Bay, Korea (한국 광양만, 슬러그 적재장내에 서식하는 암컷 피뿔고둥 Rapana venosa의 군성숙도, 산란빈도 및 난낭 산출)

  • First sexual maturity, sex ratio, spawning frequency, deposition of the egg capsules and fecundity of the female Rapana venosa(Valenciennes) inhabited in the artificially closed slag deposit area, Gwangyang Bay were investigated by histologicai and visual observations for natural living resource management. The rate of individuals reaching the first sexual maturity was 51.6% in females measuring 7.1~8.0 cm in shell height, and 100% in those > 10.1 cm. The total number of egg capsules per individual and the mean number of eggs in an egg capsule were 192~382 and 500, respectively. However, the number of eggs per individual and sizes of egg capsules under lower salinity and deficient food conditions in the closed slag deposit area were smaller than those under the optimum salinity and sufficient food conditions in the open regions. Fecundities of the species were approximately from 96,000 to 191,000 eggs/individual with two to low broods(spawning frequencies) during the spawning season. The duration of development in egg capsules was 18~19 days at about 18~2$0^{\circ}C$. R. venosa is a species whose embryos hatch as veliger larvae, not juvenile snail. The sex ratio of female : male was not significantly different from 1 : 1($\chi$$^2$= 0.23, p>0.05).

Effects of Gamma Irradiation on the Survival and Development of the Multicolored Asian Ladybird Beetle, Harmonia axyridis (Coccinellidae; Coleoptera) (감마선이 무당벌레의 생존과 발육에 미치는 영향)

  • The effects of gamma irradiation on the survival, development, fecundity, and sex ratio of the multicolored Asian ladybird beetle (Harmonia axyridis) were investigated. A dose of more than 50 Gy dose of irradiation on eggs completely inhibited hatch, and 1st and 3rd instar larva irradiated with over 40 Gy almost died. Adverse effects of gamma irradiation over 70 Gy were also observed in the survival and development of pupae and adults. Although irradiated individuals usually survived, they had longer periods of larval and pupal development than untreated individuals. Slight female-biased sex ratios were observed in some treatments. However, sex ratio differences soon equilibrated. Generally, inherited harmful effects of gamma irradiation were observed in F1 and F2 generations. Comet tail indicating DNA damage was showed in survival individuals exposed to gamma irradiation.

  • Clownfish are important and very popular fish in the ornamental aquarium industry. Demand for the fish is increasing dramatically. The present study was conducted to verify methods of broodstock management, patterns of spawning, rates of egg hatching and estimates of larval growth fur the saddleback clownfish, Amphiprion polymnus. Spawning occurred 8 times between August 2002 to June 2004 with 2 females and 1 male participating. Fertilized eggs were separated by an adhesive matrix and were oval in shape. The eggs were $2.46{\pm}0.13mm$ in size as measured along the longest axis. The percentage of fertilized eggs was 96.7%. Hatching was observed seven days post-spawning and hatching rate was 85.5%. The sizes of the newly-hatched larvae were $4.58{\pm}0.21mm$ TL (total length). Larvae had an open mouth and anus, and an oval yolk sac. At the 1 st day after hatching, the sizes of the larvae were $4.90{\pm}0.35mm$ TL. The larvae began to eat rotifers after complete yolk absorption. On the 5th day post-hatch, larvae were $5.88{\pm}0.31mm$ TL with complete fins and the survival rate was 48.6%. At 8 days after hatching, a band began to appear on head and back of the larvae indicating the beginning of metamorphosis. Metamorphosis was completed at an average TL of $15.00{\pm}2.12mm$ on the 23rd day after hatching. By the 45th day after hatching, juveniles averaged $22.76{\pm}3.22mm$ TL and survival rate was 28.4%.

Development of an in vitro culture method for harvesting the free-living infective larvae of Strongyloides venezuelensis (베네수엘라분선충 (Strongyloides venezuelensis Brumpt, 1934) 자유생활형 유충의 시험관 내 배양 기술 개발)

  • An in uitro culture technique was established for harvesting Strongwloides venezuelensis free-living infective larvae using a nutrient broth medium as a substitute for rat-feces in polyvinyl culture bags ($10{\;}{\times}{\;}12{\;}cm$). The egg hatch rate V) in sterile saline at different incubation temperatures (X) was expressed as the quadratic function, Y = $-0.192X^2$ + 8.673x - 19.550 (r = 0.901). The highest (100%) egghatch rate was observed at $25^{\circ}C$. A significant difference (p<0.05) in development rate W) of free-living infective larvae was observed between different concentrations of nutrient broth (X) which was highest (20.6%1 in 0.12% nutrient broth concentrations, incubated at $20^{\circ}C$ for 5 days [Y = $-864.032X^2$ + 245.995X- 0.560 (r = 0.875)]. Yields (Y) of infective larvae were observed relatively high when the culture medium was incubated at higher temperatures (X) which peaked at $25^{\circ}C$ (20.0%) than at lower temperatures. $15^{\circ}C$M (10.9%) and $20^{\circ}C$ (18.1%) [Y = $-0.189X^2$ + 8.387x- 72.795 (r = 0.981)]. The period W) required for the development of infective larvae decreased with higher incubation temperatures (X) [Y = $0.035X^2$ - 2.025X + 32.375 (r = 0.995)] The highest yield (19.2%) of infective larvae was obtained from culture bag inoculated with 15.000 eggs than with below and over 15,000 eggs in 0.12% nutrient broth and incubated at $25^{\circ}C$ for 4 days. The newly adapted culture method (from egg to third-stage larva) may be useful as a bio-bar/bioassay system for screening new chemical products, anthelmintics and pesticides, as well as for parasito immunological studies with Strongwloides species.

Survey the Occurrences and Establishment of Environment-friendly Control System of Ricania shanthungensis in Jeonnam Province (전남지역 갈색날개매미충 발생현황과 친환경 방제)

  • This study was carried out to establish of environment-friendly control system and to survey the occurrence status of Ricania shanthungensis in Jeonnam province from 2016 to 2017. R. shanthungensis occurred at 1,344 ha in 61 towns of 15 cities in Gwangju and Jeonnam province. At four sites in the stationary area, the densities of eggmass and pupae were lower in 2017 than in 2016, the hatching rate decreased, and the first hatching time, emergence of adults, and the start of spawning tended to be slower. Telenomus sp., an egg parasitoid of R. shanthungensis, was found in most areas. Sunchon had the highest rate of 46.1%, followed by Muan 40.6, Goksung 29.2, Gurye 25.8, and Gwangju 17, respectively. The optimum spraying time for the control of R. shanthungensis was early March. The spraying material and the dilution multiples are 10 times of the machine oil at environmental friendly cultivation and 500 times of the chlorpriphosphate wp. at normal cultivation. When sprayed these materials, 95% of egg prohibited to hatch. There was more than 80% of the insecticidal effects on the organic materials of the machine oil emulsion, the Sophora root extrect + microbial extract agent, and the castor emulsion. For adults, Sophora root extrect + microbial extract, neem extract, and machine oil were better. Four kinds of chemical pesticides such as dinotefuran wp were effective for nymph and adult control. We have developed an adult catching device using the most preferred daylight color and behavioral habits of R. shanthungensis. The capture device consisted of two daylight compact lamps (30W and 20W), a yellow plate, and a catcher using water, and caught about 700 individuals a day. Based on the above results, we have established a system for controlling and life cycle of R. shanthungensis in Jeonnam province.
