Effects of Nonylphenol and Diethylhexyl Phthalate on the Population Growth of Freshwater HABs, Microcystis and Stephanodiscus (내분비교란물질이 조류의 성장에 미치는 영향)

  • Effects of two endocrine disrupters (EDs) , nonylphenol (NP) and diethylhexyl phthalate (DEHP), on the population growth and morphology of two freshwater HABs (harmful algal blooms) , Microcystis aeruginosa and Stephanodiscus hantzschii, which frequently evoked the hazard bloom in an eutrophic lakes and reservoirs worldwide, were examined with seven different concentrations of EDs (0.01, 0.05, 0.1, 1,2, 2.5 and 3 ppm). Even at concentration below 0.01 ppm, NP strongly inhibited the algal growth of both M. aeruginosa and S. hantzschii, regardless of the algal growth phase. Morphologically, the algal cell treated with NP gradually lost green color in cytoplasm, became smaller in cell size, and then, was hardly seen in microscopic field. On the other hand, DEHP employed did not affect two algae at all concentrations, and rather stimulated the growth by about 10% with a treatment at 3.00 ppm compared to control. These results indicate that the continuous input of EDs, DEHP into the natural water system plays a crucial role to enhance or help an outbreak of algal bloom in eutrophic waters.

  • Aquatic ecosystems are receiving various harmful effects due to anthropogenic chemical pollutions. To protect wildlife, risk assessments of the chemicals are conducted using reference indexes of toxicity estimated by species-level laboratory tests and/or micro-/mesocosm community-level studies. However, the existing micro-/mesocosm communities are structurally too complicated, and it is also difficult to compare the experimental results directly with those from species-level tests. Here, we developed a procedure of a simple bi-trophic microcosm experiment which contains the common species (a green algae, Pseudokirchneriella subcapitata and a cladoceran, Daphnia magna) for testing chemical toxicities. For the proper operation of bitrophic microcosm experiment, the minimum required concentration of primary producer (P. subcapitata) is $5{\times}10^5cells\;mL^{-1}$. The microcosm system showed higher stability when the initially introduced D. magna population was composed of neonates (<24-h old) than adults and those mixture. This simple microcosm system would be an applicable tool to estimate the disturbing impacts of pollutants on plant-herbivore interactions, and linking the species- and population-/community level risk assessments in the future studies.

Formation of Phytoplankton Community and Occurrences of Odorous Compounds by Sediment Incubation (퇴적층 배양에 따른 식물플랑크톤 군집 형성과 이취미물질 발생 특성)

  • In this study, Uiam-sediment(US) and Gongji-sediment(GS) were incubated to analyze the formation of phytoplankton community and occurrences of odorous compounds from benthic cyanobacteria. For the phytoplankton, 45 and 40 taxa were found in GS and US, respectively. The phytoplankton species detected in the harmful algae included Microcystis, Anabaena and Oscillatoria. As a result, a standing crop of phytoplankton appeared to be in the range of $0.9{\sim}71.8{\times}10^3$ cells/mL in GS, $0.8{\sim}51.2{\times}10^3$ cells/mL in US, and as the incubation progressed, benthic cyanobacteria was increased. The highest number of akinetes was shown in the initial incubation, and akinetes was reduced by 10% on day 4 of incubation start. After reduction of akinetes incubated for 4 days in US and 15 days in GS, Anabaena appeared from the surface water. Benthic cyanobacteria such as Oscillatoria tenuis, O. limosa and Lyngbya hieronymusii was dominant in GS (max. $3.96{\times}10^6cells/cm^2$) and US (max. $5.77{\times}10^6cells/cm^2$), forming cyanobacterial mats in sediment surface. Cyanobacterial mats were suspended after 10~12 days incubation in GS and US. Before and after the suspension of cyanobacterial mats, the concentration of geosmin was the highest in GS(115 ng/L) and US(175 ng/L). In the second suspension of cyanobacterial mats in GS, the concentration of 2-MIB was increased to the range of 22~92 ng/L.

Characteristics of environmental condition and planktonic organisms in ship's ballast water originating from international ports of Japan (우리나라 주요 국제항에 입항하는 일본 기원 선박의 평형수내 환경 및 부유생물 특성)

  • This study investigated the environmental conditions and planktonic organisms in the ballast waters (BW) of 22 vessels originating from the international ports of Japan for the purpose of negotiating exemptions from the Ballast Water Management Convention (BWM Convention). The shortest duration of the BW was $3.33{\pm}1.87days$ in area "A", which included Kyushu and Suo Nada at Seto Inland. The total suspended solids, dissolved organic carbon, and particulate organic carbon ranged from 4.60 to 60.9 mg L-1, from 0.97 to 2.69 mg L-1, and from 0.24 to 4.51 mg L-1, respectively. A low average concentration of nutrients was measured in the BW from area "A", but that in the BW from area "C" (around central Honshu) was high, which may be related to the ballasting periods. High chlorophyll-a concentrations (>$1{\mu}g\;L-1$) were measured in four vessels, three of which carried the BW in area "A". High abundances of phytoplankton (> 50,000 cells L-1) were measured in four vessels, three of which carried the BW in area "A". The two vessels originating from Tokyuyama Bay in area "A" showed high densities of dinoflagellates, which are known to be harmful algae. Our results suggest that the negotiations for an exemption from the BWM Convention for Japan should proceed with caution.

Changes in the Water Environment Based on the Statistical Data in the Lake Paldang (통계로 보는 팔당호 물환경 변화)

  • Since the 1970s regulations against the pollution of drinking water have been introduced in Lake Paldang watershed area. To understand the effects of water environment management policies and the impacts of climate changes on Lake Paldang, a long-term comprehensive study of this watershed and the changes in its water environment is required. In this study, we analyzed changes in the weather, hydrology, sources of pollution, water quality, and algal development from 2000 to 2015 year based on the statistical data provided by several national information systems. While the population and amount of sewage in the Lake Paldang watershed increased by about 1.5 times, the amount of animal manure showed a decreasing trend during the same period. The wastewater also increased by about 1.5 times while the amount of water intakes rose by about 1.14 times. The water quality in front of the Paldang Dam, which is the representative monitoring site of the Lake Paldang, was stable. The annual average BOD concentration remained within 2 mg/L, which is a "Good (lb)" level according to the environment standards of Republic of Korea. The development of phytoplankton and harmful cyanobacteria were largely influenced by meteorological factors.

  • Paralytic shellfish poisoning (PSP) occurs when saxitoxin (STX), which is produced by harmful algae (dinoflagellates) and then accumulated in bivalve shellfish by filter-feeding, is consumed by humans. With recent advances in analysis technology, it has been reported that dinoflagellates also produce a variety of analogues such as the gonyautoxin (GTX) group and the N-sulfo-carbamoyl toxin (C toxin) group, in addition to STX. Accordingly, CODEX and the EFSA are stepping forward to manage STX and analogues as STX groups requiring safety management. In Korea, the occurrence of dinoflagellates producing STX analogues has already been reported, and contamination of analogues (GTX group, C toxin group) in live mussels has also been reported. In this study, in order to provide the basis for systematic monitoring and safety management of STX and analogues, their physicochemical characteristics, occurrence of dinoflagellates, toxicity and toxic equivalency factor, analytical method and occurrence were widely reviewed. This review is expected to contribute to strengthening the safety management of STX and its analogues.

Supplementation of Microalgae (Tetradesmus sp.) to Pre-Starter Diet for Broiler Chickens (초기 육계 사료 내 미세조류(Tetradesmus sp.) 첨가에 따른 장관환경 변화)

  • This study aimed to determine the effects of dietary microalgae (Tetradesmus sp. (TO)) on intestinal immunity and microbiota of pre-starter broilers. One hundred and twenty 1-day-old birds (Ross 308) were allocated to two dietary treatment groups with six blocks in a randomized complete block design. The two experimental diets consisted of a corn-soybean meal-based basal diet and a diet with 0.5% TO powder instead of cornstarch in the basal diet. After feeding the experimental diets for ten days, all birds' body weight and feed intake were measured, and representative eight birds were selected from each treatment group. Small intestinal lamina propria cells were isolated using flow cytometry to examine the frequency of immune cells. Cecal feces were harvested for 16s rRNA gut microbiota analysis and fecal IgA levels. Here, we found that 0.5% TO supplementation increased CD3+CD4+ T cells in the small intestine, but decreased CD3+CD8+ T cells in the small intestine. Gut microbial analysis showed that TO supplementation significantly increased the alpha diversity of the gut microbiome. Taxonomic analysis showed that TO treatment increased the abundance of Firmicutes and decreased that of Bacteroidetes at the phylum level. The distribution of Enterobacteriaceae containing many harmful bacteria at the family level, was lower in the TO group. In the LEfSe analysis, the TO group had a significantly enriched abundance of Agathobaculum at the genus level. Overall, results show that Tetradesmus sp. supplementation influences intestinal T-cell immunity and induces the expansion of beneficial gut microbes in pre-starter broiler chickens.

  • 한국농어촌공사 산하의 농업용저수지 중 3786개소에 대한 수질조사를 '19년도에 실시한 결과, TOC 기준 4등급 초과 저수지 비율은 약 20%로써, 도심 근교 저수지에서 녹조현상 빈발로 인해 수질, 악취, 미관 등의 환경문제 개선 민원이 다수 발생하고 있다. 현재 녹조 발생 사후관리를 위해 주로 사용되고 있는 대형 조류제거선은 저수심 수변부에서의 적용성에 한계가 있고, Al 기반의 응집제를 사용하여 조류를 수거해서 폐기하고 있는 실정이다. (주)이엔이티는 농어촌연구원, (주)코레드, (주)삼호인넷과 함께 호소나 정체하천의 수변지역에 적용될 수 있는 저에너지형 유해조류 포집시스템 개발과, 수거된 조류부산물을 무독화하여 농업재료로 재활용하는 방안을 연구하고 있다. 저수지나 정체수역의 녹조는 바람, 수면유동 등에 의해 수변에 집적되는 특성이 있어, 인공지능 기술로 녹조현상을 감시하여 조류 밀집구간에 접근할 수 있는 자율이동식 수상이동장치를 개발 중이다. 수상이동장치는 조류포집장치를 탑재하기 위한 부력체, 원격 운전이 가능한 무인항법장치, 수변식생대 및 저수심지역 이동을 고려한 수차방식 추진체, 전체 장치의 전원 공급을 위한 고성능 배터리 등으로 구성하여 상세 도면 설계를 진행하고 있다. 조류포집장치에는 표층에 주로 분포하는 남조류를 선택 흡입하는 포집 부표를 적용하였고, Al계 응집제 사용을 배제한 분리막 실험을 통해 침지형 막분리조 및 가압형 농축조를 설계하였다. 유해조류 포집 및 농축은 수상에서 이동체에 탑재하여 이뤄지고, 육상에서는 자원 회수가 가능하도록 회분식 응집공정으로 구분하였다. 조류 밀집지역에서 수거된 조류의 무독화 및 농업재료 자원화 타당성 평가를 위해 특용 버섯균주를 활용한 시료별 분석항목을 선정하고 실험 매트릭스에 따라 실증실험을 수행하였다. 수거조류를 전처리하여 성분 및 발열량을 분석하고 버섯재배 전후의 마이크로시스틴 독소(LR, RR, LR)를 포함한 성분 분석을 수행하여, 고체연료, 비료 및 사료로 활용방안을 검토하였다. 무인자율이동 조류포집장치는 실증화 규모로 제작하여 기선정된 테스트베드에서 현장적용성 평가를 수행할 예정이다. 본 연구를 통해 개발된 유해조류 포집 시스템은 기존의 녹조제거 방안을 보완하여 정체수역의 생태계 복원 및 친수공간의 환경개선 등에 적용되며, 무독화가 입증된 유해조류의 농업재료 자원화 기술은 고부가 상품 개발 및 환경폐기물 감축에 활용될 것이다.

Ichthyotoxic Cochlodinium polykrikoides red tides offshore in the South Sea, Korea in 2014: I. Temporal variations in three-dimensional distributions of red-tide organisms and environmental factors

  • The ichthyotoxic Cochlodinium polykrikoides red tides have caused great economic losses in the aquaculture industry in the waters of Korea and other countries. Predicting outbreak of C. polykrikoides red tides 1-2 weeks in advance is a critical step in minimizing losses. In the South Sea of Korea, large C. polykrikoides red tide patches have often been recorded offshore and transported to nearshore waters. To explore the processes of offshore C. polykrikoides red tides, temporal variations in 3-dimensional (3-D) distributions of red tide organisms and environmental parameters were investigated by analyzing 4,432 water samples collected from 2-5 depths of 60 stations in the South Sea, Korea 16 times from May to Nov, 2014. In the study area, the vegetative cells of C. polykrikoides were found as early as May 7, but C. polykrikoides red tide patches were observed from Aug 21 until Oct 9. Cochlodinium red tides occurred in both inner and outer stations. Prior to the occurrence of large C. polykrikoides red tides, the phototrophic dinoflagellates Prorocentrum donghaiense (Jun 12 to Jul 11), Ceratium furca (Jul 11 to Aug 21), and Alexandrium fraterculus (Aug 21) formed red tides in sequence, and diatom red tides formed 2-3 times without a certain distinct pattern. The temperature for the optimal growth of these four red tide dinoflagellates is known to be similar. Thus, the sequence of the maximum growth rates of P. donghaiense > C. furca > A. fraterculus > C. polykrikoides may be partially responsible for this sequence of red tides in the inner stations following high nutrients input in the surface waters because of heavy rains. Furthermore, Cochlodinium red tides formed and persisted at the outer stations when $NO_3$ concentrations of the surface waters were < $2{\mu}M$ and thermocline depths were >20 m with the retreat of deep cold waters, and the abundance of the competing red-tide species was relatively low. The sequence of the maximum swimming speeds and thus potential reachable depths of C. polykrikoides > A. fraterculus > C. furca > P. donghaiense may be responsible for the large C. polykrikoides red tides after the small blooms of the other dinoflagellates. Thus, C. polykrikoides is likely to outgrow over the competitors at the outer stations by descending to depths >20 m and taking nutrients up from deep cold waters. Thus, to predict the process of Cochlodinium red tides in the study area, temporal variations in 3-D distributions of red tide organisms and environmental parameters showing major nutrient sources, formation and depth of thermoclines, intrusion and retreat of deep cold waters, and the abundance of competing red tide species should be well understood.

Analysis of Water Quality Improvement of Ceratophyllum demersum under Laboratory Condition - by Nutrients Removal Efficiency (실험실 조건에서 붕어마름의 수질개선 효과 분석 - 영양염류 제거 효율을 중심으로)

  • To evaluate the ability of the submerged plant, Ceratophyllum demersum's (C. demersum) to remove nutrients and to inhibit growth of cyanobacteria, a total of 6 mesocosms were conducted in a batch reactor for 9 days. From the 84 hr of the experiment, C. demersum was stabilized and showed daily cycle trends according to changes in pH and DO levels. The concentration of nutrients, $NH_3{^+}$, $NO_3{^-}$ and $PO_4{^3}$ continuously decreased until 9 days of the experiment, with the rapid decrease in nutrient concentration for the first 24 hours. High correlation coefficient ($r^2{\geq}0.96$, p<0.001) between the amount of C. demersum's biomass per unit area and the nutrients removal level were derived, and greater C. demersum's biomass per unit area showed higher removal efficiency of nutrients. However, there were differences in the C. demersum's activity level between batch reactors with higher and similar density of the C. demersum, but nonetheless water purification effect appears to have a significant influence due to attached algae and microorganisms. The growth rate of harmful cyanobacteria, Microcystis aeruginosa (M. aeruginosa) with C. demersum's density of 2,500 g $fw/m^2$ (100% of cover degree) was 0.31 /day, compared to the growth rate of 0.47 /day for the control group (0% of cover degree). In terms of number of cells, the control group had 1.7 times higher number of cells than the experimental group, proving that C. demersum has the ability to inhibit the growth of harmful cyanobacteria.
