번식 장애 한우의 혈액요소태질소 및 성 스테로이드 호르몬 농도 변화 (Alteration in Concentrations of Blood Urea Nitrogen and Sex Steroid Hormone in Korean Cattle with Reproductive Disorders)

  • The objective of this study is to investigate the changes in concentrations of blood urea nitrogen (BUN) and sex steroid hormones, such as estrogen, progesterone, and testosterone in Korean cattle (Hanwoo) with reproductive disorders and to examine the relationship between BUN and body condition score (BCS) in Hanwoo. The concentration of BUN was 16.2 mg/dl, 17.8 mg/dl, 15.1 mg/dl, 17.9 mg/dl, and 28.3mg/dl in pregnancy, repeat breeding, follicular cyst, luteal cyst, and ovarian atrophy, respectively. In Hanwoo with BCS $2.0{\sim}2.9$, $3.0{\sim}3.5$ and $3.6{\sim}4.0$, the concentration of BUN was 15.8 mg/dl, 17.0 mg/dl, and 17.6 mg/dl, respectively. Fluoroimmunoassay showed that serum estrogen and progesterone levels were decreased in reproductive disorders Hanwoo, such as ovarian atrophy, endometritis, and weak estrus. The testosterone level was significantly decreased in Hanwoo with reproductive disorders compared to that in pregnant Hanwoo ($0.02{\sim}0.03\;ng/ml$ vs 0.13 ng/ml, p<0.05). The progesterone and estrogen concentrations in follicular fluid obtained from ovary with follicular cyst were significantly higher (p<0.05) than those in normal follicle fluid. These results show that there is no relationship between BUN and BCS in Hanwoo, and the concentration of sex steroid hormone in serum and follicular fluid are changed in reproductive disorders Hanwoo.

한우의 생식세포 보존에 관한 연구 II. PEG 30% FSH 투여가 한우의 체내수정란 생산에 미치는 영향 (Studies on Preservation of Germ Cells in Hanwoo II. Effects of In Vivo Embryos Production by PEG 30% FSH in Hanwoo)

  • 한우의 수정란을 보존하기 위하여 고급육계통과 다유계통의 공란우를 선발하여 과배란처리에 따른 황체의 반응, 황체수에 대한 채란된 수정란의 수 그리고 채란 수정란수에 대한 이식가능한 수정란 생산에 미치는 영향을 조사하였다. 다유계통의 공란우는 이유시 체중에 대한 육종가 상위 20%이내, 고급육 계통의 공란우는 육질에 대한 육종가 상위 20%이내의 종빈우를 선발하였다. 과배란처리 방법은 발정후 11일차에 PEG 30% FSH를 견갑부에 피하주사로 1회 투여하고 13일차에 PGF$_2$a 25mg을 투여하였으며 15일차와 16일차에 각각 2회씩 인공수정하였고 23일차에 채란하였다. 과배란처리에 따른 황체의 반응은 좌측난소가 47.1% (242/513), 우측난소가 52.8%(271/513)로써 우측의 반응이 좋게 나타났고 황체의 반응과 비교하여 채란된 수정란의 회수율은 79.9%(410/513)로 나타났다. 회수된 수정란 중에서 이식이 가능한 수정란은 86.5%(355/410)이었고 수정란의 발달단계에 있어서 상실배는 61.6%(219/355), 배반포는 38.3%(136/355)로 나타났으며 두당 이식가능한 생산수는 6.4개였다.

  • 한우에 있어서 생체내 난자 채취기술과 체외수정 기술을 접목하여 고능력 한우로부터 수정란을 대량 생산하기 위한 기초 연구로서, 발정주기에 따른 난소의 크기와 난포의 발육 양상을 초음파진단기를 이용하여 조사하였으며, 여러 마취.진정제를 조합 이용하여 마취효과를 조사하여 얻어진 결과는 다음과 같다. 1. 한우의 난소의 크기는 도축 난소의 실측치와 생체에서 초음파진단기를 이용한 측정치간에 유의적인 차이가 인정되지 않았으며, 발정주기중 난포기때와 황체기때 크기를 비교해도 유의적인 차는 인정되지 않았다. 2. 한우의 발정주기중 난소내 발육하는 소난포와 중난포의 수는 3일과 12일 째에 증가하는 한 발정주기에 2회의 난포발육기가 있었다. 3. 한우의 최적 마취조합은 0.3$m\ell$ 의 Rompun(equation omitted)을 근육주사하고 5$m\ell$의 lidocaine 을 경막외 마취한 후 lidocaine 2$m\ell$을 자궁경에 도포하는 경우였다.

Effects of culture media conditions on production of eggs fertilized in vitro of embryos derived from ovary of high grade Hanwoo

  • Background: This study was investigated the effects of culture media conditions on production of eggs fertilized in vitro of embryos from ovaries of high grade Korean native cow, Hanwoo. Methods: The IVMD 101 and IVF 100 were used for in vitro maturation of selected Hanwoo oocytes and In vitro embryo culture after in vitro fertilization, respectively. The IVMD 101 and IVD 101 were used for in vitro culture and completely free of serum. Results: The cleavage rates of 2-cell embryos in reference to Hanwoo oocytes were 86.7, 92.9, and 90.1 % in the control group, IVDM101 medium and IVD101 medium, respectively which indicates that the IVDM101 medium and IVD101 medium may result favorable outcomes. The in vitro development rates of blastocysts were 12.4, 38.4 and 32.4 % in the control group, serum free IVMD101 medium and IVD101 medium, respectively. For hatched blastocysts, it was 5.3, 33.9, and 28.6 % in the control group, serum free IVMD101 medium and IVD101 medium, respectively. Hence, more favorable results were expected for the hatched blastocysts in which the IVMD101 medium and IVD101 medium were used than the control group. Average cell numbers of blastocysts were 128.3, 165.7, and 163.6 in the groups of TCM-199 + 10 % FBS medium, IVMD 101 medium, and IVD 101 medium, respectively which clearly show that the IVMD 101 and IVD 101 medium consequence significantly higher cell numbers compared to the control group (i.e., TCM-199 +10 % FBS medium). Pregnancy rate after embryo transfer was 39.6 % when the serum free medium was used which is higher than that of the medium supplemented with serum (32.8 %). In addition, stillbirth rates were 4.9 % in the group of serum free medium whereas it was 13.6 % in the serum supplemented medium (13.6 %). Conclusions: Taken altogether, serum free media, the IVMD 101 and IVD 101 represented more favorable results in the embryo development rate of embryos, cell numbers of blastocyst, and pregnancy rate. Of note, the IVMD 101 medium showed better outcomes hence, it might be a better option for future applications for in vitro culture of bovine embryos.

한우 육질등급이 난포란의 배반포 체외생산에 미치는 영향 (Correlation of Hanwoo (Korean Native Cattle) Carcass Classification and Oocyte Donor for Blastocyst Production In Vitro)

  • These studies were conducted to establish the practical Hanwoo (Korean native cattle) improvement system through the combining of embryo transfer technology and confirming individual Hanwoo oocyte culture system and to investigate that correlation of Hanwoo carcass classification (intramuscular marbling) and oocyte donor for blastocyst production in vitro. In case of Hanwoo, the carcass meat quality grades were divided to grade $1^{{+}{+}}$, $1^{+}$, 1, 2, and 3 depends on fat distribution of longest muscle cross-sectional surface. As results, the numbers of follicular oocytes collected from individual fundamentally-registered Hanwoo yielded $1^{{+}{+}}$, $1^{+}$, 1, 2 and 3 meat quality were 9.5, 9.4, 8.5, 8.8 and 8.8 per ovary, respectively. The numbers of retrieval oocyte from follicles were significantly higher in the cattle yield $1^{{+}{+}}$, $1^{+}$ meat quality than in the cattle yield 1, 2 and 3 meat quality (p<0.05). The rates of blastocyst formation were 18.2, 21.3, 29.4, 30.9, and 31.5% in the cattle yield $1^{{+}{+}}$, $1^{+}$, 1, 2 and 3 meat quality of after in vitro maturation, respectively. It was significantly lower in the cattle yield $^{{+}{+}}$ and $1^{+}$ meat quality than in the cattle yield 1, 2 and 3 meat quality (p<0.05). In order to evaluate embryos quality, TUNNEL assay was conducted for each meat quality grade using blastocyst stage embryo on day 8. The results showed that apoptosis cell number was higher tendency in the cattle yield $1^{{+}{+}}$and $1^{+}$ meat quality (81 and 79, respectively) than in the cattle yield 1, 2 and 3 meat quality (51, 48 and 50, respectively) but there was no statistical significance in each group. After embryo transfer, the conception rate of recipient was 53.5 (23 out of 43), 52.1 (38 out of 73) and 58.0% (58 out of 100) in the meat quality of 1, $1^{+}$ and $1^{{+}{+}}$, respectively. These results showed that the conception rate was significantly higher in the cattle yield 1 meat quality than in the cattle yield $1^{{+}{+}}$, $1^{+}$, 2, and 3 meat quality (p<0.05). In summary, these results indicate that the application of confirming Hanwoo individual oocyte culture system and embryo transfer technology can make good use of the genetic resources conservation and improvement of Hanwoo. Relevance of the meat quality grade and reproductive ability of carcasses of Hanwoo will be considered to be one of the effective means for the associated research with obesity and reproduction.

번식장애 한우의 렙틴 및 인슐린 농도 변화 (Alteration in Concentrations of Leptin and Insulin in Korean Cattle with Reproductive Disorders)

  • The objective of this study is to investigate the changes in concentrations of leptin and insulin in serum of Korean cattle (Hanwoo) with reproductive disorders and to examine the relationship among leptin, insulin, and body condition score (BCS). The concentration of leptin in serum of pregnant Hanwoo showed insignificant difference from that in serum of Hanwoo with reproductive disorder, such as repeat breeding, follicular cyst, corpus luteum cyst, ovarian atrophy, and feeble estrus (p>0.05). However, the concentrations of leptin and insulin in serum were changed with different BCS value. In emaciated Hanwoo (BCS $2.0\sim2.9$), they were significantly decreased compared to BCS $3.0\sim3.4$ (p<0.05). The leptin showed different genotypes with different BCS value. In BCS $2.0\sim2.9$, C/T genotype was expressed (83.3%) more than C/C (16.7%) or T/T (0%) genotype, whereas C/C genotype was expressed (62.5%) more than C/T (25.0%) or T/T (12.5%) genotype in BCS $3.5\sim4.0$. The insulin concentration in follicular fluid obtained from ovary with follicular cyst which has follicles having diameter of $25\sim40 mm$ was significantly higher (p<0.05) than those in normal follicle fluid which has follicles having diameter of $3\sim10 mm$. These results showed that concentration of leptin and insulin in serum were related to BCS value and follicular size and suggest that the changes in concentration of leptin and/or insulin in serum could be a potent biomarker for diagnosis of bovine reproductive disorder.

한우의 정상 난포와 난포낭종 난포에서 Aquaporin7 발현 양상 (Patterns of Aquaporin 7 Expression in Normal Follicles and Follicular Cyst Follicles of Hanwoo)

  • Alteration in ion channel or transporter expression levels affects cell volume which is produced by movement of water and ion across the plasma membrane. In particular, aquaporin (AQP) channels among ion channels play a crucial role in movement of water across the cell membrane. This study was performed to identify whether AQP expression is changed in bovine follicular cystic follicles using microarray, RT-PCR and Western blotting analyses. In microarray data, AQP4 expression was decreased, whereas AQP7 was increased in cystic follicles. Additional experiments were focused on the AQP7 expression increased in cystic follicles. The microarray data was confirmed by semi-quantitative polymerase chain reaction (PCR) and Western blot analysis. AQP7 mRNA and protein expressions were significantly increased in the cystic follicles (p<0.05). Application of estrogen ($10{\mu}g/ml$) to bovine ovarian cells showed a trend of increase in AQP7 expression. From these results, we suggest that the increase in AQP7 expression in cystic follicles may play an important role in movement of water in bovine ovary. In addition, AQP7, a aquaglyceroporin permeating water and glycerol, could be a good target in development of methods for the cryopreservation of bovine ovary.

한우에 있어서 체내수정란의 생산과 이식에 관한 연구 - I 한우 수정란 생산에 영향을 미치는 요인 (The Studies on In Vivo Embryo Production and Transfer in Hanwoo- I. Factors Influencing In Vivo Embryo Production)

  • 본 연구는 기존의 수정란 이식기술을 다각적으로 분석하고 개선하여 한우 체내수정란의 생산 및 이식기술 체계를 확립하기 위하여 수행하였으며, 결과를 요약하면 다음과 같다. 1. 배란에 미치는 FSH analogue 간의 유의성은 인정되지 않았으나 수정률, 이식 가능 수정란 및 동결수정란 생산에는 처리간에 유의성(P<0.05)이 인정되었다. 특히, Super-OV는 Foll-tropin-V 및 Embryo-S에 비해서 이식 가능 수정란의 생산율이 유의적(P<0.01)으로 낮았다. 2. 호르몬의 투여시기에 따라 공란우의 배란정도 (P<0.1), 수정률, 이식 가능 및 동결 가능 수정란의 생산(P<0.05)은 각각 유의적인 차이를 나타냈다. 특히, 발정주기 11일차의 호르몬투여는 적합하지 않은 결과를 보였다. 3. 다배란처리후 황체의 등급에 따른 수정란의 수는 좌측 또는 우측에서 6개 이상의 황체를 보인 A등급(8.06∼9.49개)의 경우가 B, C 및 D등급(2.93 ∼6.28개)보다 유의적(P<0.01)으로 높았다

  • The purpose of this experiment is to compare the pregnancy rate (PR) according to the state of the ovaries and uterus, according to the number of embryos transferred from cows and heifers and to investigate the method of artificial twin induction with Hanwoo $in$ $vitro$ fertilized (IVF) embryos by embryo transfer (ET). Looking at the PR according to the condition of the ovaries and uterus, the result was not influenced by the condition of the ovaries, but was significantly influenced by the state of the uterus. The PR according to the number of embryos transferred from cows was 36.8%, 53.0%, 50.5% for 1, 2, and 3 embryos respectively, and although there was a higher frequency of twin calves with 3 embryos than with 2, the calving rate was the highest with 2 embryos. In case of heifers, the transfer of 1 embryo showed the best pregnancy and calving rate, and although the PR was similar with 2 embryos (67.7 versus 66.4), in case of 2 embryos transferred there was high frequency of embryonic loss( 6.1%) occurred when a cow was diagnosed at 28 and 53 d after ET, total loss (21.3%; sum of fetal death, abortion and stillbirth after pregnant diagnosis at 60 day).
