Effect of Body Condition Score (BCS) on In Vivo Embryo Production and Pregnancy Rate Following Superovulation in Hanwoo (한우에서 신체충실지수(BCS)가 다배란 처리 시 체내 수정란 생산과 수태율에 미치는 영향)

  • Body condition score (BCS) is a useful management tool for distinguishing differences in nutritional needs of cows in the herd. Although it is not always possible to quantify the nutrient content of the feed supplied to the donor cow, the nutritional status can be determined by the BCS. The objective of this study was to evaluate in vivo embryo production, return to estrous of donor and pregnancy rate of recipients following BCS in Hanwoo superovulation. Sixty nine Hanwoo donor cows were flushed on day 7 of estrus cycle with same FSH and artificial insemination by the same technicians. Embryos were recovered on 7 days after the third insemination by flushing the uterus with Embryo Collection Medium. The results obtained were as follows: No differences were observed in the efficiency of superovulation rates regardless of BCS ($${\leq_-}2.0$$, 2.5 to 3.0, and $${\geq_-}3.5$$). The mean number of total embryos was each $5.20{\pm}0.86$, $11.56{\pm}1.04$, and $6.23{\pm}1.07$. The mean number of transferable embryo from $${\leq_-}2.0$$, 2.5 to 3.0, and $${\geq_-}3.5$$ of BCS was $2.60{\pm}0.87$, $7.94{\pm}0.89$, and $4.75{\pm}1.32$, respectively (p<0.05). Return to estrous regardless of donor BCS was no difference. The pregnancy rates of recipient were BCS $${\leq_-}2.0$$ 11.76%, 2.5 to 3.0 40.79%, and $${\geq_-}3.5$$ 11.11%, following transfer of fresh embryos produced in vivo, respectively. These results indicate that if the Hanwoo with BCS 2.5 to 3.3 are used for donor and recipient, the embryo production and the conception rate will be greater.

Production and Embryo Transfer of In Vivo Embryos by Repeated Superovulation Treatment of Hanwoo Cattle (한우의 반복 과배란 처리에 의한 체내 수정란의 생산과 이식)

  • This study was performed to investigate the possibility of repeated superovulation treatment at interval from 27 days to 41 days in Hanwoo cattle and to compare with superovulation effect between doses of FSH 200 mg and FSH 400 mg. Different doses of FSH (200 mg or 400 mg) were injected at Day 8 after controlled internal drug release (CIDR) treatment for superovulation of Hanwoo donors following CIDR treatment (Day 8 after the estrus). Superovulation was repeated four times for one donor and number of corpus luteum (CL), number of embryos, number of transferable embryos and pregnancy rate after embryo transfer (ET) were investigated. 5 cows were used for each FSH treatment (10 cows in total). Average number of CL were $10.16{\pm}3.85$ and $11.56{\pm}2.35$ for the donors treated with FSH 200mg and FSH 400mg, respectively. Average number of embryos collected were $8.85{\pm}4.05$ and $8.30{\pm}1.73$ for the donors treated with FSH 200 mg and FSH 400 mg, respectively. Average number of transferable embryos were $5.48{\pm}2.45$ and $4.58{\pm}2.23$ for the donors treated with FSH 200 mg and FSH 400 mg, respectively. The pregnancy rate following ET with embryos collected from 200 mg FSH treated donors and 400 mg FSH treated donors were 61.9% and 53.8% respectively. The numbers of embryos tended to be decreased as the numbers of repeat of superovulation were elapsed. These results indicated that superovulation treatment by about a month to Hanwoo donors is usable and 200 mg of FSH is preferable for simple FSH treatment following CIDR treatment.

Conception Rate of Ovulation-Estrus Synchronization Method in Hanwoo (한우에 있어서 발정ㆍ배란 동기화법에 의한 수태율)

  • This objective of this experiment were to evaluate the effect of various estrus synchronization programs on estrus detection rate and pregnancy rate in Hanwoo. After Postpartum 60 Days, a total of 150 cows divided into 2 groups. Cows Group 1 were treated with one luteolytic dosage of PGF$_2$$\alpha$(25 mg, im; lutalyse. USA) on Day 0, and with a second dosage 14 d later; cows in Group 2 were treated with GnRH(l00 $\mu\textrm{g}$, im; Conceral. Korea) on Day 0, PGF$_2$$\alpha$ 7 d later, GnRH 2 d later, and then time-inseminated approximately 16 h after this second treatment with GnRH. Ovarian morphology was monitored cows by trans-rectal ultrasonography from 24 hr to 32 hrs after second GnRH injection. The result obtained summarized as follows: 1. Cows synchronization of estrus with GnRH+PGF$_2$$\alpha$+GnRH(Ov-synch) and PGF$_2$$\alpha$ were 91.3 and 40.0%, respectively. 2. Induced ovulation were 24 to 32hr after the second GnRH injection, but high induced ovulation was 28hr. 3. High conception rate were 24hr insemination after the second GnRH injection. 4. Conception rate with PGF$_2$$\alpha$, CIDR and GnRH treatment were 50.0, 36.0 and 76.9%, respectively.

Survey on the Effect of the Parity on Reproductive Traits of Korean Native Cows (산차가 한우번식에 미치는 영향에 대한 조사 연구)

  • A survey was carried out to investigate the effect of the parity on reproductive traits of Hanwoo(Korean native cows). Data on the reproductive traits of 670 Korean native cows were collected from January, 1996 to December, 1997 and analyzed by the parity. The results obtained were summarized as follows; 1. The average ages at first breeding, first conception and first calving were 443.0, 457.0 and 746.6 days, respectively. 2. The average days to first estrus after calving was 70.1 days and it was shortest in the cows with more than 5 parities (60.4 days) and longest in the cows at 4th parity (79.7 days). 3. The average number of services per conception was 1.53 and it was fewer in the cows at 2nd paity (1.43), but more in the cows with more than 5 parities (1.73). 4. The average days to first conception after calving was 91.2 days and there was a trend that it was longer as the parity of cows increased(p>0.05). 5. The average length of calving interval was 375.3 days and it was shortest in the cows at 2nd cows (370.8 days) and longest in the cows at 4th parity (395.2 days).

Regional Differences of Proteins Expressing in Adipose Depots Isolated from Cows, Steers and Bulls as Identified by a Proteomic Approach

  • Adipose tissue in the loin muscle area of beef cattle as a marbling factor is directly associated with beef quality. To elucidate whether properties of proteins involved in depot specific adipose tissue were sex-dependent, we analyzed protein expression of intramuscular adipose tissue (IMAT) and omental adipose tissue (OMAT) from Hanwoo cows, steers, and bulls of Korean native beef cattle by liquid chromatography-tandem mass spectrometry (LC-MS/MS)-based proteomic analysis, quantitative polymerase chain reaction (PCR) and western blot analysis. Two different adipose depots (i.e. intramuscular and omental) were collected from cows (n = 7), steers (n = 7), or bulls (n = 7). LC-MS/MS revealed a total of 55 and 35 proteins in IMAT and OMAT, respectively. Of the 55 proteins identified, 44, 40, and 42 proteins were confirmed to be differentially expressed in IMAT of cows, steers, and bulls, respectively. In OMAT of cows, steers, and bulls, 33, 33, and 22 were confirmed to be differentially expressed, respectively. Tropomyosin (TPM) 1, TPM 2, and TPM3 were subjected to verification by quantitative PCR and western blot analysis in IMAT and OMAT of Hanwoo cows, steers, and bulls as key factors closely associated with muscle development. Both mRNA levels and protein levels of TPM1, TPM2, and TPM3 in IMAT were lower in bulls compared to in cows or steers suggesting that they were positively correlated with marbling score and quality grade. Our results may aid the regulation of marbling development and improvement of meat quality grades in beef cattle.

  • Q fever is a zoonosis caused by Coxiella burnetti that is important not only for livestock but also for humans. This research aims to investigate infection rate of Q fever by examining the prevalence of antibody titers in cattle in Sejong City. From March to October 2018, 505 samples of Hanwoo serum and 47 samples of bulk-tank milk were collected and analyzed using ELISA. According to the result, 25 (53.2%) out of 47 bulk-tank milk samples were antibody positive and 28 (5.5%) out of 505 cows were antibody positive. The seroprevalence of C. burnetii increased with age (<1 y=0%, 1~2 y=4.1%, 3~5 y=7.5%, ≥6 y=9.6%) and was higher in females (6.5%) compared to males (0%).

Effects of Raising Farm on Genetic Evaluation for Carcass Traits in Hanwoo Cows (사육농가의 효과가 한우 암소의 도체형질 유전 평가에 미치는 영향)

  • This research was conducted to analyze the effects of raising farm on the heritability and breeding values of Hanwoo cows for their carcass traits, including cold carcass weight (CWT), back-fat thickness (BFT), eye-muscle area (EMA) and marbling score (MAR). The carcass data and pedigree data were collected from steers raised on Hanwoo farms in Pyeongchang-gun, Gangwon-do, South Korea. Three analytical models were applied for the estimation of heritabilities and breeding values. The first model (model 1) included slaughter house-year-month combination as fixed effects and age at slaughter was fitted as linear and quadratic covariates. The second model (model 2) was similar to model 1, but raising farm was additionally included as random effect. The third model (model 3) was similar to model 1 but farm effects were additionally included as fixed effect. The comparisons between the model 1 and the models including farm effect (model 2 and model 3) revealed that heritability estimates from model 2 or model 3 were smaller to those from model 1 for all carcass traits. Especially, obvious decrease of heritability was observed in CWT where heritability was 0.23 from model 1, 0.15 from model 2 and 0.18 from model 3. The maximum log likelihood of the model 2 and 3 were higher than those of model 1 for all traits. In model 2 that raising farm was included as a random effect, the ratio of farm variance to the total phenotypic variance were ranged from 4% (EMA) to 18% (CWT). Top 10% and bottom 10% of female cows were selected based on the breeding values from model 1, and the Spearman's rank correlation coefficients among models were estimated for each trait within selected group. The correlation coefficients were ranged from 0.57 to 0.95 in top 10% group and from 0.68 to 0.95 in bottom 10% group. These results show that the discrepancies in the rankings of breeding values can be based on the models applied. In conclusion, the results obtained in this study suggest that the herd effect or farm effect should be included in the analytical model when breeding values are estimated with the purpose of improvement of carcass traits of Hanwoo breeding cows.

Relationship between Transferable Embryos and Blood Urea Nitrogen, Glucose and Total Cholesterol Concentration in Hanwoo Donors (한우 공란우 이식기능 수정란수와 혈액내 요소태질소, 포도당 및 총콜레스테롤 수준과의 관계)

  • The objective of this study was to investigate the relationship between concentration of urea nitrogen, glucose, cholesterol and number of transferable embryos for the purpose of improving reproductive performance in blood of Hanwoo donors. Fifty five, at random stages of the estrous cycle, received a CIDR. Four days later the animals were superovulated with a total of 28AU FSH (Antorin, 2AU=1 ml) administered twice daily in constant doses over 4 days. On the 3th administration of FSH, CIDR was withdrawn and 25 mg $PGF_2$ ${\alpha}$ was administered. Cows were artificially inseminated twice after estrous detection at 12 hr intervals. The cows received 100 ${\mu}g$ GnRH at the time of 1nd insemination. Embryos were recovered 7 or 8 days after the 1st insemination. Cows with BUN < 10, 11~18 and ${\geq}$19 mg/dl had number of transferable embryos of $4.32{\pm}1.3$, $5.8{\pm}1.8$ and $4.7{\pm}2.1$ respectively. The mean numbers of total ova from < 10 and 10${\leq}$ of corpora lutea(CL) was 8.9 and 14.3, respectively. The number of transferable embryos differed between < 10${\leq}$ CL was 4.8 and 5.6, respectively.

Effects of Non-ionic Surfactants on Enzyme Distributions of Rumen Contents, Anaerobic Growth of Rumen Microbes, Rumen Fermentation Characteristics and Performances of Lactating Cows

  • A series of experiments was carried out to determine the possibility for the non-ionic surfactant (NIS) as a feed additive for ruminant animals. The effect of the NIS on (1) the enzyme distribution in the rumen fluids of Hereford bulls, (2) the growth of pure culture of rumen bacteria and (3) rumen anaerobic fungi, (4) the ruminal fermentation characteristics of Korean native cattle (Hanwoo), and (5) the performances of Holstein dairy cows were investigated. When NIS was added to rumen fluid at the level of 0.05 and 0.1% (v/v), the total and specific activities of cell-free enzymes were significantly (p<0.01) increased, but those of cell-bound enzymes were slightly decreased, but not statistically significant. The growth rates of ruminal noncellulolytic species (Ruminobacter amylophilus, Megasphaera elsdenii, Prevotella ruminicola and Selenomonas ruminantium) were significantly (p<0.01) increased by the addition of NIS at both concentrations tested. However, the growth rate of ruminal cellulolytic bacteria (Fibrobacter succinogenes, Ruminococcus albus, Ruminococcus flavefaciens and Butyrivibrio fibrisolvens) were slightly increased or not affected by the NIS. In general, NIS appears to effect Gram-negative bacteria more than Gram-positive bacteria; and non-cellulolytic bacteria more than cellulolytic bacteria. The growth rates of ruminal monocentric fungi (Neocallimastix patriciarum and Piromyces communis) and polycentric fungi (Orpinomyces joyonii and Anaeromyces mucronatus) were also significantly (p<0.01) increased by the addition of NIS at all concentrations tested. When NIS was administrated to the rumen of Hanwoo, Total VFA and ammonia-N concentrations, the microbial cell growth rate, CMCase and xylanase activities in the rumen increased with statistical difference (p<0.01), but NIS administration did not affect at the time of 0 and 9 h post-feeding. Addition of NIS to TMR resulted in increased TMR intake and increased milk production by Holstein cows and decreased body condition scores. The NEFA and corticoid concentrations in the blood were lowered by the addition of NIS. These results indicated that the addition of NIS may greatly stimulate the release of some kinds of enzymes from microbial cells, and stimulate the growth rates of a range of anaerobic ruminal microorganisms, and also stimulate the rumen fermentation characteristics and animal performances. Our data indicates potential uses of the NIS as a feed additive for ruminant animals.

Estimation of genetic parameters using real-time ultrasound measurements in Hanwoo (한우 암소의 생체 초음파 성적을 이용한 유전모수 추정)

  • This study was conducted to estimate genetic effects on economically important traits for genetic improvement in Hanwoo by using the real-time ultrasound measurements for longissimus dorsi muscle area (LMA), backfat thickness (BFT), and marbling score (Marb). The phenotypic data were obtained from 1,648 pedigreed cows, and general linear models were applied to test the effects of age, region, and body condition socre. The cows between 50 and 60 months of age had the greatest scores for LMA and BFT, and Marb (P<0.05). The cows in region C had the greatest scores for body condition socre, LMA and BFT, while in region J Marb was the lowest (P<0.05). There was positive relation with LMA, BFT, and Marb according to increase body condition socre. Heritabilities for LMA, BFT, and Marb were estimated as 0.136, 0.351, and 0.236, respectively. These results would provide primary information for the efficient implementation of genetic improvement schemes in Hanwoo.
