Tissue culture of medicinal plants: micropropagation, transformation and production of useful secondary metabolites

  • Plant tissue culture studies have been done for the preservation of medicinal plant resources and efficient production of pharmaceutically important secondary metabolites. Micropropagation methods for Cephaelis ipecacuanha have been established and these methods enabled much more efficient propagation of the plants than the conventional methods using seedling or layering. The C. ipecacuanha plants derived from tissue culture grew uniformly in the field and they showed higher alkaloid contents compared to the plants grown from seedlings. Hairy root cultures of C. ipecacuanha and Panax ginseng have been established by infection with Agrobacterium rhizogenes, and the production of important pharmaceuticals by these cultures have been successfully demonstrated. In the case of C. ipecacuanha, the highest alkaloid yields from the hairy roots cultured for 8 weeks were 2.75-fold cephaeline (5.5 mg) and one third emetine (0.7 mg) compared with those from the roots of one-year old plant propagated through shoot-tip culture and cultivated in a greenhouse (2.0 mg cephaeline and 2.0 mg emetine). In the case of P. ginseng, ginsenoside contents in the hairy roots optimally cultured for 4 weeks were much higher than those in the roots of 4-year old field-grown plant. Thus our medicinal plant tissue cultures demonstrate desirable properties. However, they are always exposed to danger of microbial contamination or unexpected trouble of culture facilities. Cryopreservation of plant tissue cultures is a reliable method for long-term preservation. Cryopreservation studies on these cultures are also presented.

Effect of Green-Manure Crop Rotation on Weed Control (녹비작물을 이용한 윤작이 잡초 제어에 미치는 영향)

  • This study was conducted to evaluate the effects of crop rotation on the weed occurrence in green onion and red pepper cultivation. Rye and hairy vetch were annually seeded for winter season. The reduction of weed germination was pronounced by about 50% in green onion field, but was not significant in red pepper field through rye cropping. Among weed species, the germination of Abutilon theophrasti, Cyperus iria and Digitaria ciliaris was distinctly inhibited by exudate of rye root, but not by that of hairy vetch in vitro experiment. The return of rye biomass into onion field decreased amount of weed biomass but increased index of weed diversity.

Effects of Light on the Pigment Production and Chloroplast Development of Ginseng Hairy Roots (인삼 모상근의 색소 생성 및 엽록체 발달에 미치는 광의 효과)

  • The effects of light on the pigment production and chloroplast development were examined on ginseng hairy roots cultured in 1/2MS liquid medium. The chlorophyll and carotenoid production were increased from 1,000 to 3,500 lux condition, but decreased drastically in 7,000 lux condition. The anthocyanin production was significantly increased with increment light intensity(1,000∼7,000 lux). The thylakoid membrane of chloroplast was proplastid in dark condition and it began to develop into thylakoid membrane in 1,000 lux condition and then intact thylakoid membrane was developed in 3,500 lux condition. However, the development of thylakoid membrane in 7,000 lux condition was inhibited comparing to 3,500 lux condition. The total chlorophyll production in blue light condition were high comparing to other wavelength and same as 40% of total chlorophyll on white light(3,500 lux) condition. The chlorophyll and carotenoid production by sucrose concentration were high in 3% sucrose condition and anthocyanin production was high in 4% condition. The production of chlorophyll and carotenoid by light periods was high when explants were cultured in dark condition for 1 week and then transferred to light condition for 4 weeks. Our results suggest that pigment production and chloroplast development could be accelerated by light Intensity of specific wavelength in cultures of ginseng hairy root.

Growth Characteristics and Ginsenoside Contents of 4 Years Old Korean Ginseng (Panax ginseng C.A. Meyer) by Shade Materials and Green Manure Crops (해가림자재 종류와 녹비작물 재배에 따른 4년생 인삼의 생육과 진세노사이드 함량)

  • This research carried out to figure out the effect of the green manure crop cultivated at a preparation field and the shading net on the growth, development, and quality of ginseng. Followings are results obtained from the research. Leaf width of ginseng under the shading net of a two-layered blue and two-layered black polythylene net (TBTBPN) was good at rye and hairy vetch cultured group. Leaf length of ginseng under the shading net of a three-layered blue and one-layered black polyethylene net (TBOBPN) was good at barley and hairy vetch cultured group. Meanwhile, leaf width was good at hairy vetch cultured group. Leaf length of ginseng under a blue polyethylene sheet (BPS) was good at a barley and barley + hairy vetch cultured group, but stem length was shorter compare to other shading net cultivations. Root weight of ginseng was good under the shading net of a two-layered blue and two-layered black polyethylene net (TBTBPN) at a rye and hairy vetch cultured group, and was good under the shading net of a three-layered blue and onelayered black polyethylene net (TBOBPN) at a barley + hairy vetch cultured group, but there was no significant difference under blackout screen according to manure crop varieties. Ratio of rusty root was 10.2% at the barley cultured group under the shading net of a two-layered blue and two-layered black polyethylene net (TBTBPN), and was 23.1% at hairy vetch cultured group under shading net of a three-layered blue and one-layered black polyethylene net (TBOBPN). Ratio of rusty root was the lowest at a rye cultured group regardless the shading nets. Content of the ginsenoside was the highest at the rye cultured group under the shading net of two-layered blue and two-layered black polyethylene net (TBTBPN), was the highest at the barley cultured group under the shading net of a three-layered blue and one-layered black polyethylene net (TBOBPN), and was the highest at the rye cultured group under the blackout screen.

Effect of the Contents Ratio of Panaxadiol Ginsenosides Extracted from Various Compartment of Ginseng on the Transcription of Cu/Zn Superoxide Dismutase Gene (홍삼의 각 부위에서 추출된 Panaxadiol분획의 함량비에 따른 유해산소제거효소(Cu/Zn Superoxide Dismutase) 유도효과)

  • Cu/Zn superoxide dismutase (SOD1) is a protective enzyme responsible for the dismutat ion of superoxide radicals within the cell by converting superoxide radicals to oxygen and hydrogen peroxide, which is in turn changed to oxygen and water by catalase. Previously, we reported that the panaxadiol (PD) and its ginsenoside $Rb_2$ induced the expression of SOD1 gene through AP2 binding site and its induction. Here, we examined the effect of subfractions of panaxadiol ginsenosides, which were extracted from different parts of ginseng root that possess various ratios of panaxadiol to panaxatriol, on the induction of SOD1 gene expression. To explore this possibility, the upstream regulatory region of SOD1 was linked to the chloramphenicol acetyl transferase (CAT) structural gene and introduced into human hepatoma HepG2 cells. We observed that the transcriptional activation of SOD1 was proportional to the contents ratio of panaxadiol ginsensides. Consistent with this results, the total extract portion prepared from the finely-hairy root, which contains the higher ratio of panaxadiol to panaxatriol about 2.6, increased the SODl transcription about 3 fold. This results suggest that the panaxadiol fraction could induce the SOD1 and total extract of the ginseng finely-hairy root would be a useful material as a functional food for the SOD1 inducer.

Production of Vinblastine by Chemical Coupling of Vindoline Extracted from Cultivated Plants and Catharanthine from Hairy Root Cultures in Vinca(Catharanthine roseus) (일일초 잎과 모상근으로부터 추출한 Vindoline과 Catharanthine의 화학결합에 의한 Vinblastine 생산)

  • Vinblastine, an anticancer agent was produced by chemical coupling of two different monomeric indole alkaloids, vinblastine and catharanthine in the presence of ferric ion. Vindoline was efficiently extracted from the leaves of vinca (Cafharanthus roseus) by using supercritical carbon dioxide, whereas catharanthine was chemically extracted from the in vitro cultured hairy roots. The extracted crude monomeric precursors were purified by a two-step preparative TLC. The coupling reaction was carried out in the 0.1M glycine buffer(pH 2.0, 5ml) containing 40mM FeC13 with purified vindoline(0.3mg) and catharanthine(0.3mg) at 4$^{\circ}C$. The production yields (weight %) of vinblastine and 3', 4'-anhy-drovinblastine in the products were 23.2 and 26.0, respectively. The produced vinblastine was confirmed by FAB-MS.

  • The cells of Rubia cordifolia var. pratensis were transformed by Agrobactrium tumefaciens strain 11157. Surface-sterilized young leaves and stems of the plants were cocultivated with bacterial suspensions. Crown galls induced from stems were cultured with variation of culturing conditions and compared with untransformed cells. The growth rates and production of anthraquinone pigments of cells were remarkably improved by transformation. Furthermore, hairy roots were induced by inoculation or cocultivation with Agrobacterium rhizogenes strains.

Soil Mineral Nitrogen Upteke and Com Growth from Hairy Vetch with Conventional and No-Tillage Systems

  • Winter hairy vetch (HV) can be used as green manure with conventional tillage system (CT), in which chemical N fertilizer required for cultivation of sub-sequent com could be fully saved, or as cover crop with no-tillage system (NT) in which soil could be protected from erosion, control of weed, and the reduction of N fertilizer application. This experiment was carried out to compare the enrichment of soil mineral nitrogen (SMN) at corn root zone, and the changes of com growth and N uptake according to HV amounts (winter fallow, above-ground HV removed, intact HV, and HV added from aboveground HV removed) under two tillage systems in the upland field of National Crop Experiment Station, Suwon, Korea in 1996. HV cultivation during winter decreased SMN a little at com planting. HV incorporation with CT increased SMN rapidly during early growth stage according to rapid decomposition of Hv. SMN by HV cover with NT was increased slowly and its increase was higher in the surface soil (soil layer 0-7.5cm) compared to deep soil layer 7.5-22cm. Com growth and N status at corn silking stage, com yield and N uptake at harvest were increased in proportion to aboveground HV amounts regardless of tillage system. Average hairy vetch nitrogen (HV-N) uptake efficiency by com was 10% higher with CT than with NT in which average HV-N uptake efficiency was 43 %. Corn yields were not different between two tillage systems, but corn N uptake was increased by 33 kgN/ha more with CT than with NT due to the increase of corn N concentration. The increase of SMN and com N uptake from HV cover with NT could not be disregarded though those with CT were higher than with NT

Effects of green manures in organic vegetable production (유기농 채소생산을 위한 녹비작물 도입효과)

  • Organic farming in Korea has mainly focused on producing vegetables in plastic film house and cereals in paddy field. For high productivity of vegetables and cereals, most Korean farmers have not applied crop rotation in the cropping system. Thus, this study was carried out to evaluate the effects of crop rotation on the yield of red pepper and green onion, the changes in soil fertility and the potential as green manure of rye and hairy vetch. Rye and hairy vetch were cultivated for winter season every year, and directly incorporated into the soil. The yield of red pepper fruits in organic farming using crop rotation (OFCR) decreased 23 to 36% compared with conventional farming system (CFS). Whereas, green onion showed the increased yield of about 13%. In OFCR, total carbon content of soil was higher, however available phosphate content lower than conventional farming. As a result of measuring the bulk density of soil, OFCR and CFS were 1.26 to $1.35Mg/m^3$ and 1.37 to $1.42Mg/m^3$, respectively. Carbon and nitrogen contents of microbial biomass in OFCR were obviously higher compared with the CFS. In the plot cultivated rye for winter season, the weed germination was strongly reduced (about 52 %). Balance of macro nutrient elements, nitrogen and phosphate, in the application of rye and hairy vetch had a minus value. For potassium, the output value was higher than the input one, therefore organic farming under red pepper-rye or hairy vetch systems requires applying additional potassium input. Also, we selected two hairy vetch varieties of cv. Hungvillosa and Ostsaat which can be adapt at Korea climate. In order to estimate a yield of green manures, the weight of shoot and root was measured. The ratio of shoot and root between rye and hairy vetch showed 0.88 and 1.91, respectively. The potential levels of nitrogen, phosphate and potassium which could be supplied from rye were 7.7, 7.8 and 21.9 kg/10a and from hairy vetch were 17.0, 8.6 and 22.9 kg/10a, respectively. So we recommend that cultivating hairy vetch, as a nutrient supplier, and rye, as an amendment to enhance the soil physical property, for winter season be practical method in Korea organic farming system.

Comparison of Carbon Sequestration Potential of Winter Cover Crop Cultivation in Rice Paddy Soil

  • BACKGROUND: Cultivation of winter cover crops is strongly recommended to increase land utilization efficiency, animal feeding material self-production, and to improve soil and environmental quality. METHODS AND RESULTS: Four major winter crops (barley, Chinese milk vetch, hairy vetch, and rye) having different C/N ratio were seeded in silt loam paddy soil in the November 2007 and the aboveground biomass was harvested on the late May 2008 to evaluate its effectiveness as green manure, and root biomass distribution was characterized at the different depth (0-60 cm) to study its effect on physical properties and carbon sequestration in soil. During this experiment, the naturally growing weed in the rice paddy soil in Korea, short awn foxtail (Alopecurus aequalis Sobol), was considered as control treatment. Above-ground biomass of all cover crops selected was significantly higher than that of the control treatment (2.8 Mg/ha). Comparatively higher above-ground biomass productivity of rye and barley (15.8 and 13.5 Mg/ha, respectively) suggested that these cover crops possibly had the highest potential as a green manure and animal feeding material. Root biomass production of different cover crops followed the same trend as that for their above ground biomass. Rye (Secale cereal) might have the highest potential for soil C accumulation (7893 C kg/ha) by root biomass development, and then followed by barley (6985 C kg/ha), hairy vetch (6467 C kg/ha), Chinese milk vetch (6671 C kg/ha), and control (5791 C kg/ha). CONCLUSION(s): Cover crops like rye and barley having high biomass productivity might be the most effective winter cover crops to increase organic carbon distribution in different soil aggregates which might be beneficial to improve soil structure, aeration etc. and C sequestration.
