Effects of Juvenile Hormone Analogue on Changes of Protein and Amino acids in Haemolymph of the Silkworm, Bombxy mori. (유약홀몬이 가잠의 체액단백질 및 아미노산의 변동에 미치는 영향)

  • The experiment was carried out to investigate the effects of juvenile hormone analogue on changes of protein and amino acids in haemolymph of silkworm larva. Juvenile hormone analogue was topically administered to larvae at dose of 1$\mu\textrm{g}$ 10$\mu\textrm{g}$ per gm of body weight at 60hr. of the 5th instar. The results obtained were as follows ; 1. Larval duration of the fifth instar was extended about 1 day by JHA-1 and 4 days by JHA-10 as compared with the control. 2. Cocoon weight and cocoon layer weight by topical application of JHA were heavier than those of the control, but cocoon layer ratio was decreased in JHA-10 to the exclusion of JHA-1. 3. The concentration of haemolymph protein during the fifth instar was increased remarkably by the JHA application. 4. The total content of amino acids in haemolymph proteins of JHA-10 approximately doubled that of the control, with the conspicuous increase of glycine and arginine level.

  • To determine the metabolism of leucine during the cuticle formation and the sclerotization process in Pieris rapae L., $U-^3$H-leucine or $U-^4$C-tyrosine is injected into the haemolymph of newly molted pupa through dorsal cuticle of heart area. The results show that leucine as a common amino acid participates in the synthesis of cuticle protein over the first 3 hr after ecdysis. It is also shown that leucine in the haemolymph at ecdysis is freely being moved between major internal organs during the short time period post-ecdysis, providing the evidence for some involvements including haemolymph protein synthesis and storage of fat body and gut in metabolism of leucine.

  • Isolation, purification and molecular properties of adult specific protein (ASP) were investigated in the silkworm, Bombyx mori. ASP was purified from adult haemolymph by gel filtration on Sephadex G-100 and ion exchange chromatography on CM52. The preparation was shown to be homogeneous by native-PAGE and immunoelectrophoresis. The purified protein was consisted of two different subunits with molecular weights of 19.5kDa and 17.5kDa, and contained no sugars and no lipids.

  • Electrophoretic, immunological, and column chromatography methods were used to determine the appearance and distribution of storage proteins in various organs during the metamorphosis of Bombyx mori L. Two storage proteins start to appear in haemolymph in early 5th instar stage and show the identical mobility with fat body proteins. These proteins show the high concentration in haemolymph in last instar stage but accumulate in fat body after pupation. Storage protein-2 shows the distinct pattern for general storage proteins in both male and females. This protein is involved with the formation of cuticle protein in late last instar stage and appears to be temperally deposited in midgut during the pupal stage. Also SP-2 shows the identity with vitellogenin electrophoretically and immunologically and especially the positive reaction with antibody against yolk protein during the pupal stage, demonstrating that the storage protein is closely related to the formation of yolk protein.

  • The Injection of viable Escherichia coli K-12 with fifth instar larvae of cabbage butterfly, Artogeia rapae, induced at least five groups of proteins with the antibacterial activity against certain Gram-negative and/or Gram-positive bacteria in the haemolymph. These antibacterial proteins were separated and one was purified by different types of chromatography. The purified protein was heat-stable and basic peptide, and its molecular weight was approximately 4 kDa. We propose the name hinnavins for this antibacterial peptide.

  • In order to know the similarity of the electrophoretic protein patterns, distributional aspects of the frequency in each protein band, differences of protein patterns between female and male, and the number of the total protein bands of each variety in the haemolymph of the silkworm pupae according to their geographical origin, the silkworm varieties, and both the sexes, variable electrophoretic protein patterns were observed by polyacrylamide gel electrophoresis, using the silkworm strains which have been reared in the sericultural experiment station. 1. The total number of electrophoretic protein bands classified were twenty-eight, and nearly fourteen bands were shown by each variety on an average. 2. According to their geographical origin, the mean values of the similarity were 20.0%, 28.3% and 14.3% in Japanese, Chinese and European respectively, and the highest similarity was observed between Chinese varieties. 3. The similarity of 129 varieties were tested and it was 22.5%, 23.5% in female and male respectively. 4. The electrophoretic protein patterns in 129 varieties were tested and variation was found in 109 patterns in female and 112 in male. 5. The frequency of the each specified protein band in female and male having variation in protein bands in each variety was found averagely 6.6%. 6. The distribution frequency of the classified 28 protein bands in both the sexes within each geographical origin was variable from 0% to 100%. 7. Among the classified 28 protein bands, HP 11 protein band was found common in all the varieties tested. 8. HP 3 and HP 4 protein bands in female showed high density in electropherograms than in male.

  • The "crescent-egg" a new spontaneous mutant was detected in a white egg strain k37. Studies were carried out the linkage analysis, to investigate phenotypic characteristics and biochemical analysis of haemolymph and ovarian protein. The mutant, ore was independent from 20 linkage groups P(2), Ze(3),L(4), oc(5), sn8), Ia(9), w-1(10), K(11), ch(13), U(14), bl(15), cts(16), bts(17), mln(18), nb(19), oh(20),Lan(21), or(22), tub(23) and Xan(27). The fertilization, hatchability and larval growth were not different from the those of normal eggs. The content and composition of yolk protein were similared to normal eggs. Scanning electron microscopy revealed the areal specific structure in dorsal region of egg-shell of cre mutant. Analysis of chorion protein by isoelectrofocusing(IEF), was resolved no difference in the composite of the chorion protein. We conclude that the egg mutant ere is expressed only in the egg-shape formation and region specific determination.

  • Studies were carried out to investigate phenotypic expression, mortality and biochemical analysis of haemolymph proteins of nm-d, nm-f, nm-k and nmn bib-molting mutants of the silkworm, Bombyx mori. The non-molting mutants characters were expressed in the homozygote of each mutant genes. All strains of non-molting mutants were similar with each other in physiological characteristics, but the expression varied with each strains. The larvae of nm-d, nm-i and nmn died between day 5 and day 9 after hatching without the first molt. The nm-f and nm-k mutants died between day 5 and day 16 with a slight increase of body weight and, more than 90% of the mutants larvae died before the first molt and a few of them survived to the 2nd and the 3rd instar and died. The haemolymph protein components of nm-d, nm-i, and nmn were rapidly reduced, and on the other hand those of nm-f and nm-k consistently until they died. And there were no distinguishable difference in haemolymph components of non-molting mutants, as compared to those normals.
