Osteoblast Cell Morphology, Proliferation, and Differentiation in Variation with Biomaterials (생체재료의 선택에 따른 조골세포의 형상, 증식 및 분화)

  • Osteoblast-like cell morphology, proliferation, and differentiation were examined in variation with biomaterials. Cells were cultured on TiO$_2$, Ti, 3Y-TZP, HA (Hydroxyapatite) and Thermanox was used as a control specimen. Generally, all specimens have similar cell morphology within the same time interval. However, cells on HA seem to be more thicker than those on TiO$_2$, Ti, 3Y-TZP and cell overlapping was detected very frequently on HA. In case of cell proliferation and differentiation, bioactive material such as HA could help osteoblast-like cell proliferate and provoke a sharp increase of ALP. On the other hand, whether the substrate material is a bioinert ceramics or metal, it does not so strongly affect the cell attachment, proliferation. and differentiation.

Growth of Rice Plant and Chemical Properties of Soil as Affected by Nitrogen Fertilization Level in Milk Vetch(Astragalus sinicus L.) Cultivation in Paddy Field

  • This study was carried out to determine the effects on rice growth, yields and soil improvement under the different nitrogen levels in machine-transplanted rice after plowing at the fruiting stage of milk vetch. The fresh weight of milk vetch at plowing time was 20.95 ton/ha. In dry weight, T-N and C/N were 1.58%, 21.8%, respectively. Organic matter, total nitrogen and exchangeable calcium of soil after the experiment in the plot of milk vetch were higher than those before the experiment. Leaf area and dry weight of rice plants at heading date increased as nitrogen level increased. The number of panicle and spikelets per $\textrm{m}^2$were not different except for the 110 kg/ha nitrogen level plot with milk vetch. Brown rice yield ranged from 5.45 to 6.08 ton/ha, in creasing with increased nitrogen level. So the yield increased by 1% at 77 kg/ha nitrogen level, 7% at 110 kg/ha nitrogen level plot with milk vetch compared with conventional level (rice straw 5.4 ton/ha and nitrogen 110 kg/ha).

Desorption of Adsorbed Humic Acid on Carbon nano Tubes (카본나노튜브에 흡착된 휴믹산의 탈착에 관한 연구)

  • Concerns have been raised over the impact of nano materials on soil and groundwater environment with the increasing attention to the potential applications of carbon nano materials in various fields. Particularly, carbon nano materials introduced into water environment readily make complexes with humic acid (HA) due to their hydrophobic nature, so there have been increasing numbers of studies on the interaction between HA and carbon nano materials. In this study, we investigated the solubility of HA and multiwalled carbon nanotubes (MWCNT) in three different surfactant solutions of sodium dodecyl sulfate (SDS), Brij 30 and Triton X-100, and evaluated whether the HA can be effectively desorbed from the surface of MWCNT by surfactant. The objective of this study was to determine the optimal adsorption condition for HA to MWCNT. Futhermore, sodium dodecyl sulfate (SDS), Brij 30, Triton X-100 were used to elucidate the effect of desorption and separation on adsorbed HA on MWCNT. As a result, HA solution with 12.7 mg of total organic carbon (TOC) and 5 mg of MWCNT showed the highest adsorption capacity at pH 3 reacted for 72 hrs. Weight solubilizing ratio (WSR) of surfactants on HA and MWCNT was calculated. HA had approximately 2 times lower adsorption capacity for the applied three surfactants compared to those of MWCNT, implying that the desorption of HA may occur from the HA/MWCNT complex. According to the results of adsorption isotherm and weight solubilizing ratio (WSR), the most effective surfactants was the SDS 1% soluiton, showing 53.63% desorption of HA at pH 3.

Effect of Organic Fertilizer Levels on Dry Matter Yield, Nitrogen Fixation and Transfer on the Barley and Pea Mixtures (보리.완두 포장에서 유기질비료 시용 수준이 건물수량과 질소 고정 및 이의 이동에 미치는 영향)

  • In order to study the effect of organic fertilizer on dry matter (DM) yield, nitrogen fixation and transfer from pea to barley, an experiment was carried out from May to June in 2008 in Incheon. A completely randomized block design with three replications was used for the experiment and one reference plot assigned each treatment for nitrogen fixation evaluation. Seeding mixture was 40kg barley and 80kg pea per ha. N rate of 40, 80 and 120kg/ha as organic fertilizer was applied at seeding. The equivalent of 1kg per ha as $(15NH_4)_2SO_4$ Solution at 99.8 atom N was applied to the plots ($30{\times}20cm$) at mid April. Forage was harvested from each plot in ripening stage at ground level and separated into barley and pea. Nitrogen fixation was 32.4%, 23.4% and 0% at three different organic N levels. Transfer rate were from 47.6% to 21.8% in difference method and 24.6% and 21.4% on $^{15}N$ dilution method. N Transfer amount were from 92.8kg/ha to 41.9kg/ha on difference method and 47.3kg in the 40kg N plots and 49.7kg in the 80kg N plots on $^{15}N$ dilution method but there was no N transfer in 120kg N organic fertilizer plots. Benefit from increased organic fertilizer was not clear in terms of nitrogen fixation and transfer from pea to barley in barley and pea mixtures.

Preparation of Thermo-Responsive and Injectable Hydrogels Based on Hyaluronic Acid and Poly(N-isopropylacrylamide) and Their Drug Release Behaviors

  • Copolymers composed of hyaluronic acid (HA) and poly(N-isopropylacrylamide) (PNIPAAm) were prepared to create temperature-sensitive injectable gels for use in controlled drug delivery applications. Semi-telechelic PNIPAAm, with amino groups at the end of each main chain, was synthesized by radical polymerization using 2-aminoethanethiol hydrochloride (AESH) as the chain transfer agent, and was then grafted onto the carboxyl groups of HA using carbodiimide chemistry. The result of the thermo-optical analysis revealed that the phase transition of the PNIPAAm-grafted HA solution occurred at around 30$\∼$33$^{circ}C$. As the graft yield of PNIPAAm onto the HA backbone increased, the HA-g-PNIPAAm copolymer solution exhibited sharper phase transition. The short chain PNIPAAm-grafted HA ($M_{w}$=6,100) showed a narrower temperature range for optical turbidity changes than the long chain PNIPAAm-grafted HA ($M_{w}$=13,100). PNIPAAm-grafted HA exhibited an increase in viscosity above 35$^{circ}C$, thus allowing the gels to maintain their shape for 24 h after in vivo administration. From the in vitro riboflavin release study, the HA-g-PNIPAAm gel showed a more sustained release behavior when the grafting yield of PNIPAAm onto the HA backbone was increased. In addition, BSA released from the PNIPAAm-g-HA gels showed a maximum concentration in the blood 12 h after being injected into the dorsal surface of a rabbit, followed by a sustained release profile after 60 h.

  • A novel approach to the manufacture of biocompatible ceramic scaffold for tissue engineering using micro-stereolithography system is introduced. Micro-stereolithography is a newly proposed technology that enables to make a 3D micro structure. The 3D micro structures made by this technology can have accurate and complex shape within a few micron error. Therefore, the application based on this technology can vary greatly in nano-bio fields. Recently, tissue-engineering techniques have been regarded as alternative candidate to treat patients with serious bone defects. So many techniques to design and fabricate 3D scaffolds have been developed. But the imperfection of scaffold such as random pore size and porosity causes a limitation in developing optimum scaffold. So scaffold development with controllable pore size and fully interconnected shape have been needed for a more progress in tissue engineering. In this paper, bone scaffold was developed by applying the micro-stereolithography to the mold technology. The scaffold material used was HA(Hydroxyapatite) nano powder. HA is a type of calcium phosphate ceramic with similar characteristic to human inorganic bone component. The bone scaffold made by HA is expected, in the near future, to be an efficient therapy for bone defect.

Runoff Loading of Nutrients from a Paddy Field during Non-Cropping Season (비영농기간 단일필지 논으로부터 영양물질의 유출량)

  • In intensive agriculture, exceeded chemical fertilizer application would increase the concentration of nitrate nitrogen in groundwater. Consequently, it could bring the eutrophication in lakes and streams. The present study examined runoff loading of nitrogen and phosphorus from the paddy field during non-cropping season. The runoff loading of total-N, ammonia-N, nitrate-N and total-P were 12.96kg/ha, 5.42kg/ha, 1.52kg/ha and 1.41kg/ha. When the runoff loading of nutrients was compared by runoff water and sediments. About 70-80% of total-N by runoff water and the rest 20-30% by runoff sediments were flowed into streams. But 60-70% of total-P by runoff sediments and the rest 30-40% by runoff water were flowed into streams. The phosphorus compounds, which were flowed into streams by runoff sediments and then sedimented, keep exchanging with water at water body in undelivered condition. And it moves gradually into water layer. This process can cause eutrophication continually and repeatedly in water environment. So, a sound program is needed to reduce soil erosion from farmlands.

  • The present study was carried out for 7-months (from October 1.1997 to April 30. 1998) at a rice cultivation area blocked from livestock farming, farmstead and industrial complex to investigate the runoff loading of nitrogen and phosphorus in a plot(5,000$m^2$) paddy field areas. The runoff loading of total-N, amminia-N nitrate-N and total-P were 12.96kg/ha, 5.42kg/ha, 1.52kg/ha and 1.41kg/ha , respectively. When the runoff loading of nutrients was compared by dissolved and adsorbed forms, about 66% of total-N by dissolved form and the rest 34% by adsorbed form were flowed into streams. But 56% of total-P by adsorbed form and the rest 44% by dissolved form were flowed into streams. The phosphrous compounds , which were flowed into streams by runoff sediments and then sedimented, keep exchanging with water at water body in undelivered condition. And it moves gradually into water layer. This process can cause eutrophication continually and repeatedly in water environment . So, a sound program is needed to reduce soil erosion from farmlands.

The Costume Culture in Early 17th Century Perspectives Through Excavated Letters of Hyun-Poong Kwak's Family (17세기 전기 현풍 곽씨 집안의 의생활에 대한 소고)

  • This Paper reveals the costume culture of early 17th century from the study of private letters found in the tomb of Mrs. JinJoo Ha(\ulcorner - after 1652) excavated in 1989. The tomb was located at KooJi-myon, DalSung-kun, Kyung-Poong Province. Joo Kwak(1569-1617), Mrs. Ha s husband, didn t have a title from the government, but he was a clan in HyunPoong area who had enough land and servants. However, he didn't live togther with mrs. Ha, his second wife, he had to travel between SoRye and NonKong where his wife resided. The 40 or so private letters that contains valuable information that can help us to understand and reconstruct the costume culture of early 17th century in HyunPoong area. We believe that these materials contain somewhat general information that reveals costume of KyungSangDo area even though the materials are from a specific family. By accumulating information from real material like these letters, we also believe that it will give us a better understanding on the costume culture in 17th century, Choson dynasty. Furthermore, it will certainly help us to reconstruct a more realistic lifestyle of the time.

Site Characteristics and Carbon Dynamics of the Gwangneung Deciduous Natural Forest in Korea

  • The study area, Kwangneung Experiment Forest (KEF) is located on the west-central portion of Korean peninsula and belongs to a cool-temperate broadleaved forest zone. At the old-growth deciduous forest near Soribong-peak (533.1m) in KEF, we have established a permanent plot and a flux tower, and the site was registered as a KLTER site and also a KoFlux site. In this study, we aimed to present basic ecological characteristics and synthetic data of carbon budgets and flows, and some monitoring data which are essential for providing important parameters and validation data for the forest dynamics models or biogeochemical dynamics models to predict or interpolate spatially the changes in forest ecosystem structure and function. We made a stemmap of trees in 1 ha plot and analyzed forest stand structure and physical and chemical soil characteristics, and estimated carbon budgets by forest components (tree biomass, soils, litter and so on). Dominant tree species were Quercus serrata and Carpinus laxiflora, and accompanied by Q. aliena, Carpinus cordata, and so on. As a result of a field survey of the plot, density of the trees larger than 2cm in DBH was 1,473 trees per ha, total biomass 261.2 tons/ha, and basal area 28.0 m2/ha. Parent rock type is granite gneiss. Soil type is brown forest soil (alfisols in USDA system), and the depth is from 38 to 66cm. Soil texture is loam or sandy loam, and its pH was from 4.2 to 5.0 in the surface layer, and from 4.8 to 5.2 in the subsurface layer. Seasonal changes in LAI were measured by hemispherical photography at the l.2m height, and the maximum was 3.65. And the spatial distributions of volumetric soil moisture contents and LAIs of the plot were measured. Litterfall was collected in circular littertraps (collecting area: 0.25m2) and mass loss rates and nutrient release patterns in decomposing litter were estimated using the litterbag technique employing 30cm30cm nylon bags with l.5mm mesh size. Total annual litterfall was 5,627 kg/ha/year and leaf litter accounted for 61% of the litterfall. The leaf litter quantity was highest in Quercus serrata, followed by Carpinus laxiflora and C. cordata, etc. Mass loss from decomposing leaf litter was more rapid in C. laxiflora and C. cordata than in Q. serrata litter. About 77% of C. laxiflora and 84% of C. cordata litter disappeared, while about 48% in Q. serrata litter lost over two years. The carbon pool in living tree biomass including below ground biomass was 136 tons C/ha, and 5.6 tons C/ha is stored in the litter layer, and about 92.0 tons C/ha in the soil to the 30cm in depth. Totally more than about 233.6 tons C/ha was stored in DK site. And then we have drawn a schematic diagram of carbon budgets and flows in each compartment of the KEF site.
