참치 추출물의 항암 및 면역효과 (Antitumor and Immunological Effects of Tuna Extract)

  • 본 연구는 참치의 석유 에텔 추출물로 부터 항암 효과가 있는 활성 성분을 분리 정제하고, 흰생쥐의 임파모 세포인 LI210 과 P388, 인체 장암세포인 HCT-48, HT-29 및 HRT-18를 대상으로 단계별로 얻어지는 성분의 항암 활성올 측정하고, 그 물질의 생체내에서의 작용 기전을 밝히기 위해 여러가지 면역학적 실험을 수행하였다. In vitro배양액에서의 인체 결장암세포인 H HCT-48에 대해, crude extract는 배양액 1ml강 $60\mu\textrm{g}$ 첨가시 증식이 2배로 억제되었으며(1 unit), 이를 column chromatography 수행하여 얻은 fraction D의 1 unit은 $2 22\mu\textrm{g}$이며, 다시 TLC를 하여 얻은 Spot I 의 1 unit은 $9\mu\textrm{g}$으로서 정제 했을때 277배 증가하였다. Spot I 은 GC/ M5에 의해 7가지 성분이 섞여있는 혼합물로서 이 중 4가지 생분은 $C_{14:0},\;C_{16:0},\;C_{17:1},\;및\;C_{18:0}$인 지방산으로 확인이 되었으며, 나머지 3가지는 지방 성분으로 분자식은 확인되지 않았다. 다른 인체 장암 세포인 HT-29과 HRT-18에 대한 crude extract의 1 unit은 $64\mu\textrm{g}\;및\;54\mu\textrm{g}$이었으며 frattion D의 1 unit에 해당하는 양은 두 세포 모두 약 $34\mu\textrm{g}$이었다. 또한, 생쥐의 백혈병 성 임파모 세포인 L1210과 P388의 경우 crude extract의 1 unit에 해당하는 양은 $2-2.5\mu\textrm{g}$이며, fraction D의 경우는 $1.2-1.4\mu\textrm{g}$으로서 참치 추출물의 세포 종식 억제력은 백혈병성 임파모세포에서 인체 장암세포보다 높았으며, 암세포의 종류에 따라 효과의 차이가 있었다. 참치 추출물(fraction D)을 첨가한 배양액에서 배양한 HCT-48 세포획 크기 분포도는 배양시간이 경과함에 따라 세포의 크기가 작아지며, 현미경 사진에서 보면 본래의 형태가 위축되거나, 세포의 주위가 흐트러지면서 사면 현상을 보였다. 참치 석유 에텔 추출물에는 DHA가 18.8%를 차지 하며 EPA는, 3.25% 포함되어 있었다. 대조군 생쥐에 비해 S-180을 접종한 후 참지 추출물을 투여한 생쥐에서 용혈반 형성 세포수와 혈청 단백질 중 immunoglobulin의 relative %가 현저히 증가되었다. 그러나 비장의 자연 살해 세포의 활성에는 변화를주지 않았다. 이상의 결과로 보아, 참치의 석유 에델 추출물중에 in vitro에서 인체 장암세포 및 백혈병성 임파모세포의 증식 억제 및 생체내에서 면역 증강 효과를 나타내는 성분이 존재함을 알 수 있다.

양유(洋乳)의 증숙 및 발효 추출물의 인지능 개선 활성 (Cognitive Enhancing Activity of the Steamed and Fermented Extracts of Codonopsis lanceolata Radix)

  • This study was designed to determine and compare the cognitive enhancing effect of various Codonopsis lanceolata extracts by steaming, fermentation and high hydrostatic pressure process. We prepared water extract of C. lanceolata, steamed C. lanceolata, steamed and fermented C. lanceolata and C.lanceolata by high hydrostatic pressure process and fermentation. Cognitive enhancing effect of extracts was evaluated in scopolamine-induced memory impairment mice using by passive avoidance test and Morris water maze tests. MTT assay was conducted to investigate neuroprotective effect on glutamate induced cell death in HT22 cells. Steamed and fermented C. lanceolata water extract decreased escape latency in Morris water maze test and increased the latency time of the passive avoidance test compared to other extracts. Furthermore, the steamed and fermented C. lanceolata water extract showed neuroprotective effect. These results suggest that steaming and fermentation process more improve cognitive enhancing effect of C. lanceolata than other process.

Neuroprotective and Anti-Neuroinflammatory Activities of Anthraquinones Isolated from Photorhabdus temperata Culture Broth

  • Photorhabdus temperata (PT), a gram-negative bacterium, lives symbiotically within entomopathogenic nematodes. The insecticidal compounds derived from Photorhabdus are used as biopesticides in agriculture. However, the physiological properties are not well characterized. In the course of our screening for neuroprotective and anti-neuroinflammatory substances from natural products, the culture broth of PT showed considerable activities. By activity-guided purification, five anthraquinones, namely, 3-methoxychrysazine (1), 1,3-dimethoxy-8-hydroxy-9,10-anthraquinone (2), 1,3,8-trihydroxy-9,10-anthraquinone (3), 3,8-dihydroxy-1-methoxy-9,10-anthraquinone (4), and 1,3,4-trimethoxy-8-hydroxy-9,10-anthraquinone (5), were isolated from the ethyl acetate fraction of the PT culture broth. Among the isolated compounds, $75{\mu}M$ 3 significantly protected mouse hippocampal neuronal cells (HT22) against 5 mM glutamate-induced cell death via the inhibition of reactive oxygen species production, $Ca^{2+}$ influx, and lipid peroxidation. Additionally, 3 and 4 effectively suppressed the interferon-${\gamma}$-induced neuroinflammation of mouse-derived microglial cells (BV2) at 10 ng/ml, via the reduction of nitric oxide, interleukin-6, and tumor necrosis factor-${\alpha}$. Anthraquinones 3 and 4 derived from the PT culture broth are a potential starting point to discover neuroprotective and anti-neuroinflammatory drug leads. The novel compound 5 is reported for the first time in this study.

Expression of a Fusion Protein with Cry1Ac Protein and a Scorpion Insect Toxin in Acrystalliferous Bacillus thuringiensis Strain

  • Expression of a fusion protein between B. thuringiensis crystal protein, Cry1Ac1 and a scorpion insect toxin (AaIT, Androctonus australis Hector insect toxin) in acrystalliferous B. thuringiensis strain (Cry-B strain) was examined. The cry 1Ac1 gene was cloned in B. thuringiensis-E coli shuttle vector, pHT3101, under the control of the native cry 1Ac1 gene promoter (pProAc) and a gene encoding AaIT was inserted in XhoI site in the middle of the cry 1Ac1 gene (pProAc-ScoR). B. thuringiensis Cry-B strain carrying pProAc-ScoR (PyoAc-ScoR/CB) produced an inclusion body of irregular shape and the expressed fusion protein is approximately 65 kDa in size. Sporulated cells and spore-crystal mixtures of ProAc-ScoR/CB had insecticidal activity against Plutella xylostella larvae, showing $LT_50$ of ProAc-ScoR/CB (22.59 hrs) lower than that of ProAc/CB (30.06 hrs) at $1{\times}{10^7} {CEU/cm^2}$. These results suggest that the fusion protein including a B. thuringiensis crystal protein and an AaIT may be functionally expressed in B. thupingiensis. Moreover, we verified the additive toxicity of AaIT, which is a new feasible candidate for insect control.

민들레의 Monoamine Oxidase 저해활성 (The Inhibitory Activity of the Taraxacum mongolicum on Monoamine Oxidase)

  • We examined the inhibitory activities against monoamine oxidase (MAO) of Taraxacum mongolicum in vitro and in vivo methods. Methanol extract of T. mongolicum showed significantly inhibitory activities on MAO-A and MAOB that were prepared from rat brain and liver in vitro. MAO-A and MAO-B activities were potently inhibited by chloroform fraction of T. mongolicum in vitro tests. The $IC_{50}$ values of each fraction on MAO-A are as followed; methanol extracts (0.90 mg/ml), $CHCl_3$ fraction (0.10 mg/ml), EtOAc fraction (0.36 mg/ml). and those on MAO-B are methanol extracts $(0.39{\mu}g/ml)$, $CHCl_3$ fraction $(0.18{\mu}g/ml)$, BuOH fraction $(0.22{\mu}g/ml)$. Those MAO-A and MAO-B activities in vivo tests have different tendency each other. MAO-A activity was increased by the oral administration of ethanol extract of T mon golicum, while MAO-B activity was decreased. The concentration of serotonin of brain tissue after oral administration of ethanolic extract of T. mongolicum is slightly increased in rat. This tendency is not different from the activity of deprenyl which is the well known MAO inhibitor used as a positive control. Based on these results, we can suggest that T. mongolicum may have the effects on the inhibitory activities against MAO. Thess activities of T. mongolicum is considerable for development of functional materials for the purpose of treatment and control of depressant, dementia, Parkinson' disease, stress and promoting exercise.

국민 건강보험 급여 한약 처방 56종의 치매 주요 생리지표 및 신경세포 변화에 대한 효능 비교 연구 (Screening of 56 Herbal formulas covered by the National Health Insurance Service on Dementia-related Factors)

  • Objectives: The purpose of this study was to investigate the effects of 56 herbal formulae covered by the National Health Insurance Corporation (NHIC) on dementia-related biomarkers and neuronal cell changes. Methods: The 56 herbal formulae were extracted with 70% ethanol at $100^{\circ}C$ for 2 h. The antioxidant properties was measured by radical scavenging assay using ABTS+ radical. The acetylcholinesterase (AChE) activity was tested by Ellman's assay and $amyloid-{\beta}$ ($A{\beta}$) aggregation was determined using fluorescence method. To estimate the inhibitory effects of herbal formulae on neuronal cell death and inflammation using HT22 hippocampal cells and BV-2 microglia, respectively. Results: Among the 56 herbal formulae, Dangguiyukhwangtang, Banhasasimtang, Samhwangsasimtang, Cheongwiesan, Hwangryunhaedoktang, Banhabaekchulchunmatang, Jaeumganghwatang, Cheongseoikgitang, and Hoechunyanggyuksan has a significant inhibitory effects on acetylcholinesterase (AChE) activity. Doinseunggitang and Samhwangsasimtang exerted the effect on the inhibition of $amyloid-{\beta}$ ($A{\beta}$) aggregation. Additionally, 10 herbal formulae affected AChE and $A{\beta}$ aggregation revealed antioxidant activity as well as neuroprotective and anti-neuroinflammation effects in neuronal cell lines. Conclusions: 10 herbal formulae that have been shown to be effective against the major dementia markers have been shown to have antioxidant activity, neuronal cell protection and inhibition of brain inflammation. Further investigation of these herbal formulae will need to be validated in dementia animal models.

Effect of Codonopsis lanceolata with Steamed and Fermented Process on Scopolamine-Induced Memory Impairment in Mice

  • Codonopsis lanceolata (Campanulaceae) traditionally have been used as a tonic and to treat patients with lung abscesses. Recently, it was proposed that the extract and some compounds isolated from C. lanceolata reversed scopolamine-induced memory and learning deficits. The purpose of this study was to evaluate the improvement of cognitive enhancing effect of C. lanceolata by steam and fermentation process in scopolamine-induced memory impairment mice models by passive avoidance test and Morris water maze test. The extract of C. lanceolata or the extract of steamed and fermented C. lanceolata (SFCE) was orally administered to male mice at the doses of 100 and 300 mg/kg body weight. As a result, mice treated with steamed and fermented C. lanceolata extract (SFCE) (300 mg/kg body weight, p.o.) showed shorter escape latencies than those with C. lanceolata extract or the scopolamine-administered group in Morris water maze test. Also, it exerted longer step-through latency time than scopolamine treated group in passive avoidance test. Furthermore, neuroprotective effect of SFCE on glutamate-induced cytotoxicity was assessed in HT22 cells. Only SFCE-treated cells showed significant protection at 500 ${\mu}g/ml$. Interestingly, steamed C. lanceolata with fermentation contained more phenolic acid including gallic acid and vanillic acid than original C. lanceolata. Collectively, these results suggest that steam and fermentation process of C. lanceolata increased cognitive enhancing activity related to the memory processes and neuroprotective effect than original C. lanceolata.

증숙 더덕 추출물의 인지능력 개선 효과 (Effect of Steamed Codonopsis lanceolata on Spatial Learning and Memory in Mice)

  • Alzheimer's disease is progressive neurodegenerative disorder by the loss of memory and learning abilities. Codonopsis lanceolata (C. lanceolata) is traditional medicinal plant used for the treatment of inflammatory diseases. The aim of study was to evaluate the effect of steamed C. lanceolata on scopolamine-induced memory impairment in the Morris water maze test and passive avoidance test. In addition, this study investigated the neuroprotective effects of steamed C. lanceolata on glutamate-induced cell death in HT22 cells using MTT assay. The results showed that steamed C. lanceolata (500 mg/kg body weight, p.o.) reversed spatial memory impairment by scopolamine in Morris water maze test and passive avoidance test. Steamed C. lanceolata attenuated memory impairment by scopolamine compared with common C. lanceolata. In addition, administration of steamed C. lanceolata significantly also reduced cell death. We suggest that steaming process more improve cognitive enhancing and neuroprotective effect of C. lanceolata than common C. lanceolata.

Evaluation of the Probiotic Potential of Bacillus polyfermenticus CJ6 Isolated from Meju, a Korean Soybean Fermentation Starter

  • To evaluate the probiotic potential of Bacillus polyfermenticus CJ6 isolated from meju, a Korean traditional soybean fermentation starter, its functionality and safety were investigated. B. polyfermenticus CJ6 was sensitive to all antibiotics listed by the European Food Safety Authority. The strain was also non-hemolytic, carried no emetic toxin or enterotoxin genes, and produced no enterotoxins. The resistance of B. polyfermenticus CJ6 vegetative cells and spores to simulated gastrointestinal conditions was high (60-100% survival rate). B. polyfermenticus CJ6 produced high amounts (0.36 g as a purified lyophilized form) of ${\gamma}$-polyglutamic acid (PGA). We speculate that the improved cell viability and the production of ${\gamma}$-PGA have a significant correlation. Adhesion of the strain to Caco-2 and HT-29 cells was weaker than that of the reference strain (Lb. rhamnosus GG), but it was comparable to or stronger than those of reported Bacillus spp. When B. polyfermenticus CJ6 spores were given orally to mice, the number of cells excreted in the feces was 4-fold higher than the original inocula. This suggests the inoculated spores propagated within the intestinal tract of the mice. This idea was confirmed by field emission scanning electron microscopy, which revealed directly that B. polyfermenticus CJ6 cells germinated and adhered within the gastrointestinal tract of mice. Taken together, these findings suggest that B. polyfermenticus CJ6 has probiotic potential for both human consumption and use in animal feeds.

형용자침이 발열 흰쥐의 체온하강과 중추성 면역에 미치는 영향 (A Study on Acupuncture of Spring-point(형혈); Effect on Body Temperature and Central Immune System)

  • Objective : There are many reports that acupuncture has thermoregulatory effects on human and animals. To investigate the effect and mechanism of antipyretic action of acupuncture, we observed the body temperature and cytokine expressions in the hypothalamus of rats. Methods : Lipopolysaccharide (LPS, i.p., 2.5mg/kg) was injected to conscious rats (Sprague-Dawley, male, n=4l) to cause hyperthermia and simple needling (stainless steel, 0.25 mm o.d., 5 mm insertion for 10 sec with no manipulation) was performed bilaterally with the measurement of rectal temperature. Next, we sacrificed rats to remove brain and determined the level of mRNA for interleukin-6 (IL-6), $interleukin-1{\beta}{\;}(IL-1{\beta})$, interleukin-2 (IL-2) and $interferon-{\gamma}{\;}(IFN-{\gamma})$ in the hypothalamus by using reverse transcriptase-polymerase chain reaction (RT-PCR). Resul1s : Needling on forepaw (acupoint HT8) and needling on hindpaw (acupoint BL66 and acupoint LR2) significantly inhibited LPS-induced fever of rats (P<0.01, 10 min after treatment respectively), but same treatment on proximal tail (non-acupoint) did not cause any change on fever. The levels of IL-6 and $IL-1{\beta}$ mRNA in the hypothalamus was significantly enhanced by LPS-injection, while the level of IL-6 and $IL-1{\beta}$ mRNA was markedly reduced after treatment on BL66 (P<0.01). Treatment on forepaw reduced it slightly, but not significantly. Equivalent stimulation on proximal tail did not cause any changes. Conclusions : Our results indicate that acupuncture stimulation on various body parts has differential thermoregulatory effects on LPS-induced fever of rats with site-specificity. And, we suggest that its antipyretic action might be accompanied with the suppression of hypothalamic production of pro-inflammatory cytokine of immune system, IL-6 and $IL-1{\beta}$.
