Profiling of Salivary Exosomal Micro RNAs in Burning Mouth Syndrome Patients

  • Purpose: The exact causes of burning mouth syndrome (BMS) is unclear so far. There are many studies to elucidate the relation between oral disease and genetic predisposition. In this study, we first tried to investigate salivary exosomal genetic components that could play an important role for diagnosing and elucidating the progression of BMS. Methods: We compared salivary exosomal micro RNAs (miRNAs) of BMS Patients to those of control using next generation sequencing (NGS). Unstimulated whole saliva from 15 patients with BMS and 10 control subjects were divided into two sets. Isolated exosomes and their total RNAs were subject to NGS for the screening of miRNAs. Results: There were up-regulated 10 exosomal miRNAs (hsa-miR-1273h-5p, hsa-miR-1273a, hsa-miR-1304-3p, hsa-miR-4449, hsa-miR-1285-3p, hsa-miR-6802-5p, hsa-miR-1268a, hsa-miR-1273d, hsa-miR-1273f, and hsa-miR-423-5p) and down-regulated 18 exosomal miRNAs (hsa-miR-27b-3p, hsa-miR-16-5p, hsa-miR-186-5p, hsa-miR-142-3p, hsa-miR-141-3p, hsa-miR-150-5p, hsa-miR-374a-5p, hsa-miR-93-5p, hsa-miR-29c-3p, hsa-miR-29a-3p, hsa-miR-148a-3p, hsa-miR-22-3p, hsa-miR-27a-3p, hsa-miR-424-5p, hsa-miR-19b-3p, hsa-miR-99a-5p, hsa-miR-548d-3p, and hsa-miR-19a-3p) in BMS patients comparing with those of control subjects. Conclusions: We show that there are 28 differential expression of miRNAs between the patients with BMS and those of control subjects. The specific function of indicated miRNAs should be further elucidated.

Screening of MicroRNA in Patients with Esophageal Cancer at Same Tumor Node Metastasis Stage with Different Prognoses

  • Patients at the same pathological stage of esophageal cancer (EC) that received the same surgical therapy by the same surgeon may have distinct prognoses. The current study aimed to explore the possibility of differentially-expressed microRNAs (miRNAs) underlying this phenomenon. Samples were collected from EC patients at the same tumor node metastasis (TNM) stage but with different prognoses. Paracancerous normal tissues were taken as controls. The specimens were histopathologically analyzed. Differentially-expressed miRNAs were analyzed using real-time quantitative reverse transcription polymerase chain reaction. Compared with patients with poor prognosis, those with good prognosis exhibited 88 two-fold or more than two-fold increased miRNA fragments and 4 half-decreased miRNAs. The most noticeably up-regulated miRNAs included hsa-miR-31, hsa-miR-196b, hsa-miR-652, hsa-miR-125a-5p, hsa-miR-146b, hsa-miR-200c, hsa-miR-23b, hsa-miR-29a, hsa-miR-186, hsa-miR-205, hsa-miR-376a, hsa-miR-410, hsa-miR-532-3p, and hsa-miR-598, whereas the most significantly-downregulated miRNAs were hsa-let-7e, hsa-miR-130b, and hsa-miR-103. EC patients at same TNM stage but with different prognoses show differentially-expressed miRNAs.

Expression Analyses of MicroRNAs in Hamster Lung Tissues Infected by SARS-CoV-2

  • Coronavirus disease 2019 (COVID-19), caused by severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2), is an infectious disease with multiple severe symptoms, such as fever over 37.5℃, cough, dyspnea, and pneumonia. In our research, microRNAs (miRNAs) binding to the genome sequences of severe acute respiratory syndrome coronavirus (SARS-CoV), Middle East respiratory-related coronavirus (MERS-CoV), and SARS-CoV-2 were identified by bioinformatic tools. Five miRNAs (hsa-miR-15a-5p, hsa-miR-15b-5p, hsa-miR-195-5p, hsa-miR-16-5p, and hsa-miR-196a-1-3p) were found to commonly bind to SARS-CoV, MERS-CoV, and SARS-CoV-2. We also identified miRNAs that bind to receptor proteins, such as ACE2, ADAM17, and TMPRSS2, which are important for understanding the infection mechanism of SARS-CoV-2. The expression patterns of those miRNAs were examined in hamster lung samples infected by SARS-CoV-2. Five miRNAs (hsa-miR-15b-5p, hsa-miR-195-5p, hsa-miR-221-3p, hsa-miR-140-3p, and hsa-miR-422a) showed differential expression patterns in lung tissues before and after infection. Especially, hsa-miR-15b-5p and hsa-miR-195-5p showed a large difference in expression, indicating that they may potentially be diagnostic biomarkers for SARS-CoV-2 infection.

Physical Properties of High-Solid Coatings with Acrylic Resins Containing Acetoacetoxy Group and Allophanate-Trimer (Acetoacetoxy기 함유 아크릴수지와 Allophanate-Trimer에 의한 하이솔리드 도료의 도막물성)

  • Copolymers(HSA-98-20, HSA-98-0, HSA-98+20) which we acrylic resin containing 80% solid content were synthesized by the reaction of monomers, including methyl methacrylate, n-butyl acrylate, and 2-hydroxyethyl acrylate with a functional monomer, acetoacetoxyethyl methacrylate (AAEM), which nay give improvements in cross-linking density and physical properties of films. The physical properties of prepared acrylic resins, containing AAEM, are as follows viscosities, $1420\sim5760cps$ ; number average molecular weight, $2080\sim2300g/mol$; polydispersity index, $2.07\sim2.19$ ; and conversions, $88\sim93%$. In the next step, high-solid coatings (HSA-98-20C, HSA-98-0C, HSA-98+20C) were prepared by the curing reaction between acrylic resins containing 80% solid content and isocyanate at room temperature. Various properties were examined on the film coated with the prepared high-solid coatings. The introduction of AAEM to the coatings enhanced the abrasion resistance and solvent resistance, which indicated the possible use of high- solid coatings for top-coating materials of automobile. Since the curing by viscoelastic measurement occurred in sequence of HSA-98+20C > HSA-98-0C > HSA-98-20C, it was concluded that the curing rates became faster with incresing $T_g$ values.

Epigallocatechin 3-gallate Binds to Human Salivary α-Amylase with Complex Hydrogen Bonding Interactions

  • Amylase is a digestive enzyme that catalyses the starch into sugar. It has been reported that the green tea flavonoid (or polyphenols) (-)-epigallocatechin 3-gallate (EGCG) inhibits human salivary ${\alpha}$-amylase (HSA) and induced anti-nutritional effects. In this study, we performed docking study for seven EGCG-like flavonoids and HSA to understand the interaction mechanism of HSA and EGCG and suggest new possible flavonoid inhibitors of HSA. As a result, EGCG and (-)-epicatechin gallate (ECG) bind to HSA with complex hydrogen bonding interactions. These hydrogen bonding interactions are important for inhibitory activity of EGCG against HSA. We suggested that ECG can be a potent inhibitor of HSA. This study will be helpful to understand the mechanism of inhibition of HSA by EGCG and give insights to develop therapeutic strategies against diabetes.

  • The concept of HSA (Home Service Aggregator) is derived from performing the energy IT business efficiently as well as successfully launcing a new service based on BPL (Broadband over Power Line). The HSA business using a BPL can extend the field of energy industry and an give a chance to create a new demand by consumer-oriented services. This study focuses on the exact evaluation of HSA business using BPL, and reasonable trusty evaluation should be the first step to launch the HSA business. In this study, the categories of cost are comprised of equipment (mainly RSM and MGW) cost, instalation cost, and maintenance cost. AMR (Automatic Meter Reading), internet integration billing service, integration charging service, internet service, sorority service, and electricity safety are listed for benefit. In this study, the ROI of HSA business is 0.9594, which is less than 1. However, that value does not consider the electricity safety benefit which is classified as a social benefit. Therefore, the value can be above 1 if it includes social and private benefits.

Curing Reaction and Physical Properties of High-Solid Acrylic/Isocyanate Coatings (하이솔리드 아크릴/이소시아네이트 도료의 경화반응과 도막물성)

  • In the previous study, three kinds of monomers and the functional monomer, acetoacetoxyethyl methacrylate (AAEM), which could improve the film property and cross-linkage, were polymerzied into acrylic resin copolymers (HSA-98-20, HSA-98-0, HSA-98+20) containing 80% solid content. In this study, the high-solid coatings(HSA-98-20C, HSA-98-0C, HSA-98+20C) were prepared by the curing reaction between acrylic resins containing 80% solid content and isocyanate at room temperature. Various properties were examined for the film coated with the prepared high-solid coatings. The introduction of AAEM in the coatings enhanced the abrasion resistance and solvent resistance of coatings, which indicated the possible use of high-solid coatings for top-coating materials of automobile. The curing times measured by viscoelastic measurement were 350, 264, and 212 min for HSA-98-20C, HSA-98-0C, and HSA-98+20C, respectively. This shows that the curing times become shorter with increasing $T_g$ values.

The Evaluation of the Axial Strength of Composite Column with HSA800 Grade Steel (HSA800 강재를 적용한 합성기둥의 축방향 내력 평가)

  • According to the Korean Building Code (KBC), the validity of the application of 800MPa grade steel(HSA800) to composite column should be verified by experimental or analytical method. Thus, stub column tests for encased and filled composite members with HSA800 steel were conducted, and axial strength and the validity of design compressive strength equations in KBC were evaluated. The test results show that the equation of the compressive strength of encased composite column member in KBC should be modified in order to use HSA800 steel without any reduction of specified minimum yield strength. For this purpose, it is suggested that the interval of hoop should be narrowed and the effective concrete area should be used. The equation of the compressive strength of filled composite column member in KBC is applicable to filled composite column with HSA800 steel without any modification.

Conformational Study of Human Serum Albumin in Pre-denaturation Temperatures by Differential Scanning Calorimetry, Circular Dichroism and UV Spectroscopy

  • Thermal conformational changes of human serum albumin (HSA) in phosphate buffer, 10 mM at pH = 7 are investigated using differential scanning calorimetric (DSC), circular dichroism (CD) and UV spectroscopic methods. The results indicate that temperature increment from $25^{\circ}C$ to $55^{\circ}C$ induces reversible conformational changes in the structure of HSA. Conformational change of HSA are shown to be a three-step process. Interestingly, melting temperature of the last domain is equal to the maximum value of fever in pathological conditions, i.e. $42^{\circ}C$. These conformational alterations are accompanied by a mild alteration of secondary structures. Study of HSA-SDS (sodium dodecyl sulphate) interaction at $45^{\circ}C$ and $35^{\circ}C$ reveals that SDS affects the HSA structure at least in three steps: the first two steps result in more stabilization and compactness of HSA structure, while the last one induces the unfolding of HSA. Since HSA has a more affinity for SDS at $45^{\circ}C$ compared to $35^{\circ}C$, It is suggested that the net negative charge of HSA is decreased in fever, which results in the decrease of HSA-associated cations and plasma osmolarity, and consequently, heat removal via the increase in urine volume.

Production of Human Serum Albumin in Chloroplast-Transformed Tobacco Plants

  • Human serum albumin (HSA) is the most abundant protein in plasma and is the most often used intravenous protein in many human therapies. However, HSA is currently extracted only from plasma because commercially feasible recombinant expression systems are not available. This study attempted to develop an efficient system for recombinant HSA production by chloroplast transformation of tobacco. A HSA cDNA was isolated from a cDNA library constructed with human liver tissue. Chloroplast transformation vectors were constructed by introducing various regulatory elements to HSA regulatory sequences. Vectors were delivered by particle bombardment into leaf explants and chloroplast-transformed plants were subsequently regenerated into whole plants. Southern blot analysis confirmed that the HSA cDNA was incorporated between rps12 and orf70B of the chloroplast genome as designed. Western blot analysis revealed that hyper-expression and increasing the stability of HSA were achieved by modification of the regulatory sequences using the psbA5'UTRs in combination with elements of the 14 N-terminal amino acids of the GFP and the FLAG tag. However, only plants transformed with the vector containing all of these elements were able to accumulate HSA.
