예강지탕(銳降脂湯)이 고지혈증 SHR 및 항산화에 미치는 영향 (The Effects of Yegangji-tang on Hyperlipidemic SHR and Antioxidant Activity)

  • Objective : Yegangji-tang (YJT) is noted in China as effective medicine for hyperlipidemia. This experiment was performed to determine the effects of Yegangji-tang (銳降脂湯: YJT) on antioxidant activity and hyperlipidemia induced by hypercholesterolemic diet in mice. Methods : We observed three experimental groups: normal, control, and treatment. Rats were provided a normal diet in the normal group, and the other groups were provided a hypercholesterolemic diet to induce hyperlipidemia. After 2 weeks, YJT was treated to the treatment group. We observed the change of body and liver weight, change of serum total cholesterol, LDL cholesterol, HDL cholesterol and triglycerides, lipid peroxidation and antioxidant activity of liver tissue, cholesterol gene revelation control efficiency, and histologic change of liver and abdominal aorta. Results : Total cholesterol, LDL cholesterol and triglycerides decreased significantly with YJT, whereas HDL cholesterol increased significantly. Lipid peroxidation decreased and antioxidant activity of the liver increased significantly. Gene revelation of ACAT and HMG-CoA reductase in hepatic tissue decreased significantly with YJT. We also found that tissue defects of the liver and abdominal aorta were controlled by YJT on histologic study. Conclusion : These results suggest that YJT might be effective in treatment and prevention of hyperlipidemia.

양릉천(陽陵泉) 오미자(五味子)약침이 고지방식이로 유발된 고지혈증 흰쥐에 미치는 영향 (A Study on the Effect of Herbal-acupuncture with Schizandrae Fructus Solution on Hyperlipidemia in Rats Induced by High Fat Diet)

  • Objectives : The purpose of this study is to observe the effects of herbal acupuncture with Schizandrae Fructus solution at $GB_{34}$(Yangneungcheon) on hyperlipidemia in rats. Methods : In the present study, herbal acupuncture with Schizandrae Fructus solution was administered into hyperlipidemic mice. The author performed several experimental items to analyze the levels of various components and enzymes in serum, liver, as well as the histological changes of liver and aorta. Results : 1. In the Schizandrae Fructus herbal-acupuncture group, serum total cholesterol and Atherogenic index were significantly decreased, and the ratios of HDL to total cholesterol, and phospholipid to total cholesterol in serum were significantly increased as compared with those of the control group. 2. In the Schizandrae Fructus herbal-acupuncture group, HMG-CoA reductase activity was significantly decreased as compared with those of the control group. 3. Schizandrae Fructus herbal-acupuncture reduced deposit of lipid in the liver tissue. Conclusions : From the above results, it is suggested that Schizandrae Fructus herbal-acupuncture at $GB_{34}$ has a therapeutic effect on hyperlipidemia.

Hypocholesterolemic Effect of Amaranth Squalene (Amaranth esculantus) in Rats Fed a High Cholesterol Diet

  • In experiment 1, rats (n=6) fed diet containing 10 g/kg cholesterol for 4 wk (control) with either no amaranth (control), amaranth grain (300 g/kg, AG) or amaranth oil (90 g/kg, AO). Both the AG and AO groups had lower concentration of serum and hepatic cholesterol and triglyceride than the controls (p < 0.05). Fecal excretions of cholesterol and bile acid in AO group increased about 4 fold and 2 fold, respectively, while AG affected only bile acid excretion (p < 0.05). In experiment 2, rats (n=6) were fed the cholesterol diet for 4 wk and injected intraperitoneally with saline (control) or amaranth squalene (AS) for 7d. The hypolipidemic effect of AS was evident in both serum and liver. Fecal excretions of cholesterol and bile acid were greater (p < 0.05) in AS than control. HMG-CoA (3-hydroxy-3-methylglutaryl coenzyme A) reductase activity was reduced in AS group (11.6%, p=0.13). This study suggests that the cholesterol-lowering effect of AS is mediated by greater fecal elimination of steroids through interference with cholesterol absorption.

몇 가지 채소와 약용 식물을 이용하여 제조한 식물 복합 추출물의 영양학적 특성과 생리기능성 (Nutritional Characteristics and Physiological Functionalities of Polyplant Extracts from Some Vegetables and Medicinal Plants)

  • The nutritional characteristics and functionalities of the polyplant extracts(PPE) from the extraction of various plants were assessed for the development of a new functional food. The PPE contained 42.95 g of glucose, 22.61 g of fructose, and 12.48 g of sucrose per 100 g of PPE as free sugar, and its reducing sugar contents was measured at 480.6 mg per g of PPE. Among free amino acids, proline contents was the highest at 0.58 g per 100 g of PPE, and it contained only 12.65 mg of malic acid and 6.92 mg of tartaric acid per g of PPE as an organic acid. The antioxidant activity of PPE was the highest at 91.6%, whereas SOD-like activity and xanthin oxidase inhibitory activity, which are associated with anti-aging effects, were 21.6% and 9.3%, respectively. HMG-CoA reductase inhibitory activity was 10.0% and anti-dementia acetylcholinesterase inhibitory activity was quite low(5.1%).

The author performed several experimental items to analyze the levels of various components and enzymes in serum, urine and liver, as well as the histological change of liver and aorta. Results : 1. In the Cndii Rhizoma Herbal-acupuncture group, The levels of total cholesterol were significantly decreased, and the ratio of HDL to Total cholesterol, Phospholipid/total cholesterol were significantly increased as compared with those of the control group. 2. In the Cndii Rhizoma Herbal-acupuncture group, HMG-CoA reductase activity was significantly decreased than those of the control group NP(needle prick) group and saline group. 3. In the Cndii Rhizoma Herbal-acupuncture group, phathohistological change of liver was suppressed as compared with the control group. Conclusions : From the above results, it is suggested that Cndii Rhizoma Herbal-acupuncture at $GB_{34}$ has a therapeutic effect on hyperlipidemia.

고체 배양법에 의한 Lovastatin생산 (Production of Lovastatin in Solid Culture)

  • 공시균인 A .terreus mutant로부터 lovastatin의 생산성을 극대화할 수 있는 조건을 확립하고자 액체 배양법 및 고체 배양법 을 이용하여 HPLC를 통해 분석하였다. Lovastaton의 생산을 위해 액체 배양에서는 균사체 접종법인 No. 2배지를 사용한 전배양 및 본배양법이 포자접종법인 No. 1배지를 사용한 경우보다 lovastatin의 생산이 더 우수하였다. 고체 배양법에서는 분쇄 밀의 호화전분을 사용하여 28$^{\circ}C$, 15일간 배양시 lovastatin의 생산이 가장 우수하였으며 대량배양을 위해 배양용기를 달리하여 배양하였으며 cap을 부착하는 내열성 plastic bag을 이용하여 배양한 결과 두 방향으로 cap을 부착하여 통기량을 조절한 배양에서 lovastatin의 생산이 가장 우수하게 나타났다.

HK-1001 약침이 흰쥐의 고지혈증에 미치는 영향 (The Effect of HK-1001 Pharmacopuncture on Hyperlipidemia Induced Rats)

  • Objective&Methods The purpose of this study is to observe the effects of HK-1001 Pharmacopuncture at GB34(Yangleungchean) on hyperlipidemia in rats. The author performed several experimental items to analyze the levels of various components and enzymes in serum, urine and liver, as well as the histological changes of liver and aorta. Results 1. HK-1001 Pharmacopuncture infusion solution increased the cell viability rate, DPPH radical scavenging activity and HMG-CoA reductase inhibition rate in rat liver cells. 2. The levels of total cholesterol, free cholesterol, LDL-cholesterol, phospholipid in serum and AI(atherogenic index) were decreased, and the ratio of HDL to TCL(HDL/TCL) and the level of TG in serum were increased as compared with those of the control group. 3. In the HK-1001 group, serum GOT was significantly lower than those of the HG group and the saline group, and serum ALP was significantly higher than that of the HG group. 4. Hepatic GSH and catalase activities were significantly increased as compared with those of the saline group. Conclusion From the above results, it is suggested that HK-1001 Pharmacopuncture at GB34 has a therapeutic effect on hyperlipidemia.

식이지방의 종류 및 d-Limonene 투여가 간 발암과정에 미치는 영향 (Effects of Different Dietary Oil and d-Limonene on Histopathological and Biochemical Changes in Experimental Hepatocarcinogenesis)

  • The purpose of this study was to investigate the effcts of n-3, n-6 fatty arid and d-limonene on histopathological and biochemical changes in experimental rat hepatocarcinogenesis. To attain the above objectives, weanling Sprague-Dawley female rats were intraperitoneally injected twice with a dose of diethylnitrosamine(DEN, 50mg/kg body weight) and after 1 week 0.05% phenobarbital was provided with water. Sardine oil rich in n-3 fatty acids and corn oil rich in n-6 fatty acids were fed at 15% by weight and 5% d-limonene was added to the diet in each group. Ten weeks or 20 weeks after DEN treatment, rats were sacrifirced. The formation of glutathione S-transferase placental form positive(GST-P$\^$+/) foci was significantly decreased by the treatment of either sardine oil or d-limonene HMG-CoA reductase activity was not affected by dietary oils and d-limonene. Protein kinase C (PKC) activity was decreased by either sardine oil or d-limonene. Particularly d-limonene decreased the membrane PKC activity. Membrane Cholesterol/Phospholipid(Chol/PL) ratio was significantly decreased by d-limonene in sardine oil group. The data showed that GST-P$\^$+/ foci number was positively correlated with membrane PKC activity and serum cholesterol and negatively correlated with liver cholesterol level. These results suggest informations about the correlation between histopathological and biochemical changes such as cholesterol metabolism and PKC activity in experimental hepatocarcinogenesis and thereby can elucidate the possible mechanism related to the cancer inhibition.(Korean J Nutrition 33(1) : 23-32, 2000)

청혈단(淸血丹)의 임상적인 부작용에 대한 연구 (Clinical Assessment on the Safety of Chunghyul-dan (Qingwie-dan))

  • Background and Purpose : Chunghyul-dan has been widely used in the Department of Cardiovascular & Neurologic Diseases, Kyung Hee Oriental Medical Center to prevent stroke by lowering serum cholesterol level. Previous experimental and clinical studies revealed that Chunghyul-dan had therapeutic effects on hyperlipidemia by inhibiting HMG-CoA reductase and pancreatic lipase. It was also reported that Chunghyul-dan showed an anti-oxidation effect by scavenging free radicals and inhibiting nitric oxide synthesis. Therefore, we examined the safety of Chunghyul-dan on all subjects who had been treated with it. Methods : We performed a retrospective study by reviewing the medical records of those who had been administrated Chunghyul-dan at Kyung Hee Oriental Medical Center from February 8,2001 to December 31,2002. The subjects' general characteristics (gender, age, medical history, and present illness), recorded adverse effects, and the results of laboratory findings were obtained and analyzed to assess the clinical safety of Chunghyul-dan. Results : Six hundred fifty six subjects were treated with Chunghyul-dan. Clinical adverse effects appeared in 13 subjects, the major symptom being indigestion (8 subjects). The apparent frequency of adverse effects was much lower than that in previous reports on the safety of certain medications. On investigation of laboratory findings, we could not find any hepatic or renal toxicity. Conclusion : We suggest that our results contribute towards confirming the safety of Chunghyul-dan by offering clinical evidence.

구기자 가공품의 생리기능성 및 항고혈압 활성 검증 (Physiological Functionality of Gugija Products and an In Vivo Examination on Anti-hypertension Effects)

  • To develop a new Gugija(Lycium chinensis Mill) product having increased value, the physiological functionality of various commercial Gugija products were investigated. In addition, an in vivo study was performed using spontaneous hypertensive rats(SHR) to examine the anti-hypertension effects of products. The results showed that antioxidant activity was highest in the methanol extract of Gugija leaf pickle(97.7%), and anti-hypertensive angiotensin I -converting enzyme(ACE) inhibitory activity was 80.4% in Gugija doenjang(soybean paste) water extract. Anti-cholesterolemia HMG-CoA reductase inhibitory activity was highest in the methanol extract of Gugija rice cake(66.1 %). However, SOD-like activity was below 30% in most products; and fibrinolytic activity was not detected or was very weak. Ultimately, we selected Gugija tea and Gugija wine as superior anti-hypertensive Gugija products, and subsequent in vivo testing was performed using SHR, comparing the tea and wine to Gugija fruit. Among them, the Gugija fruit demonstrated the best anti-hypertension effects in SHR.
