천년초 뿌리의 항산화 및 염증 매게 물질 조절 효과 (Antioxidant and Inflammatory Mediators Regulation Effects of the Roots of Opuntia humifusa)

  • 본 연구에서는 천년초 분획물에 따른 항산화 및 항염증 효과를 확인하고자 80% MeOH로 추출하여, 추출물을 EtOAc Fr., n-BuOH Fr. 및 $H_2O$ Fr. 로 용매 분획하였다. 각 분획물에 대하여 총 폴리페놀 함량, DPPH 및 ABTS radical scavenging 방법을 이용한 항산화 활성 측정 결과, 모든 분획에서 항산화 활성을 확인하였고, 그 중 OHRE 분획은 가장 높은 총 폴리페놀 함량(179.3 mg/g)을 나타내었고, $IC_{50}$ 값이 DPPH는 $77.0{\pm}1.38{\mu}g/mL$, ABTS는 $26.3{\pm}2.02{\mu}g/mL$으로 가장 높은 항산화 활성을 나타내었다. 또한 항염증 효과는 PMACI로 HMC-1 cells에 염증을 유발시킨 뒤 천년초 뿌리 분획물을 처리하여 TNF-$\acute{\alpha}$를 비롯해 IL-$1{\beta}$, IL-6, IL-8등의 염증 매게 물질을 효과적으로 억제하는 것을 확인하였다. 이상의 결과로부터 천년초 뿌리는 높은 항산화 활성과 및 염증 매게 물질의 생성을 억제하는 우수한 항염증 효과가 있는 것으로 확인되었다.

곰취 열수 추출물의 항산화 및 항염증 활성 (Antioxidant and anti-inflammatory activities of hot water extracts of Ligularia fischeri)

  • 본 연구는 대표적인 산채지역으로 알려져 있는 영양군 곰취 열수추출물의 총 폴리페놀 함량과 DPPH 라디컬 소거능을 측정하였으며 사람 비만세포주인 HMC-1 세포를 이용하여 PMACI을 처리하여 비만세포를 활성화시켜 세포 생존률을 확인하였고, 염증성 사이토카인인 $TNF-{\alpha}$, IL-6, IL-8의 변화량을 측정하여 곰취의 기능성 식품 소재로서의 이용 가능성을 알아보고자 실험을 실시하였다. 영양군 곰취 열수 추출물의 추출수율은 원물 건조량 대비 $24.43{\pm}1.82%$(w/w), 총 폴리페놀 함량은 $198.07{\pm}0.24mg/g$으로 다른 곰취 추출물의 연구결과와 유사한 값을 나타내고, 영양군 곰취 열수 추출물의 DPPH radical 소거 활성을 측정한 결과, $IC_{50}$ 값이 $28.2{\pm}0.04{\mu}g/mL$로 양성대조군인 ascorbic acid의 $IC_{50}$$3.5{\pm}0.01{\mu}g/mL$보다 낮은 항산화활성을 보였으나 일반 천연물 유래 추출물보다 비교적 높은 항산화활성을 나타내었다. 영양군 곰취 열수 추출물을 HMC-1 세포에 처리했을 때 MTT assay법으로 분석한 결과, 세포 생존률에는 아무런 영향을 미치지 않고, $TNF-{\alpha}$, IL-6, IL-8 생성에 대한 억제효과는 유의성 있게 나타났다. 이상의 결과를 종합해 보면 항산화력 및 세포를 이용한 in vitro 실험을 실시한 결과 영양군 곰취 열수 추출물은 항산화력이 매우 우수할 뿐만 아니라 항염증효과도 뛰어나 향후 기능성 소재로서의 이용 가능성이 높은 것으로 판단되나 보다 다양하고 진보된 연구로 곰취의 염증성 질환 치료에 대한 정확한 기전이 설명되어야 할 것이다.

  • Objective: Bee Venom(BV) has been used for various kinds of inflammatory or painful conditions in Oriental Medicine clinics, and there publishes reports on its therapeutic effects and the probable mechanism of those therapeutic effects, where CDs and cytokines plays important role. This study investigated the influences of bee venom on the expressions of CDs and cytokines of HMC cell line Methods: In this study we analysed the expression profile of HMC cell line treated with BV of 10-2ug/ml in relation to that of HMC cell line treated with vehicle by way of CD/cytokine microarray hybridization with 342 genes on it. Results: There were no upregulated genes by more than 3 fold, while there showed some downregulated genes by less than 1/3 fold as follows: colony stimulating factor 2, CD122, IL-7, CD112, TNF-alpha, CD138, CD166, TGFbetaR2, CD42b, CD62L, CD111, interleukin 10 receptor alpha, colony stimulating factor 1(macrophage), CD38 antigen(p45), CD121a, CD33 antigen(gp67), colony stimulating factor 1 receptor, B cell linker protein (SLP65) mRNA, CD94, alanyl(membrane) aminopeptidase, immunoglobulin(CD79A) binding protein 1, CD205, CD241, CD207, CDw121b, integrin alpha L(CD11a), integrin beta 1(CD29), CD91, CD42b. Conclusions: Bee venom treatment induced downregulation of some CDs or cytokines including $TNF-{\alpha}$. IL-1R with its possible implication in an antiinflammatory action of BV. Further research on expression profile changes induced by BV treatment is expected.

벌사상자 Ethyl Acetate 분획물의 항염증활성연구 (Ethyl Acetate Fraction of Cnidium monnieri(L). Cussion Suppresses PAM plus A23187-induced Inflammation Reaction through Blockade of NF-κB and MAPK activation)

  • Cnidium monnieri (L). Cussion is used as a tonic agent in traditional oriental medicine. However, the molecular mechanism of mast cell-mediated anti-inflammatory modulation has not been fully understood. The aim of the present study was to demonstrate the effects of Cnidium monnieri (L). Cussion eathyl acetate fraction on the expression of pro-inflammatory cytokines, as well as to elucidate its mechanism of action in the human mast cell line (HMC-1). Cells were stimulated with phorbol 12-myristate 13-acetate (PMA) plus A23187 in the presence or absence of Cnidium monnieri (L). Cussion eathyl acetate fraction. Cnidium monnieri (L). Cussion eathyl acetate fraction significantly inhibited the PMA plus A23187-induction of inflammatory cytokines such as tumor necrosis factor (TNF)-$\alpha$, interleukin (IL)-6 and IL-8. Moreover, EtOAc fraction attenuated cyclooxygenase (COX)-2 expression. In activated HMC-1 cells, phosphorylation of extra-signal response kinase (ERK) 1/2 decreased after treatment with EtOAc fraction. Moreover EtOAc fraction inhibited PMA plus A23187-induced nuclear factor (NF)-${\kappa}B$ activation, $I{\kappa}B$ degradation. EtOAc fraction suppressed the expression of TNF-$\alpha$, IL-6, IL-8 through a decrease in the ERK 1/2, as well as activation of NF-${\kappa}B$. These results indicated that Cnidium monnieri (L). Cussion EtOAc fraction exerted a regulatory effect on inflammatory reactions mediated by mast cells.

  • Lonicera japonica Thunb.(Caprifoliaceae) has long been known as an anti-inflammatory. In the present study, the effect of water fraction of Lonicera japonica (LJ) on trypsin-induced mast cell activation was examined. HMC-1 cells were stimulated with trypsin (100 nM) in the presence or absence of LJ (10, 100, and 1000 $\mu$ g/mL). TNF-$\alpha$ and tryptase production were measured by enzyme-linked immunosorbent assay (ELISA) and reverse transcription-PCR. Extracellular signal-regulated kinase (ERK) phosphorylation was assessed by Western blot. Trypsin activity was measured by using Bz-DL-Arg-p-nitroanilide (BAPNA) as substrate. LJ (10, 100, and 1000 $\mu$g/mL) inhibited TNF-$\alpha$ secretion in a dose-dependent manner. LJ (10, 100, and 1000 $\mu$g/mL) also inhibited TNF-$\alpha$ and tryptase mRNA expression in trypsin-stimulated HMC-1. Furthermore, LJ inhibited trypsin-induced ERK phosphorylation. However, LJ did not affect the trypsin activity even 1000 $\mu$g/mL. These results indicate that LJ may inhibit trypsin-induced mast cell activation through the inhibition of ERK phosphorylation than the inhibition of trypsin activity.

Socioeconomic impact of traditional Korean medicine, Pyeongwee-San (KMP6) as an anti-allergic inflammatory drug

  • The prevalence of allergic disease has been increasing over the past few decades in the majority of Western industrialized nations. There are some socioeconomic disparities regarding allergic disease status and management. Pyeongwee-San (KMP6) is Korean medicine for the treatment of gastrointestinal tract disease. It is known that KMP6 has an improving effect on the spleen and stomach functions in traditional Korean medical theory. Here, we hypothesized that KMP6 could be used to regulate the inflammatory reaction. We show the molecular mechanisms of Pyeongwee-San (KMP6) on inflammatory reactions. A molecular docking simulation showed that hesperidin, component of KMP6, regulate the enzymatic activity by interaction in the active site of caspase-1. KMP6 control the activity of caspase-1 in activated human mast cell line (HMC-1 cells). KMP6 reduced the expression of receptor interacting protein (RIP)-2 in HMC-1 cells. Thymic stromal lymphopoietin protein production and mRNA expression were inhibited by KMP6. In the activated HMC-1 cells, KMP6 suppressed the activation of mitogen-ativated protein kinase and nuclear factor-kappaB. In addition, KMP6 significantly inhibited the expression of inflammatory cytokines. Our findings indicate that KMP6 may attenuate allergic reactions via the regulation of caspase-1/RIP-2 signaling pathway. These studies will help advance the social welfare system.

수종의 청열약물의 미백 및 항알러지 효과에 대한 실험적 연구 (Experimental studies on the Whitening and Anti-allergic effect of various Herbal medicines that clear heat)

  • Objective : In this study, herbal medicine(GJE, Gardenia jasminoides Ellis; HCT, Houttuynia cordata Thunb.; CIL, Chrysanthemum indicum Linne; PMS,Paeonia moutan Sims, P. subfruticosa Makino; APL, Agrimonia pilosa Ledebour) were screened for their inhibitory activities against Tyrosinase and PMA plus A23187-induced $TNF-{\alpha}$, IL-6, IL-8 productions in HMC-1 cells to reveal their skin -whitening and anti-allergic effect. Method : To investigate Tyrosinase inhibition we treated Mushroom Tyrosinase(Fluka, 93898) $10{\mu}{\ell}$ and 7.5mM Tyrosine (Sigma, T3754) $20{\mu}{\ell}$ with 80% ethanol medicine extracts. Then we observed 96well micro plate extinction at 490nm. In the next experiment, to investigate Anti-allergic effect we blended cultured Human Mast Cells(HMC-1) with medicine extracts. We treated the blended solution with Phorbol 12-myristate 13-acetate(PMA) and A23187, then observed $TNF-{\alpha}$, IL-6, IL-8 by ELISA (enzyme-linked immunosorbent assay) at 450nm. Results : In inhibiting Tyrosinase the results are as follows. 1. We observed 22% inhibition of Mushroom Tyrosinase at $500{\mu}g/m{\ell}$ concentration of GJE extracts. 2. We also could observe that the decreased Mushroom Tyrosinase activities in HCT, CIL extracts. In inhibiing $TNF-{\alpha}$, IL-6, IL-8 productions in HMC-1 cells the results are as follows. 1. Of the extracts examined, HCT, PMS, APL extracts showed over 50% inhibitions of Cytokines at $200{\mu}g/m{\ell}$ concentration. 2. In particular, APL extracts showed the best inhibitory effect on Cytokine productions in a dose-dependent manner. Conclusion : These results suggest that GJE extracts contributes to the anti melanin activities and represent a potential source of whitening agent. Thus these herbal medicines suggest novel drugs on anti-allergic effects.

비빔밥 재료의 항염증 및 항산화 효과 (Anti-inflammatory Effect and Antioxidative Activities of Ingredients used in Bibimbab)

  • 본 연구에서는 우리나라 전통 음식인 비빔밥의 우수성을 검증하고자 비빔밥에 들어가는 재료인 애호박, 도라지, 숙주, 속대기, 부추, 고사리의 조리 전과 후의 항염증 및 항산화 효과를 조사하였다. 인간유래 비만세포인 HMC-1 (human mast cell)에 70% ethanol로 추출한 조리 전, 후의 다양한 비빔밥 재료의 추출물을 1.0 mg/ml의 농도로 처리한 후, PMA와 A23187로 자극하여 염증반응을 유도하였다. MTT 분석에 의한 세포 생존율 결과, 모든 구에서 82% 이상의 세포 생존율을 보여 조리 전, 후의 비빔밥 재료 추출물에 의한 세포 독성은 나타나지 않았다. HMC-1 비빔밥 재료의 추출물을 0.01, 0.1, 1.0 mg/ml의 농도로 처리한 후, 자극제로 염증반응을 유도하였을 때 그 효과를 TNF-${\alpha}$, IL-6, IL-8의 분비량으로 알아보았다. 전체적으로 cytokine의 분비가 감소하였는데, 특히 1.0 mg/ml 농도에서 조리 후의 부추 추출물이 TNF-${\alpha}$, IL-6 분비량을 각각 90%, 93% 저해하였고, 무 추출물은 IL-8 분비량을 85% 저해하여 다양한 비빔밥 재료 중 가장 높은 저해율을 나타냈다. DPPH 분석에 의한 항산화 활성결과, 조리 전, 후의 부추 추출물에서 각각 67.50%, 73.65%로 재료 중 가장 높았으며 전반적으로, 조리함으로써 항산화 활성이 향상되었다. 결론적으로 조리 전보다 조리 후의 재료에서 세 가지의 cytokine을 유의성 있게 억제하였고, 항산화 활성 또한 높게 나타나 비빔밥의 우수성을 검증하는 기초자료로 활용이 가능하다고 사료된다.

  • Acanthoic acid (AA) is a pimaradiene diterpene isolated from the Korean medicinal plant, Acanthopanax koreanum (Araliaceae), which has been traditionally used as a tonic and sedative as well as in the treatment of rheumatism and diabetes in korea. Proteinase-activated receptor-2 (PAR-2) agonist trypsin plays a role in inflammation, and human leukemic mast cells (HMC-l) express PAR-2. In the present study, the effect of acanthoic acid on production of tumor necrosis factor-${\alpha}$ (TNF-${\alpha}$) and tryptase in trypsin-stimulated HMC-1 was examined. (omitted)

건칠이 비만세포 매개 염증반응에 미치는 영향 (The Effects of Lacca Sinica Exsiccata (LSE) on the Mast Cell-mediated Inflammatory Responses)

  • Objective : Lacca Sinic: Exsiccate (LSE) extracted from Rhus vemicitlus Stokes (RVS) has been used traditionally as a remedy for inflammation in Korea, China, and Japan. However, as yet there is no clear explanation of how LSE affects the production of inflammatory cytokines. This study was to determine the effects of LSE on the mast cell-mediated inflammatory responses. Method : We measured the amount of inflammatory cytokine production induced by the phorbol myristate acetate (PMA) plus calcium ionophore(A231S7) in the human mast cell line (HMC-l) incubated with various concentrations of Laces Sinica Exsiccate (LSE). The $TNF-\alpha$, IL-6 and IL-8 secreted protein levels were measured by the ELISA assay. The $TNF-\alpha$, IL-6 and IL-8 mRNA levels were measured by the RT-PCR analysis. Nuclear and cytoplasmic proteins were examined by Western blot analysis. The NF-${\kappa}B$ promoter activity was examined by a luciferase assay. Result : LSE inhibited the PMA + A231S7-induced $TNF-\alpha$, IL-6, and IL-8 expression and suppressed NF-${\kappa}B$ activation in the stimulated-HMC-1. In addition, LSE inhibited induction of NF-${\kappa}B$ promoter-mediated luciferase activity. Conclusion : In this study, we have found that LSE is an inhibitor of NF-${\kappa}B$ and cytokines on the mast cell-mediated inflammatory responses.
