고지방식이를 급여한 마우스의 간과 HepG2 세포에서 TJGB의 효과에 대한 연구 (Effect of TJGB on the liver of high-fat diet-fed mice and the viability of HepG2 cells)

  • Objectives: This study was performed to investigate the effect of TJGB on the liver of high-fat diet (HFD)-fed mice and the cell viability of HepG2 cells. Methods: After a week adaptation, 8-week-old C57BL/6N mice were fed with a 45% HFD or normal diet for 3 weeks. For the next 9 weeks, the mice were divided into 6 groups: normal diet group; HFD group; HFD plus orlistat group; HFD plus Ephedra sinica Stapf (ES) group; HFD plus low dose of TJGB group; HFD plus high dose of TJGB group. To estimate the effect of TJGB in the liver of HFD-fed mice, the protein expressions of phospho-acetyl-CoA carboxylase (p-ACC) and liver X Receptor (LXR) were determined by Western blot assay. The cell viability of ES and TJG was also evaluated in HepG2 cells. Results: The administration of TJGB had little effect on the protein expressions of p-ACC and LXR in the liver of HFD-fed mice. And the cytotoxicity was showed above 7.8 ㎍/mL in HepG2 cells. Conclusion: Further research is needed to evaluate the mechanism of TJGB on hepatic steatosis and cytotoxicity in HepG2 cells.

고지방식이를 제공한 마우스에서 여주와 아쉬아간다 발효추출물 조합의 항비만 효과 (Anti-obesity effect of the combination of fermented extracts from Momordica charanatia and Withania somnifera in mice fed a high-fat diet)

  • 본 연구는 고지방식이(HFD)를 제공한 비만동물모델에서 Momordica charanatia (MC)와 Withania somnifera (WS) 추출물의 혈청 콜레스테롤 및 내장지방 감소효과를 분석하였다. 유산균으로 발효한 MC와 WS 추출물의 조합(FMCWS) 및 비발효 추출물(MCWS)의 조합을 HFD로 유도된 비만마우스에 8주 동안 경구 투여하였다. 실험 기간 동안 체중, 식이섭취량, 총 콜레스테롤, 중성지방, HDL-콜레스테롤 수치를 분석하였다. 체중과 총 콜레스테롤 및 중성지방 수치는 정상대조(NC)군과 비교하여 HFD를 섭취한 HFD군에서 유의하게 증가했다. 그러나, 추출물의 투여로 인해 비만마우스의 체중증가가 감소되는 경향을 확인하였고 더불어 HDL-콜레스테롤의 증가와 함께 총 콜레스테롤 및 중성지방 수치를 유의적으로 감소시켰다. 복부지방 무게와 부고환 지방조직 내 지방세포의 크기는 NC군에 비해 HFD군에서 유의하게 증가하였다. 그러나, FMCWS와 MCWS를 각각 투여한 군은 HFD군에 비해 복부지방 무게와 부고환 지방세포의 크기가 유의하게 감소하였다. 또한, HFD군의 간 조직에서 관찰되는 거대 소포성 지방구의 침착이 이들 군에서는 현저하게 감소하였다. 이러한 결과는 MC와 WS 추출물의 조합이 체중 증가에 대한 감소 경향이 있을 뿐만 아니라 내장지방 및 혈청 지질 수준을 현저히 낮추는 데 강력한 시너지 효과가 있어 HFD 유발 비만마우스에서 항비만 활성을 향상시키는 효능이 있음을 시사한다.

  • Objectives : The researcher investigated the anti-obesity effect of Platycodi Radix (P), Platycodi Radix and Cyperi Rhizoma combination water extract (PC) in mice fed a high fat diet and focused on the analysis of local area adipose tissue.Methods : Male ICR mice were divided into four groups, which were fed either a normal AIN diet (N group), a high fat diet (HFD group), or a high fat diet and orally administration with a concentraion of 300 mg/kg body weight (P group or PC group) for eight weeks.Results : Compared to mice in the HFD group, mice in the P group or PC group showed significant reductions in weight gain and relative weight of total fat. Compared to mice in the HFD group, mice in the P group showed significant reductions in relative weight of liver. In blood biochemistry analysis, AST, ALT, triglyceride, total-cholesterol and low density lipoprotein(LDL)-cholesterol, AI levels of P group or PC group were significantly lower than those of the control group AI. But serum serum high density lipoprotein(HDL)-cholesterol levels from the P group or PC group were significantly higher than those of the HFD mice in serum. And serum adiponectin levels from the P group or PC group were significantly increased that those of the HFD mice. And adipocyte number in the fat tissue from the P group or PC group was significantly higher than those of the HFD mice.Conclusions : Platycodi Radix, Platycodi Radix-Cyperi Rhizoma have an anti-obesity effect in mice and the effect is mediated by inhibition of fat gain.

C57BL/6N 마우스에서 전립선암의 발병률 및 진행에 대한 고지방식이-유도 비만의 영향 (Effect of High-Fat Diet-induced Obesity on the Incidence and Progression of Prostate Cancer in C57BL/6N Mouse)

  • 고지방식이(High fat diet, HFD)에 의해 유발되는 비만은 정상적인 마우스에서는 연구되지 않았지만 여러 유전자변형마우스의 전립선암(Prostate cancer)에 대한 강력한 위험인자 및 예후인자로 검증되었다. HFD-유도 비만이 정상적인 마우스에서 전립선암의 발생 및 진행에 영향을 미칠 수 있는지 여부를 조사하기 위해, 16주 동안 60% HFD 식이를 급여한 비만 C57BL/6N 마우스에서 전립선의 무게 및 조직학적 구조 변화와 암관련 단백질 발현을 분석하였다. 첫째, HFD 식이를 급여한 C57BL/6N 마우스는 체중, 장기의 무게, 지방축적, 혈청지질 수치의 증가 등을 포함한 비만증상을 성공적으로 유도되었다. 전립선의 무게는 No그룹에 비해 HFD-유도 비만마우스에서 유의미하게 증가하였다. 전립선의 4가지 엽들 중 외측전립선(Dorsolateral prostate, DLP)과 정낭(Seminal vesicle, SV)의 무게는 유의적인 변화가 없었지만, 복부전립선(Ventral prostate, VP)과 전방전립선(Anterior prostate, AP)의 무게는 No그룹보다 HFD-유도 비만마우스에서 증가하였다. 또한, 전립선의 조직학적 구조에서 과형성(Hyperplasia) 및 비호지킨림프종(Non-hodgkin's lymphoma, NHL)의 발생률은 HFD-유도 비만마우스에서 유의미하게 증가하였으며, 같은 그룹에서 AP의 상피두께도 증가하였다. HFD-유도 비만마우스에서 AKT (Protein kinase B) 신호경로에서 주요 단백질의 인산화수준이 유의미하게 증가했다. 따라서 이러한 결과는 HFD-유도 비만은 C57BL/6N 마우스에서 전립선암의 발생과 진행을 촉진할 수 있음을 시사한다.

Anti-obesity Effect of Steamed Soybean and Fermented Steamed Soybean in High-fat Diet-induced Obese ICR Mice

  • This study was performed to investigate the ameliorating effects of steamed soybeans (SS) and fermented SS (FSS) on lipid metabolism in high-fat diet-induced obese mice. ICR mice were divided into four groups and given the following different diets: normal diet (ND), high-fat diet (HFD), HFD with 1% SS (HFD + SS), and HFD with 1% FSS (HFD + FSS). After 14 weeks, the body weight gain was higher in the HFD group compared with the ND group but lower in the HFD + FSS group compared with the HFD group. Plasma levels of triglyceride (TG), total cholesterol (TC), low density lipoprotein-cholesterol, aspartate aminotransferase (AST), and alanine aminotransferase (ALT) were significantly higher in the HFD group compared to the ND group, but lower in the HFD + SS and HFD + FSS groups compared with the HFD group. In addition, leptin concentration in plasma was lower in the groups fed HFD + SS and HFD + FSS compared with the HFD group. The accumulation of hepatic TG and TC was significantly inhibited in the HFD + SS and HFD + FSS groups. Furthermore, SS and FSS attenuated lipid peroxidation and nitric oxide formation in the liver induced by the high-fat diet. These results suggest that soybeans, especially FSS, may be useful in preventing obesity-induced abnormalities in lipid metabolism.

패 추출물이 고지방식이로 비만이 유도된 C57BL/6J 마우스의 항비만 효과에 미치는 영향 (Anti-obesity Effects of Ishige okamurae Extract in C57BL/6J mice Fed High-fat Diet)

  • 본 연구는 패 추출물의 항비만 효과를 조사하기 위해 C57BL/6J mice에 4주 동안 고지방 식이를 공급하여 비만을 유도한 후에, 6주간 패 추출물을 첨가한 고지방 사료를 섭취시키고 체중, 간, 신장 및 부고환 지방조직 무게, 혈액과 간의 지질 농도 및 혈중 렙틴 함량을 분석하였다. 실험 결과 HFD군에서는 고지방식이로 인하여 체중이 유의하게 증가하였으나, HFDIO군에서는 HFD군보다 체중의 증가가 유의하게 적었으며, 부고환 지방조직의 무게도 HFDIO군이 HFD군보다 유의하게 감소하였다. 고지방식이로 혈중 중성지방, 총 콜레스테롤, LDL콜레스테롤의 함량은 HFD군에서 가장 높게 나타났으나 HFDIO군에서는 유의하게 낮아졌으며 반면에 HDL콜레스테롤의 함량은 HFDIO군이 HFD군보다 유의하게 높았다. 이에 동맥경화지수와 심혈관질환위험도가 HFD군에 비해 HFDIO군에서 유의하게 낮아졌으며, 혈중 렙틴의 양도 HFDIO군이 유의하게 낮아지는 결과를 얻었다. 간 기능의 지표가 되는 ALT, AST, ${\gamma}$-GTP는 HFD군에서 유의하게 높은 값을 나타냈지만 모두 정상범위 내의 값을 나타내었고, 혈중 BUN과 혈중 Creatinine의 농도도 정상 범위 내의 값을 나타내었다. 이상의 실험 결과 고지방식이와 함께 급여한 패 추출물은 체중, 간 및 지방조직의 무게 감소와 더불어 혈장 및 간의 지질농도 개선과 혈장 렙틴 농도를 감소시키는 유용한 항비만 소재가 될 수 있을 것으로 사료된다. 앞으로 패의 어떠한 성분이 이러한 작용을 하는 지에 대한 더욱 체계적인 연구가 필요한 것으로 판단된다.

Improvement of blood lipid metabolism and obesity through the administration of mixed lactic acid bacteria including Lactobacillus plantarum K-1 in mice fed a high-fat diet

  • We investigated the effects of single and combined administrations of Lactobacillus species (L. plantarum, LP; L. gasseri, LG; L. casei, LC) on blood lipid metabolism and obesity in mice fed a high-fat diet (HFD). The mice were continuously supplemented with LP, LP/LG, or LP/LG/LC, along with HFD, for 12 weeks. The consumption of HFD led to significant increases in body weight, total cholesterol, and triglyceride levels compared to the normal control group. However, administration of LP, LP/LG, or LP/LG/LC to HFD-fed mice reduced body weight gain and showed a tendency to suppress the levels of total cholesterol, triglycerides, and LDL-cholesterol, while increasing HDL-cholesterol levels. The HFD group exhibited increased abdominal fat weight and larger adipocytes in the epididymal adipose tissue compared to the NC group. However, the administered probiotics led to a significant reduction in adipocyte size with decreasing tendency in abdominal fat weight compared with the HFD group. Additionally, the deposition of giant vesicular fat cells in the liver of the HFD group considerably decreased in the probiotic-administered group. Microbiome analysis revealed an imbalance in intestinal microbes in the HFD group, characterized by lower Bacteroidetes and higher Proteobacteria ratios. However, probiotic administration tended to restore the microbial distribution by controlling the abundance of Bacteroidetes and Proteobacteria, resulting in decreased Firmicutes/Bacteroidetes and Proteobacteria/Bacteroidetes ratios. These results suggest that single and combined administration of LP and other probiotics holds enormous potential in reducing obesity in HFD-fed mice as they regulate lipid metabolism, reduce adipocyte size, and restore the balance of intestinal microbes.

Cissus quadrangularis Extracts Decreases Body Fat Through Regulation of Fatty acid Synthesis in High-fat Diet-induced Obese Mice

  • The current study investigated the anti-obesity effect of Cissus quadrangularsis extracts (CQR-300) and its molecular action mechanism on obese mice induced high-fat diet (HFD). To induce the obesity, mice were fed a HFD for 6 weeks and then fed HFD only or HFD with CQR-300 at 50 and 200 mg/kg. Then, body weight gain and white adipose tissue weights were measured. We investigated the reduction in body fat and the regulation of fatty acid synthesis was measured by dual energy X-ray absorptiometry and real-time PCR with Western blot, respectively. In vitro study, CQR-300 inhibited pancreatic lipase activity. The CQR-300 treatment was significantly decreased the body weight gain and adipocytes size as well as white adipose tissues weights in HFD-induced obese mice. Furthermore, CQR-300 reduced the body fat and fat mass with regulating of adipose tissue hormones as leptin. Treatment with 50 mg/kg CQR-300 showed effectively lower expression levels of adipogenesis/lipogenesis related genes and proteins such as CCAAT/enhancer binding protein ${\alpha}$ ($C/EBP{\alpha}$), peroxisome proliferator-activated receptor ${\gamma}$ ($PPAR{\gamma}$), Sterol regulatory element binding protein-1c (SREBP-1c), and fatty acid synthase (FAS) in white adipose tissue (WAT) as compared with the HFD fed only mice. These results suggest that the CQR-300 has an anti-obesity effect via inhibition of lipase activity, decrease the body fat mass by regulating the adipogenesis and lipogenesis related genes and proteins in epididymal adipose tissue with evaluate body fat reduce in the HFD-induced obese mice.

마 추출물이 고지방식이로 유도된 C57BL/6J 마우스의 체중 및 혈청지질 수준에 미치는 영향 (Effect of Yam Extract on Body Weight Levels and Serum Lipid Profiles in C57BL/6J Mice Fed a High Fat Diet)

  • $Dioscorea$ $batatas$ Decne is a type of yam that eaten raw and used as a traditional oriental medicine in Asian countries. In this study, we evaluated the functional effects of yam water extracts on body weight levels and serum lipid concentrations in mice fed high fat diet. Mice were divided into four groups: normal diet control (ND), high fat diet control (HFD), HFD+yam extract 100 mg/kg (HFD-Y100), and HFD+yam extract 200 mg/kg (HFD-Y200). Yam extract was administrated orally to mice fed a high fat diet for 5 weeks. Treatment with yam extract significantly reduced body weight levels and energy efficiency in a dose-dependent manner in HFD-fed mice. Yam extract also attenuated serum levels of triglyceride, total cholesterol, and LDL-cholesterol, as well as organ weights of liver and abdominal adipose tissue in mice fed a high fat diet. Moreover, blood levels of glucose, insulin, and leptin significantly decreased in a dose-dependent manner upon administration of yam extract. Blood activities of GPT, GOT, and LDH were lower in the yam extract-treated groups compared to the HFD group. These results indicate that yam water extract may reduce elevated body weight and serum lipid concentrations in mice fed a high fat diet, suggesting its usefulness as a functional food for reducing body fat and hyperlipidemia.

Dietary carnosic acid suppresses hepatic steatosis formation via regulation of hepatic fatty acid metabolism in high-fat diet-fed mice

  • In this study, we examined the hepatic anti-steatosis activity of carnosic acid (CA), a phenolic compound of rosemary (Rosmarinus officinalis) leaves, as well as its possible mechanism of action, in a high-fat diet (HFD)-fed mice model. Mice were fed a HFD, or a HFD supplemented with 0.01% (w/w) CA or 0.02% (w/w) CA, for a period of 12 weeks, after which changes in body weight, blood lipid profiles, and fatty acid mechanism markers were evaluated. The 0.02% (w/w) CA diet resulted in a marked decline in steatosis grade, as well as in homeostasis model assessment of insulin resistance (HOMA-IR) index values, intraperitoneal glucose tolerance test (IGTT) results, body weight gain, liver weight, and blood lipid levels (P < 0.05). The expression level of hepatic lipogenic genes, such as sterol regulating element binding protein-1c (SREBP-1c), liver-fatty acid binding protein (L-FABP), stearoyl-CoA desaturase 1 (SCD1), and fatty acid synthase (FAS), was significantly lower in mice fed 0.01% (w/w) CA and 0.02% (w/w) CA diets than that in the HFD group; on the other hand, the expression level of ${\beta}$-oxidation-related genes, such as peroxisome proliferator-activated receptor ${\alpha}$ (PPAR-${\alpha}$), carnitine palmitoyltransferase 1 (CPT-1), and acyl-CoA oxidase (ACO), was higher in mice fed a 0.02% (w/w) CA diet, than that in the HFD group (P < 0.05). In addition, the hepatic content of palmitic acid (C16:0), palmitoleic acid (C16:1), and oleic acid (C18:1) was significantly lower in mice fed the 0.02% (w/w) CA diet than that in the HFD group (P < 0.05). These results suggest that orally administered CA suppressed HFD-induced hepatic steatosis and fatty liver-related metabolic disorders through decrease of de novo lipogenesis and fatty acid elongation and increase of fatty acid ${\beta}$-oxidation in mice.
