Efficiency of a hybrid solid digestion-denitrification column in suspended solid and nitrate removal from recirculating aquaculture system

  • This research focused on the solid and nitrate removal efficiency in a solid digestion-denitrification column. The 20 L up-flow column consisted of 18 L acrylic column with 2 L down-comer inlet tube located in the middle. In the first part, the wastewater with high suspended solids from the Tilapia fish tank was applied into the sedimentation unit at 5 variable flow rates i.e., 11.25, 25.71, 60, 105.88 and 360 L/h. The results indicated that the flow rate of 11.25 L/h (0.57 m/h) gave the highest solid removal efficiency of $72.72%{\pm}8.24%$. However, the total suspended solids removal was highest at 360 L/h (18.13 m/h). In the second part, methanol was added as an external organic carbon source for denitrification process in a hybrid column containing settled solids. The COD:N ratios of 0.5:1, 1:1, 2:1, 3:1, 4:1, 5:1 and 6:1 were investigated and compared with control without methanol addition. This experiment was operated at the HRT of 1 h with 450 L wastewater from recirculating aquaculture pond containing 100 mg-N/L sodium nitrate. The results indicated that the COD:N ratio of 3:1 gave the highest nitrate removal efficiency of $33.32%{\pm}21.18%$ with the denitrification rate of 5,102.88 mg-N/day.

Effect of Chlorella Extract on Quality Characteristics of Yoghurt (Chlorella 추출물 첨가가 요구르트의 품질 특성에 미치는 영향)

  • Yoghurt base was prepared from skim milk added with 0.25∼2.0%(w/v) of chlorella extract powder and fermented with lactic acid bacteria(Streptococcus thermophilus : Lactobacillus casei = 1:1) at 37$^{\circ}C$ for 24 hours. Quality characteristics of the yoghurts were evaluated in terms of acid production, number of viable cell, color, texture property, and sensory property during lactic acid fermentation. The addition of 0.25% chlorella extract powder stimulated the growth of lactic acid bacteria which showed the highest number of viable cell counts(l.46${\times}$10$\^$8/ CFU/mL) after 12 hours incubation, and also enhanced the acid production which was pH 4.33 after 12 hours incubation. The L values(brightness) of the yoghurts containing chlorella extract powder were lower than control group, whereas the b values(yellowness) were higher. The hardness and gumminess of the yoghurt added with 0.25% chlorella extract powder were higher than others. Sensory scores of the yoghurt added with 0.25% chlorella extract powder were significantly higher than other groups in color, chlorella odor, sweet taste, chlorella taste, mouth feel, aftertaste and overall acceptability.

Detection of Helicobacter Pylori in Saliva of Korean Infant (한국인 소아의 타액에서 Helicobacter pylori의 검출)

  • Helicobacter pylori(H. pylori) has been associated with the cause of peptic ulcer and gastric cancer. H. pylori infection occur mostly during childhood and increase by aging. In route of transmission, Oral cavity does important role. So we employed this study to elucidate route of transmission by detection of H. pylori in infant saliva. We investigated 20 infants aged below 10 years and 20 teens aged below 20 years as study group and 71 adults aged 20 and over years as control group. H. pylori DNA was isolated from 5(25%) infants aged below 10 years, 6(30%) teens and 17(23.9%) adults by nested polymerase chain reaction(n-PCR). There was no statistically significant difference(P>0.05). The obtained results suggest that H. pylori infection is relatively common in saliva of Korean infant and oral cavity may be reservoir of H. pylori.

Physical Properties of Rapeseed (I) (유채 종자의 물리적 특성(I))

  • Some physical properties of rapeseed such as geometric properties (linear dimensions, sphericity, seed volume, surface area) and gravimetric properties (the mass of one thousand seeds, bulk density) were analyzed at five levels of moisture content of 10.03, 14.91, 20.07, 25.06 and 30.12% (w.b.). The physical properties of rapeseed were evaluated as a function of seed moisture content. In the moisture range, when the moisture content increase, sphericity decreased from 0.946 to 0.927, and geometric mean diameter, seed volume and surface area increased from 2.17 to 2.31 mm, 5.58 to $6.88 \;mm^3$ and 14.76 to $16.77\;mm^2$ respectively. Mass of one thousand seeds increased from 5.04 to 6.46 g. Bulk density decreased from 579.3 to $549.2\;kg/m^3$ due to swelling of the seed.

Changes of Flavor Components in Chewing Gum during Storage by Gas Chromatography/Mass Spectrometry (GC/MS에 의한 저장중 츄잉껌 향기성분 변화연구)

  • The changes of amount in flavor components of chewing gum during storage of various relative humidity were studied by GC/MS. The volatile components in chewing gum were extracted by sample preperation and 15 volatile components were identified by NIST/NBS library searching. According to the period of storage, quantities of major volatile components extracted by tetrahydrofuran-methanol solutions were determined by using tetradecane as internal standard. Until 4 weeks, comparing with starting peak area ratio, the decrease of chewing gum flavor components were rapid at 33, 75 than 53 R.H(%). And after 16 weeks, amount of chewing gum flavor at all storage conditions similarly decreased to $45{\sim}49%$ of the initial amount.

Cosmetic Potency of Puerariae Radix in Dermal Fibroblasts

  • Interaction between epidermis and dermis plays an important role in wound healing and hair follicle formation. This study focused on investigating the potency of ethanol extract of Puerariae Radix (EPR) as cosmetic ingredient using human dermal fibroblasts (hDFn). Our results revealed that EPR suppressed collagenase activity dose-dependently. EPR inhibited activity of $5{\alpha}$-reductase I and II at the final concentration of $25{\mu}g/ml$ in hDFn cells. Also, EPR promoted the proliferation and the ERK activation of cells. ERK phosphorylation by EPR was blocked by specific inhibitor of ERK, PD98059. EPR-induced cell proliferation was blocked by PD98059. This means that EPR could promote the proliferation of hDFn cells via the activation ERK. Collectively, these results suggest that EPR may be used as a new cosmetic ingredient.

Anti-microbial and Anticariogenic Activity of Yam and Prunella Extract against Oral Microbes (구강병인균에 대한 마와 꿀풀추출물의 항균.항우식효과)

  • Yam, Prunella was stepwise extracted with hexane, chloroform, ethyl acetate, butanol, and water. Anti-microbial activity of each extract was investigated. Hexane extract was tested for anti-microbial effect on Streptocaccus mutans, one of causative factor of dental caries. Methanol extracts of 7 plants were investigated to anti-microbial effects on S. mutans KCTC 5316, P. gingivalis KCTC 5352, S. aureus KCTC 1927 by means of agar diffusion method. Methanol extract of Yam and Prunella revealed anti-microbial activity against S. mutans, P. gingivalis, and S. aureus. Also, hexane fraction of Yam revealed anti-microbial activity against S. mutans. In sequence of hexane, chloroform, ethylacetate, butanol fraction by Prunelia acted as potent anti-microbial agent on P. gingivalis. The measured MIC of hexane fraction of Yam and Prunella on S. mutans KCTC 5316 strain was 0.25 mg/ml and 0.5 mg/ml and the MIC of hexane fraction of Prunella on S. aureus was 0.5 mg/ml. The hexane fraction of Yam and Prunella suppressed viable ceil counts(VCC) of S. mutans, especially after 24 hrs. The Prunella hexane fraction suppressed VCC of S. aureus, after 12 and 24 hrs. Tested concentrations were 0.1, 0.25 and 0.5 mg/ml. the results were compared with control (0 mg/ml). The pH of S. mutans media and GTase activity were determined to evaluate the anticariogenic activity of Yam, Prunella hexane fraction. The pH were increased from 5.6 to 7.0-7.2 in concentration of 2.0 mg/ml. Yam hexane extraction revealed 35% inhibition to GTase activity and Punella inhibited 25% of GTase. These results suggest that the hexane extracts of Yam and prunella have Antibacterial activities against S. mutans, P. gingivalis, S. aureus and have preventive effect on dental caries.

  • Lichen planus is a common, chronic inflammatory disease of the skin and mucous membrane for which no precise causes have been confirmed. But it is often connected with infections. Helicobacter pylori(H. pylori) among various bacteria has been associated with the cause of gastritis, peptic ulcer and gastric cancer. Considering the similarities of histological features between gastric ulcer and oral ulcers, it is resonable to assume that H. pylori might also be involved in the development oral mucosal ulceration. So we employed this study to investigate the possible involvement of H. pylori in the aetiology of erosive oral lichen planus. We analyzed detection rate of H. pylori in saliva of patients with erosive oral lichen planus by nested PCR. As a result, it revealed a significant difference statistically by showing positivity in 16 to 21(76.2%) saliva samples of patients group and in 11 of 44(25%) saliva samples of control group(P>0.001). We were able to suppose that H. pylori in saliva can be related to cause of erosive oral lichen planus.

Case Study of N Deficiency Symptom of Strawberry in the Soil Applied with Sea Deposit Compost (바다부유물질로 만든 퇴비를 시용한 토양에서 발생한 딸기의 질소결핍증 해결 사례연구)

  • Nitrogen content in soil is a major factor for the crop growth. Ammonium nitrogen in soils is volatilized when soil pH is high. The growth and development problem of strawberry such as color of leaves turning into purple was found when soils were treated with the compost prepared from suspended solids of high pH from the sea. From in-situ analysis it was found that soil pH was 8.33 and nitrate, phosphorous, and potassium contents were relatively low. Nitric acid was added to adjust pH of irrigating water as 1.7, 1.9, and 2.3, then $KNO_3$ $0.25g\;L^{-1}$ and $KH_2PO_4$ $0.25g\;L^{-1}$ were added. It was resulted that soils with pH 1.7 produced the most developed strawberries. Strawberry was recovered by irrigation containing the same solution. From the results, the growth and development problem of the strawberry resulted from low nitrate absorption rate. It was concluded that the growth and development of strawberries were recovered by the reduced soil pH using nitric acid.

Cell Fusion Between Xylose Fermenting Yeast and Xylanase Secreting Yeast (Xylanase 분비효모와 Xylose 발효효모의 Protoplast Fusion)

  • To improve a new yeast strain capable of converting xylan to ethanol directly, we tried protoplast fusion between xylose fermenting yeast (Candida sp. X-6-41) and xylan assimilating yeast (Crypto-coccus sp. XB-33), finally selected the most promising two fusants (XFU-1 and XFU-2). As the optimum conditions for protoplast formation, the yeast cells were cultured to exponential phase in YPD and YPX containing 0.6M KCI, respectively, and then treated with zymolyase (0.25mg/$m\ell$), cellulase(4mg/$m\ell$) and 100mM 2-mercaptoethanol at pH 8 and 3$0^{\circ}C$. The protoplasts of parental auxotrophs were fused in the presence of 20mM CaCl$_2$and 40% polyethylene glycol(M.W.4000). The physiological and morphological characteristics of the fusants, such as assimilation of carbon sources, cell size, growth rate, xylanase activity and xylan fermentation ability were investigated. Xylanase activity of fusants that cultured in chemically minimal medium was higher than that of fusants that cultured in completed medium, because xylanase producing activity of xylose fermenting yeast(X-6-41) was inhibited by isoleucine.
