Improving the Nutritive Value of Full-Fat Rice Bran for Broiler Chickens Using a Lipase-Based Enzyme Preparation

  • Two experiments were conducted to test the hypothesis that a lipase-based enzyme preparation would increase the AME content of full-fat rice bran (FFRB) by increasing fat digestibility when fed to broiler chickens. Experiment 1 used FFRB from Australia and lasted for 35 days, while Experiment 2 used FFRB from Thailand and lasted for 14 days. Rice bran was substituted in a maize-soybean diet at levels of 90 g/kg (Experiment 1) and at 90 and 180 g/kg in Experiment 2. Total collections of excreta were used for determination of AME content and fat digestibility. In Experiment 1, the enzyme increased the AME content of FFRB between days 4-7, 18-21 and 32-35 by 6.1-16.1% (p>0.05), however this was not associated with improved fat digestibility. In Experiment 2, the enzyme enhanced the AME content of FFRB between days 4-7 (10.42 vs. 9.06, p=0.107) and 11-14 (11.94 vs. 9.93, p=0.041), but again, this was not caused by increased fat digestibility. Inclusion of 180 g/kg depressed the AME content of FFRB by 7.4-11.5% (p>0.10) in conjunction with decreased (p<0.05) fat digestibility between 0-14 days of age. Improvements in bird growth with the enzyme were seen in Experiment 2 but not in Experiment 1. Increases in AME content of FFRB per se were not caused by enhanced fat digestibility, suggesting that the side activities associated with the preparation must have acted singularly or in combination to improve AME content and bird performance. These data show that the response of FFRB to the lipase-based enzyme preparation was dependent upon the geographical origin of the rice bran and the level of FFRB substituted in the basal diet.

Effects of freezing-storage temperature on the shelf life of mackerel fish (냉동 저장 온도가 고등어의 저장성에 미치는 영향)

  • The purpose of this study was to evaluate changes in the quality characteristics of mackerel fish fillet under freezing storage conditions. The changes in product quality were determined by measuring pH, volatile basic nitrogen (VBN); thiobarbituric acid (TBA), lightness ($L^*$), redness ($a^*$), yellowness ($b^*$) values, shearing force, and microbiological evaluation (Total Plate Count, TPC) during 21 days of storage at -20, -5, and $-1^{\circ}C$, and their shelf-lives were established. TPC as an effective quality indicator was used to estimate the shelf-life via linear regression analysis. The TPC increased steadily with increasing storage periods at every temperature (p<0.05) and showed a significant relationship with temperature at $-1^{\circ}C$ (r=0.949), $-5^{\circ}C$ (r=0.919), and $-20^{\circ}C$ (r=0.905). Therefore, based on TPC, the shelf life of the mackerel fillet was noted to be 32 days when stored at $-1^{\circ}C$, more than 51 days when stored at $-5^{\circ}C$, and about 2 months when stored at $-20^{\circ}C$.

Development of Value-added Hot Sauce Products with Korean Chili Peppers (Capsicum annuum L.) (국내산 고추를 이용한 핫소스 개발)

  • The purposes of this study were to develop value-added sauce (chili hot sauce) products with Korean advanced chili peppers (Capsicum annuum L.), to determine physicochemical characteristics of hot sauces, and to conduct a sensory evaluation of developed hot sauces. American chili hot sauce products were collected from the American local favorites, and were analyzed based on their compositions. The developed Korean hot sauce contained tomato (29.2%), onion (18.2%), sugar (11.5%), vinegar (10.9%), Korean chili peppers (11.0%), herbs, plum extract, and oligosaccharide, and it showed a higher overall acceptability, compared to that of the hot sauce samples. Its physiochemical analysis showed: salt $5.46{\pm}0.21%$, total acidity $6.04{\pm}0.04%$, pH $3.32{\pm}0.25$, and $^{\circ}brix$ $58.50{\pm}0.10$. This result suggests the possibility for developing a processed chili pepper products.

  • The aims of these study were to diagnose early pregnancy and reproductive disorders by using progesterone concentration and ultrasonography. The measurement of blood progesterone (P$_4$) concentration was conducted to diagnose pregnancy and to detect corpus luteum (CL) or evaluate disorder of CLs. As a result, the incidence rates of reproductive disorders were as follows : SH and EED (41.9%), inacitve ovaries (32.6%), follicullar cyst (9.3%), PCL (7.0%), endometritis (4.7%), pyometra (2.3%) and luteal cyst (2.3%). 61 Cows having P$_4$concentration 1.0 ng/ml(at the insemination) were increased to 1.0 ng/ml $\geq$ 6day after insemination. 50 cows among 61 cows were diagnosed pregnant. 8 cows among 13 HanWoos having P$_4$concentration 1.0 ng/ml at the insemination and 1.0 ng/mnl 6 day after insemination had non-ovulatory estrus and the others had P$_4$concentration 1.0 ng/ml at the insemination and 1.0 ng/ml $\geq$ 6 day after insemination, which was the error of estrus detection. All 13 cows were diagnosed non-pregnant. 47 cows diagnosed pregnant after insemination of P$_4$concentration 3.0 ng/ml were examined by ultrasonography at 30 day post-insemination. As a result, 41 cows were diagnosed pregnant (87.2%) but 14 cows having P$_4$concentration 3.0 ng/ml at 21 day after insemination was diagnosed to non-pregnancy. Calving intervals by surveying 100 cows were as follows 11~12 months (20%), 12~13 months (36%), 13~14 months (19%), 14 months $\geq$ (25%), respectively. In conclusion, hormone and ultrasonography help to detect reproductive disorders exactly and diagnose early pregnancy. This study suggest that diagnosis of early pregnancy and reproductive disorder by blood P$_4$concentration and ultrasonography improve reproduction management of HanWoo.

Antioxidant and Antigenotoxic Activities of Extracts from Anglerfish (아귀 추출물의 항산화 및 항유전독성 활성)

  • Antioxidant activities of extracts from anglerfish (Lophiiu Zitulon) were evaluated. Each part of fresh (skin, flesh, stomach, and liver) or dried (skin and flesh) anglerfish was extracted by four different solvents (methanol, ethanol, acetone, and distilled water). Antioxidant activities of the extracts were determined by radical scavenging activity (RSA) and reducing power (RP). Relatively higher RSA and RP were found in methanol and water extract of fresh anglerfish liver. Antigenotoxic effect of the extracts, which was measured by Comet assay, was shown in most of the extracts except methanol, acetone and distilled water extracts of fresh stomach sample. These results indicated that antioxidant and antigenotoxic properties of extracts from angler fish were variable depending on parts, solvent, and/or physicochemical state. The appropriate extraction process could provide some valuable bioactive materials from anglerfish.

Quality Characteristics of Sponge Cake added with Citrus peel Powder (감귤분말을 첨가한 스펀지 케이크의 품질특성)

  • This study was conducted to evaluate the effect of Citrus peel powder on the quality characteristics of sponge cake. Citrus peel powder sponge cake was prepared with different ration of Citrus peel powder(0, 10, 20, 30, 40%). The specific gravity, baking loss rate and cake weight increases as the ratio of Citrus peel powder. The sponge cake's a, b level increased with the increasing level of Citrus peel powder 40% appeared to be the highest. In terms of textural characteristics, the samples containing 10, 20, and 40% Citrus peel powder had significantly higher hardness than the control samples. The substance's level of springiness, and cohesiveness decreased as the Citrus peel powder additive increased. In sensory evaluation, overall acceptability and taste of the cake was better than others when 10% Citrus peel powder was added. The results indicated that substituting 10% Citrus peel powder to sponge cake is optimal for quality and provides a product with reasonable high overall acceptability.

Evaluation of the antioxidant potential and enzyme activities in species of Rhus (옻나무류의 항산화력 및 항산화 효소 활성)

  • Comparison of the antioxidant potential and enzyme activities of the extracts of Rhus showed considerable differences. The antioxidative activities of Rhus javanica were the highest of three species tried (7 $\mu\textrm{g}$/$m\ell$). The highest activities showed in fraction No.3 of 12 fractions by Sephadex LH-20 column chromatography and the antioxidative activity showed, in purified extract of each stem,1.5 $\mu\textrm{g}$/$m\ell$ (in Rhus verniciflua), 1.9 $\mu\textrm{g}$/$m\ell$ (in Rhus javanica) and 2.3 $\mu\textrm{g}$/$m\ell$ (in Rhus tricocarpa) respectively. These were identificated as phenolic compounds which are well known antioxidant compounds such as 2-propenoic acid (Caffeic acid), Benzoic acid (Gallic acid), 7-hydroxy- 6methoxy-2H-1-Benzopyran-2-one (Scopoletin). POD activities of stem were higher than leaf. Especially, POD activity in stem of Rhus javanica was 193 times higher than leaf. Rhus tricocarpa, however, showed very low POD activity. SOD activities of stem were higher than that of leaf in Rhus javanica and Rhus verniciflua but in Rhus tricocarpa, the activity of leaf was 25 times higher than that of stem.

Effects of an in vitro vitamin D treatment on the inflammatory responses in visceral adipose tissue from Ldlr-/- mice

  • BACKGROUND/OBJECTIVES: Atherosclerosis is associated with increased inflammation in the visceral adipose tissue (VAT). Vitamin D has been reported to modulate the inflammatory responses of stromal vascular cells (SVCs) and adipocytes in adipose tissue, but the role of vitamin D in atherosclerosis biology is unclear. This study examined the effects of in vitro 1,25-dihydroxyvitamin D3 (1,25[OH]2D3) treatment on the inflammatory responses of SVCs and adipocytes from atherosclerotic mice. MATERIALS/METHODS: C57BL/6J (B6) mice were divided randomly into 2 groups and fed a 10% kcal fat control diet (control group, CON) or 41% kcal fat, 0.21% cholesterol (high fat + cholesterol, HFC) diet (obese group, OB), and B6.129S7-Ldlrtm1Her/J (Ldlr-/-) mice were fed a HFC diet (obese with atherosclerosis group, OBA) for 16 weeks. SVCs and adipocytes isolated from VAT were pre-incubated with 1,25(OH)2D3 for 24 h and stimulated with lipopolysaccarides for the next 24 h. Proinflammatory cytokine production by adipocytes and SVCs, the immune cell population in SVCs, and the expression of the genes involved in the inflammatory signaling pathway in SVCs were determined. RESULTS: The numbers of total macrophages and SVCs per mouse were higher in OB and OBA groups than the CON group. The in vitro 1,25(OH)2D3 treatment significantly reduced macrophages/SVCs (%) in the OBA group. Consistent with this change, the production of interleukin-6 and monocyte chemoattractant protein 1 (MCP-1) by SVCs from the OBA group was decreased by 1,25(OH)2D3 treatment. The 1,25(OH)2D3 treatment significantly reduced the toll-like receptor 4 and dual-specificity protein phosphatase 1 (also known as mitogen-activated protein kinase phosphatase 1) mRNA levels in SVCs and MCP-1 production by adipocytes from all 3 groups. CONCLUSIONS: These findings suggest that vitamin D can attribute to the inhibition of the inflammatory response in VAT from atherosclerotic mice by reducing proinflammatory cytokine production.

Assessment of natural radioactivity in soil and olive mill pomace utilizing nal (TI) gamma-ray spectrometry and low background alpha/beta counting system

  • The study conducted in the northwest region of Jordan aimed to assess the levels of natural radioactivity in soil and olive mill pomace (OMP) samples. The researchers used Nal (TI) gamma-ray spectrometry to measure the activity concentrations of 226Ra, 232Th, 40K, and 137Cs in the samples. The average activity concentrations of 226Ra, 232Th, 40K, and 137Cs in the soil samples were found to be 18.624 ± 5.82, 12.276 ± 5.728, 518.33 ± 212.57, and 0.140 ± 0.09 (Bq, kg-1), respectively. In the OMP samples, the average activity concentrations of 226Ra, 232Th, and 40K were 7.272 ± 4.386, 3.454 ± 1.503, and 169.997 ± 81.873 (Bq kg-1), respectively, and no 137Cs was detected. The study also investigated fundamental parameters associated with radon, specifically the radon emanation coefficient (RnEC) and radon mass exhalation rate (Ex). The RnEC values ranged from 0.621 to 0.78 (Bq kg-1), with an average value of 0.71 ± 0.06 (Bq kg-1). The estimated Ex from the soil samples ranged from 65.83 to 124.86 (mBq kg-1h-1), with an average value of 99.74 ± 21.73 (mBq kg-1h-1). Regarding radiological hazards, the study examined various parameters, including radium equivalent activity, external and internal hazard indices, gamma and alpha indices, absorbed gamma dose rate, and excess lifetime cancer risk. All of these assessed values were found to be below the worldwide recommended limits for radiological safety. Additionally, the study analyzed the concentrations of gross alpha and gross beta radioactivities in soil and OMP samples. The soil samples had an average gross alpha activity of 4.642 ± 1.04 (Bq kg-1) and an average gross beta activity of 48.13 ± 14.50 (Bq kg-1). The OMP samples showed an average gross alpha activity of 0.32 ± 0.27 (Bq kg-1) and an average gross beta activity of 59.19 ± 12.94 (Bq kg-1). Overall, the obtained results are crucial for evaluating the radiological risks associated with natural radioactivity in the northwest region of Jordan. The findings establish baseline data for comparison and reference for radioactivity levels in the environment.

Drug Resistance Patterns of the Bacterial Strains Isolated from Rural Areas and an Urban General Hospital (무의촌균주(無醫村菌株) 및 병원균주(病院菌株)의 항균제(抗菌劑) 내성(耐性) 양상(樣相)에 관(關)한 연구(硏究))

  • Besides the benefits of antimicrobial agents in the control of various infectious diseases, widespread and prolonged use of particular antimicrobial agents has brought about the increase of drug-resistant strains in a community and the profound changes in the pattern of infectious diseases. In Korea, there are some remote villages where no clinics and drug stores are available and the residents in those areas are assumed to have fewer chances to contact with antimicrobial agents. In the present study, the differences in susceptibilities to 14 antimicrobial agents between the isolates from rural areas(R) and Seoul National University Hospital(SNUH, H) were studied. The isolates and their numbers were Staphylococcus aureus, R;55, H;68), Enterococci(R;28, H;30), Escherichia coli(R;40, H;40), Enterobacter aerogenes(R;25, H;21) and Klebsiella pneunoniae(R;58, H;67). Minimal inhibitory concentrations(MIC's) of penicillin, ampicillin, carbenicillin, cephalexin, tetracycline, oxytetracyline, doxycycline, minocycline, gentamicin, kanamycin, streptomycin, erythromycin, troleandomycin and co-trimoxazole were determined by agar dilution method. I. Comparison of MIC's and resistant strain proportions between isolates from SNUH and rural areas. MIC's and/or resistant strain proportions of the isolates from SNUH were significantly higher than those of the isolates from rural areas in the cases of 1. S. aureus to doxycycline, streptomycin and kanamycin. 2. E. coli to penicillin, ampicillin, carbenicillin, tetracycline, oxytetracycline, doxycycline, minocycline, streptomycin, kanamycin, erythromycin and co-trimoxazole. 3. E. aerogences to carbenicillin, tetracycline, oxytetracycline, doxycycline, minocycline, streptomycin, kanamycin, genaamicin and co-trimoxazole. 4. K pneunoniae to penicillin, ampicillin, tetracycline, oxytetracycline, doxycycline, monocycline, streptomycin, kanamycine, gentamicin and co-trimoxazole. However, the mean MIC and resistant strain proportion of S. aureus to tetracycline were higher in isolates from rural areas than in those from SNUH and Enterococci showed no differences in susceptibilities to the antimicrobial agents between isolates from rural areas and from SNUH. Therefore, in general, differenes in susceptibility to these antimicrobial agents between the isolates from rural areas and SNUH were remarkably greater and broader in gram negative enteric bacteria. II. Multiple drug resistance pattern. Patterns and incidences of multiple drug resistance were studied with penicillin, ampicillin, tetracycline, cephalexin, gentamicin, streptomcin, kanamycin and co-trimoxazole in Enterococci, E. coli, E. aeroges and K. pneumoniae. There appeared significant differences in the incidence of multiply drug-resistant strains and multiple drug resistance patterns between the isolates from SNUH and rural areas in Enterococci, E. coli, E. aerogenes and K. pneumoniae. However, there was no difference in the incidence of multiply drug-resistant strains between isolates of S. aureus from SNUH and rural areas but the pattern of multiple resistance of the SNUH strains of S. aureus was diverse, while that of the rural strains was predominantly confined to penicillin-tetracycline combination. The incidence of multigly drug-resistant strains and diversity of their patterns were the highest in E. coli strains isolated from SNUH and there were no multiply drug resistant strrains in Enterococci and K. pneumoniae strains isolated from rural areas. The number of drug-resistance determinants was also different between the isolates from rural areas and SNUH. Most of the multiply drug-resistant strains of E. coli, E. aerogenes and K. pneumoniae isolated from SNUH were resistant to more than 3 kinds of antimicrobial agents, most frequently to ampicillin, tetracycline and streptomycin, while multiply drug-resistant strains from rural areas were resistant to 2 kinds of antimicrobial agents among ampicillin, tetracycline and streptomycin. With drug-resistant E. coli strains, resistance to tetracycline which was used most widely since 1951 was most frequently involved as a part of mutliple drug-resistance, followed by resistance to ampicillin and streptomycin. This strongly suggests that emergence of drug-restant strains in a community is directly dependent on the selective pressure exerted by the antimicrobial agent used. III. Cross resistance. Cross resistance of bacteria was studied among tetracycline penicillin, aminoglycoside and macrolide derivatives by analyzing correlation coefficients of sucseptibilities using the least square method. In this study, there were high correlations among the susceptibilities to related derivatives. It appears that the relatively low correlations in susceptibilities present in some cases are due to intrinsic resistance of E. aerogenes to penicillin, Enterococci to aminoglycoside and E. coli E. aerogenes and K. pneumoniae to macrolide derivatives.
