To improve a new yeast strain capable of converting xylan to ethanol directly, we tried protoplast fusion between xylose fermenting yeast (Candida sp. X-6-41) and xylan assimilating yeast (Crypto-coccus sp. XB-33), finally selected the most promising two fusants (XFU-1 and XFU-2). As the optimum conditions for protoplast formation, the yeast cells were cultured to exponential phase in YPD and YPX containing 0.6M KCI, respectively, and then treated with zymolyase (0.25mg/$m\ell$), cellulase(4mg/$m\ell$) and 100mM 2-mercaptoethanol at pH 8 and 3$0^{\circ}C$. The protoplasts of parental auxotrophs were fused in the presence of 20mM CaCl$_2$and 40% polyethylene glycol(M.W.4000). The physiological and morphological characteristics of the fusants, such as assimilation of carbon sources, cell size, growth rate, xylanase activity and xylan fermentation ability were investigated. Xylanase activity of fusants that cultured in chemically minimal medium was higher than that of fusants that cultured in completed medium, because xylanase producing activity of xylose fermenting yeast(X-6-41) was inhibited by isoleucine.

This paper is dealing with the fundamental properties of planting concrete, replacing the existing cover concrete on the roof of a building. This study is to find out the physical characteristics of the planting concrete and rearing characteristics of the grass throughout the modeling experimental materials. As the results of the experiment, the physical properties of planting concrete show the following results; when the paste to aggregate ratio is 0.2~ 0.4, voids volume : 30~17%, unit weight: 1,710~2,010kg/m3, compressive strength : 45~145kgf/$\textrm{cm}^2$, its pH is more than 11, but is reduced to the proper degree for planting after being neutralized. Kentucky bluegrass covered with planting concrete is grown well. The planting concrete used with blast furnace slag cement shows a better properties at the height, the width and the covering rate by 1.1cm, 0.5mm and 7%, respectively, than those used with ordinary portland cement. Also, the less the paste to aggregate ratio is, the better the plant grows. The orders of the effects of temperature control are as follows; the system of planting concrete with grass>the system of planting concrete without grass>the system of mixed soil>the existed roof system. In case, the planting concrete is placed to the roofs of buildings instead of normal concrete slab, and a number of favorable effect can be expected such as the improvement os environmental factors, the reduction of construction cost, the saving of energy and the reduction of environment load. The future research on the change of a variety of the aggregate conditions and the application of the practical structures should be made, and also the research of the endurance also be performed.
The objective of this study was to evaluate Solpro500 (a wheat hydrolysate containing a high level of glutamine) as a replacement for spray dried porcine plasma (SDPP) in diets fed to nursery pigs. One hundred and eight pigs (Dalland, $5.39{\pm}0.80$ kg BW) weaned at 21 days were assigned to one of three treatment groups for a 28 day feeding trial. The experimental diets were based on corn and soybean meal and were supplemented with either 8% SDPP, 4% SDPP plus 4% Solpro500 or 8% Solpro500. Each treatment was fed to six pens with six pigs per pen (4 barrows and 2 gilts). The experimental results indicated no significant difference (p>0.05) in daily gain, feed intake or feed efficiency for pigs fed the three experimental diets. However, the diarrhea index for pigs fed either 4% SDPP and 4% Solpro500 or 8% Solpro500 was lower (p<0.05) than that for pigs fed 8% SDPP. No differences (p>0.05) were found in the apparent fecal digestibility of dry matter, organic matter or crude protein between pigs fed the three diets. The intestinal morphology (villous height, villous width and crypt depth) was not affected by diet treatments (p>0.05). In conclusion, Solpro500 SDPP can replace SDPP without any negative effects on nursery pig performance.

We investigated traditional knowledge about ethnobotanical uses around national parks in the northern areas of Province Gyeongsangbuk-do, with the focus on the Sobeaksan National Park area. Interviews were carried out to 138 residents at 42 places from 7 counties and/or cities, and verified species and usage information were categorized by taxonomic groups, usage, and used parts. The ethnobotanical species of the regions consisted of a total of 277 taxa: 244 species, 3 subspecies, 28 varieties, and 2 forma in 213 genera of 79 families. The significant native plants included one Endangered Species (Paeonia obovata Max.) and four endemic species (Salix hallaisanensis H. Lev., Aconitum pseudolaeve Nakai, Paulownia coreana Uyeki, and Cirsium setidens (Dunn) Nakai, Sasa coreana Nakai). About 17 Approved Species for Delivering Overseas designated by Korea Ministry of Environment have been also traditionally valuable in the regions. The main usage of the plants were edible (185 taxa) and medicinal (175 taxa). Leaves were most commonly used parts (109 taxa), followed by stems (73 taxa), fruits (69 taxa), roots (67 taxa), and wholes (57 taxa). The traditional usage of plants were diverse as those useful parts. Leaves, fruits and seeds were used as edible and medicinal; sprouts (young shoots) were mainly used as edible; but roots and flowers were used as medicinal. The study does not only provides examples of traditional uses of native plants, but also facilitates sustainable managements and developments of ethnobotanical knowledge for the contemporary society.
Korea Aerospace Research Institute (KARI) is developing an electric-driven HALE UAV in order to secure system and operational technologies since 2010. Based on the 5 years of flight tests and design experiences of the previously developed electric-driven UAVs, KARI has designed EAV-3, a solar-powered HALE UAV. EAV-3 weighs 53 kg, the structure weight is 21 kg, and features a flexible wing of 19.5 m in span with the aspect ratio of 17.4. Designing the main wing and empennage of the EAV-3 the amount of the bending due to the flexible wing, 404 mm at 1-G flight condition based on T-800 composite material, and side wind effects due to low cruise speed, V_cr = 6 m/sec, are carefully considered. Also, unlike the general aircraft there is no center of gravity shift during the flight. Thus, the static margin cuts down to 28.4% and center of gravity moves back to 31% of the Mean Aerodynamic Chord (MAC) comparing to the previously developed scale-down HALE UAVs, EAV-2 and EAV-2H, to minimize a trim drag and enhance a performance of the EAV-3. The first flight of the EAV-3 has successfully conducted on the July 29, 2015 and the test flight above the altitude 14 km has efficiently achieved on the August 5, 2015 at the Goheung aviation center.
Randomly Amplified Polymorphic DNA-Polymerase Chain Reaction (RAPD-PCR) analysis was carried out with a battery of 11 random decamer primers to study band frequency (BF), genetic identity index (I) and mean average percentage difference (MAPD) between Bhadawari and Murrah breeds of buffalo. The primers OPA04 and BG15 resolved a band of 460 bp, which was present only in animals of Bhadawari breed. Whereas, the primers OPA14, BG27 and BG28 produced Murrah specific fragments of sizes 730 bp and 1,230 bp, respectively. The estimate of genetic identity index was highest (0.845) with the primer OPA01 and the lowest (0.479) with the primer BG27. The genetic identity index pooled over the primers was 0.596${\pm}$0.037 between these two breeds. The highest MAPD estimate (53.9) between the two breeds was obtained with the primer BG27 and the lowest (14.3) with the primer OPA01. It might be concluded that the genetic identity index between these two breeds calculated on the basis of BF showed moderate level of genetic identity with the primers employed. MAPD calculated on the basis of uncommon bands also demonstrated lower to medium level of genetic difference between Bhadawari and Murrah breeds of buffalo.
Four rumen-cannulated cows (330 kg average weight at 4 years) were used to evaluate the supplement of ensiled cassava tops (ECT) (variety KM 94, 39% DM) on rumen functions, thyroid hormones and liver enzymes. The treatments, arranged in a 4 $\times$4 Latin square design, were ECT at 0, 50, 100 and 150 g CP 100 kg$^{-1}$ body weight (BW), and a basal diet of urea-treated fresh rice straw (UFRS) ad libitum and 1.1 kg dry matter (DM) cassava root meal (CRM) in each 30 day study period. The results showed a continuous decrease in dry matter intake (DMI) of UFRS with increasing level of ECT supplement (p<0.001). The highest total DMI was observed for treatment ECT$_{150}$ (2.68 kg DM 100 kg$^{-1}$ BW day$^{-1}$) followed by treatments ECT$_{100}$, ECT$_{50}$ and ECT$_{0}$, with 2.47, 2.24 and 2.06 kg DM 100 kg$^{-1}$ BW⋅day$^{-1}$, respectively. Increasing levels of ECT supplement increased the concentration of total volatile fatty acids (p<0.05) and ammonia nitrogen (p<0.05) and resulted in a decrease in pH (p<0.05). Overall average plasma triiodothyronine and thyroxine concentrations were 0.80, 0.82, 0.85 and 0.69 ng ml$^{-1}$ (p>0.05), and 50.9, 49.5, 50.7 and 42.4 ng ml-1 (p>0.05) for treatments ECT$_{0}$, ECT$_{50}$, ECT$_{100}$ and ECT$_{150}$, respectively. There were non-significant differences in alanine aminotransferase and aspartate aminotransferase among treatments. It is concluded that ECT is a valuable protein-rich feed supplement to cattle, and the highest level of on average 2.48 kg DM ECT per cow and day (28% of total DMI) did not significantly affect thyroid gland hormones and liver enzymes in cows.
To investigate the role of polysaccharide from Acanthopanax senticosus (ASPS) in preventing lipopolysaccharide (LPS)-induced intestinal injury, 18 mice (at 5 wk of age) were assigned to three groups with 6 replicates of one mouse each. Mice were administrated by oral gavage with or without ASPS (300 mg/kg body weight) for 14 days and were injected with saline or LPS at 15 days. Intestinal samples were collected at 4 h post-challenge. The results showed that ASPS ameliorated LPS-induced deterioration of digestive ability of LPS-challenged mice, indicated by an increase in intestinal lactase activity (45%, p<0.05), and the intestinal morphology, as proved by improved villus height (20.84%, p<0.05) and villus height:crypt depth ratio (42%, p<0.05), and lower crypt depth in jejunum (15.55%, p<0.05), as well as enhanced intestinal tight junction proteins expression involving occludin-1 (71.43%, p<0.05). ASPS also prevented intestinal inflammation response, supported by decrease in intestinal inflammatory mediators including tumor necrosis factor ${\alpha}$ (22.28%, p<0.05) and heat shock protein (HSP70) (77.42%, p<0.05). In addition, intestinal mucus layers were also improved by ASPS, as indicated by the increase in number of goblet cells (24.89%, p<0.05) and intestinal trefoil peptide (17.75%, p<0.05). Finally, ASPS facilitated mRNA expression of epidermal growth factor (100%, p<0.05) and its receptor (200%, p<0.05) gene. These results indicate that ASPS can prevent intestinal mucosal barrier injury under inflammatory conditions, which may be associated with up-regulating gene mRNA expression of epidermal growth factor and its receptor.
Aerosol particles with different size-cuts ($PM_{10}$, $PM_{2.5}$, and $PM_{1.0}$) were collected at Gosan Superstation on Jeju Island from August 2007 to June 2008. Mean concentrations of $PM_{10}$, $PM_{2.5}$ and $PM_{1.0}$ were $29.28{\mu}gm^{-3}$, $17.83{\mu}gm^{-3}$, and $14.30{\mu}gm^{-3}$, respectively. Soluble ions comprised 45.7%, 53.9%, and 60.3% of the total mass of $PM_{10}$, $PM_{2.5}$, and $PM_{1.0}$, respectively. While sulfate was the most dominant species of fine mode ($PM_{1.0}$), nitrate was enriched in coarse mode ($PM_{1.0-10}$). When the concentrations of coarse mode particles were greatly increased, nitrate tended to be enhanced in coarse mode with high calcium but low sulfate concentrations. During the high $PM_{1.0}$ events, however, nitrate was increased with sulfate at fine mode. Particularly, nitrate concentrations were substantially enhanced during high particle episodes, leading high ratios of nitrate to sulfate in air under northwest wind during wintertime. On the other hand, the levels of nitrate were lower than those of sulfate at average particle concentrations. The backward air mass trajectories indicated that nitrate concentrations were elevated in air arriving Gosan passing through Santung peninsula or near South Korea.
The purpose of this study was to evaluate the immunogenicity and safety of Salmonella Typhi Vi capsular polysaccharide vaccine (Vi vaccine) in Korea. The immunogenicity of a single dose of Vi vaccine was evaluated in 157 subjects (75 children and 82 adults) before and at 1, 6, and 12 months after vaccination. Immunogenicity was measured with a passive hemagglutination assay (PHA), quantified as geometric mean titers (GMTs) and seroconversion rates. The safety of the vaccine was investigated by determining adverse reactions occurring within 4h, 3 days, and 1 month after injection. The seroconversion rate for children and adults 1 month after vaccination was 96.92% and 89.02%, respectively. In the case of children, the GMTs of Vi antibodies before vaccination were $5.87{\pm}1.34\;and\;142.59{\pm}2.39$ at one month after vaccination. For adults, the GMTs before and one month after vaccination were $5.58{\pm}1.28\;and\;58.56{\pm}3.67$, respectively. Vi antibodies persisted for as long as 6 and 12 months after vaccination. All adverse reactions in adults and children were minor and did not require treatment. The Vi CPS vaccine was safe and immunogenic in adults and children older than 5 years.
