Apparent Dominance of Regenerated Primary Production in the Yellow Sea

  • The Yellow Sea is known to be a very productive region in terms of fisheries. However, its trophic status seems to be highly variable, ranging from oligotrophic to eutrophic, based on new production (NP) values. The NP and regenerated production (RP) values estimated from $^{15}N$-labelled nitrate and ammonium uptake in spring (April 1996) and winter (February 1997) during this study ranged from 0.05 to 19.8 mg $N m^{-2} d^{-1}$ and from 0.1 to 22.8 mg $N m^{-2}d^{-1}$, respectively. Our measurements and earlier observations suggested that NP in the Yellow Sea varied over the four orders of magnitude (range 0.05-180.9 mg $N m^{-2} d^{-1}$) temporally and spatially, and that RP (range 0.1-507.5 mg $N m^{-2}d^{-l}$) based on ammonium predominated during most period of the year, except in winter when both productions were low. The significant nitrogen uptake by phytoplankton below the euphotic zone and episodic entrainment of phytoplankton from below the euphotic zone into the euphotic zone, and nitrite excretion and dissolved organic nitrogen release during nitrate uptake might explain the apparent dominance of RP in the Yellow Sea.

Dyeing Properties and Functionality of Methanol Extract from Juniperus chinensis Heartwood (향나무 심재 메탄올추출물의 염색성과 기능성)

  • In this study, we report dyeing properties and functionality of cotton and silk fabrics dyed with methanol extract from Juniperus chinensis heartwood. Non-aqueous dye of Juniperus chinensis heartwood extracted with methanol was expressed reddish color. It showed relatively good affinity to silk than cotton. Dye uptake of cotton and silk continued to increase according to the increase of the dye concentration. Cotton was not affected by the dyeing temperature and time, however, dye uptake of silk was increased as the dyeing time and temperature were increased. Dye uptake of cotton and silk were the highest at pH 8 and pH 6 respectively. Dyed fabrics showed various colors depending on mordant types and mordant concentration. Washing, light and perspiration fastness were not good. However, rubbing and dry cleaning fastness showed relatively good grade. Dyed fabrics of ultraviolet-cut ability and deodorant ability were improved. Also, dyed fabrics showed very good antimicrobial abilities of 99.9%.

Development of an Mg-Based Alloy with a Hydrogen-Storage Capacity over 6 wt% by Adding Graphene

  • Graphene (multilayer graphene) was chosen as an additive to improve the hydrogen uptake and release properties of magnesium (Mg). Five weight percent of graphene was added to pre-milled Mg by milling in hydrogen (reaction-involving milling). The hydrogen uptake and release properties of the graphene-added Mg were investigated. The activation of Mg-5graphene, which was prepared by adding 5 wt% graphene to Mg pre-milled for 24 h, was completed after the second cycle (cycle number, CN=2). Mg-5graphene had a high effective hydrogen-storage capacity (the quantity of hydrogen absorbed for 60 min) of 6.21 wt% at CN=3 at 593 K in 12 bar $H_2$. At CN=1, Mg-5graphene released 0.46 wt% hydrogen for 10 min and 4.99 wt% hydrogen for 60 min. Milling in hydrogen is believed to create defects (leading to facilitation of nucleation), produce cracks and clean surfaces (leading to increase in reactivity), and decrease particle size (leading to diminution of diffusion distances or increasing the flux of diffusing hydrogen atoms). The added graphene is believed to have helped the sample have higher hydrogen uptake and release rates, weakly but partly, by dispersing heat rapidly.

Development of Media for the Cultivation of Enterobacter amnigenus GG0461 and its Nitrate Uptake (Enterobacter amnigenus GG0461 균주의 생산을 위한 배지개발 및 질산이온 흡수)

  • To remove excess nitrate from the agricultural environments, Enterobacter amnigenus GG0461 has been isolated as a bacterial strain having high capability of nitrate uptake activity. This strain was able to remove nitrate more than 3,000 ppm (50 mM) in the Pseudomonas agar F (PAF) medium. Therefore, it could be a candidate strain for a nitrate scavenger in the various contaminated environments, such as agricultural soils, livestock sewage, and industrial wastewater. In order to develop medium for the large-scale production of the strain GG0461, each component of PAF medium was replaced with the corresponding commercial product and the optimal conditions for bacterial growth and nitrate uptake activity were measured. Glycerol was replaced with the commercially available product and the nitrogen source was substituted with commercial tryptone, yeast extract, soybean meal, and fermented fish extract. Bacterial growth and nitrate uptake activity were maximal in the media containing 2% tryptone, followed by yeast extract, soybean meal, and fermented fish extract. The pH of the growth medium containing 2% tryptone was decreased by the bacterial nitrate uptake, suggesting that the nitrate uptake is mediated by a nitrate/proton antiporter. This result shows that the medium containing commercial tryptone was good enough for the physiological activity of the strain GG0461. Each component of PAF medium was successfully replaced with the corresponding commercial product except peptone. In conclusion, the composition of medium for the cultivation of the strain GG0461 was determined as 2% tryptone, 1% glycerol, plus required salts according to the composition of PAF medium.

Effects of Barley Straw Application on Soil Physico-Chemical Properties and Nutrient Uptake in Rice Paddy Field of Double Cropping (벼 2모작 논에서 보릿짚 시용이 토양이화학성 및 양분흡수에 미치는 영향)

  • This experiment was carried out to investigate the effects of amount of nitrogen application with the barley straw application on the changes in soil physical and chemical properties. nutrient uptake and percentage recovery of chemical fertilizer N in the rice plant from 1997 to 1998. The soil physical properties, such as bulk density, hardness, porosity and gaseous phase were improved by barley straw application. There was also improvement or increment in the soil chemical properties, such as pH, organic matter, T-C. T-N, available $SiO_2$, exchangeable K and cation exchange capacity, but decrease in available $P_2O_5$. The $Fe^{+{+}}$ content in soil after barley straw application was high from tillering stage to panicle forming stage, but becoming lowered toward the heading stage, while $Mn^{+{+}}$ content was increased. N uptake with barley straw application was increased in the N $126kg\;ha^{-1}$ plot, but decreased in the N $141kg\;ha^{-1}$ plot. The uptake of fertilized N was continued longer in barley straw application than none-application plot. Percentage recovery of chemical fertilizer N in rice straw was around 1% at tillering stage, but was highly increasing till maximum tillering stage, while the recovery was generally low in barley straw application. Meanwhile, fertilizer P uptake in barley straw application was high, but potassium uptake was low at all different levels of N application.

Effect of Various Pathological Conditions on Nitric Oxide Level and L-Citrulline Uptake in Motor Neuron-Like (NSC-34) Cell Lines

  • Amyotrophic lateral sclerosis (ALS) is a fatal motor neuron disorder that causes progressive paralysis. L-Citrulline is a nonessential neutral amino acid produced by L-arginine via nitric oxide synthase (NOS). According to previous studies, the pathogenesis of ALS entails glutamate toxicity, oxidative stress, protein misfolding, and neurofilament disruption. In addition, L-citrulline prevents neuronal cell death in brain ischemia; therefore, we investigated the change in the transport of L-citrulline under various pathological conditions in a cell line model of ALS. We examined the uptake of [14C]L-citrulline in wild-type (hSOD1wt/WT) and mutant NSC-34/ SOD1G93A (MT) cell lines. The cell viability was determined via MTT assay. A transport study was performed to determine the uptake of [14C]L-citrulline. Quantitative real-time polymerase chain reaction (qRT-PCR) analysis was performed to determine the expression levels of rat large neutral amino acid transported 1 (rLAT1) in ALS cell lines. Nitric oxide (NO) assay was performed using Griess reagent. L-Citrulline had a restorative effect on glutamate induced cell death, and increased [14C]L-citrulline uptake and mRNA levels of the large neutral amino acid transporter (LAT1) in the glutamate-treated ALS disease model (MT). NO levels increased significantly when MT cells were pretreated with glutamate for 24 h and restored by co-treatment with L-citrulline. Co-treatment of MT cells with L-arginine, an NO donor, increased NO levels. NSC-34 cells exposed to high glucose conditions showed a significant increase in [14C]L-citrulline uptake and LAT1 mRNA expression levels, which were restored to normal levels upon co-treatment with unlabeled L-citrulline. In contrast, exposure of the MT cell line to tumor necrosis factor alpha, lipopolysaccharides, and hypertonic condition decreased the uptake significantly which was restored to the normal level by co-treating with unlabeled L-citrulline. L-Citrulline can restore NO levels and cellular uptake in ALS-affected cells with glutamate cytotoxicity, pro-inflammatory cytokines, or other pathological states, suggesting that L-citrulline supplementation in ALS may play a key role in providing neuroprotection.

The Effect of Irrigation Control and the Application of Soil Ameliorators on Cadmium Uptake in Paddy Rice (수도의 카드뮴 흡수이행에 대한 물관리 및 개량제 시용효과)

  • In order to identify the effect of cadmium uptake by soil ameliorators in rice plants, compost, lime, silicate, and phosphate were treated. Plants were grown with irrigation water containing $0.01mg\;l^{-1}$ cadmium and treated with two irrigation systems, intermittent irrigation and continuous submersion. Soil pH was higher at the panicle formation stage compared to other growing stages. However, Eh value was decreased as growing stage was proceeded. Eh values measured at the continuous submersion plots were lower than those at the intermittent irrigation plots. Cadmium contents in the both leaves and roots of rice plants grown at the continuous submersion plots were lower than intermittent irrigation plots in the middle of growing season. Among the continuous submersion plots, cadmium uptake was highly reduced at the both silicate and lime treated plot compared to other treatments. In the harvest season, cadmium contents in the both leaves and brown rice were higher at the intermittent irrigation plots than the continuous submersion plots. Cadmium uptake was highly reduced at the compost and lime mixture plot compared to other treatments among the continuous submersion plots. Soil pH was negatively correlated with the cadmium contents of the both shoot and brown rice while Eh was positively correlated with those. The cadmium content of shoot was positively correlated with that of brown rice in the harvest season.

Mathematical Models of Respiration Rate of Cucumber Plants under Varying Environmental Conditions (환경 변화에 따른 오이의 호흡속도에 관한 수리적 모형)

  • This study was conducted to develop mathematical model of respiration rate of cucumber plant under varying environmental conditions. 8.55% of gross photosynthesis of individual cucumber plant was used as respiration. Growth respiration coefficient was estimated as 0.0935. Maintenance respiration rate was estimated as 0.00158g CH$_2$g$^{-1}$ .h$^{-1}$ at 24$^{\circ}C$ of air temperature and it increased exponentially as air temperature became higher. Respiration rate decreased poportionally as content of storage carbohydrate became lower. Ion uptake respiration rate of roots was estimated as 0.6648g C$H_2O$.(gN)$^{-1}$ .

  • The purpose of this study was to analyze the reorganization of periodontal ligament after collagenase treatment with autoradiography. The author compared the collagenase-treated experimental group and no-treated experimental group with control group. Fourty eight Sprague-Dawley rats were divided into nine groups, including normal control and immediate group. Closed coil springs were used between the upper incisors and the first molars with 100 grams. Collagenase and $^3H-proline$ were adminstered and the samples were sacrificed and sectioned. After being dipped into the NTB-3 emulsion the samples were analyzed with light microscope under H/E stain. Data were analyzed by t-test and ANOVA. The results were as follows: 1) Generally collagenase-treated groups got more $^3H-proline$ uptake than no-treated groups. 2) Compared with normal control group, collagenase-treated group had the same $^3H-proline$ uptake in amount at 21th day. 3) Among cemento-enamel junction, middle, apex areas, cementa-enamel junction area of collagenase-treated group arrived at normal control level earlier than no-treated group. 4) Cemento-enamel junction area had the most $^3H-proline$ incorporation amount in no-treated group, but apex area had the most in collagenase group.

The Relationship between F-18-FDG Uptake, Hexokinase Activity and Glut-1 Expression in Various Human Cancer Cell Lines (다양한 사람 종양세포주에서 F-18-FDG의 섭취와 Hexokinase 활성 및 Glut-1 발현과의 상관관계)

  • Purpose: To investigate the mechanisms related to F-18-FDG uptake by tumors, F-18-FDG accumulation was compared with glucose transporter-1 (Glut-1) expression and hexokinase activity in various human cancer cell lines. Materials and Methods: Human colon cancer (SNU-C2A, SNU-C4, SNU-C5), hepatocellular carcinoma (SNU-387, SNU-423, SNU-449), lung cancer (NCI-H522, NCI-H358, NCI-H1299), uterine cervical cancer (HeLa, HeLa 229, HeLa S3) and brain tumor (A172, Hs 683) cell lines were used. After 24 hr incubation of $5{\times}10^5$ cells, 37 kBq F-18-FDG was added and the uptake by cells at 10 min was measured using a gamma counter. Hexokinase activity was measured by continuous spectrophotometric rate determination. To measure mitochondrial hexokinase activity, mitochondrial fraction was separated by a high speed centrifuge. Immunohistochemical staining of Glut-1 was performed, and graded as 0, 1, 2, or 3 according to expression. Results: There was difference among F-18-FDG uptake, total and mitochondrial hexokinase activity, and Glut-1 expression with different cancer cell lines. The correlations of F-18-FDG with total hexokinase and mitochondrial hexokinase activity were low (r=0.27 and 0.26, respectively). Glut-1 expression showed a good correlation with F-18-FDG uptake (p=0.81, p=0.0015). Previously, we reported no correlation of F-18-FDG uptake with hexokinase activity in colon cancer cell lines. Thus, when colon cancer cells were excluded, F-18-FDG uptake showed higher correlation with total hexokinase and mitochondrial hexokinase activity (r=0.81, p=0.0027 and r=0.81, p=0.0049, respectively). Conclusion: Both Glut-1 expression and hexokinase activity were contributing factors related to F-18-FDG accumulation in human cancer cell lines. The relative contribution of Glut-1 expression and hexokinase activity, however, was different among different cancer cell types.
