A sensitive, selective and rapid method has been developed for the determination ${\mu}$g/L level of vanadium ion based on the rapid reaction of vanadium(V) with 2-(2-quinolylazo)-5-diethylaminophenol (QADEAP) and the solid phase extraction of the colored chelate with $C_{18}$ cartridge. The QADEAP reacts with V(V) in the presence of citric acid-sodium hydroxide buffer solution (pH = 3.5) and cetyl trimethylammonium bromide (CTMAB) medium to form a violet chelate of a molar ratio 1 : 2 (V(V) to QADEAP). This chelate was enriched by solid phase extraction with $C_{18}$cartridge and the enrichment factor of 50 was obtained by elution of the chelates from the cartridge with ethanol. The molar absorptivity of the chelate is $1.28 {\times}10^5L\;mol^{-1}cm^{-1}$ at 590 nm in the measured solution. Beer's law is obeyed in the range of 0.01-0.6 ${\mu}$g/mL. The detection limit is 0.04 ${\mu}$g/L in the original samples. This method was applied to the determination of vanadium(V) in water and biological samples with good results.
Acid dissociation constants and chelate stability constants of 8-hydroxyquinoline-5-sulfonic acid have been determined for divalent metal ions. Co (Ⅱ), Ni (Ⅱ) and Zn (Ⅱ) by means of the Calvin-Bjerrum technique at the various temperatures. The standard free energy changes for the reactions at $20^{\circ}$, $30^{\circ}$, $40^{\circ}$and $50^{\circ}C$ were calculated, and the corresponding values of ${\Delta}H^{\circ}$ and ${\Delta}S^{\circ}$ applying over this temperature range are reported. The results are interpreted on the basis of current theories of metal chelate formation in aqueous solution.
Resolution of enantiomers of DNS-amino acids has been achieved by a reversed phase liquid chromatography with an addition of a copper(Ⅱ) complex of N-benzyl-L-hydroxyproline to the mobile phase. N-Benzyl-L-hydroxyproline was prepared and used as a chiral ligand of copper(Ⅱ) chelate for the optical resolution. The pH and the concentration of copper(Ⅱ) chelate, organic solvent, and buffer agent in the mobile phase all affect the optical resolutions of dansyl amino acids. The elution orders between D and L-DNS-amino acids were different depending on the structure of the side chain of the amino acids. The retention mechanism for the chiral separation of the dansyl amino acids can be illustrated by the equilibrium of ligand exchange and by hydrophobic interaction with $C_{18}$ stationary phase. The chiral separation can be illustrated with cis and trans effect of the ligand exchange reaction.
Separation of optical isomers of dansyl amino acids by a reversed phase liquid chromatography has been accomplished by adding a copper (II) chelate of N-benzyl-L-proline to the mobile phase. The pH, the eluent composition and the concentration of copper (II) chelate all affect the optical separations. The elution orders between D and L DNS-amino acids were consistant except dansyl phenylalanine showing that D forms of DNS-amino acids elute earlier than L forms. These behaviors are different from the results obtained by the use of copper (II) proline. The retention mechanism for the optical separation of the dansyl amino acids can be explained by the equilibrium of liqand exchange and by hydrophobic interaction.
An experiment was conducted to determine the effects of supernormal level of copper (Cu) from different supplementary sources on the performance, cholesterol level in plasma and breast muscle, and accumulation of fat and Cu in broilers. In a 5 wk feeding trial, two hundred forty hatched male broiler chickens were assigned to four dietary treatments: control diet containing 10 mg/kg supplementary Cu, control diet plus 250 mg/kg Cu from $CuSO_4$ ($CuSO_4-250$), control diet plus 125 mg/kg Cu from Cu-methionine chelate (Cu-Met-125), and control diet plus 250 mg/kg Cu from Cu-methionine chelate (Cu-Met-250). Weight gain in Cu-Met-125 treatment and Cu-Met-250 treatment were not different, but they were significantly (p<0.05) greater than that in $CuSO_4-250$ treatment. Plasma total cholesterol and reduced glutathione (GSH) in blood were significantly reduced by supplementation of $CuSO_4-250$, but were not significantly affected by Cu-Met supplementations. Plasma HDL cholesterol, plasma triglycerides and breast muscle cholesterol were not significantly affected by Cu supplementation. $CuSO_4-250$ improved metabolizability of crude fat, which resulted in low abdominal fat pad weight. Cu from Cu-Met was better absorbed and accumulated more in the breast muscle and lesser in the liver compared with Cu from $CuSO_4$.
0.1~1 M의 철킬레이트 화합물을 이용한 화학흡수반응에 의한 바이오가스내 황화수소제거를 위한 실험이 수행되었다. 철킬레이트 화합물을 이용한 황화수소제거는 철킬레이트의 최적산화반응을 통해 이루어진다. 바이오가스에 존재하는 황화수소는 킬레이트농도 및 pH 등의 공정조건에 따라 효과적으로 제거될 뿐만 아니라 철킬레이트 산화반응에 의해 황화수소내 존재하는 황성분을 생성시킨다. Fe-EDTA의 농도가 증가하면 철킬레이트 화합물의 착물이 안정되어 황생성의 전환이 증가하였다. 또한 철킬레이트화합물의 안정도는 pH에 따라 변하는 중요한 인자이고 pH 9에서 최적반응을 나타냈다.
A layer experiment was conducted to determine the effects of supplementary methionine chelates (Cu, Zn and Mn), individual or in combination, on laying performance, eggshell quality, gizzard erosion, and IgG level of serum for 8 weeks. Five hundred 96-wk-old force molted ISA Brown layers were assigned to five dietary treatments. Basal diet was formulated to meet or exceed the nutrients requirements listed in NRC (1994). Five experimental diets were control, Zn-methionine chelate (Zn-Met) supplemented, Cumethionine chelate (Cu-Met) supplemented, Zn-Mn-methionine chelate (Zn-Mn-Met) supplemented and Zn-Mn-Cu-Met supplemented diet. Each treated diet was supplemented with respective mineral(s) at the level of 100 ppm in the form of methionine chelate. Egg production was increased by Cu-Met supplementation but decreased by Zn-Met supplementation. Egg weight was significantly (p<0.05) lower in Cu-Met treatment than those of the control and Zn-Met treatment. Specific gravity of eggs and eggshell strength were highest and soft egg production was lowest in Cu-Met treatment. Gizzard erosion index was significantly increased by supplementation of Cu-Met, Zn-Mn-Met or Zn-Mn-Cu-Met. Zinc content in liver significantly increased by Zn-Met, but not by Zn-Mn-Cu-Met treatment. In conclusion, 100 ppm Cu in Cu-Met chelate improved laying performance and eggshell quality but also increased gizzard erosion index. Supplementation of Zn-Met or its combination with other mineral chelates had no beneficial effects on laying performance and eggshell quality.
A broiler experiment was conducted to compare the effects of supplementary iron sources and levels on the iron content of broiler meat. Two hundred and fifty hatched Ross broiler chickens were randomly assigned to 5 dietary treatments. Each treatment had 5 replicates of 10 birds (5 males and 5 females). Birds were housed in raised floor batteries and fed traditional broiler diets ad libitum for 5 weeks. Dietary treatments were as follows: Control, Fe-Met 100 (100 ppm iron as Fe-methionine), Fe-Met 200, $FeSO_4$ 100 (100 ppm iron as $FeSO_4{\cdot}7H_2O$) and $FeSO_4\;200$. There were no significant differences among treatments in parameters related to production performance. Liver contained approximately 10 times more iron than the leg muscle which contained approximately 3 times more iron than either breast muscle or wing muscle. Significant differences in iron content in the broiler meat were observed. In the breast meat, Fe-Met treatments were significantly (p<0.05) higher than other treatments in iron content. In the leg meat, Fe-Met treatments and $FeSO_4\;200$ treatment were significantly higher than the control in iron content. In the wing muscle, Fe-Met 200 treatment was significantly higher than other treatments in iron content. Iron content in the liver was significantly influenced by source and supplementation level of iron. Fe-Met treatments were higher than $FeSO_4$ treatments and 200 ppm treatments were higher than 100 ppm treatments in iron content in the liver. It is concluded that iron-methionine chelate is more efficient than iron sulfate and 200 ppm iron supplementation as Fe-Met is recommended for maximum iron enrichment in broiler meat.
Lactic acid와 키토산을 Cu(II) 이온과 반응시켜 합성한 착물을 사용하여 유기인 유사 독성물질인 DFP (Diisopropyl fluorophosphate) 분해반응에 적용하였다. Cu(II)-lactic acid 착물의 경우 homogeneous 상태에서 분해반응 반감기가 37. 1 min으로 분해성능이 우수하였다. 1 kDa 저분자량 수용성 키토산으로 합성한 Cu(II)-LMWS chitosan 착물은 결정화 후에는 용해도가 낮아 heterogeneous 한 상태에서 분해반응이 진행되었으며 그 반감기는 32.9 h이었다. 이 결과는 기존에 연구된 18 kDa 키토산 Cu(II)착물의 분해반응속도보다 약 16배 정도 증가된 것이다. Cu(II)-LMWS chitosan 착물을 결정화하지 않고 homogeneous한 상태로 진행한 분해반응에서는 반감기가 8.75 h로 용해도에 따라 약 4배의 차이를 확인할 수 있었다.
본 연구는 무기태 크롬($ClCl_3$)와 유기태화 크롬인 Cr-methionine chelate(크라민®)을 첨가하였을 때 in vitro 조건에서 반추위내 발효성상과 반추미생물체 내 Cr과 Methionine을 분석하여 크라민®의 by-pass 여부를 간접적으로 증명하고자 실시하였다. In vitro 소화시험에 이용한 기초영양소는 Jar 당 반추미생물 기초 영양소로 시중에 유통되고 있는 배합사료 7g(DM), 볏짚 2g(DM) 및 Corn silage 2g(DM)을 동일하게 배합하였으며, 시험구로는 대조구(control), $ClCl_3$를 1000ppb 첨가한 T1구 및 Cr 농도가 1000ppb이 되도록 크라민®을 첨가한 T2구를 두었으며, 처리 당 5반복으로 시험을 수행하였다. T2의 pH는 모든 배양시간에서 대조구 및 T1구에 비하여 낮은 경향을 보였으며, 암모니아 농도는 배양 6시간 전까지는 대조구와 T1구에 비하여 T2구가 높은 경향을 보였으나, 배양 6시간 이후에는 모든 처리구가 일정하게 낮게 유지되었다. 총 휘발성지방산 농도는 모든 처리구가 배양시간이 경과함에 다라 지속적으로 증가하였으며, 대조구에 비하여 T1구와 T2구의 농도가 유의적으로 낮았다. In vitro 배양 12시간 동안 미생물체 건물 회수율은 T1구가 가장 낮은 반면에 T2구가 가장 효율적으로 미생물을 증가시켰다. 반추미생물체 내 Cr 농도는 대조구와 T2구간에는 차이가 없었으나(P>0.05), T1구는 유의적(P>0.05)으로 높게 나타났다. 반추미생물체 내 methionine 및 cyctine 농도는 대조구와 T2구간에는 차이(P>0.05)가 없었으나, T1구는 비교적 낮은 경향을 보였다. 본 시험의 결과를 종합해 보면, 크라민®의 첨가에 따른 in vitro 배양액내 pH 및 암모니아 농도를 포함한 발효특성에 대한 부의 영향은 없는 것으로 판단되며, 오히려 미생물체 단백질의 합성에 이용되어 암모니아의 생성량과 총미생물 건물 회수량을 증가시킬 수 있는 것으로 판단된다. 또한 크라민®은 반추 미생물에 의해 상당히 제한적으로 분해되기 때문에 반추위를 회피해서 소장으로 by-pass 되어 이용된 것으로 판단되었다.
