In general, shape optimization design of the flow system has done to obtain the effects, which are required in the engineering fields. Most of these designs are accomplished by empirical or numerical analysis. But, in empirical analysis case, it is difficult to obtain an optimal shape in the feasible design region. And, in numerical method case, it usually needs many design parameters, because of the required object-function. In this paper, we present a newly numerical analysis, the growth-strain method having only one design parameter. That optimizes a shape by distributing a design parameter such as dissipation energy to be uniformed in the flow system. Also, we apply this shape design process to the three-flow systems, and then we identify that the resulting shape approaches the known optimal shape in the numerical values. Consequently, we confirm that the proposed method is very efficient and practical in the shape optimization of the flow system.
Soil-borne infectious disease including Pythium aphanidermatum and Rhizoctonia solani causes severe damage to plants, such as cucumber. This soil-borne infectious disease was not controlled effectively by chemical pesticide. Since these diseases spread through the soil, chemical agents are usually ineffective. Instead, biological control, including antagonistic microbe can be used as a preferred control method. An efficient method was developed to select an antagonistic strain to be used as a biological control agent strain. In this new method, surface tension reduction potential of an isolate was included in the ‘decision factor’ in addition to the other factors, such as growth rate, and pathogen inhibition rate. Considering these 3 decision factors by a statistical method, an isolate from soil was selected and was identified as Bacillus sp. GB16. In the pot test, this strain showed the best performance among the isolated strains. The lowest disease incidence rate and fastest seed growth was observed when Bacillus sp. GB16 was used. Therefore this strain was considered as plant growth promoting rhizobacteria (PGPR). The action of surface tension reducing component was deduced as the enhancement of wetting, spreading, and residing of antagonistic strain in the rhizosphere. This result showed that new selection method was significantly effective in selecting the best antagonistic strain for biological control of soil-borne infectious plant pathogen. The antifungal substances against P. aphanidermatum and R. solani were partially purified from the culture filtrates of Bacillus sp. GB16. In this study, lipopeptide possessing antifungal activity was isolated from Bacillus sp. GB16 cultures by various purification procedures and was identified as a surfactin-like lipopeptide based on the Fourier transform infrared spectroscopy (FT-IR), nuclear magnetic resonance (NMR), high performance liquid chromatography mass spectroscopy (HPLC-MS), and quadrupole time-of-flight (Q-TOF) ESI-MS/MS data. The lipopeptide, named GB16-BS, completely inhibited the growth of Pythium aphanidermatum, Rhizoctonia solani, Penicillium sp., and Botrytis cineria at concentrations of 10 and 50 mg/L, respectively. A novel method to prevent the foaming and to provide oxygen was developed. During the production of surface active agent, such as lipopeptide (surfactin), large amount of foam was produced by aeration. This resulted in the carryover of cells to the outside of the fermentor, which leads to the significant loss of cells. Instead of using cell-toxic antifoaming agents, low amount of hydrogen peroxide was added. Catalase produced by cells converted hydrogen peroxide into oxygen and water. Also addition of corn oil as an oxygen vector as well as antifoaming agent was attempted. In addition, Ca-stearate, a metal soap, was added to enhance the antifoam activity of com oil. These methods could prevent the foaming significantly and maintained high dissolved oxygen in spite of lower aeration and agitation. Using these methods, high cell density, could be achieved with increased lipopeptide productivity. In conclusion to produce an effective biological control agent for soil-borne infectious disease, following strategies were attempted i) effective screening of antagonist by including surface tension as an important decision factor ii) identification of antifungal compound produced from the isolated strain iii) novel oxygenation by $H_2O_2-catalase$ with vegetable oil for antifungal lipopeptide production.
본 연구의 주목적은 아스팔트 혼합물의 피로균열에 대한 예측모델을 개발하는 것이다. 아스팔트 혼합물의 피로균열 시험을 위하여 응력제어 간접인장피로 시험이 수행되었다. 피로균열에 대한 예측모델 개발을 위하여 내적손상비 증가 개념이 도입되었다. 내적손상비증가 개념에서는 방출에너지 개념을 주로 사용하였으며 기준인장변형율 및 변형율 추이 요소 등이 추가로 사용되었다. 피로시험에서 나타난 방출에너지의 원인은 아스팔트 콘크리트 시료 내부의 손상증가와 재료 자체가 갖고 있는 고유의 점탄성 특성에 기인하는 것으로 판단된다. 방출에너지는 하중재하 횟수가 증가함에 따라 점차 증가함을 보였다.
This work describes a method for determining material parameters included in recrystallization and grain growth models of metallic materials. The focus is on the recrystallization and grain growth models of Ni-Fe based superalloy, Alloy 718. High temperature compression test data at different strain, strain rate and temperature conditions were chosen to determine the material parameters of the model. The critical strain and dynamically recrystallized grain size and fraction at various process conditions were generated from the microstructural analysis and strain-stress relationships of the compression tests. Also, isothermal heat treatments were utilized to fit the material constants included in the grain growth model. Verification of the determined material parameters is carried out by comparing the average grain size data obtained from other compression tests of the Alloy 718 specimens with the initial grain size of $59.5{\mu}m$.
The isolation, morphological study and growth characteristics of the algae were investigated from Lake Chuam. The isolated algae were applied the Agal Growth Potential test. The method of isolation and purification of the algae were used to Agar plating(AP), nutrient enrichment(NE), dilution(DI) and micro capillary technique(MC). Total isolated algae were 21 species. They were composed of Cyanophyceae, Dinophyceae, Bacillariophyceae, Euglenophyceae and Chlorophyceae. The numbers of algal strain by isolation technique were highest in dilution(21 species), and those of the rests were showed in order of NE > MC > AP. The sizes of isolated Selenastrum and Scenedesmus were $1.8\pm 1.4 \mu m$, $3.3\pm 0.9 \mu m$ in diameter and $6.4\pm 2.3 \mu m$, $13.6\pm 1.9 \mu m$ in length respectively. The morphology of isolated algae and NIES-collection strain was very similar each other, but the size was smaller isolated algae than that of NIES-collection. The optimum culture condition of isolated Selenastrum and Scenedesmus was about 30$\circ$C(25$\circ$C-35$\circ$C) in temperature and the maximum growth was appeared between 7,000 lux and 8,000 lux in the light intensity. The comparison of $\mu$(specific growth rate) on the concentration of nutrients such as nitrate and phosphate, isolated Selenastrum was appeared maximum it at 1.0 mg $NO_3-N/l$ but NIES-collection strain was showed 95% of maximum it at same nitrate concentration. Maximum g of isolated algae and NIES-collection strain in Scenedesmus onto nitrate concentration were very similar with the result of selenastrum. The specific growth rates of isolated algae and NIES-collection strain on the gradient concentration of phosphate were showed 0.72/day and 0.70/day at 0.02 mg $PO_4-P/l$ in Selenastrum but those of Scenedesmus were appeared 0.61/day and 0.57/day at same concentration $PO_4-P$.
Tensile, low cycle fatigue, and fatigue crack growth rate tests were conducted at RT and $300^{\circ}C$ for type 304 stainless steel. Tensile was tested under displacement control and low cycle fatigue was tested under strain control. Fatigue crack growth rate test was conducted under load control and crack was measured by DCPD method. Yield strength and elongation decreased at $300^{\circ}C$. Dynamic strain aging was not detected at $300^{\circ}C$. Low cycle fatigue life increased but fatigue strength decreased at $300^{\circ}C$. Fatigue crack growth rate increased at $300^{\circ}C$. Dislocation structures were mixed with cell and planar and did not change with temperature. Grain size did not change but plastic strain increased at $300^{\circ}C$. Strain induced martensite after low cycle fatigue test increased at RT but decreased at $300^{\circ}C$. It was concluded that the increase of low cycle fatigue life at $300^{\circ}C$ was due to the decrease of strain induced martensite at which crack was initiated.
Forming Limit Diagram(FLD) is a representative tool for evaluating formability of sheet metals. This paper presents a methodology to determine the FLD using Finite Element Method. For predicting the forming limits numerically. Previous methods such as using the thickness strain or the ductile fracture criterion are limited at plane strain domain. These results suggest that behavior of the void growth in sheet metals is different from real one. In contrast to previous methods, a more exact model which takes void growth into account is used. This result agrees with the experimental result qualitatively.
PANrP 탄소섬유의 고온연신에서의 변형 거동을 검토하기 위해서, 일정 하중하에서 내부저항 가열법에 의해 $1200^{\circ}C$부터 $2200^{\circ}C$까지 열처리 온도를 변화시키며 섬유의 strain 변화를 측정하였다. 부하된 응력이 증가함에 따라서 strain 변화가 크게 발생하며 또한 열처리 온도 약 $1700^{\circ}C$를 경계로 해서 그 온도 이상과 이하 사이에는 큰 차이를 보였다. 각 응력에 있어서의 strain 속도 변화로부터 얻어진 활성화 에너지는 70 MPa 및 322 MPa에 대하여 각각 67.46 및 52.27 kJ/mol로써, 큰 응력의 경우가 작은 응력을 부하하는 것 보다 섬유구조 발달에 있어서 더욱 효과적이라는 것을 알았다.
A double-stranded RNA (dsRNA) mycovirus was detected in malformed fruiting bodies of Pleurotus ostreatus strain ASI2792, one of bottle cultivated commercial strains of the edible oyster mushroom. The partial RNA-dependent RNA polymerase (RdRp) gene of the P. ostreatus ASI2792 mycovirus (PoV-ASI2792) was cloned, and a cDNA sequences alignment revealed that the sequence was identical to the RdRp gene of a known PoSV found in the P. ostreatus strain. To investigate the symptoms of PoV-ASI2792 infection by comparing the isogenic virus-free P. ostreatus strains with a virus-infected strain, isogenic virus-cured P. ostreatus strains were obtained by the mycelial fragmentation method for virus curing. The absence of virus was verified with gel electrophoresis after dsRNA-specific virus purification and Northern blot analysis using a partial RdRp cDNA of PoV-ASI2792. The growth rate and mycelial dry weight of virus-infected P. ostreatus strain with PoV-ASI2792 mycovirus were compared to those of three virus-free isogenic strains on 10 different media. The virus-cured strains showed distinctly higher mycelial growth rates and dry weights on all kinds of experimental culture media, with at least a 2.2-fold higher mycelial growth rate on mushroom complete media (MCM) and Hamada media, and a 2.7-fold higher mycelial dry weight on MCM and yeast-malt-glucose agar media than those of the virus-infected strain. These results suggest that the infection of PoV mycovirus has a deleterious effect on the vegetative growth of P. ostreatus.
Tin Oxide(SnO2) has been widely investigated as a transparent conducting oxide (TCO) and can be used in optoelectronic devices such as solar cell and flat-panel displays. It would be applicable to fabricating the wide bandgap semiconductor because of its bandgap of 3.6 eV. In addition, SnO2 is commonly used as gas sensors. To fabricate high quality epitaxial SnO2 thin films, a powder sputtering method was used, in contrast to typical sputtering technique with sintered target. Single crystalline sapphire(0001) substrates were used. The samples were prepared with varying the growth parameters such as gas environment and film thickness. Then, the samples were characterized by using X-ray diffraction, scanning electron microscopy, and atomic force microscopy measurements. We found that the strain evolution of the samples was highly affected by gas environment and growth rate, resulted in the delamination under O2 environment.
