Study on Pandoraea sp. BCNU 315 Isolated from Soil (토양으로부터 분리한 Pandoraea sp. BCNU 315 에 관한 연구)

  • Bacteria that antagonize plant pathogenic fungi were isolated from the sediment soil at the Ansan industrial estate. One isolate of them showed growth inhibition of Rhizoctonia solani, Botrytis cenerea, and Fusarium oxysporum. This strain was identified as Pandoraea sp. based on phenotypic and phylogenetic characteristics and termed Pandoraea sp. BCNU 315. Tryptone as nitrogen source and sucrose as carbon source were found to be most effective for the microbial growth. In addition, the optimum temperature and pH for microbial growth were $30^{\circ}C$ and pH 7.0, respectively. The substances generated from Pandoraea sp. BCNU 315 were purified and analyzed by column chromatography, HPLC, GC-MS and NMR. As a result, one compound was determined to be indole, another compound was predicted as cyclopentadecaheptene. Detailed structural clarification of the all of the rest six compounds from Pandoraea sp. BCNU 315 has to be accompanied in the further studies.

Effect of Plant Growth Regulators on Bulblet Formation and Plant Regeneration in Fritillaria thunbergii Miq. (패모조직배양에서 생장조절 물질이 자구형성 및 식물체 재생에 미치는 영향)

  • To improve the efficiency of mass propagation in vitro of Fritillaria thunbergii, bulb scale and nodes were cultured in LS medium supplemented with the combination of 2, 4-D and kinetin or NAA and BA. The number and size of bulb, the number of adventitious shoot, the ratio of callus formation, rooting, and the effects of light and dark on the culture, plant regeneration from calli, and the gelling substances were investigated. The combination of 2, 4-D and kinetin in media was more effective than the media of NAA and BA for the bulblet formation. The media supplemented with 2 mg/L 2, 4-D and 1 mg/L kinetin, $1{\sim}2\;mg/L$ 2, 4-D without kinetin and $1{\sim}3\;mg/L$ BAA only were effective in the adventitious shoot development. Callus formation and root formation, respectively were effective in the medium supplemented with 2mg/L 2, 4-D and 1mg/L kinetin. In bulb formation, the medium with 5 mg/L kinetin was effective, and the most of bulbs were formed from the axillary bud of node part. In bulb formation, shoot growth, callus and root formation, the light culture for 16 hours per day was better than that in the dark culture. Bulb was nicely formed in the medium with 0. 2 mg/L 2, 4-D, 1 mg/L kinetin. The medium without hormone was most effective for plant regeneration. The phytagel was more effective than agar in the medium as the gelling agent.

Antifungal activity of Streptomyces costaricanus HR391 against some plant-pathogenic fungi (여러 식물병원성 진균을 억제하는 Streptomyces costaricanus HR391의 항진균능)

  • In this study Streptomyces strains were isolated from soils and their antifungal activities and involved mechanisms were investigated. Among over 400 isolates of actinomycetes, Streptomyces costaricanus HR391 was selected as a potential antagonist to control several plant-pathogenic fungi. S. costaricanus HR391 inhibited mycelial growth of Fusarium oxysporum f. sp. raphani, F. oxysporum f. sp. niveum, F. oxysporum f. sp. lycopersici, and Rhizoctonia solani by 26.5, 26.2, 21.2, and 23.8%, respectively compared to those of uninoculated control after 7-day incubation on PDB medium. S. costaricanus HR391 produced $89{\mu}M$ of siderphore, and showed fungal cell wall-degrading activity including $0.46{\mu}mol/min/mg$ of chitinase and $0.83{\mu}mol/min/mg$ of ${\beta}$-1,3 glucanase. S. costaricanus HR391 secreted 87.49 mg/L of rhamnolipid, and produced 9.49 mg/L and 4.3 mM of lipopeptide, iturin A and surfactin, respectively, all they are membrane-disrupting biosurfactants. It also produced antimicrobial peptide and antibiotics phenazine. In addition to antifungal substances, S. costaricanus HR391 secreted plant growth-promoting phytohormones, zeatin, gibberellins and IAA. These results suggest that S. costaricanus HR391 may be utilized as an environment-friendly biocontrol agent against some important pathogenic fungi.

Evaluating Various Potassium Sources for Kentucky Bluegrass Growth (켄터키블루그래스의 생육을 위한 칼륨의 유형별 효과)

  • Potassium (K) is one of the essential nutrients for plants but is not a constituent of any plant compound. K is substances for activation of many enzymes and influences the process that the enzymes are involved in catalyzing. Although experimental results associated with the K effects with both benefits and no effects on turfgrass growth were reported, the limited research results of K effects on turfgrass among K sources are available in Korea. The objectives of the study was conducted to evaluate responses of Kentucky bluegrass (Poa pratensis L.) treated by fertilizer treatments with six K sources. There were six fertilizer treatments as K sources applied to Kentucky bluegrass which were potassium nitrate (13-0-44, PN), potassium chloride (0-0-60, PC), potassium sulfate (0-0-50, PS), polymer-coated potassium sulfate (0-0-45, PPS), potassium thiosulfate (0-0-25, PT), and potassium sulfate granulated with methylene urea (20-0-25, PSU). Potassium of $8g\;m^{-2}$ and N of $5g\;m^{-2}$ was applied 4 times giving a total 20 and $30g\;m^{-2}$, respectively. Although significant differences were found for turfgrass color, quality, chlorophyll index, and clipping dry weight, no consistent results among K sources were found during the one year study. Overall, K treatments didn't affect turfgrass growth in this study.

Isolation and Characterization of Alga-Lytic Bacterium HY0210-AK1 and Its Degradability of Anabaena cylindrica (남조류 분해세균 HY0210-AK1의 분리와 특성 및 Anabaena cylindrica 분해 활성)

  • To isolate alga-lytic bacteria, a number of samples were collected from Lake of Sukchon and Pal'tang reservoir where cyanobacteria blooming occurred. HY0210-AK1, which exhibited high alga-lytic activity, was isolated using Anabaena cylindrica lawn. The morphological and biochemical characteristics of the isolate HY0210-AK1 were very similar to that of the genus Rhizobium. Taxonomic identification including 16S rDNA base sequencing and phylogenetic analysis indicated that the isolate Hy0210-AK1 had a 99.1% homology in its 16S rDNA babe sequence with Sphingobium herbicidovorans. A. cylindrica NIES-19 was susceptible to the alga-lytic bacterial attack. The growth-inhibiting offset of the bacterium was not different on A. cylindrica NIES-19 when Sphingobium herbicidovorans HY0210-AK1 was in the lag, exponential, and stationary growth phase, although the alga-Iytic effect of S. herbici-dovorans HY0210-AK1 that in stationary growth phase was somewhat pronounced at the first time of inoculation. When S. herbicidovorans HY0210-AK1 was inoculated was inoculated with $1\times 10^{8}$ CFU $ml^{-1}$ together with A cylindrica NIES-19, the bacterium proliferated and caused algal lysis. A. cylindrica NIES-19 died when S. herbicidovorans HY0210 AKl was added to the algal culture but not when duly the filtrates from the bacterial culture was added. This suggests that extracellular substances are not responsible for inhibition of A. cylindrica NIES-19 and that algal Iysis largely attributed to direct interaction between S. herbicidovorans HY0210-AK1 and A. cylindrica NIES-19. The alga-lytic bacterium HY0210-AK1 caused cell lysis and death of three strain of Micro-cystis aeruginosa, but revealed no alga-Iytic effects on the Stephanodiscus hantzschii.

Effects of Supplemental Bio-Active Substances on the Growth Performance, Nutrient Utilization, Blood Characteristics, Microflora Population and Diarrhea Frequency of Weanling Pigs (생리활성 물질(쑥, 두충 및 어성초)의 첨가가 이유자돈의 성장, 영양소 이용율, 혈액특성, 장내 미생물 및 설사빈도에 미치는 영향)

  • This study was conducted to investigate the effects of a supplemented mixture of herbs (artemisia, eucommia ulmoids and houttuynia cordata) on the growth performance, nutrient utilization, blood characteristics, microflora population and diarrhea frequency in weanling pigs. Eighty-four pigs were randomly allotted into one of the four dietary treatments, each of which had 3 replicates. Experimental diets were prepared by adding (0.0, 0.5, 1.0 and 2.0%) a mixture of artemisia, eucommia ulmoids and houttuynia cordata to a commercial feed, and were given to pigs for 42 days. Daily feed intake was significantly increased in the groups supplemented with the mixture at 1.0% group and 2.0% group. Digestibility was significantly increased in 1.0% for protein and ether extract, and in 1.0% group and 2.0% group for NFE. Total bacillus and lactobacillus sp. were significantly (p<0.05) improved in 1.0% group and 2.0% group, respectively. Glucose concentrations of serum increased significantly (p<0.05) in 0.5% group and 1.0% group, and HDL increased significantly (p<0.05) in 1.0% group and 2.0% group. Diarrhea decreased by treatments except control (p<0.05). It is concluded that pigs fed the diet supplemented with 1.0% mixture of herbs can improve daily body weight gain, feed efficiency, nutrient utilizations and diarrhea frequency in pigs.

A Bioassay Method with Radish Seedlings for Bioactive Substances (무우 유묘(幼苗)를 이용(利用)한 식물생리활성물질(植物生理活性物質)의 생물검정법(生物檢定法))

  • Some experiments were conducted to utilize the raphanus test as a bioassay method for plant growth regulaters and hormonal herbicides. Among seven radish cultivars, the response of Jangbaegminong was the most sensitive and quantitative to ${\alpha}$-NAA and its coefficient of variation value was the lowest. The optimum application time of test compounds was estimated seven to nine days after seeding. After ${\alpha}$-NAA treatment the curvature angle of radish cotyledonary petiols was sharply increased to the maximum value at 24 hours after and was changed by light and temperature conditions. The curvature response was observed by application of ${\alpha}$-NAA, phenoxy and indole compounds but often plant growth regulators, $GA_3$ benzyladenine, ABA and homobrassinolide was not detected by this method. So this bioassay was useful to evaluate the activity of a compound as an auxin or a hormonal herbicide.

The Effects of BCAA and Additional OKG or Albumin Pre-Supplementation on Energy Generation Substances and Hormone Changes during Submaximal Exercise (운동 전 BCAA, OKG 및 Albumin 가중 투여가 에너지 생성 물질과 대사관련 호르몬 변화에 미치는 영향)

  • The purpose of this study is to investigate the effects of BCAA and additional OKG or albumin supplementation on energy metabolism and hormone changes in prolonged submaximal exercise. The subjects of this study were 5 male college students, and they took participated in each experiment. In each experiment (control, BCAA, BCAA+OKG, BCAA+albumin), the subject ran for 90 minutes on the treadmill. Blood was drawn and analyzed as glucose, FFA, ammonia, growth hormone, insulin, cortisol, $T_3$, and $T_4$. The obtained results were analyzed via two-way repeated ANOVA using SPSS program. A value of p<.05 was considered statistically significant. The concentration of glucose was the lowest in the BCAA supplement group, and there was significant difference between times of exercise (p<.05). In concentration of FFA, there was a significant difference between times of exercise and supplemented groups (p<.05). The concentration of blood ammonia was the lowest in the BCAA+albumin supplement group, and was the highest in the BCAA supplement group. Concentration of growth hormone, cortisol, $T_3$ and $T_4$ were increased as exercise progressed and decreased after all exercise. But concentration of insulin was decreased as exercise progressed. In these results, BCAA and additional OKG or albumin supplement with BCAA may make better effect to the increasing energy generation and inhibiting protein degradation during prolonged submaximal exercise.

Isolation of Growth Inhibition Substance on Food borne Microorganisms from Hypericum ascyron L. and Application to Food Preservation (물레나물(Hypericum ascyron L.)의 식중독 미생물 증식 억제 물질의 분리 및 식품적용)

  • The ethanol extract and n-hexane fraction from Hypericum ascyron L. showed strong growth inhibition at 25 ppm on 5 strains of Listeria monocytogenes for 72 hr at $32^{\circ}C$. The purified substance, H2-5-2 fraction, was isolated by silica gel column and preparative thin layer chromatography from n-hexane fraction of Hypericum ascyron L. The H2-5-2 fraction showed a strong bacteriostatic activity on 5 strains of L. monocytogenes at 10 ppm in tryptic soy broth, and the viable cell was reduced 1 log cycle compared to initial cell number. The n-hexane fraction of Hypericum ascyron L. showed strong growth inhibition at 25 ppm on Bacillus cereus and Staphylococcus aureus, and at 50 ppm on Vibrio parahaemolyticus for 72 hr. The purified antimicrobial substance, the H2-5-2 fraction, was assumed as high unsaturated sterol by $^1H-NMR$ and $^{13}C-NMR$. On application test using minced Alaska pollack and ground beef, the n-hexane fraction of Hypericum ascyron L. at the level of 250 ppm was applied at $32^{\circ}C$ and $5^{\circ}C$. At $32^{\circ}C$ storage condition, the antimicrobial substances did not reduced L. monocytogenes ATCC 19113, meanwhile at $5^{\circ}C$ storage condition, L. monocytogenes ATCC 19113 was reduced in viable number.

The Study of Application of Bio-Surfactant Producing Bacteria for Growing Crop in Oil Spilled Soil (기름으로 오염된 토양에서 작물생육을 위한 계면활성제 생산 Bacteria의 활용에 관한 연구)

  • Bacillus sp.LPO3 (producing emulsifying substances such as bio-surfactant) was used as a bio-control agent to degrade hydrocarbon (gasoline in oil spilled crop soil). The soil (brought from fertilizer store)was mixed with gasoline-spilled soil (made with Diatomaceous Earth, Sigma.U.S.A). The study was conducted for a period of 13 days, 13 days during which bacterial growth, hydrocarbon degradation and growth parameters of Bacillus sp.LP03 including shoot and root length were studied. We found that the effective of bacterial producing substance might bio-surfactants let the plants survive even more promote the growth of shoot and root length and showed antifungal activity against gray mold. Without the bacteria, they couldn't grow in oil-spilled soil not even survive. According to the results of the above experiments, we can see with following results, hydrocarbon in gasoline was reduced, day by day, then RNA dot blotting was done and it fit the results we had done. Finally, this Bacteria(producing bio-surfactant) were found to have effective bio-control agent for cropping in oil spilled soil and infected by gray mold.
