Genetic Monitoring of Plant Growth Promoting Rhizobacterium (PGPR), Bacillus subtilis AH18 using Multiplex PCR in Field Soil (Multiplex PCR을 이용한 생물방제균 Bacillus subtilis AH18의 토양내 Genetic Monitoring)

  • The genetic monitoring method was developed for the rapid detection of the PGPR and biocontrol agent, B. subtilis AH18 in red-pepper field soil by multiplex PCR using sid, aec and cel gene primers. The monitoring of B. subtilis AH18 in the soil was carried by amplified a 2,3-dihydro-2,3-dihydroxy benzoate dehydrogenase [EC: 1. 3. 1. 28]gene (sid - 794 bp : EF408238) which is a key enzyme of siderophore synthesis, an auxin efflux carrier gene (aec - 1,052 bp : EF408239) and a cellulase gene (cel - 1,582 bp : EF070194). The natural un sterilized soil was inoculated with B. subtilis AH18 to determine the sensitivity ($1.8\times10^5$ cfu/g) of multiplex PCR for the rapid dectection and then the strain was monitored successfully in rhizosphere or non-rhizosphere soil of red-pepper cultural soil. At 3 weeks after the treatment, density of the strain was monitored more abundantly in rhizosphere soil.

RNAi-Mediated Gene Silencing of Trcot1 Induces a Hyperbranching Phenotype in Trichoderma reesei

  • Trichoderma reesei is the major filamentous fungus used to produce cellulase and there is huge interest in promoting its ability to produce higher titers of cellulase. Among the many factors affecting cellulase production in T. reesei, the mycelial phenotype is important but seldom studied. Herein, a close homolog of the Neurospora crassa COT1 kinase was discovered in T. reesei and designated TrCOT1, which is of 83.3% amino acid sequence identity. Functional disruption of Trcot1 in T. reesei by RNAi-mediated gene silencing resulted in retarded sporulation on potato dextrose agar and dwarfed colonies on minimal medium agar plates containing glucose, xylan, lactose, xylose, or glycerol as the sole carbon source. The representative mutant strain, SUS2/Trcot1i, also displayed reduced mycelia accumulation but hyperbranching in the MM glucose liquid medium, with hyphal growth unit length values decreased to 73.0 ㎛/tip compared to 239.8 ㎛/tip for the parent strain SUS2. The hyperbranching phenotype led to slightly but significantly increased cellulase secretion from 24 to 72 h in a batch culture. However, the cellulase production per unit of mycelial biomass was much more profoundly improved from 24 to 96 h.

Development of Microbial Inoculant Using By-product of Oriental Herbal Medicine (한약재박을 이용한 미생물제제의 개발)

  • The development of microbial inoculant was conducted using a by-product of oriental herbal medicine. The constituent of the by-product, which was high in organic matter, was 11.3% of crude protein, 5.1% of crude lipid, 49.7% of NDF (neutral detergent fiber), and 33.8% of ADF (acid detergent fiber). Microorganisms isolated from the by-product of oriental herbal medicine were 35 species. Among them, 6 bacterial species, 4 fungal species, 2 actnomycetes species, and 1 yeast species were effective in the utilization of the by-products. The 13 strains screened were tested for the plant growth-promoting effect in soybean seedling. BL-333 strain was found to increase the soybean yield by about 23% as compared with control. The strain BL-333 was identified as Paenibacillus marcerans. P. marcerans BL-333 showed high anti-fungal activities against virulent fungi, especially Fusarium sp. and Collectotrichum sp. Yields of plants which were inoculated with microbial inoculant prepared with P. marcerans BL-333 and by-product of oriental herbal medicine were found to be higher than control by $3{\sim}24%$. The yield was especially promoted in lettuce, radish, chinese cabbage and cucumber plants.

The Effects of Coal Thermal Power Plant Exports on the National Economy (석탄화력발전 해외수출의 경제적 파급효과 분석)

  • Korean domestic electricity market is saturated through trying to find ways to overcome the limitations of the domestic electricity industry with overseas electricity markets. Korean electricity industry in the overseas market in order to pursue continuous growth, competitive and aggressive investment in overseas market is promoting. This paper attempts to apply input-output analysis to estimate the role of coal thermal power plant sector exports national economy. More specifically, this study shows what national economy effect of production-inducing effect, value-added inducing effect, and employment-inducing effect are explored with demand-driven model. After define coal thermal power plant sector what small sized of Input-Output table 168 sectors among 17 sectors, this study pays particular and close attention to coal thermal power plant sector by taking the sector as exogenous specification and then investigating economic impacts of it. This study uses coal thermal power plant exportation case of Vietnam project, production-inducing effect, value-added inducing effect, and employment-inducing effect are 2,853 billion won, 973 billion won and 14,761 persons, respectively.

A Study on the Status and Supporting Strategy of National R&D Programs related to the Convergence Technology (융합기술 관련 국가 연구개발 사업 현황과 효과적 지원전략에 대한 연구)

  • As science and technology which the society requires become highly advanced and complicated, the need of so-called Convergence Technology(CovT) is getting more significant. The major developed countries are presenting their own vision and strategies of CovT to take the lead of future society and global economies. In Korea, each ministries and government offices are suggesting their own directions of promoting CovT and supporting the related programs. This paper investigates the overall status of national and international R&D activities related to CovT in order to invigorate the related national R&D programs and maximize their effectiveness. It also purports to realize the growth potential and a ripple effect of CovT, such as new business creation and fulfillment of public needs, so that secure new CovT in an early stage. Based on the analysis of the overall status of national R&D programs, we redefine and re-categorize the CovT, which is especially oriented to the development of government support policy, and present the customized strategies to each categories.

A Study on the Development History and Growth Strategies of Korean Creative Buddhist Hymns as Buddhist Ritual Contents (불교 의식용 콘텐츠로서 한국 창작 찬불가의 발전사 및 성장 방안)

  • The purpose of this study is to examine the periodic characteristics of creative Buddhist hymns in our country, and confirm the role of Buddhist hymns in popularizing Buddhism and the activities and meanings of pioneers who contributed to such Buddhist hymns. The further purpose of this study is to confirm the utility of Buddhist hymns. In the present, the problems of Buddhist hymns can be pointed out as follows. First, some Buddhist hymns which were created about 50 years ago are used, so that new Buddhist hymns can not be established. Second, their degree of completion as music are not sufficient. Third, it is insufficient to study the miswriting of transmitted Buddhist hymns. Fourth, there is no systematical management of Buddhist hymns. The countermeasures for these problems can be presented as follows. First, it is necessary to expand the utilization of Buddhist hymns in Buddhist ceremonies. Second, new creative Buddhist hymns are required. Third, the religious body should reinforce its support for promoting interests in Buddhist hymns. Fourth, it is necessary to activate temple concerts in order to strengthen the popularization of Buddhist hymns. First of all, it is required to confirm the usefulness of creative Buddhist hymns for popularizing Buddhism and create and utilize new Buddhist hymns appropriate to contemporary senses.

A Study on the Linkage between Intelligent Security Technology based on Spatial Information and other Technologies for Demonstration of Convergence Technology (융복합 기술 실증을 위한 공간정보 기반 지능형 방범 기술과 타 분야 기술 간 연계 방안 연구)

  • With the recent changes in the social environment, the growth in the so-called 'five major crimes' has been highlighted as one of the causes of anxiety in Koreans' lives. Many attempts have been made to solve this problem; however, it is still difficult to secure the location information of the socially vulnerable in emergency situations and to precisely identify the features and clothing of criminals and track them using current image analysis technology. Therefore, the development of precision positioning technology and support services along with intelligent security service technology based on spatial information has been given a high priority. This study suggested measures that could be continuously applied to link technologies and services with high linkability by analyzing technologies based on spatial information and other fields. To establish measures for linkage between intelligent security technology and other technologies and services, this study analyzed the existing technologies and research trends in intelligent security technology, and reviewed linkable services according to five criteria established to evaluate their linkability. Based on this analysis, three technologies with high linkability were ultimately selected, and measures for linkage were established. It is expected that the linkage measures derived using the objective evaluation criteria will serve as a stepping stone for promoting active technology linkage and commercialization in the future, even after the completion of this study.

Burkholderia tropica as a Potential Microalgal Growth-Promoting Bacterium in the Biosorption of Mercury from Aqueous Solutions

  • The use of microalgal biomass is an interesting technology for the removal of heavy metals from aqueous solutions owing to its high metal-binding capacity, but the interactions with bacteria as a strategy for the removal of toxic metals have been poorly studied. The goal of the current research was to investigate the potential of Burkholderia tropica co-immobilized with Chlorella sp. in polyurethane discs for the biosorption of Hg(II) from aqueous solutions and to evaluate the influence of different Hg(II) concentrations (0.041, 1.0, and 10 mg/l) and their exposure to different contact times corresponding to intervals of 1, 2, 4, 8, 16, and 32 h. As expected, microalgal bacterial biomass adhered and grew to form a biofilm on the support. The biosorption data followed pseudo-second-order kinetics, and the adsorption equilibrium was well described by either Langmuir or Freundlich adsorption isotherm, reaching equilibrium from 1 h. In both bacterial and microalgal immobilization systems in the co-immobilization of Chlorella sp. and B. tropica to different concentrations of Hg(II), the kinetics of biosorption of Hg(II) was significantly higher before 60 min of contact time. The highest percentage of biosorption of Hg(II) achieved in the co-immobilization system was 95% at pH 6.4, at 3.6 g of biosorbent, $30{\pm}1^{\circ}C$, and a mercury concentration of 1 mg/l before 60 min of contact time. This study showed that co-immobilization with B. tropica has synergistic effects on biosorption of Hg(II) ions and merits consideration in the design of future strategies for the removal of toxic metals.

Effects of Plants, Rhizobacteria and Physicochemical Factors on the Phytoremediation of Contaminated Soil (오염 토양의 식물상 복원효율에 미치는 식물, 근권세균 및 물리.화학적 인자의 영향)

  • Phytoremediation is an economic and environmentally friendly technique to remediate contaminated-soil. In this study, the effects of plants, rhizobacteria and physicochemical factors on phytoremediation have been reviewed. For successful phytoremediation, the selection of plants is primarily important. To remediate soil contaminated with petroleum hydrocarbon, raygrass (Lolium multiflorum lam), white mustard, vetch (Vicia villosa), tall fescue (Festuca arundinacea), legumes, poplar, and Pine (Pinus densiflora) were mainly applied, and the removal efficiency of petroleum hydrocarbon were ranged 68 to 99%. Corn (Zea mays), raygrass (Lolium multiflorum lam), vetch (Vicia villosa), mustard, clover (Trifolium repens), and tall fescue (Festuca arundinacea) were used for the removal of polycyclic aromatic hydrocarbon, and their removal efficiencies were 50-98%. Rhizobacteria play significant roles for phytoremediation because they can directly participate in the degradation of contaminant as well as promoting plants growth. The following rhizobacteria were preferred for phytoremediation: Azospirillum lipoferum, Enterobactor cloacae, Azospirillum brasilense, Pseudomonas putida, Burkholderia xenovorans, Comamonas testosterone, Pseudomonas gladioli, Azotobacter chroococcum, Bacillus megaterium, and Bacillus subtilis. Pysicochemical factors such as pH, temperature, nutrient, electron acceptor, water content, organic content, type of contaminants are consequential limiting factors for phytoremediation.

  • Probiotics are live organisms that when administered in adequate amounts confer a health benefit on hosts. The administration of direct-fed microbials (DFM) such as lactobacilli and bacillus, may be a more direct approach to beneficially alter gastrointestinal microflora than altering dietary ingredients or supplementing with growth-promoting levels of antibiotics. It is apparent that microbes have an important influence on immune development and resistance to infection; that microbes are not static colonizers of our bodies, but are dynamic, symbiotic coresidents. And it can improve the surrounding environments; decrease the malodor caused by degrade the excrement. Recently, new paradigm such as environment protection and safe food have been settled. In domestic farm house, there is a great demand for probiotics as a substitute of antibiotics for the improvement of environmental quality and the production of a competitive goods. Probiotics circulated in a country have three categories: an animal medicine permitted by national veterinary research quarantine service (NVRQS), a support feed registered in city or country house, and not-registered goods. However, lots of unqualified goods were produced and circulated. And thus, it is in urgent need of evaluating the present situation and effect of probiotics. This study was conducted to evaluate the system of a probiotics as a livestock-environment improving agents for the alternation of antibiotics and quality control of it.
