Tobacco Growth Promotion by the Entomopathogenic Fungus, Isaria javanica pf185

  • Isaria javanica pf185 is an important entomopathogenic fungus with potential for use as an agricultural biocontrol agent. However, the effect of I. javanica pf185 on plant growth is unknown. Enhanced tobacco growth was observed when tobacco roots were exposed to spores, cultures, and fungal cell-free culture supernatants of this fungus. Tobacco seedlings were also exposed to the volatiles of I. javanica pf185 in vitro using I-plates in which the plant and fungus were growing in separate compartments connected only by air space. The length and weight of seedlings, content of leaf chlorophyll, and number of root branches were significantly increased by the fungal volatiles. Heptane, 3-hexanone, 2,4-dimethylhexane, and 2-nonanone were detected, by solid-phase micro-extraction and gas chromatography-mass spectrophotometry, as the key volatile compounds produced by I. javanica pf185. These findings illustrate that I. javanica pf185 can be used to promote plant growth, and also as a biocontrol agent of insect and plant diseases. Further studies are necessary to elucidate the mechanisms by which I. javanica pf185 promotes plant growth.

Effect of Acaromyces Ingoldii Secondary Metabolites on the Growth of Brown-Rot (Gloeophyllum Trabeum) and White-Rot (Trametes Versicolor) Fungi

  • We investigated the antifungal activities of an endophytic fungus identified as Acaromyces ingoldii, found on a loblolly (Pinus taeda L.) pine bolt in Louisiana during routine laboratory microbial isolations. The specific objectives were to determine the inhibitory properties of A. ingoldii secondary metabolites (crude extract) on the mycelial growth of a brown-rot fungus Gloeophyllum trabeum and a white-rot fungus Trametes versicolor, and to determine the effective concentration of A. ingoldii crude preparation against the two decay fungi in vitro. Results show the crude preparation of A. ingoldii from liquid culture possesses significant mycelial growth inhibitory properties that are concentration dependent against the brownrot and white-rot fungi evaluated. An increase in the concentration of A. ingoldii secondary metabolites significantly decreased the mycelial growth of both wood decay fungi. G. trabeum was more sensitive to the inhibitory effect of the secondary metabolites than T. versicolor. Identification of specific A. ingoldii secondary metabolites, and analysis of their efficacy/specificity warrants further study. Findings from this work may provide the first indication of useful roles for Acaromyces species in a forest environment, and perhaps a future potential in the development of biocontrol-based wood preservation systems.

The Antidermatophytic Potential of the Marine Isolate of Aspergillus sp. Collected from South Coast of Korea

  • This study was carried out to assess the antidermatophytic potential of the ethyl acetate(EtOAc) extract of the marine isolate of Aspergillus sp.. The fungus was isolated by serial dilution, and was identified Aspergillus sp.. The EtOAc extract of the fungus was examined to evaluate the antidermatophytic efficacy against the fungal pathogens infecting human skin using the disc diffusion and MIC(minimum inhibitory concentration) determination methods. The EtOAc extract($5{\mu}l\;disc^{-1}$) was considered to have the antidermatophytic activity based on the inhibition percentage of the mycelial growth of the fungi tested such as Trichophyton mentagrophytes KCTC 6085, Microsporum canis KCTC 6591, Microsporum canis KCTC 6348, Trichophyton rubrum KCTC 6352, Microsporum canis KCTC 6349 and Trichophyton mentagrophytes KCTC 6316. The percentage of the inhibition ranged from 54% to 81, and the MIC obtained was 62.5, 62.5, 250, 125, 125, and $125{\mu}g\;ml^{-1}$, respectively. The extract had a strong detrimental effect on the spore germination of the tested skin infectious pathogens. These findings strongly support the role of the ethyl acetate extract as a potential antidermatophytic agent.

Cylindrocladium crotalariae의 균사자람, 포자형성과 포자발아에 대한 Water potential의 효과 (Effect of water potential on mycelial growth, reproduction, and spore germination by Cylindrocladium crotalariae)

  • 콩의 흑색뿌리썩음병을 일으키는 Cylindrocladium crotalariae은 감자한천배지와 콩의 조직을 함유한 한천 배지를 서로 다르게 Water potential를 조절하여 이 병원균을 접종한 결과 균사신장과 대형분생포자형성은 -14bars에서 좋았고 $30^{\circ}C$에서는 -100bars $25^{\circ}C$에서는 -80bars $20^{\circ}C$에서는 -64bars에서 균사신장이 중지되었으며 포자형성도 -64bars에서 중지되었다. 자낭각과 미소균핵형성은 소금을 첨가하지 않은 -1.4나 -3.0bars에서 잘 되었고 이 균에 대한 포자발아율은 -1.4bars에서 -20bars의 모든 Water potential에서 똑 같이 좋았고 -20bars에서 Water potential이 낮아지므로 발아율이 떨어지다가 -60bars에서 억제되었다.

Penicillium menonorum: A Novel Fungus to Promote Growth and Nutrient Management in Cucumber Plants

  • The present study is the first report on the isolation of Penicillium menonorum from rhizosphere soil in Korea and its identification based on morphological characteristics and internal transcribed spacer gene sequence. The fungal isolate was named KNU-3 and was found to exhibit plant growth-promoting (PGP) activity through indole acetic acid (IAA) and siderophore production, as well as P solubilization. KNU-3 produced 9.7 mg/L IAA and solubilized 408 mg of $Ca_3PO_4/L$, and inoculation with the isolate significantly (p < 0.05) increased the dry biomass of cucumber roots (57%) and shoots (52%). Chlorophyll, starch, protein, and P contents were increased by 16%, 45%, 22%, and 14%, respectively, compared to plants grown in uninoculated soil. The fungus also increased soil dehydrogenase (30%) and acid phosphatase (19%) activities. These results demonstrate that the isolate KNU-3 has potential PGP attributes, and therefore it can be considered as a new fungus to enhance soil fertility and promote plant growth. Moreover, the discovery of PGP ability and traits of this fungus will open new aspects of research and investigations. In this study, plant growth promotion by P. menonorum KNU-3 is reported for the first time in Korea after its original description.

WUFI를 이용한 공동주택 붙박이가구의 층별·형태별 결로 생성 비교 평가 (Comparative Evaluation of Condensation by Type and Layer Around Built-in Furniture of Apartment Buildings)

  • Recently, as the continuously increasing of the one-person households, builders were focusing on a built-in furniture development for efficiency. Through the dead space of the space can be reduced. Also, Condensation that occurs in a built-in furnitures installation space may result in different results depending on the room's direction, floor, finish, adjacent room effect, occupant's lifestyle. In this background, it can be considered in terms of functional aspect and morphological aspect by using WUFI2D simulation system. As a result, the tendency of occurrence of condensation was examined by comparing the water content and the growth potential of fungus graph which came from WUFI2D. Results of this study are as followings: Built-in furniture, both top & bottom open, is more stable to condensation than the built-in furniture with the top or bottom open. Also, Among from the top floor built-in furniture to the bottom of it, the bottom one is more stable to condensation.

Response of Mulberry Brown Leaf Spot Fungus Myrothecium roridum to Different Plant Extracts

  • Anti-fungal potential of 5 plant extracts viz., Eucalyptus citriodora, Allium sativum, Cassia sophera, Chromolaena odorata and Datura metel on the growth of mulberry brown leaf spot pathogen Myrothecium roridum were examined. Except fur the aqueous extract of Allium bulb, ethanolic leaf extract of all other plants more efficiently reduced the colony growth of the fungus on potato-dextrose-agar, Of which, Allium and Eucalyptus extracts were more effective. Initiation of radial growth of M. roridum on solid media was deferred maximum 6 days by ethanolic Eucalyptus extract and 4 days by aqueous Allium extract at $0.4 mg.ml^{-1}$. In the liquid media amended with Eucalyptus extract ($0.4 mg.ml^{-1}$) complete inhibition of sporulation was noticed upto 8 days, and initial inhibition of mycelial bio-mass generation was considerably diminished with time and reduction was 1.3 fold 14 days after application. While, complete inhibition of mycelial growth for 6-14 days was recorded with $\geq$0.1 mg.ml$^{-1}$ commercial eucalyptus oil. However, rejuvenation of growth appeared when fungus was re-inoculated in fresh media. Post-inoculate application of different doses Of Eucalyptus and Allium extracts significantly (p < 0.05) reduced the disease severity in pot-ted mulberry. However, persistence of the effect up to 28 days was apparent at $\geq$ 1.0 mg.ml$^{-1}$ and effectively was on par with carbendazim (1 mg.ml$^{-1}$ ). Almost equal control ability of 1.0 mg.ml$^{-1}$ Eucalyptus extracts can be achieved by ca. 10 times lowered dose of commercial eucalyptus oil. It seems, the toxic principle of E. citrodora to M. roridum is fungistatic in nature and may have essential oil based origin.

감자갈색잎점무늬병균(Alternaria alternata)에 대한 버섯 배양액 추출물 15종의 생장 억제 효과 (Inhibitory effects of 15 mushroom culture extracts on the growth of Alternaria alternata causing potato brown spot)

  • The fungus Alternaria alternata, responsible for causing brown to black spotting on numerous fruits and vegetables globally, was identified in 2022 as the causative pathogen of brown spot disease in potatoes in Korea. In pursuing potential inhibitors against A. alternata growth, we evaluated 15 mushroom culture filtrates: eight from Trametes spp. and seven from Polyporus spp., known for their antibacterial and anticancer properties. Antifungal activity was assessed by exposing each filtrate to A. alternata on a paper disc. Four filtrates displayed inhibitory action against the fungus, albeit with mild effects. Our findings highlight the potential of Trametes and Polyporus fungi as emerging antifungal candidates, offering promise in preventing potato brown spots.

  • Fusarium moniliforme strain별 균사신장(菌絲伸長)은 Strain IV은 water agar나 PDA에 관계(關係)없이 -14 bars에서 좋았고 -80 bars에서도 균사(菌絲)자람이 정지되지 않았다. Strain II와 Strain IV 를 비교(比較)하여보면 Strain II가 Strain IV보다 각 water potential 에서 균사(菌絲)자람이 좋았다. 분생포자형성(分生胞子形成)은 Strain에 관계(關係)없이 소형분생포자(小型分生胞子)는 0에서 -20bars까지는 점점(漸漸) 형성(形成)이 증가(增加)하다가 -20bars에서 가장 많이 형성(形成)되었으며 -80bars에서도 아주 적은수이지만 형성(形成)이 되었다. 대형분생포자(大型分生胞子)는 -1.4bars에서 포자형성(胞子形成)이 잘 되었고 water potential 떨어짐에 따라 -32bars에서 대형분생포자(大型分生胞子)를 형성(形成)하지 않고 소형분생포자(小型分生胞子)만 형성(形成)하였다. 포자발아(胞子發芽)는 -1.4bars에서 발아율(發芽率)이 좋았고 water potential이 떨어질수록 발아율(發芽率)도 떨어졌다.

Evaluation of Potential Reference Genes for Quantitative RT-PCR Analysis in Fusarium graminearum under Different Culture Conditions

  • The filamentous fungus Fusarium graminearum is an important cereal pathogen. Although quantitative realtime PCR (qRT-PCR) is commonly used to analyze the expression of important fungal genes, no detailed validation of reference genes for the normalization of qRT-PCR data has been performed in this fungus. Here, we evaluated 15 candidate genes as references, including those previously described as housekeeping genes and those selected from the whole transcriptome sequencing data. By a combination of three statistical algorithms (BestKeeper, geNorm, and NormFinder), the variation in the expression of these genes was assessed under different culture conditions that favored mycelial growth, sexual development, and trichothecene mycotoxin production. When favoring mycelial growth, GzFLO and GzUBH expression were most stable in complete medium. Both EF1A and GzRPS16 expression were relatively stable under all conditions on carrot agar, including mycelial growth and the subsequent perithecial induction stage. These two genes were also most stable during trichothecene production. For the combined data set, GzUBH and EF1A were selected as the most stable. Thus, these genes are suitable reference genes for accurate normalization of qRT-PCR data for gene expression analyses of F. graminearum and other related fungi.
