THE INHIBITING EFFECT OF TISSUE CONDITIONER COMBINED WITH ANTIFUNGAL AGENTS ON THE GROWTH OF CANDIDA ALBICANS (항진균제를 첨가한 수종의 점막 조정제가 Candida albicans 성장 억제에 미치는 효과)

  • Tissue conditioners have been used for treatment of denture stomatitis caused by wearing of dentures. Early studies pointed out Candide albicans (C. albicans) as main etiologic factor, and antifugal agents were added for control of the species. But there is a little information about broad comparison on the effect of tissue conditioners and antifungal agents added. The purpose of the present study was to compare the inhibiting effect of four tissue conditioners and one temporary soft liner on the growth of C. albicans for treatment of denture stomatitis using gel diffusion method by measuring diameter of the zone of growth inhibition. Three antifungal agents were added to each material for evaluation of the effect of added agents. Finally, observation was made to evaluate the effect of the loss of antifungal elements by aging of the specimen. The results of this study were obtained as follows : 1. Tempo had remarkable antifungal effect showing the zone of growth inhibition as 2.35 mm at 1st day, and was most effective on End: 4th and 7th day from incubation (p<0.05). But Coecomfort, Dura conditioner, Visco-gel, Coe-soft had little antifungal effect from the 1st day of incubation. 2. Nystatin was most effective showing 9.60-12.04 mm of zone of inhibition at the 1st day from incubation. The antifungal properties were reduced to amphotericin B, chlorhexidine and materials without agent (p<0.05), and the effect was diminished by time. 3. As pretreatment with amphotericin B, nystatin, chlorhexidine, Tempo was very effective at the 1st day from incubation showing zone of inhibition as 3.65, 12.04, 4.78 mm with addition of each agent. Dura conditioner had strongest antifungal effect at the next day as 2.86, 5.33, 1.29 mm of zone of inhibition, and yielding results of Coe-comfort, Tempo, Coe-soft was shown at 4th and 7th day from incubation (p<0.05). Taken all together, tissue conditioners have little antifungal effect except Tempo. Formation of the zone of growth inhibition was due to agents amphotericin B, nystatin, chlorhexidine and nystatin was most effective. Conclusively, it is advisable to select material which is effective on the growth of C. albican and consider addition of antifungal agents for treatment of denture stomatitis.

The Effect of Fungicides on Mycelial Growth and Conidial Germination of the Ginseng Root Rot Fungus, Cylindrocarpon destructans

  • Ginseng root rot caused by Cylindrocarpon destructans is the most destructive disease of ginseng. Six different fungicides (thiophanate-methyl, benomyl, prochloraz, mancozeb, azoxystrobin, and iprodione) were selected to evaluate the inhibitory effect on the mycelial growth and conidial germination of C. destructans isolates. Benomyl and prochloraz were found to be the most effective fungicides in inhibiting mycelial growth of all tested isolates, showing 64.7% to 100% inhibition at a concentration of $10{\mu}g/mL$, whereas thiophanate-methyl was the least effective fungicide, showing less than 50% inhibition even at a higher concentration of $100{\mu}g/mL$. The tested fungicides exhibited less than 20% inhibition of conidium germination at concentrations of 0.01, 0.1, and $1{\mu}g/mL$. However, the inhibition effect of mancozeb on condium germination of C. destructans was significantly increased to 92% to 99% at a higher concentration of $100{\mu}g/mL$, while the others still showed no higher than 30% inhibition.

The Growth Inhibition Effect of L-1210 and S-180 Cancer Cell Lines by the Extract from Anemarrhena Asphodeloides (지모(知母) 추출물이 L-1210 및 S-180 암세포주 성장 억제에 미치는 영향)

Objective This study was aimed to screen the potential antitumor activity of one kinds of Korean medicinal herb extracts against cancer cell lines and to evaluate the growth inhibition effect of L-1210 and S-180 cancer cell lines. 2. Methods It confirmed Anemarrhena asphodeloides extracts to screen the potential antitumor activity. Then, it was extracted with 4 kinds of solvents ; hexane, ethyl acetate, butanol and $H_2O$, and the Growth inhibition effect of these extracts were determined against cancer cell and normal cell. The results were as follows : The IC50(50% inhibitory concentration) values of Anemarrhena asphodeloides extracts were shown to be $253{\mu}g/ml$ against L-1210 cell lines. The IC50 values of ethyl acetate extracts were shown to be $915{\mu}g/ml$ against L-1210 cell lines. The IC50 values of butanol extracts were shown to be $52.3{\mu}g/ml$, $485{\mu}g/ml$ against L-1210, S-180 cell lines, respectively. The butanol extracts were more selectively effective than other extracts to cancer cell lines. 3. Conclusion From these data, it could be concluded that the Anemarrhena asphodeloides extracts to the Growth inhibition effect of L-1210 and S-180 cancer cell lines.

Inhibitory Effect of Garlic on the Growth of Aspergillus parasiticus (한국산 마늘에 의한 Aspergillus parasiticus의 성장 억제 효과)

  • This study was performed to investigate the possible effect of garlic produced in Korea on the inhibition/reduction of growth of A. parasiticus, a toxigenic strain. The effect was studied using different concentrations of freeze-dried garlic in potato-dextrose agar (PDA) and/or in yeast-extract sucrose (YES) broth at $25^{\circ}C$ for 15 days. While inhibition of the fungal growth due to increasing the concentration of garlic was observed, the more remarkable effect was observed on the ninth day. Reduction of fungal diameter as a result of addition of garlic on PDA was observed to range between 3.4% to 20.1 % while reduction of mycelial weight in YES broth ranged from 9.9% to 30.5%. The 0.5% and 1.0% concentrations of garlic significantly reduced fungal diameter in PDA on the 9th day, while 0.1 %, 0.5%, and 1.0% concentrations of garlic significantly reduced the mycelial weight in YES broth (p<0.05). Dose-response relationships were observed between the concentration of garlic and inhibition of growth both in solid culture and in liquid culture. This study indicates that garlic could be an effective inhibitor at a human consumption level of the growth of A. parasiticus. More research is needed to study the inhibitory effects of the main active component of garlic.

Inhibition of Bacterial Growth with Silver Wire Iontophoresis (은 이온도입이 세균성장에 미치는 영향)

  • The silver cation has broad-spectrum antibiotic activity toward Gram-positive, Gram-negative, fungal. aerobic and anerobic micro-organisms. Silver has been used to care of infected wound. pyogenic arthritis, and chronic osteomyelitis. The purpose of this study was to determine whether pure silver wire iontophoresis using milliamperage direct current has an inhibitory effect on growth in vitro of 3 different species of bacteria-Staphylococcus aureus, Escherichia coli, and Pseudomonas aeruginosa. Using agarose based media, silver iontophoresis performed at 0, 1, 2, 4, 8 mA for 15 minutes. All experiments were performed in triplicate. Following iontophoresis, inhibition zone width of bacterial growth was measured with calliper. The inhibition of bacterial growth occured at the anodal silver electrode. Inhibition zone width of bacterial growth was significantly increased in all three bacterial species (p<0.05). Between bacterial species, inhibition zone width was not significantly different. Inhibition gone and amperage showed a highly significant positive linear relationship (p<0.001). The result of this study showed that pure silver wire iontophoresis with milliamperage direct current, as well as microamperage direct current, can inhibit bacterial growth in vivo.

Influence of Selected Turfgrass Herbicides, Simazine, Pendimethalin, Propyzamide and Asulam on the Growth of Zoysia matrella (Zoysia matrella의 생장에 미치는 잔디용 제초제 Simazine, Pendimethalin, Propyzamide, Asulam의 영향)

  • This study was conducted to investigate the influence of herbicides, simazine, pendimethalin, propyzamide and asulam on the growth of Zoysia matrella when they are treated at different growth stages of the plant. The reaction of Zoysia matrella to herbicides differed depending on kinds of herbicides, their concentcations, and time of treatment. Simazine at the higher concentration of 600g /l0a exhibited inhibitory effect on leaf dry matter and growth of runner when treated on April 5, 1991. The highest inhibition of leaf and runner was observed in the plot applied on May 23, 1990. The sprouting and growth of Zoysia matrella was not affected by all the concentrations of the herbicides applied on October 23,1991. Pendimethalin did not influence the growth of Zoysia matrella at he treatment of April 5. However, at the treatment of May 23, the number and growth of runner was inhibited although the plant height and leaf dry matter was not affected. When pendimethalin was treated on October 23, the growth of runner was very slightly reduced in the next year. In case of propyzamide, there was no inhibitory effect on the growth of Zoysia matrella regardless of treatment of times, methods, and dosages. Greater inhibition in the growth of Zoysia matrella was observed at higher dosage(1,ll0g /l0a)-treated plot on April 5 and all the concentrations treated on May 23. But, no inhibition was observed on the October 23 treatment.

In vitro Screening of UVA Phototoxicity Inhibitors using the Natural Products (In vitro 실험법에 의한 천연물 중의 UVA 광독성 억제제 검색)

  • The phototoxicity inhibitory activity of 15 natural products having antiinflammatory effect was screened by three in vitro methods: yeast growth inhibition test with Candida albicans, RBC photohemolysis and MTT assay. We induced phototoxic reaction by irradiating UVA (365 nm) on chlorpromazine (CPZ) that has been widely documented as phototoxic agent in clinical and experimental studies and then observed the effects of the natural products after treating them with CPZ. In yeast growth inhibition test, X. stramonium showed the inhibitory effect on the UVA phototoxicity and E. officinalis, Yeast, P. suffruticosa showed phototoxicity inhibitory effect in that their % hemolysis compared with control were 36.14${\pm}$ 2.69, 42.82${\pm}$1.35, 36.41${\pm}$0.48 on UVA. In MTT assay, all tested natural products increased cell viability compared with the control.

Effect of Ethanol Extract of Herbal Medicine on Crop Growth Inhibitor (한약재 주정 추출물이 종자발아와 유묘생장에 미치는 영향)

  • BACKGROUND: Herbal extracts have been screened for their inhibitory effect of seed germination and root development on weeds, but there is a scarcity of reports for crop growth regulation. The objective of this research was to develop a growth inhibitor on Brassica campestris, and its effective extraction method from herbal medicine extract. METHODS AND RESULTS: Eighty four herbal medicine extracts were tested for their plant growth inhibition activity on B. campestris. The alcohol extracts of Artemisia annua, Cinnamomum cassia, and Mentha arvensis inhibited over 30% of germination and the extract of A. annua, and C. cassia inhibited over 70% of radicle growth at 0.1 % w/w treatment. The partially purified extracts of A. annua, and C. cassia with dichloromethane and hexane showed stronger radicle growth inhibition than the crude extracts on B. campestris. The diethyl ether extract of A. annua showed a similar 50% radicle growth inhibition ($RI_{50}$ = 45 mg/L) to its partially purified extract with dichloromethane or hexane, but the diethyl ether extract of C. cassia showed a worse $RI_{50}$ than the purified extract. CONCLUSION: The alcohol extracts of A. annua, and C. cassia showed potent radicle growth inhibition properties on B. campestris. Diethyl ether proved to be a good solvent for simple extraction from A. annua.

Growth Inhibition Effect of Environment-friendly Agricultural Materials in Botrytis cinerea In Vitro (친환경 유기농자재의 잿빛곰팡이병 병원균의 생장 억제 효과)

  • Inhibition effects on spore germination and mycelia growth for gray mold (Botrytis cinerea) were investigated in vitro using environment-friendly agricultural materials as well as environment-friendly pesticides. The inhibition effect on mycelia growth of gray mold is the highest when the gray mold mycelia were treated with a pesticide (commercial name: Koreayeok, Jihabudea KM, Sootingtan, Sootingstar) that contains a mixture of Bacillus subtilis, resulting in 100% inhibition of the mycelia growth. Meanwhile, the range of less than 20% inhibition effects on the growth of gray mold mycelia was observed with other commercial agricultural materials. The significant inhibition effects on spore germination of gray mold fungus were shown in vitro with two water dispersible pesticides containing sulfur [BTB (97.7%) and SulfurStar (92.3%)], respectively. These in vitro results of inhibiting of the spore germination and mycelia growth together cannot found. It remains to be determined whether the selected environment-friendly agricultural materials in effective control of gray mold in vitro can be used to control gray mold in field.

Growth Inhibition of Garlic (Allium sativum L.) Juice on the Microorganisms (마늘즙의 미생물증식 억제효과)

  • In order to investigate antimicrobial substances from natural spices, the effects of garlic juice and ginger juice on the growth of microorganisms were investigated. It was showned that garlic juice inhibited the growth of bacteria, yeasts and fungi but ginger juice did not inhibit the growth of bacteria and yeasts. As result of concentration effect of garlic juice on the growth of microorganisms, the higher concentration of garlic juice was employed, the more growth inhibition was obtained. Comparing the specific growth rate at various concentration of garlic juice, the inhibition effect on yeast appeared much higher than that on bacteria.
