Effects of Water Stress on Leaf Water Potential, Photosynthesis and Root Development in Tobacco Plant (수분 스트레스가 담배의 잎 수분 포텐셜, 광합성 및 뿌리발달에 미치는 영향)

  • Development of shoot and root, leaf water potential and photosynthetic rate affected by water stress in early growing stage of tobacco were surveyed to interpret stress response in terms of plant physiological and agricultural aspects. The growth of shoot and root was highly suppressed by water stress and the difference in dry weight by rewatering was smaller in root than in shoot. The total root length was highly decreased by water stress and the lengths of root for water stress and non-stress were 74m and 84m, respectively, after rewatering. The root growth treated by water stress was increased between 2nd and 3rd day after treatment indicating that temporary water stress at early growing stage might have increased of root zone activity for early growth stage. The leaf water potentials were decreased to -7.63MPa, -9.47MPa, -11.89MPa, -13MPa at the 2nd, 3rd, 4th and 5th day by water stress. The relative water contents were 75%, 62% and 57% at the 3rd, 4th and 5th day after treatment. Photosynthesis was reduced largely by water stress. The photosynthetic rate after treatment at 2nd day and 3rd day was dropped to 18.15$\mu$mol. $CO_2$/$m^2$ㆍsec$^{-1}$ and 9.35$\mu$mol. $CO_2$/$m^2$ㆍsec$^{-1}$. It was never recovered to the normal, even after rewatering. Stomatal conductance had been reduced since 2nd day after treatment and increased after rewatering.

The Growth of Human Osteoblasts in Culture Dishes Made with Poly-glycolic Acid Containing Fetal Bovine Serum (우태아 혈청이 포함된 Poly-glycolic Acid 배양판에서 인간 조골세포의 성장)

  • Purpose: An ideal bony construct can be divided into two broad categories: (1) the design and fabrication of biodegradable, biomimetic scaffolds that provide correct signals to induce osteogenesis: (2) the identification of an ideal source of osteoprogenitor cells to seed onto the scaffold. We selected poly-glycolic acid as a synthetic scaffold among various scaffolds because of these properties. Meanwhile, culture medium is supplemented with fetal bovine serum(FBS): such serum contains essential elements such as proteins, hormones, growth factors and trace minerals. The composition of FBS can be ideal for various cell growth in vitro. We supposed that we could enhance bone growth at a fractured site if FBS was mixed with synthetic scaffold-PGA. Methods: We cultured human osteoblasts in five different prepared culture dishes made with FBS and PGA mixture. The mixtures contained different ratio of FBS, that is, 0, 1.5, 3, 7, and 10%. We cultured human osteoblasts for seven days and examined the growth and attachment of the cells at the 1st, 3rd, 5th, 7th days, respectively. Results: In the mixture of 0% FBS and PGA, the growth of the cells lasted for one day. In 1.5 and 3% FBS and PGA, the growth of the cells was examined at the 3rd day, then minimally declined at the 5th and 7th days. In 7% FBS and PGA, the growth of the cells lasted for 5 days, then declined at the 7th day. In 10% FBS and PGA, the growth of the cells lasted for 5 days, then declined at the 7th day. Staining status of the osteoblasts with alkaline phosphatase showed pale pink color in 0% FBS and PGA groups, but bright pink color in 1.5, 3, 7, 10% FBS and PGA groups, especially in 3%, 7%. Conclusion: In consequence, the growth of human osteoblast was higher in the mixture of FBS and PGA groups than in pure PGA ones. It is assumed that the mixture of FBS and PGA affects the proliferation of human osteoblasts.

  • 1. The analysis of the shoot height growth of Phyllostachys edulis A. et C. Rivi$\acute{e}$re which stand is growing at the middle part of South Korea is presented in the present paper. The shoot height was carefully measured twice a day, 10 am and 4 pm. With this, six hours from 10 am to 4 pm is regarded as day-time-length and 18 hours of the rest as night-length. 2. The measurement were made in 1954. The very young shoots were classed into three diameter groups basing at the ground surface level to see the the afterwards influences of shoot size on height growth. 3. The hourly mean height growth in day time was higher than that in night time. The standard deviations and coefficients of variations of the daily growth of the shoot are presented in Figures 2 and 3 respectively. 4. The hourly mean height growth of the shoot by diameter groups and by day and night period are presented in Fig. 4 and 5 respectively. 5. The growth percentages of the shoot by diameter groups and by day and night is graphed in Fig. 6.

Effects of Temperature and Salinity on Early Development, Survival and Growth Rate in Seabass, Lateolabrax Japonicus (농어, Lateolabrax Japonicus의 초기 발달, 성장 및 생존율에 미치는 수온과 염분의 영향)

  • Effects of temperature (10, 12, 14, 16, 18 and 2$0^{\circ}C$) and salinity (22.0, 24.5, 27.0, 29.5, 32.0 and 34.5 ppt) were studied on incubation period, hatching success, survival and growth of alevin and juvenile seabass, L. japonicus. Embryonic development was accelerated with increasing temperature but it was not influenced by salinity. Hatching success was the highest at 14$^{\circ}C$ and 34.5 ppt. Higher temperature also accelerated the development of mouth opening, absorption of yolk and oil globules, and alevin growth. Survival of the 5-day old Juvenile was accelerated in the following order : 14<16<18<20<12$^{\circ}C$. Rearing experiment of the juvenile for 30-day indicated the faster growth at 13, 20 and 27 ppt than at 34 ppt.

  • This study was to present these results as basic data for the effects of Ayurvedic oil and the development of alopecia treatment programs. The results were as follows: 1) After Ayurvedic treatment, the hair density of vertex and occiput were 264.2 hair/$cm^2$ and 299.8 hair/$cm^2$ each. There was the increase of total 29.2% in hair density. 2) The hair growth rates on vertex and occiput after Ayurvedic treatment were 310.8 ${\mu}m$/day and 345.8 ${\mu}m$/day respectively, there was the increase of total 79% in hair growth.

  • This study examined the effects of growth factors in TCM199 on bovine 1-cell embryos development in vitro. After 6 day to 11 day in culture, 15.8%(19/120), 15.3%(20/130), 21.8%(35/160), 27.0%(56/207), 26.3%(53/201) and 30.7%(40/130) of the 1-cell embryos developed into expanding blastocysts in su, pp.ementing TCM199 with control, insulin, IGF-I, IGF-II, FGF and EGF, respectively. Hatching rate of 1-cell embryos in su, pp.ementing TCM199 with FGF, EGF and IGF-II were 21.4%(53/247), 20.3%(42/206) and 16.8%(41/243), respectively. The beneficial effect of growth factors on embryo development in vitro could be duplicated. These data indicate that the presence of FGF, EGF or IGF-II in the culture medium is beneficial for embryo development in vitro and accelerate cell differentiation.

  • Studies were made to investigate effects of the scale-up of stirred tank bioreactors on cell growth and alkaloids production for suspension cultures of Eschscholtzia californica. In the 1.5 L STR, cell lysis was observed at 110 rpm or higher agitation speed. The agitation speed of 30 L STR was 43.7 rpm to maintain the same shear stress developed in 1.5 L STR of 100 rpm. As a result of scale-up from 1.5 L to 30 L STR, the specific growth rate was decreased from 0.12 to 0.07 day-1. The alkaloids productivity was also decreased from 0.24 to 0.14 mg/L-day. Changes of mixing performance and oxygen transfer were studied to explain the decrease of cell growth and alkaloids production. Decreased oxygen transfer rate coefficient(KLa) and increased mixing time by the scale-up was observed at various aeration rates.

The Production of Algal Hydrocarbons in Outdoor Cultivations of Dunaliella salina 1650

  • In 12:12 hour light/dark cycle cultivation of D. salina 1650, maximum specific growth rate of 0.59 (l/day) and 0.35 (g-crude hydrocarbons/l/day) were obtained. The cell growth was inhibited at above 15$\times$$10^{-4} (kcal/cm^2/h)$ of light intensity in an outdoor cultivation. It was also showed that temperature is one of the critical growth parameters in the outdoor cultivation. The hydrocarbon production from D. salina 1650 seems to be partially growth related production process, and these algal hydrocarbons can be used for subsituting petroleum directly or through cracking processes. The value of weight fraction carbon of D. salina 1650 was similar to that of Botryococcus braunii and so was the hydrocarbon productivity.

Effect of Suckling on Serum Insulin-like Growth Factor- I Levels in the Primiparous Rat (Rat 의 포유조절이 혈중 Insulin-like Growth Factor- I 수준에 미치는 영향)

  • This experiment was conducted to investigate how restricted-lactation and regulation of weaning-time influence the levels of insulin-like growth factor-I (IGF-I) in primiparous rats during the suckling period. All the rats were raised in the individual cage from a few days before parturition through the whole suckling period. The restrictedlactation (RL) and weaned (W) groups were subdivided into 5 subgroups as RL0, RL5, RL10, RL15 and RL20 as well as W0, W5, Wl0, W15, and W20 according to the day of onset of suckling only 4 pups in restriction lactation and of suckling no pups in weaned group, respectively, in contrast to suckling 8 pups per litter in normal lactation (NL) group. The results obtained were summarized as follows: 1. The serum IGF- I concentration of NL rats was 750.59$\pm$3.52ng/$m\ell$ on Day 0, which was not changed until Day 15, and then it was increased through the subsequent suckling period to 1690.20$\pm$4.42ng/$m\ell$ on Day 25. 2. The IGF-I concentrations of early restricted lactation (RL0, RL5) were 1395.90$\pm$3.45ng/$m\ell$ and 1351$\pm$3.23ng/$m\ell$ on Day 10, respeotively. It was significantly higher (P<0.05) than NL group (745.96$\pm$2.24ng/$m\ell$), and then was not different between group from day 15 of lactation. 3. The IGF- I concentration of W-group in the early lactation was higher (P<0.05) than NL group during the first week of lactation. But it was decreased to the similar concentration as NL group on Day 10. These data show that lactation stimulus may regulate the IGF-I concentration.

Measurement of Growth and Chlorophyl in Barley Exposed by X-ray (X선에 노출된 보리의 생장과 엽록소 측정)

  • The purpose of this study is to measure of growth and chlorophyl in barley exposed by X-ray. Barley seed was soaked 24h duration in water, then was classified into two group; pre-seed germination group (Pre-G) or post-seed germination group (Post-G). Also, divided as control subgroup and experimental subgroup(10Gy, 20Gy, 30Gy) in each group. Experimental subgroups were exposed by X-ray using linear accelerator (Clinac IS, VERIAN, USA). Expose condition was 6 MV X-ray, SSD 100 cm, 18×10 cm, 600 MU/min. Length was measured every day for 10 days and 10th day for weight. Chlorophyl was analyzed using spectrophotometer(uv-1800, shimadzu, japan) in l0th day. Data analysis was performed using SPSS ver 22.0(Chicago, IL, USA), ANOVA test (Dunnett_T3) between control subgroup and experimental subgroup in group and Independent T-test between Pre-G and Post-G in subgroup. In Pre-G, length of barley was significantly difference between control and 30Gy in 4th day (4.3 vs. 1.5, p= 0.011). Length of 30Gy was statistical difference with control(10th day; 14.4 vs. 6.3, p < 0.01), and was not in 10Gy or 20Gy in all day. In experimental subgroup, length was shorter as increasing radiation dose. In Post-G, length of barley was not difference statistically between control and experimental subgroup in first day, but more difference between two subgroup with increasing duration after exposing. Length of experimental subgroup was shorter significantly compared with control in 10th day, and no significant difference between experimental subgroup. Density of chlorophyl was increasing with increasing radiation dose in Pre-G and Post-G. Chlorophyl density of control was lower than 30Gy; 0.26ppm in Pre-G, 0.29ppm in Post-G). Growth and chlorophyl of barley was effected by X-ray. It is expected to be used as basic data for future radiobiological research.
