Manufacturing Fermented Rapeseed Meal Compost using Two Microbial Agents and the Effect of Their Application (유용 미생물 제제 이용 발효 유채박 비료 제조 및 시용 효과)

  • Rapeseed meal, which is a byproduct of rapeseed oil extraction, improves crop productivity by supplying nutrients to the soil. The present study aimed to manufacture fermented rapeseed meal compost using two effective microbial agents and evaluate their efficiency as fertilizer. To types of fermented rapeseed meal, manufactured using either a bio-carrier or microbial agent, showed no differences in pH, electrical conductivity (EC), and total nitrogen content. However, the contents of $NH_4-N$ and $NO_3-N$ as inorganic nitrogen were increased by 5.6 times and 1.5 times, respectively, after 5 d of fermentation. Rapeseed meal fermented for 5 d was applied to tomato a basal fertilizer and after eight weeks, the plant height increased in all fermented rapeseed treatments compared to that in the chemical fertilizer treatment, and also the quantum yield of photosystem II (PS II) showed the same trend. The total nitrogen content of tomato leaves treated with a microbial fermented rapeseed meal was twice as high as that of that treated with a chemical fertilizer. It was confirmed that the increase in the tomato height was an effect of the rapeseed meal containing inorganic nitrogen, which can easily be absorbed by plants. From these results, it is considered that fermented rapeseed meal manufactured with an effective microbial agent for 5 d showed the highest inorganic nutrient content and greatest growth enhancement in tomato.

  • The multiple ear and tiller line (MET), which was selected from a local maize collection and has been selected for earliness by selfing for five generations, was compared its plant characteristics with four U.S. leading hybrids. 1. The general plant growth was a little retarded by poor soil fertility and structure. 2. The fresh weight of the MET line was about 97.8% of the U.S. check varieties. 3. The dry weight of the MET line was also about 94.5% of the check U.S. hybrids (1,695 kg / 10a.). 4. The tillering and eating habits of the MET line had contributed toward increasing fresh and dry weight of the MET line. 5. The average plant height of the MET line was much less than those of the check varieties, due to the selfing depression manifested by the MET line, (234.7cm. of the MET vs 250.7cm. of the check) 6. Apparent differences of disease resistance was observed among varieties studied. The degree of plants infected with the black stripe mosaic virus was over 20% for the check U.S. hybrids, while less than 5% of the MET line was infected with the disease. 7. The grain yield per 10 a of the MET line was far below the check hybrids due to the inbreeding depression of the MET line. However, the kernel number per unit area of the MET line with small size kernel was much greater than that of the check hybrids. The 100 kernel weight of the MET line was around 10 grams. 8. The average fresh and dry matter weight of the three ($sorghum{\times}sudan$ grass) varieties was very much alike with those of the MET line. 9. In conclusion, the use of the MET line for silage production may have dual adventages because of its low seed price and of its small size kernels, which may reduce the amount of seeds required for planting in an unit area.

  • Experiments on the physiological root activity and its related characteristics of rice varieties were carried out in order to obtain some basic informations for the application of the results obtained to a rice breeding program. A significant positive correlation was found not only among the various characteristics related to conducting and ventilating systems which connects top and root of rice plant, but also between these characteristics and root activity. On the other hand, a significant difference in physiological root activity was recognized among different varieties and also between different groups of recognized 7 rice varieties differing in the their origin. It was also found that varieties with higher root activity (root activity indices) after ear formation stage tended to have more number of lower green leaves and consequently resulted in higher grain yield. Therefore, it may be possible to diagnose indirectly the root activity by examining the number of green leaves of the rice plant at later growth stage when breeders make selections of parent material for crossing or of hybrid lines in pedigree nurseries.

Application Amount of Anaerobic Digestion Waste Water from Methane Fermentation of Pig Manure on Rice (벼에 대한 돈분뇨 혐기성 소화액비의 시용적량 구명)

  • This study was carried out to evaluate the proper application amount of anaerobic digestion waste water and the environmental influence on rice. The waste water collected after methane fermentation process of pig manure was used as a liquid manure. Liquid manure 100%+chemical fertilizer 30%(LM 100%+CF 30) treatment was the most favorable at all growth stages of rice. The LM 100%+CF 30% treatment was applied to 100% amount of liquid manure which was correspond to the same amount of nitrogen for the standard application amount on rice, with adding 30% amount of chemical fertilizer(urea) at tillering stage. The yields of rice in the treatments of 100%(LM 100%) and 150% amount(LM 150%) of liquid manure were similar or a little higher than NPK treatment but LM 100%+CF 30% treatment was less than the NPK treatment due to the increase of straw weight and plant lodging. In periodic changes of the $NH_4-N$ and $NO_3-N$ contents, the LM 70%+CF 30% treatment in paddy soil was the highest in all treatments. The NPK and the LM 100% treatments in irrigation water quality were higher than other treatments. In infiltration water quality, $NH_4-N$ content was leached out much in the LM 150% treatment and $NO_3-N$ content was in the LM 100%+CF 30% treatment. The proper application amount of anaerobic digestion waste water as a liquid manure must be to analyse the nitrogen content of the waste water and to apply the same amount of nitrogen for the standard application amount on rice.

  • Experiments were conducted by moderate season culture of each of 7 early, medium and late maturing varieties which were considered to be of strong salt tolerance in non-salty, low- and high-salty reclaimed areas (salt content: 0.5% and 1% at the end of April). The results of the experiments revealed that the early maturing varieties were suffered greatly from salt damage in such reclaimed salty area which is the "fall better type" in the year-round changes of soil salt concentration in Korea. Better yielding varieties in the salty area showed generally a high yield ratio in the salty area compared with the non-salty area. This indicates that they are strong salt tolerant varieties. It seemed that the salt tolerance could be measured by the number of delaying days in heading, the growth rate of culm length, number of panicles and number of grains per panicle in the salty area to compare with the non-salty area. The effects or salt damage on the yields of each variety at maturing stage and agronomic characteristics in the salty area were compared and discussed with the non-salty area.alty area.

  • 'Goami 3' is a new japonica rice cultivar developed from a cross between Suweon464 and Daeanbyeo by the rice breeding team of National Institute of Crop Science, RDA. 'Goami3' has about 130 days growth duration from transplanting to heading in central plain area of Korea. It has a good semi-erect plant type and resistance to lodging of about 79 cm in culm length. 'Goami 3' had 15 panicles per hill and 104 spikelets per panicle. 'Goami 3' has very high amylose (29.5%) and high non-dietary starch compared with Hwaseongbyeo. This rice variety has slow senescence and 47% tolerance to viviparous germination during the ripening stage. 'Goami 3' is susceptible to leaf blast, bacterial blight, virus disease and insect pest. The yield performance of this cultivar in milled rice was about 3.92MT/ha by ordinary season culture in local adaptability test from 2005 to 2007. 'Goami 3' is adaptable to central and southern plain area of Korea.

  • Haiami is a new Japonica rice variety developed from a cross between 'Jinmibyeo' TR treated with ethyl methane sulfonate (EMS) EMS and 5-methytryptophan, and 'Gyehwabyeo' in order to develop a new premium quality rice variety by a rice breeding team of National Institute of Crop Science, Rural Development Administration in 2008. This variety has about 138 days of growth duration from transplanting to harvesting in central plain area of Korea. The heading date of this vareity was on $15^{th}$, August. The 'Haiami' has good semi-elect plant type and resistant to lodging with strong culm. The number of panicles/hill of 'Haiami' is more than that of 'Hwaseongbyeo'. This variety shows slow leaf senescence and considerable tolerance to viviparous germination. It is susceptible to leaf blast, bacterial blight, and insect pests, but resistance to rice stripe virus. The milled rice of this variety exhibited translucent, clear non-glutinous endosperm and short grain shape. The essential amino acid properties of 'Haiami' have more than 31% that of 'Hwaseongbyeo' in polished rice. This variety has premium palatability of cooked rice. The yield performance of this rice cultivar was about 5.38 MT/ha in milled rice in local adaptability test for three years from 2006 to 2008. 'Haiami' is adaptable to central and southern plain areas of Korea.

Physiological Response of Potato Variety to Soil Salinity (토양염농도에 따른 감자 품종들의 생장특성)

  • This study was conducted to examine the basis for the information to select the suitable potato varieties grown in new reclaimed land. The potatoes of five varieties were planted in the port with 4 different electrical conductivities of saturated extracts of soil taken the Saemangeum reclamation area, which was made of non-treatment salt and three concentrations of salt treatment, 1.6 dS $m^{-1}$, 3.2 dS $m^{-1}$, 4.8 dS $m^{-1}$, respectively. All of the potato varieties were uniformly emerged without missing plant in all treatment groups, even 4.8 dS $m^{-1}$ treatment group. According to the salt concentration of soil, required date to the emergence of the potato comparing to non-treatment salt was delayed 3-4 days in 1.6 dS $m^{-1}$, 6-10 days in 3.2 dS $m^{-1}$, 7-13 days in 4.8 dS $m^{-1}$, respectively, and the number of its branch decreased by 14-58% comparing to non-treatment salt depending on varieties. Since the increase of the salt concentration of the soil was more serious the decrease of the number of its branch, but plant height tended to increase when branch number per plant was small, which was depending on more number of its branch than salt concentration. Fresh tuber yield of potato comparing to non-treatment salt were decrease 33.7% in 1.6 dS $m^{-1}$, 59.5% in 3.2 dS $m^{-1}$, 79.3% 7-13 days in 4.8 dS $m^{-1}$, respectively. The threshold EC starting the growth inhibition of fresh weight decreased was 1.2 dS $m^{-1}$ for Chudong, 1.8 $m^{-1}$ for Chubeak, 1.9 $m^{-1}$ for Chugang and Chuyeong, and 2.0 $m^{-1}$ for Sumi, and EC which decreased 50% of dry weight index was 2.4 dS $m^{-1}$ for Chubaek, 2.45 dS $m^{-1}$ for Chudong, 2.81 dS $m^{-1}$ for Chugang, 3.03 dS $m^{-1}$ for Chuyeong, and 3.29 dS $m^{-1}$ for Sumi. The present results suggest that Sumi is considered to the suitable potato variety grown on saline soils.

  • trans-Resveratrol(3,5,4'-trihydroxystilbene) is phenolic compound present in grapes, wines, and peanuts, has been reported to have health benefits including anticarcinogenic effects, protection against cardiovascular diseases and reduced cancer risk. A simple method for the quantitative extraction of trans-resveratrol from peanut has been developed. Optimal conditions for extraction were investigated. Type of solvent, time, and temperature assayed influenced trans-resveratrol yield. Adequate extraction condition was decided to ethanol/water (80:20v/v) maintained at $25^{\circ}C$ for 45 min. After extraction, the protocol consists of sample preparation using a $\textrm{C}_{18}$ solid-phase extraction (SPE) cartridge after concentrate with rotary evaporator and quantified by reversed phase HPLC using a $\textrm{C}_{18}$ column at 308 nm. Analytical methods for measuring trans-resveratrol in peanut were adapted to isolate, identify, and quantify trans-resveratrol in 11 peanut varieties by HPLC (high-performance liquid chromatography) with UV detector, The 11 peanut varieties content ranged from 0.018 to 1.125 $\mu\textrm{g}/\textrm{g}$ with an average of 0.289 $\mu\textrm{g}/\textrm{g}$. The contents were higher in the seeds with than without testa, regardless of varieties. The trans-resveratrol content was Higher in 110, 130 days after sowing than that of other period.

Quantification of Isoflavone Malonylglucosides in Soybean Seed during Germination (콩 발아 중 isoflavone malonylglucosides의 함량 변이)

  • Soybean produces three major types of isoflavones, daidzein, genistein, and glycitein aglycones and their glucosides and malonylglucosides. It has been known that malonylated glucosides are rapidly converted to their corresponding aglycones due to the unstable thermolabile glucoside malonates; therefore, the analytical study of malonylated glucosides has been insufficient. In this study, we analyzed the malonylglucoside content in soybean seeds. Isoflavone analysis of three soybean cultivars revealed that 81.5~90.0% of the total isoflavones were malonylglucosides, whereas aglycones were rarely detected. Moreover, the total isoflavone content increased during a 5-day germination period where growth regulators and coumaric acid treatments tended to yield higher isoflavone content than the normal germination treatment, however the differences were not significant; notably, the isoflavone accumulation trend continued with additional germination days. The content of malonylglucoside was higher than that of other isoflavones, which was 83.7~86.6% of the total isoflavone content in seeds with a 3-day germination period. Furthermore, isoflavones were significantly accumulated in the hypocotyl of seedlings with a 5-day germination period. The content of isoflavone in the hypocotyl of the Pungsannamul-kong was 10,240 ug/g when treated with coumaric acid, which was considerably higher than that of other cultivars and treatments. Additionally, soybean seeds heated at $60^{\circ}C$ for 1 hour produced higher isoflavone content than non-heated soybean seeds. Our results show that it is possible to increase the isoflavone content in soybean seeds through various treatments.
