시판되고 있는 녹차잎을 polyethylene 필름으로 진공포장하여 전자빔을 $0{\sim}30\;kGy$로 조사하였다. 조사된 녹차잎을 $75^{\circ}C$의 물에서 10분간 추출하여 녹차를 제조하고 물리화학적 특성을 분석하였다. 전자빔 조사는 녹차의 총 페놀 함량(TPC), 총 플라바놀 함량(TFC), 아스코르브산 함량(AAC)을 감소시켰다. 20 kGy의 전자빔 조사는 녹차의 TPC, TFC, AAC를 대조구와 비교하여 각각 223.46, 32.50, 6.03 mg/g에서 202.88, 31.16, 5.57 mg/g으로 감소시켰다. 전자빔 조사는 또한 카테킨, 카페인, 아질산염 소거능도 감소시켰다. 그러나 전체적인 색도 및 라디칼 소거능은 전자빔 조사에 의해 영향을 받지 않았다.
본 실험에서는 DPPH radical 소거능을 통하여 차잎의 수확시기 및 발효정도에 따른 항산화 활성에 있어서 변화를 비교하고 일상의 녹차 음용으로 섭취되는 항산화 물질을 EGCG 등의 함량으로 추정하고자 하였다. 우전과 같이 수확시기가 빠른 것이 다소 높은 항산화 활성을 보였고, 특히 볕가림 재배한 옥로가 일반 녹차에 비 해 적어도 비슷한 활성을 나타냈다. 중국의 반발효차에 있어서 발효정도에 따라 항산화 활성이 감소한데 비해 완전히 발효된 흥차의 경우 항산화 활성이 녹차의 수준으로 유지하였고, 이러한 홍차의 항산화 물질에 대해서는 보다 체계적 인 연구가 필요하리라 생각된다. 분량 2그램의 녹차를3회 우려 마시는 경우 이로부터 섭취할 수 있는 항산화 물질은 EGCG 기준으로 109∼147 mg정도이고 비타민 C로는 142∼168 mg에 해당한다. 시판되고 있는 녹차티백은 제품에 따라 다소 차이를 보였으나 한 티백으로부터 52∼86 mg정도의 EGCG에 해당하는 항산화 활성을 보였는데 비타민 C로는 63∼96 mg에 해당한다. 증열과정이 긴 냉녹차나 차잎의 분량이 적은 현미 녹차가 낮은 항산화 활성을 보였고, 감잎차, 뽕잎차, 솔잎차에서도 적으나마 항산화 활성이 보인 반면 식 물의 뿌리 인 둥굴레 차는 항산화 활성 이 거의 없다. 전통적 인 방법으로 차를 3회 우려 마시는 경우 차잎 속에 포함된 항산화 물질의 절반정도가 용출되는 셈인데 가능한 분말차로 사용되는 것이 경제적일 수 있다. 세번째 우린 찻물에도 상당량의 항산화 물질이 함유된 반떤, 녹차 티백의 경우 3분이면 충분한 항산화 물질이 용출되는 것을 알 수 있다. 식품 속의 플라보노이드가 섭취되어 실제로 어느 정도 흡수되는가에 대해서 아직 이견이 있고 플라보노이드의 임상적인 효과에 대해 확실하지 않으나 역학적인 근거로는 긍정적이다. 천연의 항산화 물질을 자연스럽게 섭취하는 방법으로서 차의 음용을 생활화하는 것은 퇴행성 또는 노화관련 질환의 예방차원에서 바람직하리라 본다.
본 연구에서는 추출보조공정으로서 초음파 처리에 따른 녹차의 성분특성 변화를 분석하기 위하여,$60^{\circ}C$에서 1, 5, 30, 60분간 열수추출하고, 동일온도에서 초음파 추출하여 각각 가용성 고형분, vitamin C, 총 페놀성 화합물, flavonols, catechins, caffeine, free amino acids 함량을 분석하였다. 가용성 고형분 함량은 초음파 처리에 의해 약 4~5%의 추출수율이 증가하였다. Viamin C 함량은 1, 5, 30분에서는 각각 약 0.21, 0.16, 0.31 mg/g 증가하였지만, 60분 처리구에서는 대조구와 비교해 2.47 mg/g에서 2.22 mg/g으로 감소하였고, 총 페놀성 화합물 함량이 약 10~13 mg/g 증가하였다. Flavonols는 myricetin이 4.77 mg/100g에서 9.94 mg/100g, quercetin은 10.97 mg/100g에서 20.84 mg/100g, kaempferol은 7.95 mg/100g에서 17.33 mg/100g으로 초음파 처리에 의해 약 2배 이상 증가함을 알 수 있었다. 초음파처리에 의해 녹차 침출액의 EGCG, EGC, ECG, EC, (+)-C 함량이 증가하는 경향을 보였다. 이러한 경향과는 달리, 유리아미노산의 경우는 초음파 처리에 따른 영향이 거의 없었고, 추출시간이 길어짐에 따라 증가하는 경향을 보였다. 결론적으로 초음파 처리에 의해 녹차의 vitamin C, 총 페놀 화합물, flavonol류, catechins의 추출이 증가함을 알 수 있었고, 따라서 녹차 가공에 있어 초음파 처리가 매우 효율적인 추출보조공정으로 사용될 수 있으리라 사료된다.
This study was designed to evaluated the nitrite scavenging activity and protective effect of the Puerariae Radix and green tea extract on lead acetate and cadmium-induced liver damage in mice. The quantitative analytical method for major antioxidants, isoflavones, puerarin, catechine and caffeine in galgun (Puerariae Radix) and green tera extract were established by HPLC. Contents of isoflavones, such as daidzin, genistin, daidzein and genistein were 4.23g/100g, 0.13g/100g, 0.07g/100g, and 0.03g/100g, and puerarin contents was 8.99g/100g, respectively. The total catechins and caffeine contents of green tea extract were 49.24g/100g and 6.53g/100g. The nitrite scavenging ability of galgun extract (pH 1.2, 100mg/ml) was 98.07% and it was higher than those of other extracts. It was the highest at the pH 1.2 and more than 64% in 25~100mg green tea extract, and was dependents on pH and concentration of the samples. The hepatoprotective effects of an aqueous extract from the root of gal gun and standard puerarin were evaluated against lead acetate and cadmium-induced liver damage in mice. Galgun extract and standard at a dose of 100mg/kg and 10mg/kg, 50mg/kg were administered orally once daily for successive 5 days and then a lead acetate and cadmium were orally at 3 hrs after the every day administration of galgun. The substantially elevated serum enzymatic activities of alanine and aspartate aminotransferase were due to lead acetate and cadmium treatment was dose dependently restored to the near normal level. In addition, galgun extract also significantly prevented the elevation of hepatic malon-dialdehyde formation in the liver of lead acetate and cadmium intoxicated mice in a dose-dependent manner. The results of this study clearly indicated that green tea and galgun extracts had nitrite scavenging ability and galgun extract had potent hepato-protective effects against lead acetate and cadmium-induced hepatic damage in mice and standard puerarin was also showed similar to the results of the galgun extracts.
A cumulative evidence indicates that consumption of tea catechin, flavan-3-ol derived from green tea leaves, lowers the risk of cardiovascular diseases. However, a precise mechanism for this cardiovascular action has not yet been fully understood. In the present study, we investigated the effects of different green tea catechins, such as epigallocatechin-3 gallate (EGCG), epigallocatechin (EGC), epicatechin-3 gallate (ECG), and epicatechin (EC), on angiotensin II (Ang II)-induced hypertrophy in primary cultured rat aortic vascular smooth muscle cell (VSMC). [$^3H$]-leucine incorporation was used to assess VSMC hypertrophy, protein kinase assay, and western blot analysis were used to assess mitogen-activated protein kinase (MAPK) activity, and RT-PCR was used to assess c-jun or c-fos transcription. Ang II increased [$^3H$]-leucine incorporation into VSMC. However, EGCG and ECG, but not EGC or EC, inhibited [$^3H$]-leucine incorporation increased by Ang II. Ang II increased phosphorylation of c-Jun, extracellular-signal regulated kinase (ERK) 1/2 and p38 MAPK in VSMC, however, EGCG and ECG , but not EGC or EC, attenuated c-Jun phosphorylation increased by Ang II. ERK 1/2 and p38 MAPK phosphorylation induced by Ang II were not affected by any catechins. Ang II increased c-jun and c-fos mRNA expression in VSMC, however, EGCG inhibited c-jun but not c-fos mRNA expression induced by Ang II. ECG, EGC and EC did not affect c-jun or c-fos mRNA expression induced by Ang II. Our findings indicate that the galloyl group in the position 3 of the catechin structure of EGCG or ECG is essential for inhibiting VSMC hypertrophy induced by Ang II via the specific inhibition of JNK signaling pathway, which may explain the beneficial effects of green tea catechin on the pathogenesis of cardiovascular diseases observed in several epidemiological studies.
Cancer preventive activities of green tea and its main constituent, (-)-epigallocatechin gallate (EGCG) have been extensively studied by scientists all over the world. Since 1983, we have studied the cancer chemopreventive effects of EGCG as well as green tea extract and underlying molecular mechanisms. The first part of this review summarizes groundbreaking topics with EGCG and green tea extract: 1) Delayed cancer onset as revealed by a 10-year prospective cohort study, 2) Prevention of colorectal adenoma recurrence by a double-blind randomized clinical phase II trial, 3) Inhibition of metastasis of B16 melanoma cells to the lungs of mice, 4) Increase in the average value of Young's moduli, i.e., cell stiffness, for human lung cancer cell lines and inhibition of cell motility and 5) Synergistic enhancement of anticancer activity against human cancer cell lines with the combination of EGCG and anticancer compounds. In the second part, we became interested in cancer stem cells (CSCs). 1) Cancer stem cells in mouse skin carcinogenesis by way of introduction, after which we discuss two subjects from our review on human CSCs reported by other investigators gathered from a search of PubMed, 2) Expression of stemness markers of human CSCs compared with their parental cells, and 3) EGCG decreases or increases the expression of mRNA and protein in human CSCs. On this point, EGCG inhibited self-renewal and expression of pluripotency-maintaining transcription factors in human CSCs. Human CSCs are thus a target for cancer prevention and treatment with EGCG and green tea catechins.
Sj$\"{o}$gren's syndrome (SS) is an autoimmune disorder that affects the salivary glands, leading to xerostomia, and the lacrimal glands, resulting in xerophthalmia. Secondary SS is associated with other autoimmune disorders such as systemic rheumatic diseases and systemic lupus erythematosis (SLE), which can affect multiple organs, including the epidermis. Recent studies have demonstrated that green tea polyphenols (GTPs) possess both anti-inflammatory and anti-apoptotic properties in normal human cells. Epidemiological evidence has indicated that, in comparison to the United States, the incidence of SS, clinical xerostomia and lupus is considerably lower in China and Japan, the two leading green tea-consuming countries. Thus, GTPs might be responsible, in part, for geographical differences in the incidence of xerostomia by reducing the initiation or severity of SS and lupus. Consistent with this, molecular, cellular and animal studies indicate that GTPs could provide protective effects against autoimmune reactions in salivary glands and skin. Therefore, salivary tissues and epidermal keratinocytes could be primary targets for novel therapies using GTPs. This review article evaluates the currently available research data on GTPs, focusing on their potential application in the treatment of the oral manifestations of SS and skin manifestations of SLE.

Green tea (GT) polyphenols undergo extensive metabolism within gastrointestinal tract (GIT), where their derivatives compounds potentially modulate the gut microbiome. This biotransformation process involves a cascade of exclusive gut microbial enzymes which chemically modify the GT polyphenols influencing both their bioactivity and bioavailability in host. Herein, we examined the in vitro interactions between 37 different human gut microbiota and the GT polyphenols. UHPLC-LTQ-Orbitrap-MS/MS analysis of the culture broth extracts unravel that genera Adlercreutzia, Eggerthella and Lactiplantibacillus plantarum KACC11451 promoted C-ring opening reaction in GT catechins. In addition, L. plantarum also hydrolyzed catechin galloyl esters to produce gallic acid and pyrogallol, and also converted flavonoid glycosides to their aglycone derivatives. Biotransformation of GT polyphenols into derivative compounds enhanced their antioxidant bioactivities in culture broth extracts. Considering the effects of GT polyphenols on specific growth rates of gut bacteria, we noted that GT polyphenols and their derivate compounds inhibited most species in phylum Actinobacteria, Bacteroides, and Firmicutes except genus Lactobacillus. The present study delineates the likely mechanisms involved in the metabolism and bioavailability of GT polyphenols upon exposure to gut microbiota. Further, widening this workflow to understand the metabolism of various other dietary polyphenols can unravel their biotransformation mechanisms and associated functions in human GIT.
분말 녹차를 상대습도(23, 69, 81%)와 온도(-20, 4, $20^{\circ}C$)를 달리하여 3달간 저장한 후, 1.5 g에 100 mL을 첨가하여 추출물을 제조하여 총 페놀 함량, 총 플라바놀 함량, 아스코르브산 함량, 카테킨 함량의 변화를 조사하였다. 상대습도 23%, $4^{\circ}C$의 조건에 저장한 경우에서 총 페놀 함량, 총 플라바놀 함량, 아스코르브산 함량이 가장 높았는데 각각 287.1mg/g, 44.9 mg/g, 36.9 mg/g의 값을 보였다. 이는 저장 초기의 각각의 측정값인 267.5 mg/g, 49.4 mg/g, 24.2 mg/g에 비해 총 페놀과 아스코르브산은 증가하였고, 총 플라바놀 함량은 감소하였다. 카테킨류의 경우에도 상대습도 23%, $4^{\circ}C$에 저장하였을 때 가장 높았는데 주된 카테킨인 EGC와 EGCG는 초기의 16.9와 27.3 mg/g에서 3달 후에는 각각 24.3과 36.5 mg/g으로 증가하였다. 또한 카페인도 같은 조건에서 초기의 20.7 mg/g에서 3달 후에는 30.7 mg/g으로 증가하였다. 그러나 상대습도가 높고 온도가 $-20^{\circ}C$나 $20^{\circ}C$에 저장한 경우에는 상대적으로 각 성분들이 감소하는 경향을 나타내었다. 이러한 결과는 분말 녹차의 상대습도와 저장 온도가 녹차의 품질 보존에 매우 중요하며, 낮은 습도에서 냉장 상태로 저장하는 것이 녹차의 품질 유지에 유리함을 의미한다.
