Gene Cloning and Characterization of an ${\alpha}$-Amylase from Alteromonas macleodii B7 for Enteromorpha Polysaccharide Degradation

  • Enteromorpha polysaccharides (EP) extracted from green algae have displayed a wide variety of biological activities. However, their high molecular weight leads to a high viscosity and low solubility, and therefore, greatly restrains their application. To solve this problem, bacteria from the surface of Enteromorpha were screened, and an Alteromonas macleodii strain B7 was found to be able to decrease the molecular weight of EP in culture media. Proteins harvested from the supernatant of the A. macleodii B7 culture were subjected to native gel electrophoresis, and a band corresponding to the Enteromorpha polysaccharide lyase (EPL) was detected by activity staining. The enzyme identity was subsequently confirmed by MALDI-TOF/TOF mass spectrometry as the putative ${\alpha}$-amylase reported in A. macleodii ATCC 27126. The amylase gene (amySTU) from A. macleodii B7 was cloned into Escherichia coli, resulting in high-level expression of the recombinant enzyme with EP-degrading activity. AmySTU was found to be cold-adapted; however, its optimal enzyme activity was detected at $40^{\circ}C$. The ${\alpha}$-amylase was highly stable over a broad pH range (5.5-10) with the optimal pH at 7.5-8.0. The highest enzyme activity was detected when NaCl concentration was 2%, which dropped by 50% when the NaCl concentration was increased to 16%, showing an excellent nature of halotolerance. Furthermore, the amylase activity was not significantly affected by tested surfactants or the presence of some organic solvents. Therefore, the A. macleodii strain B7 and its ${\alpha}$-amylase can be useful in lowering EP molecular weight and in starch processing.

Beryllium(II) Recognition by Allosteric Effects in 1,2-Ethylenedioxybenzene Based Ditopic Receptors

  • Efficient ditopic receptor, uranyl(II) N,N'-(ethylenedioxy)benzenebis(salicylideneimine) (3) for beryllium ion has been obtained upon functionalization of 1,2-ethylenedioxybenzene (1) with a uranyl-salphen (salphen = N,N'-phenylenebis(salicylideneimine)) unit. Binding affinities of the receptor, 3 in AN-DMSO (v/v 95:5) solution have been measured for alkali and alkaline earth metal ions by conductometry comparing 1. The results showed that both monotopic 1 and ditopic receptor 3 were selective for $Be^{2+}$ ions over other cations, while especially 3 that can complex both with cations (coordinated to basic oxygen of ethylenedioxybenzene) and anions (coordinated to the Lewis acidic uranyl center) results in an increase of the stability constants by a factor of $10^{2.42}$ with respect to 1. Furthermore, the $Be^{2+}$-3 interactions are demonstrated by $^1H$ NMR experiments in highly polar solvent medium, DMSO-$d_6$. Higher selectivities were also observed for $Be^{2+}$ when the ditopic receptor, 3 was incorporated into PVC membranes and tested as ion selective electrodes at neutral pH.

Identification and Characteristics of Lactic Acid Bacteria Isolated from Shellfishes (패류로부터 젖산 세균의 분리 및 특성)

  • Lactic acid is an important product arising from the anaerobic fermentation by lactic acid bacteria (LAB). It is used in the pharmaceutical, cosmetic, chemical, and food industries as well as for biodegradable polymer and green solvent production. The poly lactic acid (PLA) is an important material for bio-plastic manufacturing process. For PLA production by new LAB, we screened LAB isolates from shellfish. A total of 28 LAB were isolated from various shellfishes. They were all Gram positive, oxidase and catalase negative. Based on API 50CHL kit, 7 strains among the 28 isolates were identified as Lactobacillus plantarum, 6 strains as Lactobacillus delbrueckii, 5 strains as Leuconostoc mesenteroides, 3 strains as Lactobacillus brevis, 2 strains as Lactococcus lactis, 1 strain as Lactobacillus salivarius, 1 strain as Lactobacillus paracasei, 1 strain as Lactobacillus pentosus, 1 strain as Lactobacillus fermentum and 1 strain as Pediococcus pentosaceu. Also, we examined the amount of total lactic acid produced by these new strains by HPLC analysis with Chiralpak MA column. One strain E-3 from Mytilus edulis was indentified as Lactobacillus plantarum and found to produce 20.0 g/L of D-form lactic acid from 20 g/L of dextrose. Further studies are underway to increase the D-lactic acid production by E-3.

Analysis of metabolites in wheat roots in response to salinity stress

  • Salinity stress is one of the most important abiotic stresses and severely impairs plant growth and production. Root is the first site for nutrient accumulation like as $Na^+$ in the plant. To investigate the response of wheat root under salinity stress, we executed the characterization of morphology and analysis of metabolites. Wheat seeds cv. Keumgang (Korean cultivar) were grown on the moist filter paper in Petri dish. After 5 days, seedlings were transferred to hydroponic apparatus at 1500 LUX light intensity, at $20^{\circ}C$ with 70% relative humidity in a growth chamber. Seedlings (5-day-old) were exposed to 50mM, 75mM, 100mM NaCl for 5 days. Ten-day-old seedlings were used for morphological characterization and metabolite analysis. Root and leaf length became shorter in high NaCl concentration compared to following NaCl treatment. For confirmation of salt accumulation, wheat roots were stained with $CoroNa^+$ Green AM, and fluoresce, and the image was taken by confocal microscopy. $Na^+$ ion accumulation rate was higher at 100mM compared to the untreated sample. Furthermore, to analyze metabolites in the wheat root, samples were extracted by $D_2O$ solvent, and extracted sample was analyzed by 1H NMR spectroscopy. Fourteen metabolites were identified in wheat roots using NMR spectroscopy. Methanol and ethanol were up-regulated, whereas formate, aspartate, aminobutyrate, acetate and valine were down-regulated under salinity stress on roots of wheat. Fumarate had no change, while glucose, betaine, choline, glutamate and lactate were unevenly affected during salinity stress.

Solution-processible Inorganic-organic Hybrid Bipolar Field Effect Transistors

  • Solution-processible hybrid bipolar field effect transistors (HBFETs) with balanced hole and electron mobilities were fabricated using a combination of the organic p-type poly (3-hexylthiophene) (P3HT) layer and inorganic n-type ZnO material. The hole and electron mobilities were first optimized in single layer devices by using acetonitrile as a solvent additive to process the P3HT and annealing to process the ZnO layer. The highest hole mobility of the P3HT-only-devices with 5% acetonitrile was 0.15 cm2V-1s-1, while the largest electron mobility was observed in the ZnO-only-devices annealed at $200^{\circ}C$ and found to be $7.2{\times}10-2cm2V-1s-1$. The inorganic-organic HBFETs consisting of P3HT with 5% acetonitrile and ZnO layer annealed at $200^{\circ}C$ exhibited balanced hole and electron mobilities of $4.0{\times}10-2$ and $3.9{\times}10-2cm2V-1s-1$, respectively. The effect on surface morphology and crystallinity by adding acetonitrile and thermal annealing were investigated through X-ray diffraction and atomic force microscopy (AFM). Our findings indicate that techniques demonstrated herein are of great utility in improving the performance of inorganic-organic hybrid devices

Metabolism of YH1885 by Rat, Dog, Monkey and Human Liver S9 Fractions

  • YH 1885 (5,6-dimethyl -2-(4-fluorophenylamino)-4-(1-methyl -1,2,3,4-tetrahydroisoquinolin -2- yl) pyrimidine hydrochloride) was developed as an antiulcer drug. The objective of this study was to examine a comparative metabolism of YH1885 in rat, dog, monkey and human liver tissues and to determine the metabolite profiles produced by the four species. YH1885 was metabolized by liver 59 fractions from all four species. Control incubations containing 59 fraction but no cofactors, contained essentially no metabolites. Metabolism of YH1885 apparently became saturated in the concentration range studied because the % of YH 1885 metabolized decreased with increasing drug concentration for all four species. Six to nine metabolite peaks were detected in the incubations and the particular profile of metabolites varied with species. The total amount of metabolites formed by liver microsomes from human and monkey were less than microsomes from rat or dog. The major metabolite peak formed by rat liver 597actions fluted near the solvent front on the HPLC or remained at the origin in TLC, indicating that it contained one or more polar metabolites. Dog liver 59 fractions incubations contained four major metabolites that each accounted for about 15 to 20 % of the total radioactivity at the low concentration of YH1885. The metabolite profiles of YH1885 appeared to be similar in incubations with rhesus monkey and human liver 59 fraction. The amount of metabolites formed by rhesus monkey liver preparations was greater than that of human liver that contained prominent metabolite peaks with approximate relative retention time of 0.14 and 0.43.

  • The representation of color with natural dyes is closely related with the modern well-being life. This study is focused on the representation of target color of the blue, yellow, and red color system, these color sources were originated from the green tea, gardenia, and black rice respectively. The quantitative numerical value of brightness (L), color coordinates (a, b) were designated for the target color, the optimum color nearest to the target value was extracted with the solvent pH, temperature, and extraction time. The longer the extraction time had more thickening color in case of the gardenia. The L-a-b coordinate of extracted color were analyzed with the color difference meter and compared with the target color. The experimental variables were optimized to extract the color of the smallest ${\Delta}E$ with the target color. As a result, the CIE standard value was proposed and the color was represented.

Electron Trapping and Transport in Poly(tetraphenyl)silole Siloxane of Quantum Well Structure

  • A new kind of organic-inorganic hybrid polymer, poly(tetraphenyl)silole siloxane (PSS), was invented and synthesized for realization of its unique charge trap properties. The organic portions consisting of (tetraphenyl)silole rings are responsible for electron trapping owing to their low-lying LUMO, while the Si-O-Si inorganic linkages of high HOMO-LUMO gap provide the intrachain energy barrier for controlling electron transport. Such an alternation of the organic and inorganic moieties in a polymer may give an interesting quantum well electronic structure in a molecule. The PSS thin film was fabricated by spin-coating of the PSS solution in THF organic solvent onto Si-wafer substrates and curing. The electron trapping of the PSS thin films was confirmed by the capacitance-voltage (C-V) measurements performed within the metal-insulator-semiconductor (MIS) device structure. And the quantum well electronic structure of the PSS thin film, which was thought to be the origin of the electron trapping, was investigated by a combination of theoretical and experimental methods: density functional theory (DFT) calculations in Gaussian03 package and spectroscopic techniques such as near edge X-ray absorption fine structure spectroscopy (NEXAFS) and photoemission spectroscopy (PES). The electron trapping properties of the PSS thin film of quantum well structure are closely related to intra- and inter-polymer chain electron transports. Among them, the intra-chain electron transport was theoretically studied using the Atomistix Toolkit (ATK) software based on the non-equilibrium Green's function (NEGF) method in conjunction with the DFT.

Anti-inflammatory and Anti-cancer Effects of Sterol-rich Fraction from Nannochloropsis oculata by using Saponification (해양미세조류(Nannochloropsis oculata)로부터 saponification을 통한 Sterols 분획물의 항염증 및 항암 효능 평가)

  • The green microalga of the genus Nannochloropsis (class Eustigmatophyceae) is a leading candidate for biofuel production due to its ability to accumulate high oil content (28.7% of cellular ash-free dry weight). We investigated the anti-inflammatory and anticancer activities of sterol-rich fraction from nannochloropsis oculata n-hexane (NOH) extract after saponification of the microalga. Among the fractions with n-hexane, chloroform and ethyl acetate, the n-hexane fraction showed the highest anti-inflammatory activity in LPS-stimulated RAW 264.7 macrophage as well as anticancer activity against human leukemia HL-60 cells without the cytotoxity. And the sterol-rich fraction was obtained from the n-hexane fraction by open silica column under the gradient solvent condition with 100% hexane (1L), hexane : ethyl acetate (20 : 1, 10 : 1, 5 : 1, 1 : 1, v/v). Among the four fractions (NOH-1~4), especially NOH-1 contained the highest content of sterols. NOH1 showed the highest HL-60 (about 85%) and NO inhibitory activities at the concentration of $100{\mu}g/mL$. These results demonstrated that the sterol-rich fraction from N. oculata might be a useful candidate as anti-inflammatory and anticancer agents for anti-inflammatory and anticancer activity.

  • In recent, it is worldwide issued that nanoscale science and technology as a solution have supported to increase the sensing performance in carbon nanotube based biosensor system. Containing material chemistry in various nanostructures has formed their high potentials for stabilizing and activating biocatalyst as a bioreceptor for medical, food contaminants, and environmental detections using electrode modification technologies. Especially, the large surface area provides the attachment of biocatalysts increasing the biocatalyst loading. Therefore, nano-scale engineering of the biocatalysts have been suggested to be the next stage advancement of biosensors. Here, we would like to study the electrical mechanism depending on the exposure methods (soaking or dropping) to the sample solution to the assembled carbon nanotubes (CNTs) on the gold electrodes of biosensor for a simple and highly sensitive detection. We performed various experiments using polar and non-polar solutions as sampling tests and identified electrical response of assembled CNTs in those solutions.
