THE EFFECTS OF PROSTAGLANDINS AND INDOMETHACIN ON OVULATION IN THE PERFUSED FOWL OVARY

  • The effects of prostaglandins (PG) and indomethacin, a PG synthesis inhibitor, on ovulation in the perfused fowl ovary were studied. Laying hens were killed by cutting the jugular vein 18-19 h before expected ovulation of the second follicle of a clutch sequence. The cannulated ovary was dissected free and connected with a recycling perfusion apparatus. Agents to be studied for their effects on ovulation were added to the perfusion fluid. $PGF_{2{\alpha}}$ at 0.1 and 1 mg/l induced ovulation, with a success rate of 25% and 30%, respectively. The same doses of $PGE_2$ were effective at 60% and 63%, respectively. Indomethacin partially blocked gonadotrophin-induced ovulation. It is suggested that PGs may play a supportive role in the process of follicular rupture in the domestic fowl.

  • KiSS1-derived peptide receptor, a GPCR protein, binds with the hormone Kisspeptin plays a major role in the neuroendocrine regulation of reproduction. It is important in the onset of puberty and triggers the release of gonadotrophin-releasing hormone. It is a potential drug target for the disorders related to GnRH, hence, analysing the structural features of the receptor becomes important. The three dimensional of the receptor modelled in a previous study was utilised. In this study, we have analysed the protein - protein interaction of the receptor with Kisspeptin 10 and 15. The study revealed the important residues which are involved in the interaction. The result of this study could be helpful in understanding the mechanism of Kiss1 receptor activation and the pathophysiology of the disorders related to the receptor.

Effect of human chorionic gonadotrophin injection after artificial insemination on pregnancy establishment in dairy cattle

  • The objective of this study was to evaluate the effect of treating dairy cattle with exogenous human chorionic gonadotrophin (hCG), five (5) days post artificial insemination (AI) on serum progesterone (P4) concentration and pregnancy rate. In this experiment, five days after AI, cows were assigned randomly to two groups namely: a) treated group (67) which were administrered with 1500 IU hCG (Chorulon) and b) control group (61), which received no treatment. On day 5, 10, 15 and 20 after the artificial insemination, blood samples from a total of 8 cows (4 from each group) were collected and were analyzed for serum P4 concentration. Cows were detected for estrus according to standing heat by visual observation. Cows that were detected still in estrus after days 18-24 were re-inseminated and recorded as not pregnant (open). Pregnancy diagnosis was conducted by ultrasonographic examination and transrectal palpation of the uterus on approximately 60 days in cows that observed to be not in estrus. The conception rate in hCG treated and control groups were 52.5 and 36.1%, respectively. The results proved that there were no significant differences in conception rate between two groups (p=0.0568). However, pregnancy rates were reduced by hCG treatment. Average serum P4 concentrations did not differ between Hcg-treated and control groups on day 5 (0.377 versus 0.375 ng/ml). On day 20 serum P4 concentrations were greater in the treated group compared with the control group (3.085 versus 2.010 ng/ml). The treatment with hCG seemed to increase P4 level compared with the control. In conclusion, the results of this study showed that 1500 IU of hCG administered on 5 day post AI increased conception rate in dairy cows. This was supported by the results on serum P4 concentration which was greater in hCG treated group.

Effects of Human Chorionic Gonadotrophin, Flunixin Meglumin, Lidocane on Pregnancy Rate with Hanwoo IVF Embryo Transfer

  • This study was carried out to confirm the effects of luteotrophin, human chorionic gonadotrophin (hCG), and an anti-luteolytic agent, flunixin meglumin (FM), on pregnancy rates in Hanwoo with in vitro produced (IVP) embryo transfers (ET), and to research the effects on the estrus cycle. Treatments included hCG and FM administration 3~10 minutes prior to ET. Also, pregnancy rates were compared with lidocane treatment and FM treatment prior to ET. The results are shown below. 30-day pregnancy rate was 76.7% in the hCG-treated group and 75.7% in the FM-treated group. Both rates were higher than the 70% rate for the control group. 42-day pregnancy rate was 76.7% in the FM-treated group. This was higher than 66.7% recorded for both the hCG-treated and control groups. The pregnancy rate of the hCG-treated group was high at Day 30 (76.7%) but low at Day 40 (66.7%), and there were no differences from the FM-treated and control groups. The recurrent estrus rate of infertile individuals at 2 weeks after ET was 36.4% in the hCG-treated group, under 71.4% in the FM-treated group and 80.0% in the control group. The non-pregnancy rate of individuals without recurrent estrus was 18.2% in the hCG-treated group, which was higher than the 0% rate in both the FM-treated and control groups. The pregnancy rates were higher in the FM-treated group than the Lidocane-treated group with 72.3% versus 67.5% in the heifers and 48.9% versus 43.6% in the cows. From the above results, the FM treatment proved more effective than the hCG treatment and no treatment whatsoever in increasing pregnancy rates after ET. In addition, hCG treatment was shown to be undesirable due to the deviations it caused in the reproductive physiology of the hCG-treated recipients. Therefore, in our study, the FM treatment resulted in a higher pregnancy rate than either lidocaine treatment or no-treatment in the trials of ET.

Effects of Progestagen and Pmsg on Estrous Synchronization and Fertility in Kivircik Ewes during Natural Breeding Season

  • An experiment was conducted using indigenous Kivircik ewes to evaluate the effect of intravaginal progestagen sponges, containing 30 mg of fluorogestone acetate (FGA), followed by administration of pregnant mare serum gonadotrophin (PMSG) on inducing synchronized oestrus in the season and fertility. Three times of PMSG administration relative to sponge withdrawal (24 h before (n = 30), at (n = 29) or 24 h after (n= 29)) and two routes of PMSG administration (intramuscular (n = 46) and subcutaneous (n = 42) were compared for estrous response, number of multiple births and fecundity rates. There were no significant differences in terms of estrous response, due to differences in the time and route of PMSG administration. Lambing percentage, proportion of multiple births and fecundity were 75.6, 51.6 and 114.6%, respectively. The administration had a significant effect on lambing (p<0.05), multiple birth and fecundity rates (p<0.01). The subcutaneous administration of PMSG resulted in a significantly higher lambing rate (p<0.05) and fecundity rate (p<0.01), compared to the intramuscular injection of the PMSG.

  • Fry of a Siamic catfish Clarias batrachus was brought to Korea on August 14,1979 and exper-imentally reared for a whole life cycle in a small water recycling aquarium. After 29 days since start of feeding the fry were measured 5 cm in 1ength and 1.43 g in body weight and after 260 days they were measured 205.7 g average. Chorionic gonadotrophin (2 IU) was injected to a healthy female (224 g) and after 63 hours fertilized eggs were obtained in a small water recycling aquarium. About 1,300 fry were hatched out 20 hours after spawning at $30^{\circ}C$ constant water temperature. The fry began to feed on water fleas 53 hours after hatching.   • As an preliminary experiment for making transgenic animals producing human follicle stimulating hormone (hFSH), we tried to express recombinant hFSH gene in vitro. hFSH is a heterodimeric glycoprotein hormone produced in the anterior pituitary gland. The hormone is essential in the regulation of reproductive processes, such as follicular development and ovulation. Genes encoding the common gonadotrophin alpha subunit and FSH-specific beta subunit were inserted into retroviral vectors under the control of the rat beta actin promoter. Gene transfer to the Chinese hamster ovary (CHO) cells was done by infection of the retroviruses harvested from PT67 packaging cells transfected with recombinant retrovirus vector DNA. After selection with G4l8, PCR and RT-PCR analyses of the G4l8-resistant CHO cells showed successful transfer and expression of both ${\alpha}$ and ${\beta}$ fragments of the FSH gene.

Factors Affecting on In Vivo Embryo Production in Hanwoo (한우에서 체내수정란 생산에 영향을 미치는 요인에 관한 연구)

  • This study was carried out to establish an effective embryo production system by analyzing several factors ifluencing in vivo embryo production in Hanwoo. The results are summarized as follows : 1. The number of total, fertilized and transferble embryos by gonadotropins were no significant difference among PMSG, FSH and FSH+PMSH. 2. The numbers of recovered and transferable embryos by type of FSH were no significant difference in both FSH-P and FOLLTROPIN-V but significant difference with SUPER-OV(p<0.01). 3. The numbers of recovered and transferable embryos by age of donor were highest in 5 years old. 4. The recovered number of transferable embryos was highest in the cows at the 1 st and 3rd parity but significantly lower in hifers and the cows at more than 6th parities(p<0.01). 5. The numbers of trasferable embryos by postparient were more<270 days than > 270 days in significant difference(p<0.01). 6. Embryo production after treatment of repeated super ovulation was no significant difference. 7. Recovered and trasferable embryos production after treatment of repeated super ovulation interval was highest 81∼100 days and significant difference(p<0.01). 8. The numbers of recovered and transferable embryos by seasons were no significant difference.

  • Hormonal induction of sex reversal was examined by using sex steroid hormones in serranid fish, Epinephelus septemfasciatus. Young fish were collected from the coastal area of Cheju Island, and reared for 2 years before fish were used for the experiments. Without any hormonal treatment, gonads of fish ($1,000\~2,800$ g in body weight) were occupied by oocytes of the perinucleolus stage and bundles of protogonial cells in the area of germinal epithelium. When the induction of sex reversal was attempted by daily oral administration of $17\alpha$-methyltestosterone (0.5 mg/kg fish) for 90 days, active spermatogenesis was induced, and spermatogonia and spermatocytes and spermatids were appeared in all gonads we examined. However, after daily, oral treatment of $17\beta$-estradiol (0.5 mg/kg fish) to. 50 days with the following injection of human chorionic gonadotrophin ($1,000\~1,500$ IU/kg fish) mature oocytes were not induced in fish gonad.

Early Development of Loach Oocytes Activated by Parthenogenetic Agents (미꾸라지 난자의 활성화에 의한 처녀발생 유기)

  • We examined early development in loach(Misgurnus mizolepis) embryos with parthenogenetic agents well-known in mammals. Female loach was superovulated with an intraperitoneal injection of 15 IU human chorionic gonadotrophin (hCG) per gram body weight. After 13 h of hCG injection, the oocytes were obtained from the abdomen. The oocytes were activated with 10% ethanol in tap water or fish Ringer's solution for 5, 10 and 15 minutes(eTW5, 10, 15 and eFRS5, 10, 15), respectively. The activation rates were 29% and 10% in eFRS10 and eFRS15, 5% and 6% in eTW10 and eTW15 by judging the cleaved blastomeres. Whereas, no parthenogenetic embryo was produced by tap water or fish Ringer's solution alone. The activation rate with the fish Ringer's solution was higher than that of tap water. No embryonic development was observed by calcium ionophore, A23187, at concentrations of 10, 20, 40 and 100$\mu$M when treated for 1, 2.5 and 5 minutes, respectively. The activation agents did not cause early development as in mammalian eggs. Therefore, the results suggest that fresh water fish may have a different egg activation pathway from that of mammals.
