직장인의 만성질환 위험 요소별로 구성된 영양 교육의 효과 비교 연구 (Effects of Workplace Nutrition Education Program Tailored for the Individual Chronic Disease Risks)

  • The incidence of chronic disease is continuously increasing in Korea. Especially, office workers have higher risk of chronic disease because of their dietary habit and lifestyle. The study aimed to investigate the effect of tailored nutrition counseling on improving chronic disease risk factors. Ninety-nine male workers (age $46.9{\pm}7.0$ yrs) volunteered for 12 weeks of nutrition program containing dietary intake and physical activity adjustment. Five individualized programs were performed with the main theme of weight loss (WL, n = 16), blood pressure lowering (BL, n = 34), normalizing blood glucose (GL, n = 21), lipid lowering (LL, n = 13) and reducing MS risk factors (ML, n = 15). Anthropometric data, blood-pressure, self-reported questionnaire, blood profiles were measured before and after 12weeks of nutrition education. The education program included 5 times of 1:1 interview. Compared to 0 week, anthropometric data (weight, BMI, fat, visceral fat, waist, SBP, DBP) were significantly decreased after 12 weeks (p < 0.001). Fasting blood glucose and total cholesterol were decreased (p < 0.05). The primary outcomes with individually tailored programs showed to be more effective than one general nutrition program.

Antihyperglycemic activity of Biophytum sensitivum (L.) DC in alloxan diabetic rats

  • The study was to evaluate the antihyperglycemic activity of Biophytum sensitivum (L.) DC in different extracts. Albino Wistar rats with alloxan hydrate induced diabetes were divided into 7 groups of 6 each. Both aqueous and methanolic extract of Biophytum sensitivum were prepared and given individually at different doses to different batches of rats (both normal and diabetic rats) after an overnight fast. Methanolic extract at the dose of 200 mg/kg body weight showed maximum blood glucose lowering effect in diabetic rats. The same dosages did not produce any hypoglycemic activity in normal rats. The antihyperglycemic activity of Biophytum sensitivum was compared with a standard drug Glibenclamide, an oral hypoglycemic agent. The above results suggest that maximum hypoglycemic effect was found only with a dose of up to 200 mg/kg b.w. of methanolic extract which is therefore the optimum dose for hypoglycemia and was used in all the experiments of the present study.

Effects of Silkworm Extract on Disaccharidase Activities of Small Intestine and Blood Glucose-Lowering in C57BL/6J Mice

  • This study examined the anti-diabetic effect of a silkworm extract in C57BL/6J mice, an ob/㏈ model, fed a high fat diet for 8 weeks. The body weight was not significantly different with the silkworm-extract supplement, nor did food intake and body weight gains also did not differ significantly among the high-fat diet groups. However, the water intake by the silkworm-extract supplemented groups increased significantly compared with that by the distilled-water supplement group, nonetheless, the FER did not differ significantly. For all groups, the blood glucose increased the most after 30 minutes and yet returned to a fasting level within 90 minutes. The fasting time and resulting glucose tolerance for the silkworm-extract supplemented groups were significantly decreased compared to that for the high fat diet with distilled water supplement group, while the level of blood glucose in silkworm-extract supplemented groups was significantly decreased compared with than in the diabetic control group. The HbA1c and insulin levels were no different among the groups. The sucrase and lactase activities in the proximal small intestine were significantly decreased in the silkworm-extract supplement groups compared to that in the diabetic control group. There was no significant difference in the glycogen contents in the liver and muscle among the groups. In conclusion, it was found that the silkworm-extract supplement repressed the disaccharidase activity in the small intestines mucosa of the C57BL/6J mice.

구아바 잎 열수추출물의 당류분해효소 저해 효과 및 OGTT에 미치는 효과 (Effect of Aqueous Extract of Guava (Psidium guajava L.) Leaves on the Oral Glucose Tolerance Test and Inhibition of Glycoside Hydrolase)

  • Guava ($Psidium$ $guajava$) contain a great deal of polyphenol compound and work on the treatment of $Diabetes$ $mellitus$ effectively. In this study, the bioactivities of aqueous extract (GLEx) of guava leaf were investigated. Total phenolic contents of GLEx was 266.9 mg tan/g. The effects of GLEx on digestive enzymes, ${\alpha}$-amylase, maltase and sucrase were investigated. $IC_{50}$ values of GLEx against ${\alpha}$-amylase, maltase and sucrase were 0.65 mg/$m{\ell}$, 2.0 mg/$m{\ell}$ and 3.5 mg/$m{\ell}$ respectively. The effect of GLEx on oral glucose tolerance test (OGTT) was performed in normal ICR mouse, control (dstilled water) and GLEx (aqueous extract of Guava leaf) were co-administered orally with glucose, showed reducing effect on the blood glucose level. The guava is likely to useful for prevention or improvement of hyperglycemia by lowering the blood glucose level and inhibiting glycoside hydrolase activity.

식이 내 곡류 종류와 지방수준이 성장기 이후 비만유도 흰쥐의 당대사와 항혈전능에 미치는 영향 (The Impact of Kinds of Dietary Grain and Dietary Lipid Level on the Glucose Metabolism and Antithrombogenic Capacity of Full Grown Obesity Induced Rats)

  • This study was designed to evaluate impact of kinds of dietary grain and dietary lipid level on the glucose metabolism and antithrombogenic capacity in obesity induced rats. Total of 80 Sprague-Dawley male rats were raised for one month with control diet containing $50\%$ (w/w) well-milled rice powder and $20\%$(w/w) of dietary lipids. The rats were blocked into 8 groups and raised for two months with diets containing well-milled rice, brown rice, black rice, or glutinous barley powder and 8 or $20\%$(w/w) of dietary lipids. The contents of total dietary fiber in experimental grains were in following order; glutinous barley > black rice > brown rice > well-milled rice. Weekly food intake were lower in glutinous barley group among all experimental groups. Body weight gain was high in high level of fat groups ($50\%$w/w) than medium level of fat groups ($8\%$ w/w). Plasma glucose concentration was not different significantly in each groups. But brown rice group was a little lower than others. Plasma insulin concentration was lower in black rice and glutinous barley group than rice group. Plasma glucagon concentration did not differ significantly among all experimental groups. Hexokinase activities in skeletal muscle are different significantly according to level of dietary fat and grain variety factors. Brown rice group was significantly highest among all experimental groups in hexokinase activity. Plasma $TXB_2$ concentrations in black rice and glutinous barley groups were lower as compared to rice and brown rice groups. Plasma 6-keto-$PGF_{1\alpha}$ concentrations in glutinous barley group was higher as compared to others. In conclusion brown rice has a little lowering effect glucose concentration. Black rice and glutinous barley intakes enhance antithromboenic capacity. It is suggested that the intakes of mixed gains are recommend.

Effects of a mobile healthcare service provided by public health centers on practicing of health behaviors and health risk factors

  • BACKGROUND/OBJECTIVES: This study evaluated whether a mobile health (mHealth) application can instigate healthy behavioral changes and improvements in metabolic disorders in individuals with metabolic abnormalities. SUBJECTS/METHODS: Participants were divided into an mHealth intervention group (IG), which used a mobile app for 24 weeks, and a conventional IG. All mobile apps featured activity monitors, with blood pressure and glucose monitors, and body-composition measuring devices. The two groups were compared after 24 weeks in terms of health-behavior practice rate and changes in the proportion of people with health risks, and health behaviors performed by the IG that contributed to reductions in more than one health risk factor were analyzed using multiple logistic regression. RESULTS: Preference for low-sodium diet, reading nutritional facts, having breakfast, and performing moderate physical activity significantly increased in the mHealth IG. Furthermore, the mHealth IG showed a significant increase of eight items in the mini-dietary assessment; particularly, the items "I eat at least two types of vegetables of various colors at every meal" and "I consume dairies, such as milk, yogurt, and cheese, every day." The proportion of people with health risks, with the exception of fasting glucose, significantly decreased in the mHealth IG, while only the proportion of people with at-risk triglycerides and waist circumference of females significantly decreased in the control group. Finally, compared to those who did not show improvements of health risks, those who showed improvements of health risks in the mHealth IG had an odds ratio of 1.61 for moderate to vigorous physical activity, 1.65 for "I do not add more salt or soy sauce in my food," and 1.77 for "I remove fat in my meat before eating." CONCLUSIONS: The findings suggest that the additional use of a community-based mHealth service through a mobile application is effective for improving health behaviors and lowering metabolic risks in Koreans.

국내산 벌개미취 잎 추출물의 α-glucosidase 억제능 및 항산화 활성 평가 (Antioxidant and α-Glucosidase Inhibitory Activities of the Extracts of Aster koraiensis Leaves)

  • Background: The plant Aster koraiensis has long been used as an ingredient in folk medicine. It has been reported that Aster koraiensis extract (AKE) prevents the progression of diabetes-induced retinopathy and nephropathy. However, although these beneficial effects of AKE on diabetes complications have been identified, the antidiabetic effects of AKE have not yet been completely investigated and quantified. In the present study, the glucose-lowering and antioxidant effects of aqueous and ethanolic AKEs were evaluated. Methods and Results: The glucose-lowering effects of aqueous and ethanolic (30%-, 50%-, and 80%-ethanol) AKEs were investigated via ${\alpha}$-glucosidase inhibitory assays. The mode of inhibition by AKEs on ${\alpha}$-glucosidase was identified through kinetic analysis. The total antioxidant capacity of each of the 4 AKEs was evaluated by assessing their conversion rate of $Cu^{2+}$ to $Cu^+$. The content of chlorogenic acid and 3,5-di-O-caffeoylquinic acid, the bioactive compounds in AKE, in each extract were analyzed by high performance liquid chromatography (HPLC). The AKEs showed potent ${\alpha}$-glucosidase inhibitory activity with mixed inhibition mode, and significant antioxidant capacity. Conclusions: These results of this study suggested that the AKEs tested had ${\alpha}$-glucosidase inhibitory and antioxidant effects. Among the extracts, the 80% ethanol extract showed the most significant ${\alpha}$-glucosidase inhibitory activity, with a half maximal inhibitory concentration ($IC_{50}$ value) of $1.65{\pm}0.36mg/m{\ell}$ and a half maximal effective concentration ($EC_{50}$ value) for its antioxidant activity of $0.42{\pm}0.10mg/m{\ell}$. It can therefore be used as a source of therapeutic agents to treat diabetes patients.

표고버섯 균사체의 생산과 당뇨 유발 흰쥐에 대한 혈당강하 효과 (Production of Lentinus edodes Mycelia in Submerged Culture and It's Hypoglycemic Effect in Diabetic Rats)

  • 표고버섯 균사체가 항당뇨 효과에 미치는 영향을 검토하기 위해 당뇨가 유발된 흰쥐에 표고버섯 균사체을 7일간 급여시킨 후 혈장 glucose, total cholesterol, triglyceride 및 ALT, AST 활성을 관찰하였고, 표고버섯 균사체 생장의 최적 배양 조건을 조사하였다. 균사체 생장을 위한 최적의 pH와 온도는 5와 $30^{\circ}C$로 나타났으며, 발효조배양시에는 최대 균사체 생장(8.13g/l)이 진탕배양일 때보다 6일 단축되었다. Hypoglycemic 효과에서 혈장 glucose는 당뇨 유발군에서 표고버섯 균사체 급여군이 대조군보다 23.0%가 유의하게 감소하였으며, 혈장 total cholesterol과 triglyceride는 20.3%와 27.1%로 낮게 나타났다. 또한 혈장 ALT와 AST 활성은 당뇨 대조군에 비해 표고버섯 균사체 급여군이 29.6%와 34.3% 낮은 활성을 보였다.

Effect of Azadirachta indica flower extract on basal and experimentally elevated blood glucose in rats

  • Azadirachta indica A. Juss (N.O. Meliaceae), popularly known, as 'Neem' is an indigenous tree widely available in India. Almost every part of the tree has long been used in Unani system of medicine for the treatment of a variety of human ailments. The flowers have been mentioned as a remedy useful in controlling diabetes mellitus. The present study had been designed to investigate the hypoglycemic/anti-hyperglycemic effects of the methanolic extract of the flowers of A. indica (Gule-Neem) and its different fractions on normal, glucose fed hyperglycemic, adrenaline induced hyperglycemic and alloxan induced diabetic rats. The methanolic extract was resolved into water soluble and water insoluble fractions. Water soluble portion of the methanolic extract was found to possess significant blood sugar lowering effect in glucose-fed and adrenaline-induced hyperglycemic rats but it did not show such effect in normal and alloxan induced mild and severe diabetic rats. Water-soluble portion was fractionated by employing the polarity criterion with ethyl acetate and butanol. The ethyl acetate fraction was further fractionated into phenolic and non-phenolic fractions. Hypoglycemic effect of these fractions was also evaluated. The results suggest that the flowers of A. indica contain at least two different constituents, responsible for the said activity. These investigations validate the use of flowers of A. indica in diabetes by Unani physicians.

택란 분획물이 당뇨유발 흰쥐에서의 혈당강하에 미치는 영향 (The antidiabetic properties of fractions of Lycopus lucidic Turcz in streptozotocin diabetic rats)

  • This study was carried out to evaluate the effects of fractions of methanol(MeOH) extracts of Lycopus lucidic Turcz on hyperglycemia and energy metabolites in streptozotocin(STZ) diabetic rats. Diabetes mellitus was induced in male Sprague-Dawley rats weighing 200-220 g by an injection of STZ dissolved in a citrate buffer into the tail vein at a dose of 45 mg/kg of body weight, and the rats were divided into 7 groups, that is, one normal group and 6 diabetic groups: STZ-control, hexane, chloroform(CHCl$\sub$3/). ethylacetate(EtOAc), butanol(BuOH) and H$\sub$2/O fraction-fed groups. All groups were fed an AIN-93 diet and the fractions of Lycopus lucidic Turcz were administered orally with 2 % Tween 80 for 14 days after the STZ injection. Body weight, diet intake and organ weights were monitored. The plasma levels of blood glucose, insulin and protein were determined. The plasma concentrations of cholesterol, HDL-cholesterol, triglycerides and free fatty acid were assayed. The plasma activities of aspartate aminotransferase (AST) and alanine aminotransferase(ALT) were also measured. Body weight losses were observed by feeding the fractions of Lycopus lucidic Turcz in STZ experimental groups, and the kidney weight was increased. The extent of blood glucose decrement was significantly greater in the hexane and BuOH fraction-fed groups than STZ-control group. The plasma protein level was significantly lower in the H$\sub$2/O fraction-fed group. The plasma cholesterol level was decreased in BuOH and H$\sub$2/O fraction-fed groups compared with the STZ-control group. The levels of free fatty acids in the CHC1$\sub$3/ and H$\sub$2/O fraction-fed groups were significantly decreased(p<0.05). ALT activitiy of BuOH fraction-fed group was lower than control but it was not significantly different. These results suggest that the fractions of Lycopus lucidic Turcz are capable of lowering blood glucose and fat metabolites concentrations when administered to STZ-treated rats, and AST/ALT activity and insulin levels show the possibility of therapeutic use to diabetes mellitus.
