Effect of Buan Mulberry on Metabolic Improvement in Streptozotocin-Induced Diabetic Rats (Streptozotocin으로 유발된 당뇨쥐의 신진대사 증진에 대한 부안 오디의 효과)

  • This study was designed to evaluate the anti-diabetic effects of Buan mulberries by using an insulin-dependent diabetes mellitus animal model. Several studies have shown that mulberries have metabolism-improving, antioxidant, and lipid-lowering properties in rats with streptozotocin (STZ)-induced diabetes. In this study, Sprague-Dawley male rats were randomly assigned to 1 normal control group and 5 STZ-induced diabetes groups: rats that had STZ-induced diabetes and did not receive any agents (diabetic group; negative control), rats that had STZ-induced diabetes and received insulin (insulin group; positive control), rats that had STZ-induced diabetes and received 0.5% mulberry extract (0.5% mulberry group), rats that had STZ-induced diabetes and received 1.0% mulberry extract (1.0% mulberry group), and rats that had STZ-induced diabetes and received 2.0% mulberry extract (2.0% mulberry group). Mulberry extracts were administered to the diabetic animals for 4 weeks. The rats that received mulberry extracts showed lower body weights and insulin levels, as well as higher kidney weights, blood glucose levels, urine quantities, and water intake in comparison with the normal controls. Further, the insulin concentrations in the mulberry-fed animals were higher than those in the diabetic group, and the kidney weights, blood glucose levels, urine quantities, and water intake in the mulberry-fed animals were lower than the corresponding values in the diabetic controls. These results suggest that mulberry may be an effective functional food to prevent diabetes-related complications.

The Effect of Mixing Beverage with Aralia continentalis Kitagawa Root on Blood Pressure and Blood Constituents of the Diabetic and Hypertensive Elderly (땅두릅뿌리를 이용한 혼합음료가 당뇨 및 고혈압 환자의 혈압과 혈액 성상에 미치는 영향)

  • Studies were carried out to observe the influence of the Aralia continentalis Kitagawa root on blood pressure and blood constituents of the 24 hypertensive elderly(10 men and 14 women aged over 60). Total subjects were divided into three groups according to taking antihypertensive medications(A), antihypertensive medications and mixing beverages with the Aralia continentalis Kitagawa root(B) or mixing beverages with Aralia continentalis Kitagawa root(C). The systolic blood pressure of C group was significantly lower than that of A and B groups. The diastolic blood pressure of C group was lower than A and B groups. The blood glucose level of C group was a little lower than that of A and B groups. The level of TG increased more in A group than C group and decreased in B group. The levels of T-cholesterol and LDL-cholesterol lowered in B and C groups and HDL-cholesterol level was higher in C group. The GOT, GPT and T-bilirubin levels of C group were significantly higher than others. Mixing beverages with Aralia continentalis Kitagawa root did not affect on blood protein and non-protein nitrogens. In conclusion, mixing beverages with Aralia continentalis Kitagawa root had an effect on lowering blood pressure, blood glucose, T-cholesterol and LDL-cholesterol concentrations.

Effect of Naringin on Lipid Metabolism and Antithrombotic Capacity in Rat (랫드에서 Naringin이 지방대사 및 항혈전능에 미치는 영향)

  • Naringin, major citrus flavonoids, has been identified to exert antioxidative, antidiabetic, and lipid lowering effects. In this study, we examined the effect of 0.2 g/kg, 0.5 g/kg naringin supplementation for 3 times/week for 5 weeks on lipid metabolism and antithrombotic capacity in rat. Eighteen five week-old Sprague Dawley(SD) female rats, which had initial body weights of $246{\pm}9g$, were randomly divided into three groups: Control (non naringin group); Low (0.2 g/kg naringin-supplemented group); High (0.5 g/kg naringin-supplemented group). Three groups of rats were supplemented with three experimental diets for 5 weeks and we investigated antithrombotic capacity before sacrifice. Naringin did not significantly alter the body weight gain, relative organ weight. However, the level of serum triglyceride, serum free fatty acid, serum total lipid and serum glucose levels were significantly lowered compared to those of control. The high group (0.5 g/kg naringin-supplemented group) was showed significantly increased bleeding time compared to control group. These results suggest that naringin supplemental diets reduces the level of hypertension, glycosuria and fatness on the female SD rats, when orally administered below the dosage 0.5 g/kg for 5 weeks.

Chemical Features of Solid Residues Obtained from Supercritical Water Treatment of Populus alba×glandulosa (현사시나무 목분의 초임계수 처리 공정으로부터 유래한 미분해 고형성분의 화학적 특성)

  • After supercritical water treatment of poplar wood meals (passed through 60 mesh) for 60s between 325 and $425^{\circ}C$ at the fixed pressure at $220{\pm}10atm$, some solid residues were present in the degradation products. They mainly consisted of chemically modified lignin and fibrous materials. Glucose and xylose were identified as main sugar components of fibrous materials, and the highest ratio of glucose/xylose was achieved at the highest reaction temperature. As reaction temperature was elevated, the portion of fibrous materials decreased in the solid residues, while lignin was further accumulated. The H : G : S ratio of lignin in solid residues was estimated by analytical pyrolysis. Irrespective of reaction temperatures, the H:G:S ratios were not significantly changed in the lignin in solid residues. Compared to poplar milled wood lignin (MWL), it was remarkable that H type monomers were further lowered, while portion of S type monomers increased. The amount of G type monomers were relative stable. In presence of HCl catalyst, lowering H type as well as enhancing S type was further distinguishable. According to the result of nitrobenzene oxidation (NBO), ca. 265 mg of vanillin and syringaldehyde was yielded from poplar MWL as main products. However, remarkably reduced amount of NBO products were determined from solid residues by raising operating temperature as well as by the addition of HCl catalyst. These results strongly indicate that $\beta$-O-4 linkage could be easily cleaved during supercritical water treatment, so that the lignins in the solid residues seem to be condensed phenol polymers, which are mainly formed by carbon-carbon linkages rather than $\beta$-O-4 linkage.

  • Silkworm (Bombyx mori) pupae have been widely worked in wound dressing, hepatoprotective activity, antigenotoxicity, control of glucose level and anti-cancer activity. To investigate the anti-obesity activity of ethanol extract of silkworm pupae powder fermented with Cordyceps militaris (ESfC), the free glycerol release and cAMP concentration as well as fat accumulation were measured in the primary adipocytes of SD (Sprague Dawley) rats and high fat diet (HFD)-treated C57BL/6 mice treated with 12 weeks. Firstly, the presence of the cordycepin with lipid lowering effect was confirmed in ESfC using HPLC analysis. The level of free glycerol and cAMP concentration was significantly increased in the primary adipocytes treated with high dose of ESfC ($400{\mu}g/ml$) although these levels were consistently maintained in other dose ESfC treated groups. In HFD-induced obesity model, the increased fat weight and size of adipocytes in HFD+Vehicle treated group was recovered in HFD+ESfC treated group. Also, the liver weight and the number of lipid droplets were higher in HFD+Vehicle treated group than No treated group. But, this level was significantly decreased in HFD+ESfC treated group compared with HFD+Vehicle treated group. Furthermore, a similar recovery was detected on the phosphorylation of periliphin and HSL, and ATGL expression. Overall, the results of the present study provide some scientific evidences that ESfC can stimulate lipolysis in primary adipocytes and prevent fat accumulation in HFD-treated obesity model, and therefore have the potential for use as anti-obesity agents to treat obese patient.

Anti-hyperlipidemic effect of Agaricus brasiliensis fruiting bodies in rats fed with high cholesterol diet (고콜레스테롤 식이의 급여에 의해 고지혈증이 유도된 흰쥐의 신령버섯 자실체의 항고지혈증 효과)

  • In this study, we aimed to investigate the effects of dietary supplementation with fruiting body of Agaricus brasiliensis (AB) mushroom on the lipid profiles of serum and histological patterns of liver of high cholesterol-fed rats. Five-week-old, female Sprague-Dawley albino rats were divided into three groups of 8 rats each, including a normal control-diet (NC) group, a high-cholesterol diet (HC) group, and a group fed high-cholesterol diet supplemented with 5 % powder of Agaricus brasiliensis fruiting bodies (HC+AB). Total serum cholesterol, low density lipoprotein (LDL), and triglyceride (TG) concentrations in the HC+AB group were significantly reduced when compared with those in the HC group. Body weight in the HC+AB group was significantly lower than that in the HC group, whereas no adverse effects were observed on the levels of plasma albumin, creatinine, blood urea nitrogen, uric acid, glucose, and total protein. In the HC+AB group, liver enzyme activities related to liver function, such as GOT and GPT, presented values lower than those in the HC group and were very similar to the ones in the NC group. Excretion of total lipid and cholesterol in feces in the HC+AB group was significantly higher than that in the NC and HC groups, indicating that mushroom feeding inhibits the absorption of lipid cholesterol in the intestine. Liver histopathological analyses revealed that rats fed with HC diet developed fat liver disease, whereas only small amounts of fat were deposited in the livers of the HC+AB group. In conclusion, the results suggest that fruiting body powder of A. brasiliensis provides health benefits to high-cholesterol-fed rats by lowering body weight and the risk of atherogenic lipid profile.

Stress in Olive Flounder (Paralichthys olivaceus) and Fat Cod (Hexagrammos otakii) by the Sudden Drop and Rise of Water Temperature (수온의 급하강과 급상승이 넙치 (Paralichthys olivaceus)와 쥐노래미(Hexagrammos otakii)에 미치는 스트레스)

  • The effects of sudden changes of water temperature (WT) on the stress response and physiological change of the cultured olive flounder in large (FL) and small (FS) size, and fat cod (FC) were examined by manipulating WT (2 types) in a flow through seawater culture system with 6 tanks (water vol. 270 L/tank). The WT was decreased from $20^{\circ}C$ to $10^{\circ}C$ within 5 hours ($2^{\circ}C/hour$) and maintained at $10^{\circ}C$ for 21 hours (Exp. I), and it was raised from $20^{\circ}C$ to $30^{\circ}C$ within 5 hours and maintained at $30^{\circ}C$ for 21 hours (Exp.II). In Exp. I, the levels of blood hematocrit at 5 hours ($10^{\circ}C$) in FS was significantly decreased from $13.5\pm2.0\%\;to\;11.3\pm2.3\%$, but FC at 2.5 hours ($15^{\circ}C$) ($19.0\pm0.3\%\;to\;23.2\pm3.8\%$) was increased, The blood hemoglobin concentration of all fish in Exp, II was significantly increased until 8 hours after raising WT from $20^{\circ}C$ to $30^{\circ}C$. In Exp. I and II , the levels of plasma cortisol in FL, FS and FC was changed from $5.2\pm8.5ng/mL,\;4.4\pm4.5ng/mL\;and\;2.7\pm0.4ng/mL$, respectively, before sudden drop and rise of WT. The levels of plasma cortisol of in FL ($164.0\pm53.1ng/mL$) and FC ($207.9\pm25.4ng/mL$) were significantly increased by the lowering WT sharply during whole experiment. The FL ($12.6\pm2.0ng/mL$) and FS ($4.0\pm3.9ng/mL$) showed no significant differences in cortisol level according to sudden rise of WT (5 hours). But it in FC ($44.7\pm18.2ng/mL$) was increased. In Exp. I, the plasma glucose levels of all fish groups were decreased after 5 hours ($10^{\circ}C$), The plasma lactic acid concentration of FL and FS showed no significant differences until 5 hours after raising WT from $20^{\circ}C$ to $30^{\circ}C$, But it in FC was significantly increased with WT raise.

Effects of Early Parenteral Nutrition for Extremely Low Birth Weight Infants (초극소 저출생 체중아(${\leq}$1,000 g)에서 조기에 시작하는 정맥 영양공급의 효과)

  • Purpose: The object of this study was to evaluate the efficacy of early total parenteral nutrition with early amino acid in extremely low birth weight infant (ELBWI). Methods: We retrospectively analyzed the medical records of all ELBWIs who were born and admitted to Samsung Medical Center from January 2003 to December 2003 and January 2009 to December 2009 and alive at the time of discharge. Data for nutritional status and morbidities were compared between period 1 (2003, n=22), in which parenteral nutritional support was started gradually over several days and period 2 (2009, n=38), in which parenteral nutrition with amino acid was started as soon as possible after birth. Results: Compared to period 1, birth weight and Apgar score were lower in period 2. The intake amount of glucose, amino acid and total calorie was higher and the level of blood urea nitrogen was increased more from 7th day to 14th day after birth in period 2 when compared than period 1. The weight gain velocity was faster at 7th and 14th postnatal day in period 2. There were no differences in the incidence of necrotizing enterocolitis, moderate to severe bronchopulmonary dysplasia, and severe intraventricular hemorrhage (${\geq}$ Gr III) between two periods but, the incidence of periventricular leukomalacia(PVL) was significantly lower in period 2. Conclusion: Early initiation of total parenteral nutrition with early amino acid in ELBWIs was beneficial at weight gain with lowering catabolism and increasing anabolism. And it could be related with reducing the incidence of PVL.

Effect of Cynanchi wilfordii Radix Extracts on Lipid Compositions and Blood Pressure in Spontaneously Hypertensive Rats (백하수오(Cynanchi wilfordii Radix) 추출물이 선천성 고혈압 흰쥐의 지질성분 및 혈압에 미치는 영향)

  • The purpose of this study was to observe the effects of Cynanchi wilfordii Radix extracts (CRE) on the improvement of the lipid compositions and blood pressure level in spontaneously hypertensive rats (SHR) fed an experimental diet for 5 weeks. The rats were divided into 3 groups: a control group, a 0.5% CRE treated group, and a 1% CRE treated group. Consumption of CRE extract for 5 weeks in SHR significantly suppressed blood pressure rise with aging (p<0.05). After eating the experimental diets, the triglycerides in serum was significantly lower in the CRE group than that in the control group. The fasting glucose levels of the 0.5% and 1% CRE group had a tendency to be lower compared with those of the control group. Total cholesterol, HDL-cholesterol and HTR (HDL-cholesterol/total cholesterol) of the SHR in the 0.5% and 1% CRE diet were significantly increased compared to the control diet. Thus, long term consumption of CRE might be beneficial in lowering high blood pressure and the improvement of lipid metabolism in SHR rats.
