Insecticidal Activity and Stability by Freeze-drying of Entomopathogenic Bacteria, Photorhabdus temperata M1021 (동결건조에 따른 살충성 세균 Photorhabdus temperata M1021의 안정성과 살충성 평가)

  • In order to develop eco-friendly biopesticide, an entomopathogenic bacterium Photorhabdus temperata M1021 has been lyophilized via freeze-drying along with protective agents such as skim milk, starch, sodium alginate, glucose and sodium glutamate to protect cells from lysis. Freeze-drying powder of P. temperata M1021 containing 7% skim milk (w/v) showed highest survival rate of 63% among all the protective agents used in trials. Furthermore, the freeze-dried microbial powder showed 75% of survival rate after stored at $4^{\circ}C$ for 4 weeks at air contact conditions. Injection toxicity of the freeze-dried sample was tested against larvae of Galleria mellonella. A dose of $2.0{\times}10^1$ cells of P. temperata M1021 killed 100% of the G. mellonella larvae within 4 days after injection. Moreover, $2.0{\times}10^0$ cells caused 50% mortality within the 4 days after injection. Freeze-dried P. temperata M1021 strains exhibited effective insecticidal activity and could be a better candidate for being used as a biopesticide.

Synthesis and Biocompatibility Study of Hydrogel for Patch Sensor in Non-invasive Glucose Monitoring System (무채혈 혈당 측정시스템의 Patch Sensor용 수화젤의 합성 및 생체적합성에 관한 연구)

  • This study aims to verify for humans the suitability of the enzyme-fixed hydrogel used for the patch sensor of the blood sugar testing system without blood sampling, which utilizes reverse iontophoresis. Using acrylate monomers, hydrogel was synthesized to which a certain unit of enzyme is fixed. In order to analyze the material property of the synthesized hydrogel, a structural analysis was performed using FT-IR spectroscopy, while the DSC was used to verify the thermal stability. In addition, with the UV-Vis spectrophotometer, it was verified that the degree of active enzyme is at least 50% greater than the standard product. The SEM was used to verify secure fixation of the enzyme onto the surface. As a result, it was observed that the enzyme is successfully fixed to the surface. Since the hydrogel makes direct contact with a patient's skin, it is essential to evaluate the toxicity when making direct contact with the skin. For that purpose, various sets of tests were undertaken according to the ISO 10993-cytotoxicity, intracutaneous reactivity, skin irritation test and maximization sensitization. Consequently, it was successfully verified that the enzyme-fixed hydrogel have bioavailability.

Toxic Effects on Lethal Concentration 50%, Heamatological Parameters and Plasma Components of Crucian carp, Carassius carassius by Acute Waterborne Nickel Exposure (수인성 니켈 급성 노출이 붕어, Carassius carassius의 반수치사농도, 혈액학적 성상 및 혈장 성분에 미치는 독성 영향)

  • Crucian carp, Carassius carassius (Weight 39.7±3.1 g, Length 14.8±0.5 cm) were exposed to waterborne nickel at 0, 10, 20, 40, 80 and 160 mg Ni2+/l. The lethal concentration 50 (LC50) of C. carassius exposed to waterborne nickel was 117.69 mg Ni2+/l. In hematological parameters, RBC counts was significantly increased at 48 hours, whereas a significant decrease was observed at 96 hours. The MCV and MCH were significantly increased in the concentration of 80 mg Ni2+/l at 96 hours. The plasma components such as calcium, magnesium, glucose, cholesterol, total protein, AST, ALT and ALP were significantly changed by waterbonre nickel exposure. The results of this study suggest that the nickel exposure to C. carassius affects the survival rates, hematological parameters and plasma components as toxicity

Effect of Red Ginseng with Processed Sulfur Extracts on Serum Lipids Concentration and Metabolic Variables in Diabetic Rats (홍삼의 법제유황 처리가 당뇨쥐의 혈중지질 및 대사지표물질에 미치는 영향)

  • Objectives : The purpose of this study was to evaluate effects of processed sulfur with red ginseng on streptozotocin(STZ) induced diabetic rats for expansion of processed sulfur internal application. Methods : We prepared red ginseng with non processed sulfur extracts(GS), red ginseng with processed sulfur I extracts(GPS I) and red ginseng with processed sulfur II extracts(GPS II). In the present study, we examined about contents of crdue saponin, antioxidant activity, $\alpha$-glucosidase inhibitory activity, and effects of STZ induced diabetic rats. Results : Contents of crude saponin increased by processed sulfur, and GPS II was shown highest contents in crude saponin and sulfur compared with another groups. Electron donating ability of GPS II was shown highest activity compared with GS and GPS I, SOD-like activity showed same tendency as electron donating ability at 1 $mg/m\ell$ concentration. Inhibitory activity of $\alpha$-glucosidase was approximately same level in acarbose and GPS II. Blood glucose level of GPS II group was decreased 18.34% compared with DC(diabetes control) group and maintained stability range in glucose level. but GS and GPS I showed high level compared to GPS II. Serum triglycerides concentration also showed lowest level in GPS II. The activity of ALT, AST and ALP was shown high level in diabetic induced groups, and lowest level in GPS II. Creatinine was shown non-significantly difference in each groups and GPS II was shown lowest level in BUN. Conclusions : These results suggested that processed sulfur with red ginseng have improvement effects on diabetes and internal application of processed sulfur with red ginseng have no specific toxicity in liver and kidney.

Effects of Dietary Supplementation of Coffee Meal on Intestinal Enzyme Activity, Biochemical Profiles and Microbial Population in Broiler Chicks (커피박 첨가가 육계의 소장 효소 활성도, 생화학 지표 및 장내 미생물 균총에 미치는 영향)

  • The current study was performed to investigate the effects of dietary supplementation of dried coffee meal (CM) on growth performance, intestinal and blood biochemical index, intestinal enzymes, and cecal microbial populations. A total of 162, 3-day-old male broiler chicks were randomly allocated into three dietary groups: control group (CON), basal diet added with 0.5% CM (CM I), and basal diet added with 1.0% CM (CM II). Dietary supplementation of CM did not change bird performance and the relative weight of intestinal mucosal tissues. The birds fed the diet supplemented with CM (0.5 and 1.0%) significantly decreased mucosal glucose concentration (P<0.05) without affecting blood glucose level compared with those fed control diet. The level of blood aspartate aminotransferase (AST) significantly increased in CM II group (P<0.05) without affecting ${\gamma}$-glutamyl transpeptidase (${\gamma}$-GTP) compared with that in the CON group. The specific activity of intestinal maltase, leucine aminopeptidase (LAP) and alkaline phosphatase (ALP) were not affected by dietary supplementation of CM, whereas sucrase activity in birds fed the diet supplemented with CM was decreased (P<0.05) compared to that in the control birds. The colony forming units (CFU) of E. coli in the cecum of CM-fed birds was significantly decreased (P<0.05) compared with that of control birds without changing the CFU of Lactobacillus. In conclusion, dietary supplementation of lower level of CM (0.5%) can be used as a beneficial feed resource without liver toxicity in broiler chicks.

Toxicological Studies on Aucubin(I) -Acute Toxicities and Effects on Blood Serum Enzymes- (Aucubin의 독성연구(I) -급성독성 및 혈청효소에 미치는 영향 -급성독성 및 혈청효소에 미치는 영향-)

  • Aucubin, an iridoid glucoside which was previously reported to exhibit liver-protective activities against $CCl_4$ and ${\alpha}-amanitin$ induced liver damages, was subject to toxicological studies. To measure the lethal dose, the doses of 100mg/kg, 300mg/kg, 600mg/kg and 900mg/kg were administered intraperitoneally to experimental mice. No death was observed 24 hrs later, but serum GOT and alkaline phosphatase activities were deceased slightly at the doses of 300mg to 900mg/kg, and the triglyceride contents were slightly increased. To investigate acute toxicity of aucubin itself, multiple dosages(20 mg/kg, 40 mg/kg and 80 mg/kg for four times a week) were injected intraperitoneally into mice, then serum enzymes activities and chemistries were assayed; no significant change of the enzyme activities of alkaline phosphatase, GPT, GOT in the test groups were observed in comparison with those of the control group, and the contents of triglyceride, glucose, urea nitrogen and total proteins in the test group serums appeared to be almost same levels as those of the control group were. Histological examiation on liver biopsy samples indicated no gross changes between the control group and the test group were noted. Therefore, aucubin appears to be apparently low toxic substance and its minimum lethal dose in mouse seems to be more than 0.9 g.

Toxicity of ivermectin in Jindo-dogs 1. Clinical and hematological observation (진돗개에서 ivermectin의 독성 I. 임상증상과 혈액학적 변화 관찰)

  • Ivermectin is a synthetic derivative of the naturally occurring avermectin $B_{1a}$ (22, 23-dihydroavermectin $B_{1a}$) and $B_{1b}$ (22, 23-dihydroavermectin $B_{1b}$), It is widely used as antiparasitic and pesticidal agents because of its remarkably potent and broad spectrum of antiparasitic activity. Although the drug has shown excellent anthelmintic effects, development of toxicosis in some animals such as the Collie species of dog is well documented. However, no studies have been reported on the toxic effects of the drug in Korean native animals such as the Jindo dog. The toxic effect of ivermectin was evaluated in 25 Jindo dogs divided into five groups which were orally administered with ivermectin at dosage levels of $200{\mu}g/kg$, $300{\mu}g/kg$, $600{\mu}g/kg$ and $2,500{\mu}g/kg$ of body weight, respectively. Toxic signs were not observed in the groups receiving $200{\mu}g/kg$ and $300{\mu}g/kg$ B.W. ivermectin. One dog developed mild clinical signs of toxicosis in the group receiving $600{\mu}g/kg$ dosage of ivermectin. In the group with $2,500{\mu}g/kg$ dosage, all dogs developed mild (salivation, drooling, vomiting, mydriasis, and/or confusion) and/or moderate (ataxia and tremors) clinical signs of toxicosis. Hematologic changes were not observed in the groups receiving $200{\mu}g/kg$, $300{\mu}g/kg$ and $600{\mu}g/kg$ B.W. ivermectin. In the groups receiving $2,500{\mu}g/kg$ B.W., total erythrocyte counts, total and differential leukocyte counts and hemoglobin levels were not affected by drug. Aspartate aminotransferase levels were increased after administration of ivermectin, while serum cholesterol and blood glucose levels were decreased.

Studies on Antitumor Components of Flammulina velutipes of Korea (I) (팽나무 버섯의 항암(抗癌) 성분(成分)에 관한 연구(硏究)(제 1보)(第 1報))

  • To find antitumor components with low toxicity in the Basidiomycetes of Korea, the carpophores of Flammulina velutipes (Fr.) Singer were extracted with hot water for eight hours. The extract was purified by dialyzing through Visking tube and a protein-bound polysaccharide fraction was obtained as pale brownish amorphous powder after freeze-drying. The fraction was examined for antitumor activity against sarcoma 180 implanted subcutaneously in the left groin in ICR mice. The inhibition ratio of this fraction against the tumor was 62.3% at the dose of 10mg/kg/day for the period of ten days. The tumors in three of the ten treated mice were completely regressed. The chemical analysis of the antitumor component by anthrone and Lowry-Folin methods showed that it consisted of a polysaccharide (42.4%) and a protein (24.5%). The hydrolysis of the polysaccharide moiety with 3% HCl-MeOH and trimethysilylation of the hydrolyzate yielded five monosaccharides which were identified by G.L.C. Several amino acids were identified by an amino acid autoanalyzer in the acid hydrolyzate of the protein moiety.

The Effects of Dai-saiko-to (Da-Chai-Hu-Tang) on 3T3-L1 Preadipocytes and High-Fat Diet-Induced Obese Mice (대시호탕(大柴胡湯)이 3T3-L1 지방전구세포와 고지방식이 유도 비만쥐에 미치는 영향)

  • Objectives This experimental study was designed to investigate the effects of Dai-saiko-to (DSH) on differentiation of 3T3-L1 preadipocytes and body weight, serum lipid levels in high-fat diet-induced obese mice. Materials and Methods Cells were incubated with DSH at an indicated concentration (0.01-1 mg/ml) for 24h, then the growth rate was assessed by MTS assay. 3T3-L1 preadipocytes were incubated in DMEM for 2 days with the indicated concentrations of DSH. On Day 6, the cells were fixed and the cellular lipid contents were assessed by Oil-Red-O staining. The expression of peroxisome proliferator-activated receptor ${\gamma}$ ($PPAR{\gamma}$) and cytidine-cytidine-adenosine-adenosine-thymine (CCAAT)/enhancer-binding proteins ${\alpha}$ ($C/EBP{\alpha}$) as adipocyte-specific proteins were determined by real time RT-PCR and western blotting. Four-weeks old mice (wild-type C57BL/6) were used for all experiments. Body weight gain and serum lipid levels were measured in the obesity-induced mice. Results DSH did not show toxicity even at the concentration of 1 mg/ml and DSH significantly inhibited the differentiation of 3T3-L1 preadipocytes in a dose-dependent manner. Also, DSH significantly reduced the expressions of $PPAR{\gamma}$ and $C/EBP{\alpha}$ in a dose-dependent manner. Furthermore, DSH significantly reduced body weight gain, serum glucose, total cholesterol and LDL-cholesterol contents in obesity-induced mice. Conclusions These results demonstrated that DSH inhibited 3T3-L1 preadipocyte differentiations and high-fat diet-induced obesity in mice.

Effects of Carthamus Tinctorius Extract on Adipogenic Differentiation of Mouse Bone Marrow-Derived Mesenchymal Stromal Stem Cells (홍화 추출물이 생쥐 골수 유래 중간엽 줄기세포의 지방분화에 미치는 영향)

  • Objective: This study investigated the effect of purified Carthamus tinctorius (C. tinctorius) extracted with a hot water and ethanol method on adipogenic differentiation of mouse bone marrow-derived mesenchymal stromal stem cells (mBMSCs). Methods: The C. tinctorius was extracted using hot water and ethanol. The samples were concentrated by a rotary evaporator and were then dried using a freeze-dryer. The mBMSCs were cultured and maintained in a minimum essential medium eagle alpha (${\alpha}-MEM$) supplemented with 10% FBS and 1% antibiotic antimycotic solution. To induce adipogenic differentiation, the cells were treated with Dulbecco's modified eagle's medium-low glucose (DMEM-LG) containing 1 mg/mL insulin, 1 mM dexamethasone, and 0.5 mM 3-isobutyl-1-methylxanthine. To evaluate the adipogenic differentiation ability, oil-red O staining was performed after adipogenic differentiation for 21 days. The mRNA expression and protein level of adipogenic-related genes were quantified by quantitative real-time PCR and western blotting, respectively. Results: In the results of the MTT assay, no concentrations of C. tinctorius extracts showed toxicity on mBMSCs, so we fixed the treatment concentration of the extract at 100 ng/mL. In oil-red O staining, the water-C. tinctorius extract treatment significantly decreased adipogenic differentiation compared with the control and ethanol extract groups. The water-C. tinctorius extract group in particular showed reduced mRNA and protein expression of Peroxisome proliferator-activated receptor gamma ($Ppar{\gamma}$) and CCAAT/enhancer-binding protein alpha ($C/ebp{\alpha}$), which are adipogenic-related transcription factors. Conclusion: These data suggest that extract of C. tinctorius decreased the adipogenic differentiation of mBMSCs, while only water-C. tinctorius extract had an effect on different adipogenesis in mBMSCs. The C. tinctorius will be a useful therapeutic reagent for the prevention of obesity-related diseases such as diabetes, hyperlipidemia, coronary artery disease, and osteoporosis.
