Fundamental Investigation of Non-invasive Determination of Glucose by Near Infrared Spectrophotometry (근적외선 분광법을 이용한 비침투적 혈당 분석법 개발에 관한 기초 연구)

  • This study is to improve the diagnosis of diabetes mellitus and the self-monitoring of blood glucose in people with diabetes by providing a non-invasive method of monitoring blood glucose. A near-infrared (NIR) spectrophotometer was used to measure absorption spectra of 80 glucose samples ranges from 1 mg/dL to 200 mg/dL, and shows the standard error of prediction 1.8 mg/dL. Also, to investigate the effect of interference in blood, NaCl and sand were added in glucose and found the standard error of prediction of 2.8 mg/dL and 3.8 mg/dL, respectively. A new and more accurate calibration system for the spectrophotometer was developed from systematic study of light scattering, which cause nonlinear spectrophotometer response.

Study of the mechanisms underlying increased glucose absorption in Smilax china L. leaf extract-treated HepG2 cells (청미래덩굴 잎 물추출물이 처리된 HepG2 세포에서의 포도당흡수기전 연구)

  • Purpose: Previous studies have shown that treatment with Smilax china L. leaf extract (SCLE) produces antidiabetic effects due to ${\alpha}$-glucosidase inhibition. In this study, we examined the mechanism underlying these antidiabetic effects by examining glucose uptake in HepG2 cells cultured with SCLE. Methods: Glucose uptake and glucokinase activity were examined using an assay kit. Expression of glucose transporter (GLUT)-2, GLUT-4, and HNF-$1{\alpha}$ was measured by RT-PCR or western blot. Results: Treatment with SCLE resulted in enhanced glucose uptake in HepG2 cells, and this effect was especially pronounced when cells were cultured in an insulin-free medium. SCLE induced an increase in expression of GLUT-2 but not GLUT-4. The increase in the levels of HNF-$1{\alpha}$, a GLUT-2 transcription factor, in total protein extract and nuclear fraction suggest that the effects of SCLE may occur at the level of GLUT-2 transcription. In addition, by measuring the change in glucokinase activity following SCLE treatment, we confirmed that SCLE stimulates glucose utilization by direct activation of this enzyme. Conclusion: These results demonstrate that the potential antidiabetic activity of SCLE is due at least in part to stimulation of glucose uptake and an increase in glucokinase activity, and that SCLE-stimulated glucose uptake is mediated through enhancement of GLUT-2 expression by inducing expression of its transcription factor, HNF-$1{\alpha}$.

Improvement in Enzyme Immobilization of Polypyrrole Enzyme Electrode using Radical Transfer (Radical Transfer 반응을 이용한 Polypyrrole 효소전극의 효소고정화 향상)

  • In the case of immobilizing of glucose oxidase into polypyrrole (PPy) using electrosynthesis, the glucose oxidase (GOx) forms a coordinate bond with the polymers backbone. However, because of intrinsic insulation and net-chain of the enzyme, the charge transfer and mass transport are obstructed during the film growth. Therefore, the film growth is dull. We synthesized the enzyme electrode by electropolymerization added some organic solvent. A formative seeds of film growth is delayed by adding ethanol. The delay is induced by radical transfer between ethanol and pyrrole monomer. The radical transfer shares the contribution of dopant between electrolyte anion and GOx polyanion. This may lead to increase amount of immobilized the enzyme in PPy. For the UV absorption spectra of synthetic solution before synthesis and after, in the case of ethanol added, the optical density was slightly decreased for the GOx peaks. It suggests amount of GOx in the solution was decreased and amount of GOx in the film was increased. We established qualitatively that amount of immobilization can be improved by adding a little ethanol in the synthetic solution. It is due to radical transfer reaction. The radical transfer shares the contribution of dopant between small and fast electrolyte anion and big and slow GOx polyanion.

Varietal Variations in Physicochemical Characteristics and Amylopectin Structure of Grain in Glutinous Rice

  • Thirty-eight glutinous rice varieties were classified into nine groups on the scatter diagram by the upper two principal components (56% contribution to the total information) based on eleven physicochemical characteristics including the viscograms and physical properties of cooked rice. The first principal component was the factor mainly associated with the viscogram characteristics of rice flour emulsion and the second was the factor chiefly related to the physical properties of cooked rice and water absorbability of rice grain. Indica glutinous rices were clearly distinguished from japonica ones by the first principal component score. Javanica glutinous rices were widely distributed on the intermediate zone between indica and japonica or on several japonica rice groups. Significant positive or negative correlations were found among water absorption rates of rice grain, physical properties of cooked rice, and viscogram characteristics of rice flour. Especially in japonica glutinous rices, the breakdown and setback viscosities of rice flour were closely associated with the alkali digestion value of milled rice and the stickiness of cooked rice. The frequency ratio of short glucose chains (A-chain) to intermediate glucose chains (B-chain), the ratio of B- chains to long glucose chains (C-chain) and the relative frequency of A- or B-chain fractions representing the amylopection structure of rice starch was closely associated with the breakdown and setback viscosities of rice flour.

Antioxidant and Anti-hyperglycemic Effects of a Sanghwang Mushroom(Phellinus linteusau) Water Extract (상황버섯 열수추출물의 항산화 활성과 식후 혈당 상승 억제 효과)

  • The inhibitory activities of a water extract of Sanghwang mushroom(Phellinus linteusau)(SWE) against ${\alpha}$-glucosidases were evaluated in this study. Inhibiting these enzymes involved in the absorption of disaccharides significantly decreases the postprandial increase in blood glucose level after a mixed carbohydrate diet. Oxygen radical absorbance capacity and 1,1-diphenyl-2-picryl hydrazyl scavenging activities of the SWE were evaluated to investigate the antioxidant activity of the SWE associated with complications of long-term diabetes. Furthermore, the postprandial blood glucose lowering effect of SWE was compared to a known type 2 diabetes drug($Acarbose^{(R)}$) in a Sprague-Dawley rat model. SWE significantly reduced the blood glucose increase after sucrose loading. These results suggest that SWE, which has high ${\alpha}$-glucosidase inhibitory activity and high antioxidant activities, has the potential to contribute to a useful dietary strategy for controlling postprandial hyperglycemia.

In vitro Study on the Functionality in Digestive Tract of Chitin and Chitosan from Crab Shell (게껍질 Chitin 및 Chitosan의 소화관내 기능성에 관한 in vitro 연구)

  • Chitin and chitosan samples prepared from crab's shells under different conditions were compared for their physicochemical properties; and functionality in gastrointestinal tract by in vitro test. Their bulk density was in the range of $127{\sim}208\;mg/ml$, and their viscosity was $80{\sim}581\;cP$ in 0.1 chitin and $80{\sim}3,670\;cP$ in 0.5% chitosan solution, showing a wide variation. The degree of deacetylation in chitosan samples as determined by IR spectral analysis was relatively high, showing $81{\sim}93%$. At the same alkali concentration and reaction temperature, a longer reaction period gave an increased degree of deacetylation and lower viscosity. The water holding capacity of chitic substance became greater at higher soaking temperature; chitosan D at $37^{\circ}C$ showed the greatest value. Chitic substance with lower bulk density showed the higher water holding capacity. The retardation effect toward glucose absorption was higher in critic substances of lower density and higher water holding capacity; chitosan D showed the highest value of 38%. The retardation index toward bile acid absorption after 1 hour dialysis was $15{\sim}34%$ in chitic substances, 39% in pectin and 9% in cellulose. The retarding effect showed the highest value of 34% in chitosan D at 3% concentration.

A Study on the Effect of Sangbaegpitang & Supungsungiwhan on the Glucose Metabolism of db/db Mice (상백피탕(桑白皮湯)과 수풍순기환(搜風順氣丸)이 db/db Mice의 당대사(糖代謝)에 미치는 영향(影響))

  • In this study, body weight levels of glucose, insulin and triglyceride in blood and glucosidase activity of the small intestine were investigated to determine the effect of Sangbaegpitang and Supungsungiwhan on the glucose metabolism of db/db mice. The GLUT4 mRNA of muscle tissue and the Acetyl CoA Carboxylase and the activation rate of GLUT2 mRNA of liver tissue were measured by the reverse transcription-polymerase chain reaction method and by the vitro transcription. The results were obtained as follows: 1. In the Sangbaegpitang administration group, (1) The level of triglyceride was decreased significantly and the glucosidase activity of the small intestine was inhibited remarkably, (2) The amounts of the GLUT4 mRNA in muscle tissue and Acetyl CoA Carboxylase mRNA in liver tissue were increased significantly. (3) Though glucose level in both fasting and non-fasting, were decreased and the insulin level in blood was increased, the results showed no statistical significance. 2. In the Supungsungiwhan administration group, (1) The levels of glucose and triglyceride were decreased significantly in the blood of non-fasting animals. (2) The glucosidase activity of small intestine was inhibited markedly and the amounts of GLUT4 mRNA of muscle tissue and GLUT2 mRNA of liver tissue were increased significantly. (3) The glucose levels in the fasting group were reduced, while insulin level was increased but showed no statistical significance, Based on the above results, our conclusions are as follows: Sangbaegpitang & Supungsungiwhan are thought to be capable of inhibiting the activity glucosidase, the enzyme which influences carbohydrate metabolism in the small intestine of db/db mice(the experimental diabetic model) and delaying the absorption of carbohydrate, thus proving effective on inhibiting the increase of non-fasting glucose level effectively. Futhermore Sangbaegpitang and Supungsungiwhan are though: to be capable of preventing the composition of free fatty acids by restoring the production of GLUT4 mRNA of muscle tissues and GLUT2 mRNA of liver tissues. Those results suggests that above prescriptions can be applied to non-insulin dependent diabetes mellitus in order to improve insulin resistance.

The non-saponin fraction of Korean Red Ginseng (KGC05P0) decreases glucose uptake and transport in vitro and modulates glucose production via down-regulation of the PI3K/AKT pathway in vivo

  • Background: The non-saponin fraction of Korean Red Ginseng has been reported to have many biological activities. However, the effect of this fraction on anti-diabetic activity has not been elucidated in detail. In this study, we investigated the effects of KGC05P0, a non-saponin fraction of Korean Red Ginseng, on anti-diabetic activity in vitro and in vivo. Methods: We measured the inhibition of commercially obtained α-glucosidase and α-amylase activities in vitro and measured the glucose uptake and transport rate in Caco-2 cells. C57BL/6J mice and C57BLKS/Jdb/db (diabetic) mice were fed diets with or without KGC05P0 for eight weeks. To perform the experiments, the groups were divided as follows: normal control (C57BL/6J mice), db/db control (C57BLKS/Jdb/db mice), positive control (inulin 400 mg/kg b.w.), low (KGC05P0 100 mg/kg b.w.), medium (KGC05P0 200 mg/kg b.w.), and high (KGC05P0 400 mg/kg b.w.). Results: KGC05P0 inhibited α-glucosidase and α-amylase activities in vitro, and decreased glucose uptake and transport rate in Caco-2 cells. In addition, KGC05P0 regulated fasting glucose level, glucose tolerance, insulin, HbA1c, carbonyl contents, and proinflammatory cytokines in blood from diabetic mice and significantly reduced urinary glucose excretion levels. Moreover, we found that KGC05P0 regulated glucose production by down-regulation of the PI3K/AKT pathway, which inhibited gluconeogenesis. Conclusion: Our study thereby demonstrated that KGC05P0 exerted anti-diabetic effects through inhibition of glucose absorption and the PI3K/AKT pathway in in vitro and in vivo models of diabetes. Our results suggest that KGC05P0 could be developed as a complementary food to help prevent T2DM and its complications.

Facilitation of Glucose Uptake by Lupeol through the Activation of the PI3K/AKT and AMPK Dependent Pathways in 3T3-L1 Adipocytes (3T3-L1 지방세포에서 PI3K/AKT 및 AMPK 경로의 활성화를 통한 루페올의 포도당 흡수촉진 효과)

  • Lupeol is a type of pentacyclic triterpene and has been reported to have pharmacological activities against various diseases; however, the effect of lupeol on glucose absorption has not been elucidated yet. This study aimed to investigate the effect of lupeol on glucose uptake in 3T3-L1 adipocytes. Lupeol significantly facilitated glucose uptake by translocating glucose transporter type 4 (GLUT4) to the plasma membrane of the 3T3-L1 adipocytes, which was related to activation of the phosphoinositide 3-kinase (PI3K)/protein kinase B (AKT) and 5 'adenosine monophosphate-activated protein kinase (AMPK) pathways. In the PI3K/AKT pathway, lupeol stimulates the phosphorylation of insulin receptor substrate 1 (IRS-1), which activates PI3K. Its activation by lupeol promotes the phosphorylation of AKT, but not the atypical protein kinase C isoforms ζ and λ. Lupeol also promoted the phosphorylation of AMPK. The activation of AMPK increased the expressions of the plasma membrane GLUT4 and the intracellular glucose uptake. The increase in the glucose uptake by lupeol was suppressed by wortmannin (PI3K inhibitor) and compound C (AMPK inhibitor) in the 3T3-L1 adipocytes. The results indicate that lupeol can facilitate glucose uptake by increasing insulin sensitivity through the stimulation of the expression of plasma membrane glucose transporter type 4 via the PI3K/AKT and AMPK pathways in the 3T3-L1 adipocytes.

Natural Indigo Dyeing by Using Glucose Reduction (포도당 환원을 이용한 천연 인디고 염색)

  • Dyeing process of the natural indigo powder onto ramie and silk fabrics was investigated by using glucose and calcium hydroxide as a reducing system. Effect of reduction and dyeing conditions such as temperature and time of reduction/dyeing, and concentrations of glucose and calcium hydroxide on the dyeing process were explored. Indigo powder was obtained by drying the conventional niram paste in an oven at $50^{\circ}C$. Color strength of the dyed fabrics was evaluated by K/S value measured at the wavelength of maximum absorption(${\lamda}$max). Munsell color coordinates(H V/C) were used to compare fabric colors of ramie and silk. Ramie fabric showed purple-blue color for all the temperature and time. On the contrary, silk fabric showed wide range of color including brown, brown-green, green at the different temperature. With the increase of K/S value, the coordinate of value(lightness) decreased for both of ramie and silk fabrics. The coordinate of hue(shade) changed drastically with the increase of K/S value for silk fabric, compared with that of ramie fabric which showed nearly constant value at the whole range of K/S value. Optimum concentrations of calcium hydroxide were for 6 g/L for ramie and 4 g/L for silk at $60^{\circ}C$ and 50 min. K/S value increased with the indigo concentration. Maximum K/S value was shown at $10{\sim}12$ g/L of glucose concentration. For both of ramie and silk fabrics, the colorfastness of washing and light was lower than that of rubbing. All the colorfastness values were improved with the increase of color strength.
