For the information on micellization at each ginsenoside level aqueous solution of purified saponin of Panax ginseng root was dialyzed through dialysis tubing (MW 12,000) or eluted through Bio-Gel P-2 (MW 200-2,000) and analysed for ginsenosides by high performance liquid chromatography. Ginsenosides can be classified into three groups depending upon molecular aggregation pattern and spatial arrangement of hydrophilic parts in molecule. Group I that is large micelle former(aggregation number: above 10) and one side hydrophilic part (HP) includes $ginsenoside\;Rb_1$, $Rb_2$, Rc and Rd (diols). Group II thai is small micelle former (aggregation number:>10-1) and semi-two sales HP includes $Rg_2$, Rf (triol) and $Rg_3$ (diol). Group III that is no micelle former (aggregation number: 1) and two sides HP includes Re and $Rg_1$ (triol).
Contents of essential oil and its composition in aerial part of Agastache rugosa were investigated. Essential oil was obtained from the dried stems, leaves and flowers by steam distillation and fractionated into hydrocarbon and oxygenated hydrocarbon by silica gel column chromatography. Each isolate or fraction was identified by GC and GC-MS. The contents of essential oil were 0.29% in leaves of Agastache rugosa on dry basis, 0.38% in flowers. Major components were ${\beta}-caryophyllene(59.3%)$, limonene(13.1%), ${\delta}-cadinene(10.7%)$ among 13 kinds of components confirmed in hydrocarbon fraction of essential oil isolated from leaves. Also, major components in oxygenated hydrocarbon fraction were methyl chavicol (79.1%) and cis-3(1-propenyl) phenol(4.5%) among 44 kinds components confirmed. The highest content among the components identified was methyl chavicol in both leaves and flowers. On the other hand, limonene, trans-2-hexenal, 1-octen-3-one, 6, 10, 14- trimethyl pentadecane- 2-one and phytol were detected typically in leaves, but jasmone and ${\rho}-methoxyacetophenone$ were detected mainly in flowers with small quantity.

Angiogenesis is important for promoting cardiovascular disease, wound healing, and tissue regeneration. We here investigated the pharmacological effects of Korea red ginseng water extract (KRGE) on angiogenesis and its underlying signal mechanism. This study showed that KRGE increased in vitro proliferation, migration, and tube formation of human umbilical endothelial cells, as well as stimulated in vivo angiogenesis. KRGE-induced angiogenesis was accompanied by phosphorylation of ERK1/2, Akt, and endothelial nitric oxide synthase (eNOS) as well as an increase in NO production. Inhibition of PI3K activity by wortmannin completely inhibited KRGE-induced angiogenesis and phosphorylation of Akt, ERK1/2, and eNOS, indicating that PI3K/Akt activation is an upstream event of KRGE-mediated angiogenic pathway. The MEK inhibitor PD98059 completely blocked KRGE-induced angiogenesis and ERK phosphorylation without affecting Akt and eNOS activation. However, the eNOS inhibitor NMA effectively inhibited tube formation, but partially blocked proliferation and migration as well as ERK phosphorylation without altering Akt and eNOS activation, revealing that eNOS/NO pathway is in part involved in ERK1/2 activation. This study first demonstrated the critical involvement of both ERK1/2 and eNOS activation in KRGE-induced angiogenesis, which lie on downstream of PI3K/Akt. Thus, these results indicate that KRGE requires activation of both the PI3K/Akt-dependent ERK1/2 and eNOS signal pathways and their cross-talk for its full angiogenic activity.

Objectives: The purpose of this study was to characterize the effect of the fraction of Korean mountain ginseng folium (FKG) on the learning and memory impairments induced by scopolamine. Methods: The memory ameliorating effect of FKG was investigated using a passive avoidance test, the Y-maze test, and the Morris water maze test in mice. Drug-induced amnesia was induced by treating animals with scopolamine(1mg/kg, i.p.). Results: FKG (2 or 4mg/kg, p.o.) administration significantly reversed scopolamine-induced cognitive impairments in mice by the passive avoidance test and the Y-maze test(P<0.05), and also improved escape latency in the Morris water maze test at 2 or 4mg/kg(P<0.05). Although FKG has little inhibitory activity for AChE (IC50 value; 1847 ${\mu}g/ml$) in an invitro study, phosphorylated extracellular signal-regulated kinase(pERK) was increased by the administration of FKG inhippocampus on immunohistochemistry. Conclusions: These results suggest that FKG may be a useful cognitive impairment treatment, and its beneficial effects are mediated, in part, via activation of ERK pathway.
Dynamic parameters of biochemistry including the POD (peroxidase) activity, contents of DNA and RNA, isozymes of POD and ES (esterase) in American send (Panax quinquefolium L.) seed are reported in the present paper. The dynamic changes of POD activity proved that the PAS (physiological afterripening stage) is a stage in which some substances are prepared for seed germination. The POD activity correlated with ER (embryo ratio) significantly DNA content changed little only within 0.0036∼0.013 mg/ml, which did not correlate with ER. RNA content changed from 0.1539 to 1.0313 mg/ml and correlated significantly with RE during all of the embryo afterripening. None of the POD isozyme band was obtained in ESGS (embryo slowly growth stage), but five bands in ERGS (embryo rapidly growth stage) and six bands in PAS. Four bands of ES isozymes were obtained in ESGS, but six bands in ERGS, particularly, the content of ES isozymes increased in PAS. All of these may provide some information for understanding the dormancy mechanisms of American song seed.

Black ginseng (BG) obtained by a 9-fold steaming process of Panax ginseng has been reported to have anti-oxidative, anti-obesity, and anti-diabetes effects. The current study evaluated the protective effect of BG by steaming time in an HCl/ethanol-induced acute gastritis model. BG was divided into four samples according to steaming-drying processing (Gin1, Gin3, Gin6, and BG). High performance liquid chromatography analysis, free radical scavenging activity, and total phenol and flavonoid contents were examined in ginseng and four BG samples. Compared with ginseng, BG showed a stronger radical scavenging effect and higher contents of total phenol and flavonoids. To evaluate the anti-gastritic effect of BG, mice were distributed into five groups: normal mice (N), acute gastritic mice with distilled water (CON), acute gastritic mice with 100 mg/kg of ginseng (Gin0), acute gastritic mice with 100 mg/kg of BG (BG), and acute gastritic mice with 10 mg/kg of sucralfate (SC). After 1 hour of pre-treatment with water, extracts (Gin0 and BG), or drug (SC), experimental groups except for N were orally administered 0.5 mL of 150 mM HCl/60% ethanol (v/v) mixture. Blood was collected 1 hour later from the heart, and gastric tissue was harvested. Reactive oxygen species (ROS) levels were measured in serum, and related protein expression was examined by Western blot assay. In HCl/ethanol-induced acute gastritic mice, treatment with ginseng or BG improved mucosal damage in the histological evaluation. The serum ROS level significantly decreased in the BG-treated group compared with the CON group. Furthermore, expression of inflammatory cytokines significantly decreased in the BG-treated group compared with the CON group. Based on these results, antioxidant and anti-gastritic activities of ginseng were enhanced by streaming-drying processing, in part due to an increase in biological active compounds.
These experiments were carried out to define the effects of NAA, 2, 4-D and Benzyladenine on the callus induction and the organ differentiation from the explants and to find out the vegetative propagation method of Korean ginseng. The results obtained are summarized as follows; 1. NAA was significantly effective in forming roots from the ginseng stem segment and the number of roots was increased by increasing NAA concentration in the medium. The roots were formed from both distal and proximal ends of the ginseng stem segments grown on the medium containing more than 2mg/L of NAA. 2. The amount of callus growth increased proportionatly with NAA concentration in the range of 4.0mg per liter in the medium. The callus was easly induced from stem segment than leaf segment and 2, 4-D was more effective in callus induction and growth than NAA. 3. The benzyladenine showed the significant inhibition effect in forming roots from ginseng explant. The callus was not induced with BA alone, but in BA and 2, 4-D or BA and NAA added medium, the callus was easily induced and its growth was also accelerated. The interaction effects between 2, 4-D and BA on the callus induction and growth were significantly higher than those between NAA and BA. 4. As the ginseng embryos were cultured on the M.S. medium supplemented with 2mg per liter NAA, number of shoots was significantly increased and the percentage of embryo which had shown more than 4 shoots later was 22.2%. On the medium containing 8mg per liter NAA, the ginseng embryo showed the normal growth of shoots and leaves, but increased roots and callus induction on the basal part of shoots. 5. When the shoots with 3 leaflets were cut in 1.5cm long and grown on the Blayde's medium containing NAA 1.0mg per liter, roots were formed at the proximal end of shoot, and a new ginseng seedling was successfully obtained.
On May of 2002, the 34 isolates of Rhizoctonia solani were isolated from the symptom of damping-off on basal stems of 2-year-old to 6-year-old Panax ginseng which were cultivated in the 17 fields in Kyunggi-do, Chungcheungnam-do and Jeollabuk-do province in Korea. All isolates were identified as anastomosis group 2-1. Pre-emergence damping-off occurred on underground part of stem of 2-year-old ginseng in the pot trial with artificial inoculation. However, in the 4-year-old ginseng field with artificial inoculation, post-emergence damping-off occurred. The severe incidence of damping-off was found in the 6-year-old ginseng field in Kimje-si, Jeollabuk-do province on June 5 of 2003, the rate of which showed $18.6{\%}$ of area in the field by spread of the disease since 2-year-old. The sclerotia of R. solani, started to be formed after 7 days incubation on potato dextrose agar at $25^{\circ}C,$ were grayish brown, spherical to irregular and about $500{\mu}m$ in diameter, which became dark brown after 14 days incubation. The temperature range for the mycelial growth of R. solani isolates was $5\~30^{\circ}C,$ and the optimal temperature was $25^{\circ}C,$ their growth were very poor at $5\;or\;30^{\circ}C$. The isolates grew at the range of pH $4.5\~8.1$ tested and optimal pH for growth was pH 4.5$\~5.8%, whereas their growth were very poor above the pH 7.2.

Korean red ginseng is known to have anti-stress and memory enhancing effects. Recent studies suggested that stress-induced inhibition of adult neurogenesis in hippocampus may contribute, in part, to decreased negative feedback inhibition of HPA axis. In order to elucidate the mechanism of Korean red ginseng in anti-stress and memory enhancing effects, we observed the effects of repeated treatment of Korean red ginseng total saponin (GTS, 50 mg/kg, i.p.) in response to repeated unpredictable stress for 10 days. Male Sprague-Dawley rats (230 - 260 g) received with either GTS (50 mg/kg, i.p.) or vehicle (1 ml/kg, i.p.) 1 h before stress for 10 days. Rats were injected with bromodeoxyuridine (BrdU, 50 mg/kg, i.p.) 16-18 he after last stress procedure, and were sacrificed 2 hr later by perfusion. Immunohistochemistry of BrdU was done to measure proliferation of neural progenitor cells in hippocampus, which was used as an index of neurogenesis. Repeated GTS treatment for 10 days increased neurogenesis in subgranular zone area of dentate gyrus (SGZ), but not hilus, compared with vehicle-treated rats. Repeated unpredictable stress did not affect the neurogenesis compared with controls, while repeated GTS treatment increased neurogenesis in SGZ in repeated unpredictable stress-exposed group. BDNF mRNA was also measured in subregions of hippocampus by in situ hybridization. BDNF mRNA expression in CA3 and CA1 pyramidal cell layer was increased by repeated GTS treatment but not in dentate granule cell layer. Repeated unpredictable stresses significantly decreased BDNF mRNA expression in all subregions of hippocampus, but repeated GTS treatment did not prevent stress-induced BDNF mRNA downregulation. Given that repeated GTS treatment increased proliferation of neural progenitor cells in repeated unpredictable stress-exposed rats in the presence of decreased BDNF mRNA expression in dentate granule cell layer, it raise the possibility that BDNF may not playa significant role in GTS-mediated increase of neurogenesis in adult rat hippocampus. Also, these results suggest that repeated GTS treatment increased neurogenesis of SGZ and BDNF mRNA expression, which may account for memory enhancing effect of Korean red ginseng. In addition, repeated GTS treatment appears not to have anti-stress effects in terms of neurotrophin, but GTS-mediated increase of neurogenesis in hippocampus may contribute to increase negative feedback inhibition of HPA axis.

The origins and the functions of the multi vesicular bodies and the various structures of the membranes related to the cytolysomes were studied in the mycelium cells of Rhizopus nigricans, Aspergillus niger and A. ochraceus, in the hymenium and basidium cells of Agricus bisporus sand Rhizopogon rubesecens, in the cells of assimilation tissue of Marchtantia polymorpha and Pogonalum inflexum and in the mesophyll cells of Pteridium aqiulinum, Pinus densiflora, Ginkgo biloba and Panax ginseng fixed with glutaraldehyde-paraformaldehyde-$ OsO_4$. In Rhizopus nigricans, Aspergillus niger, A. ochraceus, Agricus bisporus sand Rhizopogon rubescens, the concentric multilamellar, multivesicular, myelin-vesicle-tubular and concentric parallel-lamellar complexes were originated from the plasmalemma, while in Marehantia polymorpha, Pogonatum inflexum, Pteridium aquilinum, Pinus densiflora, Ginkgo biloba and Panax ginseng, they were originated from plasmalemma and the cytoplasm. The structures originated from the plasmalemma may be grouped into multi vesicular body and myelin-like structure, both forming the secondary vacuoles or protruding into the central vacuoles and finally degrading, In some cases, endoplasmic reticulum within the cytoplasm encloses some part of the cytoplasm to form a circle where the membranous lamellae increase in number, while the enclosed cytoplasm decrease to be eventually replaced by the multilamellar structure which is released into the vacuoles and subsquently degraded. The structures originated from the cytoplasm are believed to be the cytosegresomes or cytolysomes closely related to the differentiation of the vacuoles. The possible fate of these structures are also discussed.
