  • The important biological activities of estrogen were reproduction and physiological processes in a number of tissues, including liver, bone, brain, blood vessels, adipose tissue and so on. The regulation of estrogen level is important a prevention of estrogen-related disease. Ginkgo biloba extracts (GSE) are extracted from leaves of the Ginkgo biloba tree. Peroxyl Radical Scavenging Capacity of the Flavonolignan Silybin, Ginkgo Biloba Extract EGb 761, American Green Tea and a Series of Germacranolides

  • We report on the applicability oj a method recently developed in our laboratory for measuring the antioxidant potential of isolated chemicals and extracts derived from natural products. Peroxyl radicals generated by thermal homolysis of 2,2'-azobis-amidinopropane (ABAP) oxidize $\alpha$-keto-${\gamma}$-methiolbutyric acid (KMBA) to ethylene, which is monitored by gas chromatography. Inhibition of ethylene formation in the presence of antioxidants that compete with KMBA for peroxyl radicals is the basis of the Total Oxyradical Scavenging Capacity Assay (TOSCA; Winston et al., 1998). Antioxidative activities of water-soluble extracts of American green tea, the anti-hepatotoxic flavonolignan from milk thistle (Silybum marianum) silybin, Ginkgo biloba extract EGb 761, and a series of naturally occuring sesquiterpene lactones (all ger-macranolides found in in fungi, liverworts, and plants) were studied. The specific TOSC value per $\mu$M silybin was 5.2, which is essentially comparable to that of Trolo $x^{ⓡ}$, a water-soluble vitamine E analog. Tea and Ginkgo extracts exhibited potent peroxyl radical scavenging capacity with values, respectively of =1700 and 1000 $\mu$mols Trolo $x^{ⓡ}$ equivalent per gram dry matter. The known anti-inflammatory activity of some germacranolides prompted study of their antioxidant capacity. None of the lactones exhibited antioxidant capacity toward peroxyl radicals comparable to Trolo $x^{ⓡ}$; costunilide, the most lipophilic, had a TOSC value = to glutathione. The potential role of peroxyl radicals in lipidperoxidation, other cellular damage, and var-ious disease states suggest a possible preventive role for silybin, green tea and Ginkgo biloba in oxidative stress caused by these free radical species.ecies.

은행나무 종자 추출물의 미백효능 분석 (The Analysis of Whitening Effects on Extracts from Ginkgo (Ginkgo biloba L.) Seeds)

  • 은행나무 종자인 백과를 70% 에탄올로 추출하여 미백 효능을 규명하고 화장품 소재로서의 응용 가능성을 확인하고자 하였다. 백과 에탄올추출물 (GBE)을 B16F10 melanoma cells에 처치하여 멜라닌 생성과 tyrosinase 활성을 확인한 결과 유의한 수준의 멜라닌 생성 저해가 관찰되었고, 멜라닌 생성과정에 관여하는 주요 효소인 tyrosinase의 활성이 농도 의존적으로 억제됨이 관찰되었다. 멜라닌 생성 관련 주요 인자들의 단백질 발현과 mRNA 수준을 관찰한 결과, tyrosinase, tyrosinase-related protein-1, -2 (TRP-1, -2), microphthalmia-associated transcription factor (MITF)의 단백질 발현과 유전자 수준이 GBE의 처리에 의해 유의한 수준으로 저해되었다. 이 결과를 통해, 본 연구의 백과 에탄올추출물은 멜라닌 세포의 멜라닌 생성 관련 핵심 전사인자인 MITF의 조절을 통해 멜라닌 생성 억제 효과를 나타내는 것으로 보인다. 이에 따라 백과 에탄올추출물을 화장품 미백 기능성 소재로 활용 가능성이 있을 것으로 기대된다.

3종의 식물 추출물에 의한 백점충의 in vitro 구제효과 (In vitro eliminative effects three sorts of herbal extracts of against Ichthyophthirius multifiliis)

  • 3종의 식물성 추출물 및 포르말린의 시험관 내에서의 백점충에 대한 살충효과를 조사하였다. 복합생약제는 500배 희석액에서 1시간 이내에 충을 완전히 죽일수 있었으나, 여지초발효액은 100배의 희석액에서 1시간 이내에 모든 충을 죽일 수 있었다. 은행잎 추출물은 5,000배의 희석액으로 1시간 이내에 모든 충을 죽일 수 있었다. 대조약제로서 포르말린은 100ppm의 농도로 1시간 이내에 충을 모두 죽일 수 있었다. 이 결과로 은행잎 추출물이 살충효과가 가장 좋았다. 충에의 밀도에 따른 살충효과의 차이는 없었다. 또한 은행잎 추출물의 5,000배 희석액으로 2시간 동안 약욕한 후의 어류의 아가미 조직에 있어서 변화가 없었다.

포도상구균에 대만 에탄올 농도별 은행잎 추출물의 항균효과에 관한 연구 (A Study on the Antimicrobial Effect of Ginkgo biloba Leaves Extracts according to Concentrations of Ethanol for staphylococcus aureus)

  • 본 연구에서는 은행잎 추출물의 Staphylococcus aureus에 대한 항균효과를 검증하기 위해 먼저 에탄올 농도별로 bilobalide와 ginkgolide A, B의 성분을 분석한 결과 40% 에탄올 용매를 최적 활성농도로 결정하였다. Disc diffusion test, Optical density test을 통한 S. aureus 항균실험 결과 에탄올 추출농도가 증가할수록 항균효과가 증가하나 40% 에탄올 이상에서는 항균활성에 큰 차이가 없었다. 주사전자현미경을 통하여 은행잎 에탄올 40% 추출물 16배 농축액을 처리한 균의 세포표면을 확인한 결과 심하게 손상되었음을 확인할 수 있었다. 투과전자현미경을 이용하여 관찰한 결과 은행잎 추출물로 처리한 균주에서는 세포벽이 관찰되지 않았으며, 이는 은행잎 추출물의 주성분인 bilobalide와 ginkgolide A, B가 세포벽 합성을 저해하는 것으로 보여진다.

Inhibitory Aromatase Effects of Flavonoids from Ginkgo Biloba Extracts on Estrogen Biosynthesis

  • Ginkgo biloba extract (GBE) is a popular phytomedicine and has been used for disorders of the central nervous system, cardiovascular, renal, respiratory, and circulatory diseases. Although GBE is a complex mixture of over 300 compounds, its major components are 24% flavonoids and 6% terpene lactones. In this study, we tested the inhibitory effects of the three major flavonoids (kaempferol, quercetin, and isorhamnetin) from GBE, independently and as mixtures, on aromatase activity using JEG-3 cells (human placental cells) and recombinant proteins (human placental microsome). In both systems, kaempferol showed the strongest inhibitory effects among the three flavonoids; the flavanoid mixtures exerted increased inhibitory effects. The results of exon I.1-driven luciferase reporter gene assays supported the increased inhibitory effects of flavonoid mixtures, accompanied by suppression of estrogen biosynthesis. In the RT-PCR analysis, decreased patterns of aromatase promoter I.1 mRNA expressions were observed, which were similar to the aromatase inhibition patterns of flavonoids and their mixtures. The present study demonstrated that three flavonoids synergistically inhibit estrogen biosynthesis through aromatase inhibition, decrease CYP19 mRNA, and induce transcriptional suppression. Our results support the usefulness of flavonoids in adjuvant therapy for breast cancer by reducing estrogen levels with reduced adverse effects due to estrogen depletion.

Phenolic plant extracts are additive in their effects against in vitro ruminal methane and ammonia formation

  • Objective: The methane mitigating potential of various plant-based polyphenol sources is known, but effects of combinations have rarely been tested. The aim of the present study was to determine whether binary and 3-way combinations of such phenol sources affect ruminal fermentation less, similar or more intensively than separate applications. Methods: The extracts used were from Acacia mearnsii bark (acacia), Vitis vinifera (grape) seed, Camellia sinensis leaves (green tea), Uncaria gambir leaves (gambier), Vaccinium macrocarpon berries (cranberry), Fagopyrum esculentum seed (buckwheat), and Ginkgo biloba leaves (ginkgo). All extracts were tested using the Hohenheim gas test. This was done alone at 5% of dry matter (DM). Acacia was also combined with all other single extracts at 5% of DM each, and with two other phenol sources (all possible combinations) at 2.5%+2.5% of DM. Results: Methane formation was reduced by 7% to 9% by acacia, grape seed and green tea and, in addition, by most extract combinations with acacia. Grape seed and green tea alone and in combination with acacia also reduced methane proportion of total gas to the same degree. The extracts of buckwheat and gingko were poor in phenols and promoted ruminal fermentation. All treatments except green tea alone lowered ammonia concentration by up to 23%, and the binary combinations were more effective as acacia alone. With three extracts, linear effects were found with total gas and methane formation, while with ammonia and other traits linear effects were rare. Conclusion: The study identified methane and ammonia mitigating potential of various phenolic plant extracts and showed a number of additive and some non-linear effects of combinations of extracts. Further studies, especially in live animals, should concentrate on combinations of extracts from grape seed, green tea leaves Land acacia bark and determine the ideal dosages of such combinations for the purpose of methane mitigation.

Screening of Antimicrobial Activity against Enterohemorrhagic escherichia coli 0157 : H7 from Plants in Korea

  • We screened the methanol extracts from 133 plant species growing in Korea for antimicrobial activity against enterohemorrhagic Escherichia coli 0157 : H7. Those are selected from three plant grouping ; traditional medicinal herbs, edible plants, and flowers. They were tested by disk diffusion assay. From evaluation of the inhibition zone diameter of microbial growth, we found that the flower extract of Rhododendron Schilpenbachii Max had the most significant antimicrobial activity against this bacteria. Extracts from most of the vegetables and plants did not show antimicrobial activity except for the leaves of Ginkgo biloba L. and seeds of Prunus Dallicina L. did not show antimicrobial activity except for the leaves of Ginkgo biloba L. and the seeds of Prunus sallicina L.

The Combined Effects of Ginkgo Biloba Extracts and Aspirin on Viability of SK-N-MC, Neuroblastoma Cell Line in Hypoxia and Reperfusion Condition

  • Objective: The purpose of this study is to investigate the combined effects of ginkgo biloba extract, ginkgolide A and B and aspirin on SK-N-MC, human neuroblastoma cell viability and mRNA expression of growth associated protein43 (GAP43), Microtubule-associated protein 2 (MAP2), B-cell lymphoma2 (Bcl2) and protein53 (p53) gene in hypoxia and reperfusion condition. Methods: SK-N-MC cells were cultured with Dulbecco's Modified Eagle's Medium (DMEM) media in $37^{\circ}C$, 5% $CO_2$ incubator. The cells were cultured for 8 hours in non-glucose media and hypoxic condition and for 12 hours in normal media and $O_2$ concentration. Cell survival rate was measured with Cell Counting Kit-8 (CCK-8) reagent assay. Reverse transcriptase polymerase chain reaction (RT-PCR) was used to estimate mRNA levels of GAP43, MAP2, Bcl2, and p53 genes. Results: The ginkgolide A and B increased viable cell number decreased in hypoxic and reperfused condition. The co-treatment of ginkgolide B with aspirin also increased the number of viable cells, however, there was no additive effect. Although there was no increase of mRNA expression of GAP43, MAP2, and Bcl2 in SK-N-MC cells with individual treatment of ginkgolide A, B or aspirin in hypoxic and reperfused condition, the co-treatment of ginkgolide A or B with aspirin significantly increased GAP43 and Bcl2 mRNA levels. In MAP2, only the co-treatment of ginkgolide A and aspirin showed increasing effect. The mRNA expression of p53 had no change in all treating conditions. Conclusion: This study suggests that the combined treatments of Ginkgo biloba extracts and aspirin increase the regeneration of neuroblastoma cells injured by hypoxia and reperfusion.

  • Effects of Ginkgo biloba leaf extract (GBE) and its components kaempferol-coumaroyl glucosyl rhamnoside (Kc), quercetin-coumaroyl glucosyl rhamnoside (Qc), ginkolide (G) and bilobalide (B) on blood pressure and on the levels of neurotransmitters in the spontaneously hypertensive rat brain were examined. Blood pressure decreased about 10 mmHg after administration of GBE but without significance. The concentration of norepinephrine increased (3 times) and that of DOPAC was decreased (38%) after administration of Qc. And 3-MT concentration was increased (36%) by Kc administration in striatum. While Qc administration increased the concentration of Ach (260%) and Ch(29%) significantly, B administration increased Ch concentration in cerebral cortex.
