  • Comparative karyological analyses of the Korean land snail, Diplommatina (Sinica) paxillus and Diplommatina (Sinica) changensis, were peformed by the Giemsa-staining and air-drying method. The karyotypes of both species were the same (2n = 26). However, the chromosome lengths and arm ratios, and relative chromosome lengths of the two species were distinctly different.

Characterization of haemocytes in the surf clam Mactra veneriformis

  • Haemocyte characterization in the surf clam, Mectra veneriformis was carried out based on morphological, cytochemical, and phagocytic characteristics. The haemocytes were classified into two cell types, granulocytes and agranulocytes on the basis of presence of cytoplasmic granules. Granulocytes were then classified again into 2 types, large eosinophilic granulocytes and small eosinophilic granulocytes after staining with May-Grünwald Giemsa. In electron microscopy, both types of granulocytes contained electron-dense and electron-lucent cytoplasmic granules. Agranulocytes (hyalinocytes) were also divided into two cell types, large agranulocytes and small agranulocytes based on their sizes. Both cell types did not have granules in cytoplasm. Granulocyte and agranulocyte were negative for the enzyme activities of alkaline phosphatase, peroxidase, and $\beta$-glucuronidase but positive for phenoloxidase and acid phosphatase activities. Both types of haemocytes have phagocytic activity, with the exception of small agranulocyte, and granulocytes seemed more active in this respect than agranulocytes. This present study is the first study to characterize haemocytes of M. veneriformis.

  • None of the numerous published canis idiogram and karyotypes has yet been generally accepted as a standard one because the dog has 76 acrocentric autosomes of similar size and shape. The karyotypes of DongGyeongi dog were analysed by conventional trypsin/Giemsa staining (GTG-banding techniques), and were compared with one another. There were no variations in karyotypes which were analysed by conventional GTG-banding techniques, but differences were observed in G-banding patterns with sapsaree (or canis familiaris strains). It is not clear that these disagreements in G-banding patterns between strains of dog were caused by chromosome polymorphism or a difference in interpretation.

  • The chromosomal analyses of Dendmcopos major hondoensis and D. leucotos Ieucotos(Picidae:Piciformes) in Korea were performed by conventional Giemsa staining and C-banding method. The diploid number of two species was 2n=90-92 and arm number was AN= 92-96.The conventional karyotypes were very similiar but distribution of constitutive heterochromatin were differ in the first chromosome. The second and several pairs of macro-telocentric chromosbmes have telomeric constitutive heterochromatin.

  • A survey on the Eperythrozoon infection of cattle was conducted by means of examinations of Giemsa staining blood films obtained from 2,263 cattle which had been fed in the middle and south parts of Korea. On the other hand, continuous hematological examinations to five splenectomized calves were performed. The results obtained were summarized as follows: 1. Eperythrozoon wenyoni were detected in the blood films of five cattle from Kyungpook and Cheju areas. 2. The infections of Eperythrozoon wenyoni were observed in four of five splenectomized calves from Taegu and Sungwhan areas. 3. From these results it is considered that the infections of Eperythrozoon wenyoni in cattle are quite common in the middle and south parts of Korea.

  • Edwardsiella (E) tarda belongs to the Enterobacteriaceae family and is a motile, gram-negative, rod-shaped, facultative anaerobe regarded as an opportunistic and food-borne pathogen in animals and humans. A 21-year-old male Eurasian brown bear (Ursus arctos arctos) died suddenly without any preliminary signs. Necropsy performed according to standard protocol revealed swollen abdomen with hemorrhagic congestions of the gastroenteric organs, ascites, and hemorrhagic exudates around the mouth. The liver showed discoloration, along with a severely swollen and multiple hemorrhages of the spleen, an elongated gallbladder, and a congested cortex and medullar lesion of kidneys. The stomach contained semi-liquid exudates and undigested chicken exuding a decayed odor. The stomach membranes were dark-gray in color with several cysts in the fundus lesions. Rod-shaped bacteria were found in the major organs by Giemsa staining, identified as E. tarda using a biochemical rapid diagnostic identification kit.

Studies on the Morphology and Stainability of Chicken Spermatozoa (닭 정자(精子)의 형태(形態)와 염색성(染色性) 조사(調査))

  • The present studies were designed to investigate the morphology and stainability of the chicken spermatozoa. Semen samples were collected by abdominal massage from 10 cocks of Arbor, Acres strain (egg breed) and 10 cocks of white Cornish strain (meat breed). The semen samples were diluted with Sarker's solution and were washed. Some of the semen smear slides were stained with seven differential stain methods and was compared with one another by light microscope. In addition to the staining already compared, the length of heads, middle pieces and tails of 400 spermatozoa of two chicken breed was measured with micrometer. The results obtained from these, studies were as follows: 1. Eosin stain appeared to give good results than hematoxylin, pre-treated protease and eosin or hematoxylin stain, pre-treated protease and hematoxylin-eosin stain, carbol-fuchsin, stain and Giemsa 9 technique in differential staining of spermatozoal three portions and pre-treated protease and eosin stain appeared as good staining methods for middle piece of spermatozoa. 2. The average length of chicken spermatozoa was $90.4{\pm}4.0{\mu}m$, and the average length of the head, middle piece and tail of spermatozoa was $13.0{\pm}0.5{\mu}m$, $3.8{\pm}0.2{\mu}m$ and $73.6{\pm}3.8{\mu}m$ lesoectively. 3. The average length of spermatozoa of Arbor Acres strain was $89.2{\pm}5.0{\mu}m$ and the average length of the head, middle piece and tail of spermatozoa was $12.9{\pm}0.5{\mu}m$, $3.8{\pm}0.2{\mu}m$ and $72.5{\pm}4.7{\mu}m$ respectively. The average length of spermatozoa of with Cornish was $91.6{\pm}3.0{\mu}m$ and the average length of the head, middle piece and tail of spermatozoa was $13.1{\pm}0.5{\mu}m$, $3.8{\pm}0.2{\mu}m$ and $74.7{\pm}2.8{\mu}m$ respectively.

  • A case of cutaneous leishmaniasis developed in a 48 year old Korean male who returned from middle east was studied by light and electron microscopic observations. Light microscopically, the lesion consisted of heavy chronic ill-defined granulomatous inflammation involving entire thickness of the dermis, composed of mainly histiocytic and small mononuclear cell infiltrations without evidence of necrosis or giant cell formation. Giemsa staining revealed numerous intracellular micro-organisms within histiocytes, showing dark stained central dot surrounded by light stained cytoplasm. Electron microscopically, the organisms were observed mostly ovoid in shape and frequently binary mitotic features within the host cells. follicle consisted of double unit membranes and microtubules, which are immediately below these membrnae. A long kinetoplast was noted within a very elongated mitochondrion at the center of the organisms and a flagella rose in front of the kineoplast but ended within the cytoplasm. Large numbers of free ribosomes, occasional Golgi complex and SER were also noted, but RER was seldom found. These ultrastructural features corresponded to promastigote stage of Leishmania tropica. In principle, leishmaniasis is a tropical disease and can not be found in temperate zone. However, travel to mideast by many Koreans may contract this disease while they are in endemic regions.

The Induction of Human Corneal Epithelial Apoptosis by Serum-free Medium (무 혈청배지에 의한 각막상피 세포의 세포고사)

  • The corneal epithelium is constantly shed and apoptosis may play an important role in this turn-over. We sought to define that serum-free medium was able to induce apoptosis of corneal epithelial cells. SV-40 transfected human corneal epithelial(HCE) cells were grown to 70% confluency in culture. Serum-free medium was added to cells and the cells incubated for 1, 2, 3, or 6 days. Apoptosis of cells at different times was assessed by staining cells with Giemsa or Hoechst 33342 and measuring DNA fragmentation using the TUNEL assay. HCE cells exposed to serum-free medium demonstrated a high incidence of apoptosis, which increased over time to $50{\pm}4%$ after 3 days. They also stained positively with TUNEL assay. Serum-free medium caused time dependent apoptosis of HCE cells. Thus, serum-like nutrient might be important in corneal epithelial cell homeostasis.

Computerized Image Analysis of Micronucleated Reticulocytes in Mouse Bone Marrow (컴퓨터 이미지 분석법을 이용한 마우스 골수세포에서 소핵의 계수)

  • The present study was performed to validate an automated image analysis system (Loats Automated Micronucleus Scoring System) for the mouse bone marrow micronucleus assay, comparing with conventional microscopic scoring. Two studies were conducted to provide slides for a comparison of micro-nucleated polychromatic erythrocytes (MNPCEs) values collected manually to those collected by the auto-mated system. Test article A was used as an example of a compound negative for the induction of micronuclei and test article B was wed as a micronucleus-inducing agent to elicit a positive response. Cyclophosphamide was included to provide an positive control in two studies. Bone marrow samples were collected 24 h after administration of test article A and B in male ICR mice. The cells were fixed with absolute methanol and stained with May-Grunwald and Giemsa. The number of MNPCEs was determined by the analysis of 1000 total PCEs per bone marrow sample. In addition to micronucleus scoring, an index of bone marrow toxicity based on PCE ratio (% of PCEs to total erythrocytes) was determined for each sample. The automated and manual scoring was similar when the MNPCEs incidence induced by each test article was less than 10. However manual scoring was able to effectively enumerate micronucleated PCEs in mouse bone marrow when MNPCEs incidence was more than 10, such as cyclophosphamide treatment. Conversely, PCE ratio was superior in computer-assisted image analysis. Taken together, it is suggested that improvement of the automated image analysis may be necessary to render the automatic scoring as sensitive as manual scoring for routine counting of micronuclei, especially because it is superior in objectivity and high throughput scoring.
