Effects of Propylene Glycol on Milk Production, Serum Metabolites and Reproductive Performance during the Transition Period of Dairy Cows

  • The objective of this study was to investigate the effects of an oral drench of propylene glycol (PG) on milk production, serum metabolites and reproductive performance during the transition period of animals. Twenty-four 2-3 multiparous Holstein cows (average body weight 565 kg, body condition score about 3.6, at the $9^{th}$ month of gestation) were selected, blocked, and then randomly assigned into a PG and a control group. The control and the PG group cows were orally drenched with water or 50 ml sugarcane molasses mixed with 500 ml PG from 7 days pre-partum to 30 days post-partum, respectively. Experimental results indicated that the oral drench PG had no effect on dry matter intake (DMI). The milk yield of the PG group was significantly higher than that of the control group (p<0.05), whereas milk fat content, milk protein and somatic cell counts (SCC) were not significantly different between groups. Concentration of plasma glucose in the PG group was significantly higher than that of the control group (p<0.05). Conversely, the concentrations of non-esterified fatty acids (NEFA), and blood urea nitrogen (BUN) in the PG group were lower than those of the control group (p<0.05). Concentrations of insulin and ketone bodies were not significantly difference between groups. Body condition score (BCS) in the PG group was significantly higher than that of the control group (p<0.05). In reproductive performance there was no difference between groups. The experimental results indicate that supplementation of PG during the transition period of dairy cows can supply energy rapidly, resulting in reduced catabolism of body tissue and increased milk yield.

Characterization of a Cell Line HFH-T2, Producing Viral Particles, from Primary Human Fetal Hepatocytes Infected with Hepatitis B Virus

  • A primary culture of human fetal hepatocytes was obtained through a therapeutic abortion process at 26 weeks of gestation period. More than $10^8$ cells were seeded on a plastic plate. These hepatocytes were infected with hepatitis B virus (HBV). The HBV was purified from serum of one chronic HBV carrier. Transformed hepatocytes were subcultured in a 10% FBS-supplemented medium. The morphology of the transformed cell was epithelial-like. The cells from the first pass showed signs of early proliferation and had a latent period of more than 3 months after 6-7 passages. After the rest period, the transformed cell proliferated actively and they were subcultured every three days. Transformed hepatocytes were characterized by detection of the HBV transcript by RT-PCR. The secretion of virions from transformed cells was investigated by PCR with the cell medium. Two types of virions secreted into the culture medium were examined by using the transmission electron microscope. Another approach to study the secretion of virions in to culture medium was carried out with HBV antibody. HBsAg was detected in the culture medium of transformed cells using ELISA and Western blot analyses. These data suggested that the human fetal hepatocyte cell line has been established by infection of HBV, in which this cell line secreted viral particles into the culture medium.

Fertility and Reproductive & Developmental Toxicity Study on Recombinant Human Epidermal Growth Factor (rhEGF, DWP401) in Rats (재조합 인간상피세포 성장인자(rhEGF, DWP401)가 랫드의 수태능, 태자와 신생자 발달 및 모체기능에 미치는 영향)

  • This study was conducted to investigate for its effects on reproductive and developmental toxicity of recombinant human epidermal growth factor (rhEGF) in Sprague-Dawley rats. Male rats were administered rhEGF at doses of 1, 10, 100, and 1000$\mu$g/kg/day, respective1y, by subcutaneous injection from 63 days before and throughout to mating period until the day before sacrifice. Female rats were administered rhEGF at the same doses from 14 days before mating to day 20 of gestation or to day 21 of lactation. We examined the male and female fertility indices and maternal toxicity of F0 parental animals. Also, we examined the external, visceral, or skeletal malformation of fetuses, growth and development, behavior, and/or reproductive performance of F1 animals. At the highest dose (1,000 $\mu$g/kg), the mean body weights of F0 animals were significantly increased in males and females at 3 or 2 weeks after treatment, respective1y. No clinical signs and food intakes were observed at any time during the experimental period by rhEGF treatment. In autopsy examination, the relative and absolute liver weights significantly increased in both sexes of 1,000 $\mu$g/kg. At the highest dose (1,000 $\mu$g/kg), there was a statistically significant increase of pregnancy period and the number of dead fetuses. Moreover, significant increase of mean fetal body weight and decrease of number of live fetuses, which related to the difficult dilivery were observed in highest dose group. In Fl examination, no adverse effects on external, visceral, and skeletal malformation, physical and functional development, behavior or reproductive ability of Fl animals were observed in any group. Also, there was no significant difference between control and treated groups in copulation or fertility indices of Fl animals. These results indicate that rhEGF had no adverse effect on fertility and reproductive ability of Sprague-Dawley rats.

Sows fed with synergistic blend of short- and medium chain organic acid has a carryover effect on post-weaning growth rate

  • This study investigated the effect of a synergistic blend of free and buffered organic acid (FMP) on the performance of piglets born to sows supplemented with a blend of short- and medium-chain organic acids (SGG) during the late gestation and lactation period. A total of 150 multiparous sows (n = 50/treatment, Landrace × Yorkshire) were blocked (2.4 parity) and assigned to 1 of 3 dietary treatments: CON - corn-soybean meal-based basal diet, SGG-Low - CON+ 1.5 kg/ton SGG, and SGG-High - CON + 3kg/ton SGG. During weaning, 600 piglets (6.72 ± 0.5kg) which weaned from sows supplemented with 3 levels of SGG were allocated to 2 weaner diets (Control and FMP - 3kg/ton) following 3 × 2 factorial arrangement. Supplemental effects on performance were measured at d0-d21 and d 21-42, and the entire period. Pigs fed with FMP and born to sows supplemented with SGG-High gained more weight and ate more (p < 0.05) compared with those in the CON group in both phases, and with SGG-Low in the second phase. Over the entire post-weaning period, piglets born to sows supplemented with SGG-Low and SGG-High had a higher average daily gain (ADG) and body weight (BW) (p < 0.05). Regardless of sow treatment, pigs fed with an FMP diet had higher ADG (p < 0.001), BW (p = 0.045), and a lower feed conversion ratio (p = 0.033). Also, feeding FMP diets reduced the fecal Escherichia coli and Clostridium perfringens counts at d42. The current study indicates that sows fed SGG supplement had a positive carry-over effect on the post-weaning growth rate, and FMP supplement enhances the growth performance and reduced the number of C. perfringens and E. coli. Thus, the application of 3 kg/ton of SGG in sows' diet and subsequent feeding of piglets with FMP would be an effective strategy to improve growth rate and reduce pathogenic bacteria in post-weaned piglets.

  • First sexual maturity, monthly changes in gonadosomatic index and reproductive cycle of viviparous teleost surfperch, Ditrema temmincki were investigated under photomicroscopy. Samples were collected from May 1992 to August 1993 in the vicinity of Suyoung Bay, Pusan, Korea. Body length at the first sexual maturity in female and male were 12.5cm and 13.5cm, respectively. Gonadosomatic index(GSI) in female reached the maximum in June(just before parturition) and male GSI reached the maximum in October(just before copulation). The testis is composed of a number of seminiferous tubules, and spermatogonia are originated from the germinal epithelium on the seminiferous tubule. The ovary consists of several ovigerous folds, and oogonia are originated from the inner surface of the germinal epithelium within the ovigerous fold. Annual reproductive cycle was classified into the following successive periods : in male, growing period from May to August, maturation period from September to October, copulation period from November to December, and degeneration and resting period from January to April, and in female, growing period from September to October, copulation and maturation period from November to December, gestation period from January to June, and resting period from July to August. Hepatosomatic index(HSI) in male appeared to be negatively correlated with gonadosomatic index, but that in female showed to be positively correlated.

The Comparison of Protein Patterns of Sera in Non-Pregnant and Pregnant Women (非姙娠 및 姙娠한 女子의 血淸蛋白質 패턴의 比較)

  • For the comparative study of protein patterns of the sera of pregnant women, the protein in sera of normal male subjects, non-pregnant women, pregnant women and women delivered of children were analyzed by using the methods of SDS/polyacrylamide gel electrophoresis, two-dimensional electrophoresis and amino acid analysis. The results were as follows: 1. When the protein patterns of sera in normal male ranging from 10, 000 to 110, 000 daltons were compared to non-pregnant women by SDS/polyacrylamide gel electrohporesis, their protein patterns were same each other numerically but bands 3(22, 000 dalton) and 6(39, 000) were less in male than in non-pregnant women quantitatively. When the protein patterns in the pregnant women in which serum were collected two week intervals were compared with non-pregnant women, there was increased or decreased in several bands quantitavely. The protein patterns of sera in pregnant women were compared with those of non-pregnant women; band 3(22, 000) showed similar patterns each other until the 16th week but the quantity of protein was decreased continously from the 18th week to the third trimester of pregnancy. Contary, bands 4(24, 000), 9(69, 000), 10(70, 000), 12(80, 000), 14(86, 000), 15(91, 000) and 16(94, 000) were gradually increased in quantity from the begining of gestation, and band 7(51, 000) was increased until the 32th week of gestation only but somewhat decreased after this time. The quantities of bands 12(80, 000), 15(91, 000) and 16(94, 000) were relatively increased when the protein patterns of delivered women were compared with those of the third trimester of pregnancy. Women who were dilivered female children showed more increase in bands 4(24, 000), 7(51, 000) and 10(70, 000)than one who were delivered male chilren. 2. When the protein patterns of sera in normal males were compared with those of nonpregnant women by two-dimensional electrophoresis, three spots of spot a group were not appeared in the males and the spot c group in the males was less than in the non-pregnant women. In the pregnant women, albumin was significantly decreased during the 10-12 week of gestation but recovered after these times. And spot f(70, 000) was decreased in the 10th week of gestation but increased from this time. 3. Glutamic acid, lysine, aspartic acid, leucine and valine in pregnant women were large in quantity while methionine, isoleucine and glycine were small in quantity by amino acid analysis. The total amino acids were increased remarkably in the second trimester of pregnancy but began to decrease in the third trimester of pregnancy. As mentioned, this present paper deat with that proteins which consist of maternal serum were increased with specific period in pregnancy and that the change of characteristic protein patterns were identified in the serum protein of each trimester in the pregnant women. And furthermore, the study should be preformed for the sex-identification of a fetus in the early pregnancy.

Studies on the Changes of Sex Hormone Levels throughout the Estrous Cycle and Pregnancy in the Gilts (미경산돈(未經産豚)의 발정주기(發情週期) 및 임신기간(妊娠期間)에 따른 성(性)Hormone 수준(水準)의 변화(變化)에 관(關)한 연구(硏究))

  • The present study was carried out to determine the changes of the sex hormone levels in serum throughout the estrous cycle and the gestation period on the Landrace gilts. The blood samples were taken from the vein of six gilts. LH, FSH, prolactin, progesterone, $estradiol-17{\beta}$ and cortisol in serum were analyzed by the radioimmunoassay methods. The results obtained on this study were summarized as follows; 1. The age at puberal estrus was 179.5 days, the weight at puberal estrus was 88.2kg, the length of estrous cycle was 21.3days, the gestation length was 114days and the litter size was 9.5 head in the Landrace gilts. 2. During the estrous cycle, the serum LH and prolactin concentrations were below 1.56mIU/ml and 2.4ng/ml, respectively, under the limit of detection of the assay. The FSH concentrations ranged from 1.50 to 2.20mIU/ml for day 6~15 after the estrus and they were below 1.25mIU/ml from day 3 to day + 3, with day 0 being the first day of the estrus. 3. Progesterone concentrations were 1.90ng/ml at day 0 of the estrus and increased about 13.1ng/ml at day 3 of the estrus, and reached peak levels at day 9. $Estradiol-17{\beta}$ concentrations were below 27.2pg/ml throughout the luteal phase, and reached about 27.2pg/ml at day 0 and day 18. Cortisol concentrations reached peak levels at dey 0 and ranged from 24.65 to 28.57ng/ml throughout the luteal phase. 4. During the gestation period, the concentrations of LH, FSH and prolactin ranged of 3.10~4.37mIU/ml, 1.30~1.80mIU/ml and 2.60~6.70ng/ml, respectively. 5. Progesterone concentrations declined from 38.90~16.85ng/ml throughout the pregnancy to 1.90ng/ml at the time of parturition. $Estradiol-17{\beta}$ concentrations increased from 27.20pg/ml at 15 days after the pregnancy to 620.17pg/ml at the time of parturition. Cortisol concentrations reached peak levels at the time of parturition and ranged from 13.58 to 22.31ng/ml throughout the pregnancy.

Development of a Prediction Model for Postpartum Depression: Based on the Mediation Effect of Antepartum Depression (산후우울 예측모형 개발: 산전우울의 매개효과 검증을 중심으로)

  • Purpose: This study was done to develop a prediction model for postpartum depression by verifying the mediation effect of antepartum depression. A hypothesized model was developed based on literature reviews and predictors of postpartum depression by Beck. Methods: Data were collected from 186 pregnant women who had a gestation period of more than 32 weeks and were patients at a maternity hospital, two obstetrics and gynecology specialized hospitals, or the outpatient clinic of K medical center. Data were analysed with descriptive statistics, correlation and exploratory factor analysis using the SPSS/WIN 18.0 and AMOS 18.0 programs. Results: The final modified model had good fit indices. Parenting stress, antepartum depression and postpartum family support had statistically significant effects on postpartum depression, and defined 74.7% of total explained variance of postpartum depression. Antepartum depression had significant mediation effects on postpartum depression from stress in pregnancy and self-esteem. Conclusion: The results of this study suggest that it is important to develop nursing interventions including strategies to reduce parenting stress and improve postpartum family support in order to prevent postpartum depression. Especially, it is necessary to detect and treat antepartum depression early to prevent postpartum depression as antepartum depression can affect postpartum depression by mediating antepartum factors.

Repeated bovine abortion associated with Neospora caninum in Korea (Neospora caninum에 의한 젖소의 반복유산)

  • A case of repeated bovine abortion caused by Neospora caninum is described. The 3-year-old Holstein cow, which was seropositive to N caninum antibody by indirect immunofluoresent assay, aborted consecutively within a 10-month period. Two fetuses of 6- or 5-month of gestation were aborted in June 1996 and March 1997, respectively. Histologically, multifocal necrotizing encephalitis, nonsuppurative periportal hepatitis, myocarditis, and myositis were observed in both fetuses. Tachyzoites or tissue cysts detected in the brain, heart and liver were positive to N caninum but negative to Toxoplasma gondii by immunohistochemical method. The results of our study demonstrate that repeated abortion due to N caninum actually occurs in Korea. This is believed to the first report of repeated abortion associated with N caninum in Korea.

Effects of a Yoga-focused Prenatal Program on Stress, Anxiety, Self Confidence and Labor Pain in Pregnant Women with In Vitro Fertilization Treatment (요가중심 산전프로그램이 체외수정 시술 임산부의 스트레스, 불안, 분만 자신감 및 분만통증에 미치는 효과)

  • Purpose: The purpose of this study was to identify the effects of a Yoga-focused prenatal program on the stress, anxiety, self confidence and labor pain of pregnant women who had in vitro fertilization (IVF) treatment. Methods: A quasi experimental study with a non-equivalent control group pretest-posttest design was used. The data collection period and meditation program were between January 9 and August 31, 2009. Forty-six women who were pregnant following IVF, and were between 12-20 weeks gestation, participated in the study (23 experimental group, 23 control group). Data were analyzed using Chisquare test, Mann-Whitney U Test, ANCOVA, and Cronbach's alpha coefficients with the SPSS 12.0 for Windows Program. Results: Although the sample size was limited, women who participated in the program showed statistically significant improvements in stress, anxiety, labor pain, and labor confidence for women pregnant after IVF. Conclusion: The result indicate that this 12-week Yoga-focused educational program can be utilized for women pregnant following IVF to reduce their stress, anxiety, and labor pain, and to increase delivery confidence. It is suggested that the Yoga-focused educational program be offered to every pregnant woman.
