Comparison of Germination Characteristics and Daily Seed Germinating Pattern in New Cultivars of The Third Generation of Creeping Bentgrass Grown under Alternative and Natural Room Temperature Conditions (자연실온 및 변온조건에서 제3세대 크리핑 벤트그래스 신품종의 발아특성 및 일일 발아패턴 비교)

  • This study was conducted to investigate early germination characteristics and germination pattern of new cultivars of creeping bentgrass (CB, Agrostis pall/stris Huds.) under natural room temperature conditions and to analyze differences between alternative and natural conditions. Seven cultivars of CB were tested in the study. Daily and cumulative germination patterns were measured and analyzed on a daily basis. Significant differences were observed in germination pattern, days to the first germination, days to 50% germination, days to 85% germination, and germination rate among cultivars under different environments. The first germination in all entries was initiated between 4 and 5 days after seeding (DAS) under alternative conditions, while between 6 and 8 DAS under natural conditions, being 2 to 3 days later as compared with that of alternative conditions. As for the first germination percentage, it was generally 5 to 25% under alternative conditions. However, 'Penn A-1', 'Penncross', and 'Penn A-4' cultivars showed a vigorous germination, producing 50 to 70%. Under natural room temperature conditions, it was 5 to 10% for the most entries, except 'Penncross', 'L-93', and 'Penn A-4' cultivars which vigorously produced 20 to 50%. Days to the 50% germination in CB resulted in 4 to 10 DAS under alternative conditions, while 6 to 15 DAS under natural conditions, being 2 to 5 days later than under alternative conditions. Days to the 85% germination were 5.90 to 11.75 DAS under alternative conditions and 7.95 to 12.50 DAS under natural conditions. 'Penn A-1' and 'L-93' were the fastest cultivars under alternative and natural conditions, respectively. Regardless of the temperature conditions, the longest cultivar in germination was 'T-1', being over 11 DAS. In-depth comparative data on germination characteristics and germination patterns among CB cultivars would be usefully applied for a practical CB turf establishment in golf course construction etc.

Germination and Viability of Green Manure Crop Seeds Produced from Domestic and Foreign Countries (국내 채종 및 수입 녹비작물 종자의 발아 및 활력 특성 구명)

  • Seed germination and dormancy of three green manure seeds such as hairy vetch, crimson clover and narrow-leaved vetch (Vicia angustifolia L.) produced in Milyang, southern part of Korea were investigated to provide a basic information on the stability of seedling establishment in green manure seeds. In addition, the effect of seed storage duration on the germinability of imported hairy vetch and crimson seeds was also investigated. To determine the seed maturity time of three green manure seeds, the seeds were harvested manually at May 25, May 30, and June 4, and the 100-seed weight, germinability and percentage of hard seed were evaluated. Freshly harvested seeds of three green manure crops were strongly dormant because of hard seed coat. Germination percentage of the three green manure crops varied depending on the seed harvest time ranging from 30 - 52% in hairy vetch, 16 - 35% in crimson clover and 2 - 61% in V. angustifolia, respectively. Of the three crops, crimson clover and V. angustifolia matured early on May 30 to June 4 (around 35 to 40 days after flower) but hairy vetch seed did not mature until June 4 based on the seed germinability. The matured seed of crimson clover and V. angustifolia germinated less than 20% while scarification on seed coat significantly overcome the innate dormancy. On the other hand, the seed germination of hairy vetch and crimson clover was lower under dark than in the presence of light. The germination percentage of the imported hairy vetch and crimson clover seeds showed high with 77 - 79%, 94 - 95%, respectively, but the seed germination significantly declined by 21 - 32%, 30 - 40% after one-year of storage under natural conditions. The seeds germinated only 8 - 13% for hairy vetch but crimson clover did not germinate at all after two years storage. These results indicate V. angustifolia and crimson clover mature at May 30 to June 4 but hairy vetch did not. The imported hairy vetch and crimson clover seeds should be used within one-year after import for stable seedling establishment in green manure-rice cultivation cropping system.

Seed Dormancy Type and Germination Characteristics in Tiarella polyphylla D. Don Native to Korea (한반도 자생식물 헐떡이풀 종자의 휴면유형과 발아특성)

  • Tiarella polyphylla D. Don is a native plant distributed only in Ulleung Island in Korea and has been traditionally used for medicinal purposes, although it is also used ornamentally. This study was conducted to determine the requirements for dormancy break and germination and to classify the type of seed dormancy. The experiments were performed with cold stratification (0 or 12 weeks at $5^{\circ}C$), warm stratification (0, 4, 8, or 12 weeks at $23^{\circ}C$, followed by 8 weeks at $5^{\circ}C$, and then incubation at $23^{\circ}C$), and $GA_3$ treatments (0, 10, 100, or 1000 mg/L). The treated seeds were incubated on aseptic media at room chamber ($23^{\circ}C$, a 16h photoperiod of fluorescent lamps with $40{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$). The seeds were dispersed in nature as underdeveloped embryos with no physical barrier to absorb water to prevent water absorption. However, the seeds did not germinate for 30 days after sowing without any pre-treatments. Thus, the seeds had morphological dormancy (MD) and physiological dormancy (PD). The final germination percentage following cold stratification (0 or 12 weeks) was 66.7% and 45.6%, respectively. The cold stratification delayed seed germination by about 3 weeks. In the warm stratification experiment (0, 4, 8, or 12 weeks), the final germination percentage was 21.1%, 27.8%, 41.1%, and 57.8%, respectively, 20 weeks after sowing. The embryos of the T. polyphylla seed grew in relatively warm temperatures ($23^{\circ}C$). $GA_3$ application overcame seed dormancy and promoted germination. Following $GA_3$ treatment (0, 10, 100, or 1000 mg/L), the final germination percentage was 33.3%, 45.0%, 42.5%, and 72.5%, respectively. These results suggest that the T. polyphylla seeds had non-deep simple morphophysiological dormancy (MPD) and $GA_3$ treatment could be used as a substitute for warm stratification for breaking seed dormancy. To our knowledge, this is the first report of seed dormancy characteristics of the genus Tiarella native to Korea.

Effects of Planting Date and Accelerated Aging on Seed Germination-related Traits of Lipoxygenase-Lacking Soybean (Lipoxygenase가 결여된 콩의 파종기 및 노화처리에 따른 종실특성과 발아력)

  • Lower field germination was observed in lipoxygenase- lacking soybean genotypes. This study was performed to understand seed-related traits after accelerated aging in response to planting date of soybeans. Two soybean cultivars, Jinpumkong (null lipoxygenase-2,3) and Jinpumkong 2 (lacking lipoxygenase-1, 2, 3) were evaluated for germination, and were compared with the cultivar, Taekwangkong containing lipoxygenase-1,2,3 isozymes. Greater seed coat cracking was shown in Jinpumkong and Jinpumkong 2 than Taekwangkong. Regardless of soybean genotypes, earlier planting resulted in greater seed coat cracking. After accelerated aging, seed fracturability and hardness of Jinpumkong and Jinpumkong 2 were lower than those of Taekwangkong. There was significant difference in germination percentage among soybean genotypes in response to planting date. Seeds obtained from late planting showed better germination ability. Accelerated aging resulted in lower germination percentages of Jinpumkong and Jinpumkong 2 than that of Taekwangkong, and showed higher electric conductivity in Jinpumkong and Jinpumkong 2 than Taekwangkong. After accelerated aging at 4$0^{\circ}C$ for thirty five days, germination percentages of Jinpumkong and Jinpumkong 2 were lower than that of Taekwangkong. Electric conductivity was increased continually as accelerated aging time became longer, and Jinpumkong and Jinpumkong 2 showed higher electric conductivity than Taekwangkong. Even though there were significant genotype differences in seed traits, further studies are needed to determine whether seed lipoxygenase ability is associated with germination ability.

Effect of Storage, Temperature and Pre-treatment on Germination of Melia azedarach L. Seed (저장방법, 온도 및 전처리가 멀구슬나무 종자의 발아에 미치는 영향)

  • This study was performed to investigate the effects of storage, temperature, and pre-treatments on the germination of Melia azedarach seeds collected from Buan, Jeonju, and Jeju provenance. M. azedarach seeds stored with or without pericarp in the ground, which collected from Buan provenance evidenced the highest germination percentage (PG, %) and the shortest time to first germination (days). The seeds collected from Jeonju and Jeju provenance were placed at both six continuous temperatures (15, 20, 25, 30, 35, and $40^{\circ}C$) and two alternating temperatures ($20{\leftrightarrow}30^{\circ}C$ and $25{\leftrightarrow}35^{\circ}C$) for seed incubation. The results showed a significant effect for temperature of seed incubation. The seeds incubated at $35^{\circ}C$ had the highest PG among the continuous temperatures and germinated significantly more at the two alternating temperatures than at $35^{\circ}C$. Concerning mean germination time (MGT), the seeds incubated at $35^{\circ}C$ evidenced the shortest germinations among the continuous temperatures while those at the alternating temperatures germinated for a shorter period than those at $35^{\circ}C$. The germination rate (GR) and germination performance index (GPI) were similar to PG. The seeds collected from Jeonju provenance were treated using five pre-sowing treatments (scarification, scarifcation+$GA_3$, scarification+$KNO_3$, $GA_3$, and $KNO_3$) prior to the germination experiments. Compared with the intact seeds (control), most of the pre-treatments were significantly (especially scarification+$GA_3$ 100 ppm and scarification+$KNO_3$ 1.0%) higher in PG, GR, and GPI, as well as shorter in MGT.

Diaspore, seed dispersion and seed germination characteristics of two myrmecochrous spring ephemerals -Jeffersonia dubia and Corydalis remota- (개미가 종자를 산포하는 춘계단명식물 깽깽이풀과 현호색의 전파체, 종자산포 및 발아 특성)

  • This study examined the germination characteristics by collecting fruits and pretreatment of the diaspore from April 2014 to June 2015 to investigate the diaspore characteristics, seed dispersion, and seed germination characteristics of two myrmecochorous spring ephemerals (MCSE): -Jeffersonia dubia (Jb) and Corydalis remota (Cr)-. The diaspore of Jb was about 5mm-long yellow-dark brown, oblong seeds with the attached amorphous white elaiosome. The mean weights of diaspore, seed, and elaiosome were 15.86mg, 13.46mg, and 2.40mg, respectively, and the elaiosome ratio was 15.13%. The diaspore of Cr was about 1.2mm diameter and glossy black ovoid seeds with the attached white spatula-shaped elaiosome. The mean weights of diaspore, seed, and elaisome were 2.58mg, 2.05mg, and 0.53mg, respectively, and the elaiosome ratio was 20.54%. Camponotus niponensis and Formica japonica transported the diaspore of Jb while Formica japonica and Lasius japonicus transported the diaspore of Cr. The germination percentage of Jb seeds was statistically significant and had the significance level of 1% with the pretreatment and date of sowing. However, it was independent of attachment of elaiosome. The mean germination percentages of Jb seeds was 65.0% during sowing on June 20, 17.5% during sowing on August 19, and 0% during sowing on October 20. The germination percentage of Cr was statistically significant and had the significance level of 5% and 1% with the attachment of elaisome and date of sowing, respectively. The mean germination rates were 54.17% and 35.0% in the non-treatment section and the treatment section with elaisome detached, respectively. The mean germination percentages of Cr seeds was 75.0% during sowing on June 20, 53.75% during sowing on August 19, and 5.0% during sowing on October 20. Considering the fact that the ants transported the diaspores to the ant house when the fruits of MCSE were ripened and dropped the seeds, the direct seeding right after collecting may be most suitable to the characteristics of the evolution of these plant species and may be the best method to obtain the highest germination percentages. Since the ants distribute their seeds, the MCSE produces and attaches the elaiosome to the seed to maintain the symbiotic relationship with ants. The ants then transport the seed to the ant house where the environment is controlled for suitable temperature and humidity, and then the MCSE succeeds in germination after the embryo grows sufficiently in the next spring.

Effect of Seed Priming of Carrot, Lettuce, Onion, and Welsh Onion Seeds as Affected by Germination Temperature (발아온도에 따른 당근, 상추, 양파 및 파 종자의 Priming 효과)

  • To compare germinability of primed seeds with that of water imbibed and nonprimed seeds at different temperatures, percent germination, number of days to attain 50% of the final germination percentage (T50) and mean number of days to germination (MDG) were evaluated at various temperatures ranged from $10^{\circ}C$ to $35^{\circ}C$. Primed carrot seeds in -0.50 MPa PEG 8000 showed higher percent germination at $10^{\circ}C$ or $35^{\circ}C$. Reductions in T50 and MDG values were observed at all germination temperatures when primed with the PEG. The PEG combined with 100 mM $K_3PO_4$ was not as effective as the PEG alone in improving the germinability of carrot seeds. Nonprimed lettuce seeds germinated only 2% at $30^{\circ}C$ or $35^{\circ}C$. However, priming with 50 mM $K_3PO_4$ increased germination to 40% at $30^{\circ}C$ and to 21% at $35^{\circ}C$. In contrast to the single treatment of 50mM $K_3PO_4$, priming lettuce seeds in a mixture of 50mM $K_3PO_4$ and 200 mM $KH_2PO_4$ resulted in a lower percentage of germination, but a significant greater reduction of T50 and MDG. Percent germination was increased when 200 mM $KH_2PO_4$-primed onion seeds and 100 mM $Ca(NO_3)_2$-primed Welsh onion seeds were germinated at $10^{\circ}C$ or $35^{\circ}C$. Combined treatments with $Ca(NO_3)_2$ for onion and with $KH_2PO_4$ for Welsh onion seeds did not improve percent germination, but they significantly reduced T50 and MDG.

Comparison of Germination Characteristics and Daily Seed Germinating Pattern in Fine-textured Fescues (세엽형 훼스큐속 잔디의 발아특성 및 일일 발아패턴 비교)

  • Research was initiated to investigate early establishment characteristics and germination pattern of fine-textured fescues (FF). Six varieties from Chewings fescue ($Festuca$ $rubra$ L. ssp. $commutata$ Gaud., CF), creeping red fescue ($F.$ $rubra$ L. ssp. $rubra$ Gaud., CRF), hard fescue ($F.$ $ovina$ ssp. $longifolia$ Thuill., HF) and sheep fescue ($F.$ $ovina$ L., SF) were evaluated in the study. An alternative environmental condition requiring a FF germination test by International Seed Testing Association (ISTA) was applied in the experiment, consisting of 8-hr light at $25^{\circ}C$ and 16-hr dark at $15^{\circ}C$ (ISTA conditions). Daily and cumulative germination patterns were measured and analyzed on a daily basis. Significant differences were observed in germination pattern, days to the first germination, days to 50% germination, days to 60% germination, and germination rate. The final germination percentage was variable with species and varieties, being 40.25 to 82.00% at the end of study. There were considerable variations in early germination characteristics and germination pattern among FF species. The first germination in all entries except HF was initiated between 5 and 6 DAS (days after seeding) under ISTA conditions, while HF between 6 and 7 DAS, being 1 day later. It was 8 to 10 DAS in days to the 50% germination, which was 2 to 4 days after the first germination date. Days to the 60% germination were 9.10 to 14.80 DAS under ISTA conditions, being 5.70 days in differences among the entries. CF 'Jamestown II' and 'Shadow II' and HF 'Aurora Gold' were the fast varieties. The slowest one was HF 'Rescue 911'. Among FF species, turf establishment speed was becoming faster in CRF, SF, HF and CF in this order. Information on differences in germination characteristics and pattern from this study would be usefully applied for golf course design and construction, when established with FF.

Effects of Application of Rhodopseudomonas sp. on Seed Germination and Growth of Tomato Under Axenic Conditions

  • Purple nonsulfur bacteria were isolated from river sediments and their growth promoting capabilities on tomato were examined. Isolated strains KL9 and BL6 were identified as Rhodopseudomonas spp. by 16S rDNA sequence analysis. Rhodopseudomonas strain KL9 maximally produced 5.56 mM/min/mg protein and $67.2\;{\mu}M/min/mg$ protein of indole-3-acetic acid (IAA) and 5-aminolevulinic acid (ALA), respectively, which may be one of the mechanisms of plant growth enhancement. The germination percentage of tomato seed, total length, and dry mass of germinated tomato seedling increased by 30.2%, 71.1%, and 270.8%, respectively, compared with those of the uninoculated control 7 days after inoculation of strain KL9. The lengths of the root and shoot of germinated seedling treated with 3 mM tryptophan, a precursor of IAA, increased by 104.4% and 156.5%, respectively, 7 days after inoculation of strain KL9. Rhodopseudomonas KL9 increased 123.5% and 54% of the root and shoot lengths of germinated seedling, respectively, treated with 15 mM glycine and succinate, precursors of ALA. This plant growth promoting capability of purple nonsulfur bacteria may be a candidate for a biofertilizer in agriculture.

Study on Waxy Maize Hybrid -III. Distribution of the Major Characteristics for the Collected Genetic Resources in Maize (교잡종 찰옥수수에 대한 연구 -III. 국내 옥수수 유전자원 수집종에 대한 주요 특성 분포)

  • Germination ratio on the 300 collected genetic resources showed 80 to 100 percentage, but those of some accessions were very low as below 30 percentage. Early growth phase of most of these lines were very poor while some collected lines were very vigor. Range of stem height showed from 150 to 199cm, but that of some accessions showed below 100cm as five percentage and some of them was over 250cm as eight percentage. Number of tillers per plant appeared less one but some collected lines have two to three tillers per plant. In days to tasseling, some accessions showed more four per plant. Ear length of these accession showed above 60 percentage as 11 to 15cm, while that of some collected lines showed over 10 percentage as 16 to 20cm.
