Allelopathic Potential Evaluation of Rice Varieties on Echinochloa crus-galli (돌피(Echinochloa crus-galli)에 대한 벼품종의 알레로파시 잠재성 평가)

  • Rice (Oryza sativa L.) contains water-soluble substances that inhibit the germination of Echinochloa crus-galli. This study investigated the allelopathic potential of rice in which rice had been grown with E. crus-galli in the field. Extracts of forty-seven rice varieties were screened for allelopathic potential in the laboratory. Double distilled water was used as a control. Based on the germination percentage, the varieties may be classified in the following order of decreasing inhibition : Namweon(36%) and Gyehwa(38%) hulls extracts and Sambaeg(43%) and Seoan(46%) leaves extracts. In the comparison of allelopathic activity test between leaves and hulls extracts, hulls extracts contain more allelopathic toxic substances. In the effect of concentration treatment on the six levels of extract solutions(3, 6, 12, 25, 50, 100%, v/v) germination percentage was significantly inhibited as the extract concentration increased. The highest concentration(100%) caused the greatest reduction in E. crus-galli. Allelopathic potential effects were significantly different among the various varieties and between '94 and '95 year extracts. The results indicate that there are genetic differences among varieties for allelopathic potentiality on E. crus-galli. Allelopathic potential in the ecological aspects can be decided through variety selection.

The Antidermatophytic Potential of the Marine Isolate of Aspergillus sp. Collected from South Coast of Korea

  • This study was carried out to assess the antidermatophytic potential of the ethyl acetate(EtOAc) extract of the marine isolate of Aspergillus sp.. The fungus was isolated by serial dilution, and was identified Aspergillus sp.. The EtOAc extract of the fungus was examined to evaluate the antidermatophytic efficacy against the fungal pathogens infecting human skin using the disc diffusion and MIC(minimum inhibitory concentration) determination methods. The EtOAc extract($5{\mu}l\;disc^{-1}$) was considered to have the antidermatophytic activity based on the inhibition percentage of the mycelial growth of the fungi tested such as Trichophyton mentagrophytes KCTC 6085, Microsporum canis KCTC 6591, Microsporum canis KCTC 6348, Trichophyton rubrum KCTC 6352, Microsporum canis KCTC 6349 and Trichophyton mentagrophytes KCTC 6316. The percentage of the inhibition ranged from 54% to 81, and the MIC obtained was 62.5, 62.5, 250, 125, 125, and $125{\mu}g\;ml^{-1}$, respectively. The extract had a strong detrimental effect on the spore germination of the tested skin infectious pathogens. These findings strongly support the role of the ethyl acetate extract as a potential antidermatophytic agent.

  • Populus. caspica L. is an Iranian indigenous poplar species which naturally distributed in the northern part of country. Unfortunately, overuse has removed many of the stems of better form, so that natural stands now usually appear small and crook. Therefore genetic variation for selection of new superior clone of this species is needed. Conventional hybridization system is currently used to induce genetic variation in poplar species but incompatibility barriers have been observed between them. In vitro ovule embryo culture was used to overcome incompatibility obstacle for interspecific hybridization between Populus caspica L. with Populus deltoids L.62/75. Female flowers of Populus caspica L. have artificially been pollinated with pollen grain of P. deltoides 62/75 in one direction using twig and pot crossing system. Ovaries at different ages (7, 14 and 21 days after pollination) were disinfected through 70% ethanol for 1 minute, 5% of sodium-hypochlorite solution for fifteen min followed by three time rising with sterile distil-water. Isolated ovaries were then transferred to MS hormone free medium containing 30 and 60 g/L sucrose for embryo development and germination. Collected data have been analyzed by two factorial experimental designs. The results indicated that there were significant differences between age of embryos for development and germination at ${\alpha}=0.01%$. Highest embryo germination (45%) was observed from 21 days old ovaries. No significant differences were observed between MS culture media containing 30 and 60 g/L for percentages of ovary-embryo germination and number of germinated embryo per ovary at ${\alpha}=0.05%$. Fourteen percentage of embryo germination obtained in MS medium supplemented with 60 g/L sucrose, while only 35% of isolated ovaries were able to germinate in MS containing 30 g/L sucrose. Induced plantlets in 4 cm height were transferred into pots containing soilless (1:1:1 peat, per lit and vermiculite) medium for acclimatization. After successful acclimatization, plants were delivered to nursery.

Water Absorption and Germination Ratio of Sprout-soybean Varieties Affected by Different Planting Date (나물용 콩의 재배시기에 따른 수분흡수율과 발아율)

  • The water absorption ratio was not affected by the planting dates. Hannamkong showed higher water absorption ratio than any sprout -soybean varieties. Soybean seeds produced from later planting showed higher germination percentage than those planted May 25. Pungsannamulkong, Eunhakong, Doremikong, and Someongkong showed over $90\%$ germination after two days, while Tawonkong and Hannamkong were lower, The germination ratio test of the soybean seeds at $25^{\circ}C$ is usually for 7 days. But, germination ratio test of seeds used for sprout-soybean is suitable for 3 days at $25^{\circ}C$.

Seedcoat removal and seed germination in Helianthus tuberosus L. (종피제거와 돼지감자종자의 발아)

  • Pericarp and seedcoat removal treatments were tested to determine their effectiveness in the hard-to-germinate botanical seeds of jerusalem artichoke. Fresh seed of five Helianthus tuberosus L.varieties were (A)untreated (B)water soaked overnight (C) soaked overnight followed by renoval of pericarp or (D) soaked overnight followed by renoval of pericarp and seedcoat. The results indicate that treatments which removed the pericarp and seedcoat were the very effective, giving germination of over 90%. A considerable increase in germination did not follow only the peri-carp removal treatments. The factors inhibitory to germination of fresh jerusalem artichoke seed is associated withthe seedcoat. The removal treatment of pericarp and seed-cost is recommended despite its complexity because it givesa high germination percentage and varies least from varietyto variety. Heliarthus tuberosus L., a biomass potential crop, is a member of the family compositae. The genus Helianthushas provided man with two food plants, the sunflower (H.annus) and the jerusalem artichoke or topinambour (H. tube-rosus) . (3) The jerusalem artichoke grown for its tubers, has always been an extremely minor crop, but it is stillgrown in many places as a food for man or livestock and forthe production of alcohoL. Though tubers are used for pro-pagation jerusalem artichoke also flower and produce seedin head-like in florescences. Flowers are developed acro-petaLly on flattened receptacles such that outermost flowers are oldest. Each of these epigynous flowers may develop an achene-type fruit in which outer layers of the overy wallpersist, while inner layers become disorganized. Insidethe ovary wall of mature fruit, there is a papery seedcoat, probably composed of compact cells from endosperm, integu-ments, and nucellus.In general, the efforts to improving this crop havebeen hampered by the hard-to-germinate botanical seed.Seeds did not germinate for at least IL months after harvest.Fresh seeds of some varieties require one year more to gar-minate. (5) Since the time factor between generations isof concern in a prospective breeding program of jerusalemartichoke , these observations led to investigation of thenature of delayed seed germination in jerusalem artichokeas a biomass potential crop.

Seed Germination and Dormancy Breaking of Thalictrum rochebrunianum var. grandisepalum (H. Lev.) Nakai

  • This study was carried out to develop an effective seed propagation method for Thalictrum rochebrunianum var. grandisepalum (H. Lev.) Nakai by analyzing seed dormancy types and germination characteristics. Seeds were collected between September to October at Gangwon province, and well-selected seeds were used while being dry-stored at 4±1℃. The seed size ranged 4.52 × 1.58 ㎜ and the weight of thousand seeds were 1,603.5 ± 0.02 ㎎. The moisture content was 7.2%. Seeds were achene type, and morphology characters showed an elliptical shape and rough texture, and light brown in color. Moist-chilling treatment was conducted for dormancy breaking because the seeds had an undeveloped embryo of liner type. The embryo had developed during a moist-chilling period, constantly, and fully developed in 10 weeks. Consequently, it seemed to be non-deep complex or intermediate complex type of morphophysiological dormancy, and embryo dormancy was broken by wet-chilling for 10 weeks. After 10 weeks of wet-chilling treatment, seed germination began. Germination percentage was higher in dark condition raher than light condition and recorded the maximum at 25℃ in the dark (16.3%). A pre-soaking treatment with a combined plant growth hormones promoted germination and shortened T50. Specifically, seed germination of 84.5% was achieved by pre-soaking of seeds with a combined solution of 500 ㎎/L GA3 and 10 ㎎/L kinetin for 24 h after a wet-chilling treatment for 10 weeks. Thus the effect of plant growth hormones coupled with chilling temperature on seed breaking dormancy provide asubsequent growth of seedlings for successful plantation.

  • For the development of ecological sound proof wall, successful seed germination and growth of plants are very important. In our present study, we observed germination of thirty five different seed species in growth chamber at $24^{\circ}C$, humidity 60% and alternation of 12 hours photoperiods. Germination of same seeds were carried out at room temperature on 6:4, 7:3, 8:2, and 9:1 proportion of pure and mineral soils respectively. Out of studied species, maximum germination was recorded for Perennial penant (92%), followed by Brassica napus (86%), and Silene armeria (82%). The measurement of root/shoot length showed, maximum root/shoot length for Perennial penant, followed by perennial sonata and Rebolsentri sp. Based on germination percentage and root/shoot length of germinating seeds, we considered Perennial penant, perennial sonata and Rebolsentri sp. were proper species for the construction of ecological sound proof walls

Effect of Seed Size and Mucilaginous Substance on Seed Germination of Peony(Paeonia lactiflora Pall.) (작약(芍藥) 종자(種子)의 형태적(形態的) 특성(特性)과 점액물(粘液物)이 발아(發芽)에 미치는 영향(影響))

  • The experiments were conducted to determine the influence of seed size and mucilaginous substance on seed germination of herbaceous peony collected from farmers' fields in Korea. No significant difference was recognized between the size of seed and the rale of germination. The maximum percentage(68%) of seed germination was obtained from the seeds with thin brown colour treated at $20^{\circ}C$. The amount of mucilaginous substances exuded from peony seeds was the highest in the seeds with black brown colour. Germination rate of peony seeds was negatively correlated with the amount of mucilaginous substance exuded from the seeds. High frequency(93%) of seed germination was obtained from the seeds which treated in the mixture(1:1) of sands and activated charcoal.

Effects of Some Environmental Factors on the Germination of Seeds in Ambrosia artemisiifolia var. elatior (몇가지 환경요인이 돼지풀의 종자발아에 미치는 영향)

  • In order to analyze the life cycle of A. artemisiifolia var. elation dormancy and some environmental factors inducing germination of the seeds were examined. The results were as follows : Dormancy of fertile seeds was broken in part within a month after seed collection in case of adequate moisture and alternating temperature was also effective in breaking dormancy. The temperature range, which allow germination was 12℃ ∼ 32℃. Optimum temperature for germination was 24℃. The seed of A. artemisiifolia var. elatior was light-independent. The difference of storage period appeared to have no particular effect on the viability of seeds at any time during the 9-month storage period. In the increasing temperature(IT) regime, A artemisiifolia var. elatior seeds started to germinate at 16℃, showing the higher temperature the greater germination rate, the final germination percentage was 99.34%. On the other hand, in the decresing temperature(DT) regime, seeds began to germinate at 20℃ with the 1.34% germination. An induced dormancy occurred at 12℃ making the 5.34% fecal germination in the DT regime. Low temperature was more effective to break dormancy than higher temperature Seeds of A. artemisiifolia var. elatior seems to be germinated in mid to late autumn or germination delayed until following spring. The above results suggest these variation of germination response in diverse environmental factors seems to be a physiological strategy to maintain their existence and to reproduce in the extreme thermal variation.

Effect of Osmotic Priming and Solid Matrix Priming to Improved Seed Vigor and Early Growth of Pepper and Tomato Seeds (고추와 토마토 종자의 발아력 증진과 초기생육에 미치는 Osmotic Priming 및 Solid Matrix Priming 처리 효과)

  • Osmotic and solid matrix priming treatments enhanced germination performance. We compared osmotic with solid matrix priming to determine the more effective treatment for improving seed germination in pepper and tomato. Seed hydration was immediately observed after osmotic priming and solid matrix priming treatment. The moisture content of solid matrix primed seeds was lower than that of osmotic primed seeds in the two vegetable crops. Osmotic priming and solid matrix priming did not increased percent germination, but showed shorter number of days to 50% of the final germination percentage ($T_{50}$) compared with untreated seeds, regardless of germination temperature. $T_{50}$ value was reduced in osmotic or solid matrix primed pepper seeds about 6.0, 5.0, 4.6 and 4.0 days compared with untreated seeds at 15, 20, 25, and $30^{\circ}C$, respectively. While, that in tomato seeds was reduced about 3.3, 5.0, 4.6 and 4.0 days compared with untreated seeds at 15, 20, 25, and $30^{\circ}C$, respectively. The effectiveness of osmotic priming or solid matrix priming in reducing the $T_{50}$ was greater when the seeds were germinated at $15^{\circ}C$ than at temperature of higher than $20^{\circ}C$. Solid matrix primed seeds germinated faster than osmotic primed seeds at all temperature in pepper. However, there was no significant difference on the percentage germination between solid matrix and osmotic primed seeds in tomato. After priming, dried-bark seeds showed faster germination than surface-dried seeds in pepper. On the other hand, there was no significant difference in tomato. Emergence of pepper and tomato seeds was markedly enhanced by osmotic priming or SMP treatment although the final emergence percentage was not significantly influenced. On the other hand, early growth was not significantly influenced by osmotic priming or SMP treatment of pepper and tomato seeds.
