Studies on the Morphological and Anatomical Characteristics of Genus Abies in Korea (한국산(韓國産) Abies 속(屬)의 내외형태학적(內外形態學的) 특성(特性)에 관한 연구(研究))

  • Some morphological and anatomical characteristics of four Abies species in Korea were investigated to find the characteristics and the taxonomical relationship between the species. The results were summarized as follows: There was no significant difference in needle thickness and number of hypodermic cell layer among 18 morphological and anatomical characteristics of cone and needle between the species. A. koreana and A. nephrolepis were similar in above fourteen characters and A. holophylla and A. firma were also similar in the 14 characters. A. koreana and A. firma showed similarity in above two characters. Main resin canals of needles in A. koreana and A. nephrolepis were situated in leaf margin (L-LM), those of A. holophylla, in L, L-M and M, and that of A. firma fully in other locations except L.

Comparative Morphology of Pollen Grains from Some Korean Gymnosperms (한국산 나자식물 화분의 비교형태)

  • Pollen grains of 19 taxa belonging to 12 genera in 5.families of the Korean gymnosperms were investigated by LM, SEM and TEM, and their morphological characters were described and palynological keys were made for each family. From the taxonomic viewpoint, palynological characters are very available to intergeneric and intrageneric classifications since their distinctive differences in pollen shape, size, condition of surface, sculpture pattern and presence of air bladders and projection except for the intrageneric classification of the genus Pinus since the overlapped characters such as pollen size and sculpture pattern. Pollen grains of Picea koraiensis in the family Pinaceae are distinguished from thc genera Pinus and Abies for the reasons that shape of air bladders are irregular, air bladders are bigger than pollen body, and pollen wall is thin. Compared with the genera Abies and Pinus that have air bladders, Picea koraiensis is closely related to Abies in that pollen body wall become gradually thin from pole of proximal face to air bladder, surface of pollen body is not rough, and pollen size is similar to that of the genus Abies. The presence of ektexine and enexine in proximal face, and the connections of the former to air bladder wall and the latter to distal face were elucidated by the investigation of pollen grains with air bladders through TEM. Endexine of the families of Taxaceae, Taxodiaceae and Cupressaceae was also obse-rved as a pile of laminae. From the observation by SEM, sculpture pattern of air bladder was foveolate with fine pits on surface. Sculpture pattern of air bladder is reticulate under LM, but observed by TEM, it resulted from the transmission of netted structure in air bladder.

Diversity and community structure of ectomycorrhizal mycorrhizal fungi in roots and rhizosphere soil of Abies koreana and Taxus cuspidata in Mt. Halla

  • In this study, the roots and rhizosphere soil of Abies koreana and Taxus cuspidata were collected from sites at two different altitudes on Mt. Halla. Ectomycorrhizal fungi (EMF) were identified by Illumina MiSeq sequencing. The proportion of EMF from the roots was 89% in A. koreana and 69% in T. cuspidata. Among EMF in rhizosphere soils, the genus Russula was the most abundant in roots of A. koreana (p < 0.05). The altitude did not affect the biodiversity of EMF communities but influenced fungal community composition. However, the host plants had the most significant effect on EMF communities. The result of the EMF community analysis showed that even if the EMF were isolated from the same altitudes, the EMF communities differed according to the host plant. The community similarity index of EMF in the roots of A. koreana was higher than that of T. cuspidata (p < 0.05). The results show that both altitude and host plants influenced the structure of EMF communities. Conifers inhabiting harsh sub-alpine environments rely strongly on symbiotic relationships with EMF. A. koreana is an endangered species with a higher host specificity of EMF and climate change vulnerability than T. cuspidata. This study provides insights into the EMF communities, which are symbionts of A. koreana, and our critical findings may be used to restore A. koreana.

Pollen Analytical Study on Humus Accumulated at Plant Communities of Mt. Seolag (설악산 식물군락 유기물층의 화분분석학적 연구)

  • A pollen analytical study was carried out on the accumulated humus samples collected from 27 sites under the different plant communities of Mt. Seolag. These pollen spectra were then compared with the actual vegetation. The aboreal pollen(AP) and non-abnoreal pollen(NAP) were 1 Family 25 Genus and 5 Family 5 Genus, respectively. Among the aboreal pollen identified, the pollen of Pinus, Quercus, Betula and Acer were appeared in abundance and the coniferous pollen of Pinus was occupied at least 20%-65% through all the sites studied. The pollens of Abies, Betula, accer, Quercus, Carpinus, Corylus, Fraxinus, Styrax, Prunus and Ericaceae were in accordance with the present vegetation but the other pollens were inconsistent with the actual vegetaton. It suggests that it was caused by the difference between Wehseolag and Namseolag of subalpine zone in Mt. Seolag was 200 m by pollen spectra. Quercus was distributed at the lower part of Namseolag and the subalpine boreal elements were increased with increasing altitudes. The pollen spectra of each site was put together into 5 groups of pollen assemblage by cluster analysis and the community coefficient (CC) was over 60% between all sites studied.

  • The vascular plants in Manbokdae-Yeowonjae section of the Baekdudaegan were consisted of 502 taxa; 104 families,305 genus,439 species, 60 varieties, 3 forma. From the floristic point of view, the Baekdudaegan of Manbokdae-Yeowonjae section belongs to the southern province in Korea. The rare and endangered plants designated by Korea Forest Service were Abies koreana, Pedicularis verticillata, Patrinia saniculaefolia, Hololeion maximowiczii, Lilium callolsum, Paeonia obovate, Tricyrtis dizutata and VioEa diamantica, and Cymbidium goeringii. The Korean endemic species were Abies koreana, Patrinia saniculaefolia, Vicia unijuga, Stewartia koreana, Cornus walteri, Weigela subsessilis, Saussurea seoulensis, Tyicyrtis dilatata, Hosta capitata, Clematis trichotoma et at.

Community of Endophytic Fungi from Alpine Conifers on Mt. Seorak

  • Endophytic fungi occupy various ecological niches, which reinforces their diversity. As few studies have investigated the endophytic fungi of alpine conifers, we focused on four species of alpine conifers in this study-Abies nephrolepis, Pinus pumila, Taxus cuspidata var. nana, and Thuja koraiensis-and examined them for endophytic fungi. A total of 108 endophytic fungi were isolated. There were four taxa in A. nephrolepis, 12 in P. pumila, 18 in T. cuspidata var. nana, and 17 in T. koraiensis; these were divided into five classes: Agaricomycetes (3.2%), Dothideomycetes (29.0%), Leotiomycetes (15.0%), Sordariomycetes (41.9%), and Orbiliomycetes (1.6%). The most prevalent fungi were Sydowia polyspora (22.7%) and Xylariaceae sp. (22.7%) in P. pumila, Phomopsis juglandina (16.1%) in T. cuspidate var. nana, and Thuja-endophytes sp. 1 (70.0%) in T. koraiensis. However, there was no dominant species growing in A. nephrolepis. Some host plants were analyzed using next-generation sequencing. We obtained 4618 reads for A. nephrolepis and 2268 reads for T. koraiensis. At the genus level, the top three endophytic fungi were Ophiostomataceae_uc (64.6%), Nectriaceae_uc (15.5%), and unclassified organism (18.0%) in A. nephrolepis and Nectriaceae_uc (41.9%), Ophiostomataceae_uc (41.8%), and Magnaporthaceae_uc (9.2%) in T. koraiensis. Our results show that there are different communities of endophytic fungi among different host plants, even if the host plants are in the same region. Such ecological niches are important in terms of the ecological restoration of alpine conifers.

  • The genus Cohnella, which belongs to the family Paenibacillaceae, inhabits a wide range of environmental niches. Here, we report the complete genome sequence of Cohnella sp. HS21, which was isolated from the rhizospheric soil of Korean fir (Abies koreana) on the top of Halla Mountain in the Republic of Korea. Strain HS21 features a 7,059,027 bp circular chromosome with 44.8% GC-content. Its genome contains 5,939 protein-coding genes, 78 transfer RNA (tRNA) genes, 27 ribosomal RNA (rRNA) genes, 4 noncoding RNA genes (ncRNA), and 90 pseudogenes. The bacterium contains antibiotic-related gene clusters and genes encoding plant cell wall-degrading enzymes.

Karyological Relationship of Genus Abies in Korea (한국산(韓國産) Abies 속(屬)의 핵학적(核學的) 유연관계(類緣關係))

  • Karyotype analysis of somatic chromosome was attempted to find out the relationship among the 3 species of Abies growing in Korea, and A firma introduced from Japan The results were summarized as follows: 1) The number of somatic chromosome was equal for all species, 2n=24. 2) When somatic chromosome was arranged by descending order of the short arm length, the long arm of number 9 chromosome of A. holophylla was much larger than that of the other species. A. koreana and A. nephrolepis had a similarity in b/a value and A. holophylla and A. firma were also similar. 3) When it was arranged by descending order of the short arm length, eight chromosomes of A. koreana and A. nephrolepis showed a similar in b/a value, and two chromosomes of A. holophylla and A. firma did in its value. 4) When it was arranged by descending order of total length, chromosome numbers 7 and 8 in A. koreana were particularly similar to those in A. nephrolepis. 5) When it was arranged by descending order of total length, six somatic chromosomes of A. koreana and A. nephrolepis were similar in b/a value, and two somatic chromosomes of A. holophylla and A. firma similar in its value. 6) When it was arranged by descending order of long arm length, in the of b/a value of chromosome number one in A. koreana and A. nephrolepis was much larger, and that in A. holophylla and A. firma was much smaller than that in the others.

Genetic Variation of Korean Fir Sub-Populations in Mt. Jiri for the Restoration of Genetic Diversity (유전다양성 복원을 위한 지리산 구상나무 아집단의 유전변이)

  • To provide a ecological restoration strategy considering genetic diversity of Abies koreana in Mt. Jiri, the genetic diversity and the genetic differentiation among sub-populations such as Banyabong, Byeoksoryeong, and Cheonwangbong were investigated. The average number of alleles (A) was 7.8, the average number of effective alleles ($A_e$) was 4.9, observed heterozygosity ($H_o$) was 0.578, and expected heterozygosity ($H_e$) was 0.672, respectively. The level of genetic diversity within sub-populations ($H_e=0.672$) was lower than those of both population ($H_e=0.778$) and species ($H_e=0.759$) level. However, the level of genetic diversity was high compared those of Genus Abies. Genetic differentiation was 0.014 from F-statistics ($F_{ST}$) and was 0.004 from AMOVA analysis (${\Phi}_{ST}$). There was no almost genetic differentiation among sub-populations in Mt. Jiri from bayesian clustering. Therefore, If the seeds are sampled sufficiently by selecting the parameters from three sub-populations, it is possible that we could obtain genetically appropriate materials for ecological restoration.

An Unrecorded Genus Lambertella Höhn. (Rutstroemiaceae) and Its Unrecorded Species in Korea

  • Lambertella corni-maris Höhn., an endophytic fungus that is an unrecorded species in Korea, was isolated and cultured from alpine coniferous Korean fir. The Lambertella sp. was found in Korean pine in Yeongwol-gun but no Lambertella spp. were recorded elsewhere in Korea. In this study, the morphological characteristics and molecular barcode sequence of L. cornimaris were compared to its related species. We tried to explain the cultural characteristics and microstructures, such as the conidia of L. corni-maris, compared to closely related taxa and present it.
