한국인에서 치주질환과 관상동맥질환의 관련성에 대한 염증표지자와 IL-1 유전자 다변성의 영향 (Association between Periodontitis and Coronary heart disease in Korea : Inflammatory markers and IL-1 gene polymorphism)

  • Recently epidemiologic studies have indicated that the patients with periodontitis may have increased risk of ischemic cardiovascular events, and have suggested the important roles of blood cytokines and acute reactant proteins in the systemic infection and inflammatory response. Periodontitis and coronary heart disease (CHD) may share the common risk factors and the genetic mechanism associated with interleukin(IL)-1A, B and RA genotype may be involved in the production of IL-1. This study was aimed to investigate the relationship between angiographically defined CHD and periodontitis as chronic Gram-negative bacterial infection and to determine whether the IL-1 gene polymorphism is associated in both diseases. Patients under the age of 60 who had undergone diagnostic coronary angiography were enrolled in this study. Subjects were classified as positive CHD (+CHD, n=37) with coronary artery stenosis more than 50% in at least one of major epicardial arteries, and negative CHD (-CHD, n=30) without significant stenosis. After recording the number of missing teeth, periodontal disease severity was measured by means of plaque index (PI), gingival index (GI), bleeding on probing (BOP), probing depth (PD), clinical attachment level (CAL), and radiographic bone loss around all remaining teeth. Gingival crevicular fluid (GCF) was collected from the 4 deepest periodontal pockets and assessed for cytokine ($IL-1{\beta}$, IL-6, IL-1ra, tumor necrosis $factor-{\alpha}$, and prostaglandin $E_2$). Additionally, blood CHD markers, lipid profile, and blood cytokines were analyzed. IL-1 gene cluster genotyping was performed by polymerase chain reaction and enzyme restriction using genomic DNA from buccal swab, and allele 2 frequencies of IL-1A(+4845), IL-1B(+3954), IL-B(-511), and IL-1RA(intron 2) were compared between groups. Even though there was no significant difference in the periodontal parameters between 2 groups, GCF level of $PGE_2$ was significantly higher in the +CHD group(p<0.05). Correlation analysis showed the positive relationship among PD, CAL and coronary artery stenosis(%) and blood $PGE_2$. There was also significant positive relationship between the periodontal parameters (PI, PD, CAL) and the blood CHD markers (leukocyte count, C-reactive protein, and lactic dehyrogenase). IL-1 gene genotyping showed that IL-1A(+3954) allele 2 frequency was significantly higher in the +CHD group compared with the -CHD group (15% vs. 3.3%, OR 5.118,p=0.043). These results suggested that periodontal inflammation is related to systemic blood cytokine and CHD markers, and contributes to cardiovascular disease via systemic inflammatory reaction. IL-1 gene polymorphism might have an influence on periodontal and coronary heart diseases in Korean patients.

큰느타리버섯 폐배지에서 분리한 Xylanase 생성 Bacillus subtilis CS9 (Xylanase-producing Bacillus subtilis isolated from spent mushroom (Pleurotus eryngii) substrates)

  • 경남지역의 큰느타리버섯재배사에서 채취한 버섯폐배지로부터 그람양성이며 내생포자를 형성하는 간균을 분리하여 이를 CS9라 명명하였다. 분리균 CS9는 통성혐기성균으로 운동성이 있으며 xylanase를 생성하였다. 분리균의 최적 생육조건은 $40^{\circ}C$, pH 6.0이었고, 지방산 조성분석 결과 세포벽을 이루는 주요지방산은 anteiso-C15:0, anteiso-C17:0, iso-C17:0이었으며, DNA G+C함량은 45 mol%로 Bacillus 속의 G+C함량인 43-46 mol%의 범위에 속하는 것으로 확인되었다. 분리균의 16S rDNA 염기서열을 분석한 결과, 분리균 CS9은 Bacillus subtilis YB1 균주와 96.8%의 유사도를 나타내었고, Bacillus속의 다른 균주들과는 92-94%의 유사도를 나타내었다. 이상의 생리적, 생화학적, 유전적 특성을 근거로 큰느타리버섯폐배지로부터 분리한 xylanase를 생성하는 CS9는 Bacillus subtilis CS9로 동정되었다.

한국인 자폐증과 Chromosome 5p14에 존재하는 CDH9, CDH10 유전자 다형성의 연관성 연구 (Polymorphisms of CDH9 and CDH10 in Chromosome 5p14 Associated with Autism in the Korean Population)

  • Objectives : The region of chromosome 5p14 is known to be associated with autism spectrum disorder (ASD). The cadherin9 (CDH9) and cadherin10 (CDH10) genes are located in the region of chromosome 5p14 and reported to be associated with ASD in the Caucasian population. We performed an association study to identify if single nucleotide polymorphisms (SNPs) located on the CDH9 and CDH10 genes are associated in the Korean population. Methods : Genomic DNA was extracted from the blood of 214 patients with ASD and 258 controls. SNPs selected from two genes were genotyped using an Illumina Golden-Gate Genotyping assay with VeraCode technology. Statistical analysis was performed using SAS and Plink software. Results : All controls and ASD patients were in Hardy-Weinberg equilibrium. In the results of logistic regression analyses for the genotype model and the chi-square test for the allele model, we found that SNPs on the CDH9 and CDH10 genes were not associated with ASD. Conclusion : Our data suggests that the CDH9 and CDH10 genes are not associated with ASD in the Korean population.

한국인 아동에서의 IL-1 유전자 다변성 연구 (The IL-1 Gene polymorphisms in Korean Children)

  • The severe form of chronic periodontitis(CP) has been reported to be strongly associated with the presence of allele 2 of composite IL-1B(+3954) and IL-1A(+4845) genetic polymorphisms(genotype positive). However, other studies have reported conflicting findings. These might have resulted from differences in ethnic background and disease entities. The aim of this study was to determine the distribution of IL-1A(+4845), IL-1B(+3954), IL-1B(-511), and IL-1 RN(VNTR) genetic polymorphisms in children as a future Korean population. The study population consisted of 92 children from the Dept. of Pediatric Dentistry, Chonnam National University Hospital. Genomic DNA was obtained from buccal swab. The IL-1A(+4845), IL-1B(+3954), and IL-1B(-511) genes were genotyped by amplifying the polymorphic region using multiplex polymerase chain reaction(PCR), followed by restriction enzyme digestion and gel electrophoresis. IL-1 RN(VNTR) polymorphism were then evaluated by PCR amplification and fragment size analysis in agarose gel. The allele 2 frequency was 41.3%, 4.3%, 47.8%, and 9.9% for IL-1A(+4845), IL-1B(+3954), IL-1B(-511), and IL-1 RN respectively. The frequency of genotype with allele 2 carriage for IL-1A(+4845), IL-1B(+3954), IL-1B(-511), and IL-1 RN was 77.1%, 7.6%, 63.0%, and 15.2% respectively. The allele 2 frequency in IL-1B(+3954) was significantly higher in female than in male population(p<0.05). The negative association was shown between the presence of allele 2 in IL-1B(-511) and in IL-1B(+3954), and the carriage rate of IL-1B(+3954) allele 2 tended to lower in IL-1B(-511) allele 2(P=0.056). Only 7.3% of children carried the composite genotype of IL-1A(+4845) and IL-1B(+3954). These results suggest that the polymorphism of IL-1B(+3954) and the positive composite genotype was relatively rare in Korean population.

초위성체 DNA를 이용한 제주마 집단의 품종특성 및 개체식별 체계설정 (Establishment of Genetic Characteristics and Individual Identification System Using Microsatellite Loci in Jeju Native Horse)

  • 본 연구는 서로 다른 상염색체에 위치하고 있는 초위성체유전 표지를 활용하여 제주마 집단의 타 품종과의 차별적 유전특성분석과 제주마 집단에서 활용할 수 있는 효율적인 개체 식별 시스템 설정을 위해 수행되었다. 공시재료로서는 5품종에서 총 191두가 사용되었으며 13종의 좌위에 대한 개체별 유전자형을 분석하였다. 이들 13종에서 출현된 총 대립 유전자수는 제주마의 경우 155종이 나타났다. 한국 제주마 집단에서 나타난 평균 이형접합도는 0.317-0.902였으며 marker 다형성 정보량은 0.498-0.799로 나타났다. 제주마 집단에서 나타난 대립 유전자 발현 특성은 다른 외래 품종 집단과 매우 상이한 결과를 나타냈다. ATH4 좌위의 경우 제주마 집단에서는 5종의 대립 유전자가 고른 분포를 나타낸 반면 QUA종의 경우와 THO종집단에서 특정 좌위의 극단적 발현 빈도를 나타냈다. 품종특이성을 나타내는 분자표지의 대립유전자 발현양상이 확인되었으며 이러한 품종특이성 발현 분자표지는 집단 내 개체에 대한 품종식별 유전자표지로 활용이 가능한 것으로 나타났다. 9종의 초위성체 유전 표지를 활용할 경우 누적 개체 식별력은 99.9%를 나타냈으며 두 마리의 서로 다른 개체가 서로 같은 유전자형을 가질 짝확률은 $0.60\;{\times}\;10^{10}$으로 추정되었다. 따라서 9종의 선정된 유전표지는 제주마 품종 집단에서 적정 신뢰도를 제공할 수 있는 개체 식별 시스템을 설정할 수 있을 것으로 생각된다.

Brca1 결손 세포주에서 nocodazole 처리에 의한 spindle checkpoint 활성화 연구 (Impaired Spindle Checkpoint Response of Brca1-deficient Mouse Embryonic Fibroblasts (MEFs) to Nocodazole Treatment)

  • 항암유전자 Brca1의 변이는 유방암 및 난소암에 대한 감수성을 증가시키며, Brca1은 DNA손상신호후 세포주기 조절에 필수적인 역할을 한다. 연구결과, Brca1이 세포주기 S기와 G2/M 조절점에서 중요한 역할을 담당함이 밝혀졌다. 그러나, Brca1의 spindle checkpoint 관여여부는 알려져 있지 않다. 본 연구에서는 spindle checkpoint를 활성화시키는 nocodazole를 처리하여 야생형, $p53^{-/-}$ 그리고 $p53^{-/-}\;Brca1^{-/-}$ 세포주의 세포주기 변화를 조사하였다. 야생형과 $p53^{-/-}$ 세포주는 신속한 mitosis기 정지가 나타난 반면, $p53^{-/-}\;Brca1^{-/-}$ 세포주의 경우 모든 세포가 M기에서 정지하지 않았다. Double-thymidine block 기법에 의한 세포주기 동조화후 nocodazole 처리시에도 $p53^{-/-}\;Brca1^{-/-}$ 세포주에서는 일부세포가 M기 조절점을 통과하여 계속 G1기로 진행하였다. 형태학적 분석에서도 nocodazole 함유배지에서 계속 증식하는 세포형태가 관찰되었다. 이와 같은 결과들은 Brca1이 spindle checkpoint가 정상적으로 작동하는데 중요한 역할을 담당한다는 것을 의미하고 있다.

Bacillus ginsengihumi sp. nov., a Novel Species Isolated from Soil of a Ginseng Field in Pocheon Province, South Korea

  • A Gram-positive, aerobic or facultative anaerobic, non motile, endospore-forming bacterial strain, designated Gsoil $114^T$, was isolated from a soil sample of a ginseng field in Pocheon Province (South Korea), and was characterized taxonomically by using a polyphasic approach. It grew well on nutrient agar medium and utilized a limited number of organic substrates as sole carbon sources, including D-xylose and some other carbohydrates, but did not utilize L-amino acids and organic acids. The isolate was positive for oxidase test but negative for catalase, and negative for degradation of macromolecules such as starch, cellulose, xylan, casein, chitin, and DNA. The G+C content of the genomic DNA was 41.8 mol%. The predominant isoprenoid quinone was menaquinone 7 (MK-7). The major fatty acids were $anteiso-C_{15:0}$ (32.1%), $iso-C_{15:0}$ (30.5%), and $anteiso-C_{17:0}$ (30.2%). Comparative 16S rRNA gene sequence analysis showed that strain Gsoil $114^T$ fell within the radiation of the cluster comprising Bacillus species and joined Bacillus shackletonii LMG $18435^T$ with a bootstrap value of 95%. The highest 16S rRNA gene sequence similarities were found with Bacillus shackletonii LMG $18435^T$ (97.6%), Bacillus acidicola DSM $14745^T$ (96.9%), Bacillus sporothermodurans DSM $10599^T$ (96.5%), and Bacillus oleronius DSM $9356^T$ (96.5%). The phylogenetic distance from any other validly described species within the genus Bacillus was less than 96%. DNA-DNA hybridization experiments showed that the DNA-similarities between strain Gsoil $114^T$ and closest phylogenetic neighbors were less than 39%. On the basis of its phenotypic properties and phylogenetic distinctiveness, strain Gsoil $114^T$ (=KCTC $13944^T$=DSMZ $18134^T$) was classified in the genus Bacillus as the type strain of a novel species, for which the name Bacillus ginsengihumi sp. nov. is proposed.

Agrobacterium tumefaciens를 이용한 bialaphos 저항성 형질전환 벼의 개발 (Development of Bialaphos-Resistant Transgenic Rice Using Agrobacterium tumefaciens)

  • 비선택성 제초제인 bialaphos는 고등식물에 있어서 glutamine 합성을 억제 하여 식물체를 고사시킨다. Acetyltransferase에 의해 encoding된 bialaphos저항성 유전자는 세균 Streptomyces hygroscopicus SF1239로부터 cloning된것을 사용하였다. Bialaphose 저항성 유전자를 Agrobacterium 감염법을 이용하여 국내에서 재배되는 벼(동진)에 도입한 결과 약30%정도의 형질전환 식물체를 얻을 수 있었다. $\textrm{T}_{1}$ 세대의 17개체는 hygromycin과 bialaphos에 대한 저항성 유전형질이 3:1로 분리되었다. 또한 Southern 분석을 실시한 결과 wild type의 식물체에서는 Bar 유전자의 검출을 볼 수 없었으나 형질전환 식물체의 경우 Bar 유전자의 검출이 가능하였다. $\textrm{T}_{3}$세대의 형질전찬 식물체와 wild type의 식물체를 포장상태에서 비선택성 제초제인 바스타를 살포하고 3주 후에 관찰한 결과 형질전환 식물체는 외형적으로 아무런 피해를 받지 않고 정상적으로 생장하였으나, wild type의 식물체와 잡초는 모두 고사하였다. 이상의 결과를 종합해보면 hygromycin과 bialaphos 저항성 유전자는 Agrobacterium감염법을 이용하여 단자엽 식물인 벼에 도입할 수 있다는 것을 보여준 것이며, 또한 bialaphos 저항성 유전자가 식물에 도입됨으로써 비선택성 제초제에 대한 저항성 식물을 개발 할 수 있다는 것을 보여 주었다.

Bacillus subtilis를 이용한 대두 발효식품의 혈전용해능

  • A strain producing strongly fibrinolytic enzyme was isolated from soil and was identified to be Bacillus subtilis by biochemical and physiological characterization. The optimal culture conditions for the production of fibrinolytic enzyme was determined to be 1.0% tryptone, 1.5% soluble starch, 0.5% Peptone, 0.5% NaCl, $(NH_{4})_{3}PO_4.3H_{2}O, and MgSO_{4}.7H_{2}O.$ Initial pH and temperature were pH 8.0 and $30^{\circ}C$ , respectively, The highest enzyme production was observed at 30 hours of cultivation at $30^{\circ}C$ The fibrinolytic enzyme was purified to homogeneity by DEAE Sephadex A-50 ion exchange column chromatography, 70% ammonium sulfate precipitation, Sephadex G-200 and G-75 gel filtration column chromatography. The molecular weight of the purified enzyme was 28,000 as determined by sodium dodecyl sulfate-polyacrylamide gel electrophoresis. A gene encoding the fibrinolytic enzyme was cloned into a plasmid vector pBluescript, transforming E.coli XL-1 Blue. The clone was able to degrade fibrin, This indicated that the gene could encode a fibrinolytic enzyme. The nucleotide sequence of the 2.7 kb insert was determined in both direction. One open reading frame composed of 1023 nucleotides was found to be a potential protein coding region. There was the putative Shine-Dalgano sequence and TATA box upstream of the open reading frame. The homology search data in the genome database showed that both the 2.7 kb insert and 1 kb open reading frame carried no significance in the nucleotide sequence of known fibrinolytic enzyme from Bacillus serovars. The recombinant cell harboring the novel gene involved in fibrinolysis was subjected to protein purification. The molecular mass of the purified fibrinolytic enzyme was determined to be 31864 Dalton, which was highly in accordance with the molecular mass(33 kDa) of the fibrinolytic gene deduced from the insert. The fibrinolytic enzyme was Purified 50.5 folds to homogeneity in overall yield of 10.7% by DEAE Sephadex A-50 ion exchange, 85% ammonium sulfate precipitation, Sephadex G-50, Superdex 75 HR FPLC gel filtration. In conclusion, a novel fibrinolytic gene from Bacillus subtilis was identified and characterized by cloning a genomic library of Bacillus subtilis into pBleuscript. For the soybean fermented by this strain, it is found that there increased assistant protein about 20% compared to the soybean not fermented and increased about 30% according to amino acid analysis and, in particular, essential amino acid increased about 40%. When keeping this fermented soybean powder at room temperature for about 70days, it showed very high stability maintaining almost perfect activity and, therefore, it gave us great suggestion its possibility of development as a new functional food.

공동배양과정의 배지조성과 배양조건이 벼 형질전환효율에 미치는 영향 (The Effects of Co-cultivation Medium and Culture Conditions on Rice Transformation Efficiency)

  • 1. 본 연구에서는 공동배양 배지에 Agrobacterium 성장 억제물질인 silver nitrate를 첨가하고 변온과 여과지처리를 추가하여 공동배양 기간을 7일로 늘였으며, 또한 항산화 물질 3종을 공동배양 배지에 첨가하여 세포의 oxidative burst를 최소화함으로써 벼 형질전환효율을 높일 수 있었다. 또한 이 방법을 적용하여 형질전환이 어려운 품종을 대상으로도 형질전환 식물체를 작성할 수 있었다. 2. 벼 형질전환체의 70%에서 도입유전자 수가 1copy인 것으로 나타나, 적은 수의 유전자가 안정적으로 도입됨을 확인 하였다. 3. 이러한 결과를 바탕으로, 새로운 공동배양 방법을 사용하여 우수한 농업적 형질을 가진 벼 육종 소재 및 품종을 신속하게 개발할 수 있을 것으로 기대된다.
