Cytological and Cytochemical Studies of the Hybrids in Fresh Water Teleosts

  • Genetical characters and abilities will be appeared by the special gene or gene group on the chromosomes. We have two different stand-points to this problem. (I) If these genes become homozygotic, the genetic presentation will be emphasized and fixed as a pure line. (II) The genetic presentation will be emphasized by the heterozygotic genes and, on the contrary, homozygotic genes only make no efficiency to emphasis in genetical characters. The problem which is by far the better of the two standpoints, should be evaluated and determined by the purposes of human beings.

Monitoring of Genetical Modified Soybean Sold at Local Open Market in Western Gyeongnam (서부경남 재래시장을 대상으로한 유전자 재조합 대두의 모니터링)

  • The objective of this study was monitoring of genetically modified soybean by PCR and ELISA. We collected 60 soybean samples from the open markets located in Western Gyeongnam (Sacheon, Hamyang, Hadong, Sanchung, Uiryung, Geochang, and Hapcheon). A total of 60 soybeans was examined and 14 genetical modified soybean (GMS) were detected by PCR. The GMS rate of selling soybean in Uiryung, Hadong, Sacheon, and Hapcheon was 50.0%, 37.5%, 33.3% and 25%, respectively. The 7 of 14 GMSs were positive by ELISA and most of positive samples were below 3% GMS but 1 (Uiryung 1) of the positive samples was over the 3% which is maximum permit limit in Korea. These results mean that merchants of open market did not express for selling soybean mixed with GMS, so consumers did not recognize GMO. Therefore, we thought that education of GMO for merchant of open market need to recognize about GMO maximum permit limit.

Genetical and Biochemical Studies on White Egg Strains of Silkworm, Bombyx mori (누에 백란 계통의 유전적 특성구명을 위한 유전.생화적 연구)

  • To elucidate genetical and biochemical characters of the white egg strains preserved in National Sericultural and Entomology Research Institute(NSERI), RDA of Korea, the genetic segregation ratios in egg colours were investigated by crossing test, and egg pigment precursors were also analyzed by paper chromatography and UV-spectrophotometer scanning. The result obtained by crossing test between the white egg strains and normal one illustrated that the most of white egg strains showed typical segregation ratio of white egg-2(w-2), while maternal inheritance which can be seen in white egg-1(w-1) was not found in any white egg strains. Paper chromatographic analysis showed that egg extracts of all the white egg strains contained 3OH-kynurenine, while kynurenine which is known to existed in white egg-1(w-1) could not be detected at all. From the results of these experiments, it was found that all the white egg strains preserved in NSERI were classified as the white egg-2(w-2) strain.

  • To analyze the mode of inheritance of this gene in Fb strain, carried out cross experiment between Fb and several Japanese and Chinese origin strains. The results of crossing experiments, feeding ratio in F1 hybrids were nearly the mid of parents, F2 and backcrossed hybrids(BF1) were approximately intermediate between Fb and other strains. Hence, the feeding habit of cabbage leaves on Fb strain is assume to be incomplete dominance.

  • Physiology and genetics of early maturity in cereals are the subject of practical as well as scientific interest for agronomist and plant breeders, Thorough understanding of the true nature of such a complex character requires physiological and genetical knowledge about the internal factors, which are closely bound up with and react to some particular external or environmental factors. From the practical point of view. experiments should be conducted under controlled conditions. especially the day length and temperature, so that the genotypic differences pertaining to these factors may be discerned. Takahashi and Yasuda (1958, 1970) maintained that at least three physiological factors were responsible for determining earliness in barley. namely. (1) spring and winter habit of growth or vernalization requirement, (2) ogitioeruiduc response or sensitivity to short-day, and (3) earliness factor in a narrow sense or minimal vegetative growth. The same situations were true in common wheat also (Yasuda and Shimoyama, 1965), In this report. physiology and genetics of internal factors and their relations to the time of heading in the field will be presented with some problems concerning differences in mechanism of early maturity between barley and wheat.

  • These researches were carried out to investigate the morphology of different species of Trichoderma and the possibilities of differentiation of the species of Trichoderma by electrophoretic methods. Variations between the isolates of a species of Trichoderma indicate the genetical differences, also isozyme and protein patterns will be useful to investigate genetical variations betweens the isolates. It might be possible that distinct bands of isozymes of esterase, phosphotase, catalase, catalase differentiate species of Trichoderma.

Approach Toward the Creation of Nonnarcotic Opium Poppy - Morphological and Genetical Analysis on 'Thebaine Poppy' -

  • Opium poppy (Papaver somniferum L.) is one of the most important medicinal plants, which is used as a sole commercial source of narcotic analgesic, morphine. The transformant of opium poppy we have established by infection of Rhizobium rhizogenes (formerly Agrobacterium rhizogenes) strain MAFF03-01724 showed aberrant morphology and altered opium alkaloid composition. The major alkaloid produced by this transformant was thebaine (16.3%, opium dry weight) instead of morphine. It is likely that this 'thebaine poppy' phenotype was caused by the integration of T-DNA(s) into the poppy genome DNA, and their inserted loci are of great interest. To gain an insight into the mechanism of nonnarcotic thebaine accumulation for the further approach toward the creation of 'codeine poppy' which produces codeine as a major alkaloid, the genetical and morphological analyses on the transformant was carried out. Here we report the results of the detailed analysis on the T-DNA inserted loci of T0 transfromant and the correlation between opium alkaloid composition and segregated T-DNA integration pattern in the self-pollinated T1 transformants.
