Development and Characterization of New Tall fescue Variety 'Greenmaster4ho' (톨 페스큐 신품종 '그린마스터4호'의 품종 특성 및 수량성)

  • A new variety of tall fescue (Festuca arundinacea Schreb.) named 'Greenmaster4ho' was developed during the cultivation year 2010-2017 at Grassland and Forages Division, National Institute of Animal Science, Rural Development Administration, Cheonan, Korea. In order to produce this new variety, 5 superior tall fescue lines including 12XFa07, 12XFa15, 12XFa24, 12XFa46, and 12XFa48 were polycrossed. The new variety Greenmaster4ho was evaluated in field test (Cheonan, Pyeongchang, Jeju, and Jinju) for determining the agronomic growth characteristics and forage production capability during 3 years (2015-2017). The dry matter yield (16,236 kg/ha) of Greenmaster4ho was 5 % higher than Fawn, but the nutritive value as forage crops was not significantly different with Fawn. Development of new tall fescue variety with excellent adaptability to changing unfavorable environment would be useful for forage cultivation and yield in Korean environment.

  • Recently, the rapid growth of the companion animal market has led to the development of animal disease diagnosis kits. Therefore, the utility of the introduction of biomarkers for the development of animal molecular diagnostics is being reevaluated. A good biomarker should be precise and reliable, distinguish between normal and diseased states, and differentiate between different diseases. Recently reported genetic markers, tumor markers (cell free DNA, circulating tumor cells, granzyme, and skin tumors), and others (brucellosis, programmed death recovery-1, symmetric dimethylarginine, periostin, and cysteinyl leukotrien) have been developed. The biomarkers are used for risk prediction or for the screening, diagnosis, and monitoring of disease progression. The most important criteria for related biomarkers are disease specificity. Many potential biomarkers have emerged from laboratory and test studies, but they have not been validated in independent or large-scale clinical studies. Candidate biomarkers evaluate disease associations, verify the effectiveness of biomarkers for early detection and disease progression, and incorporate them into humans and animals. In the future, it will be necessary to reevaluate the utility of well-structured biomarker-based research and study the development of kits that can be used in on-site tests in accordance with the trends introduced in the diagnosis of animal diseases.

rvH1N1 Neuraminidase Inhibitory Activities of Phenolics from Perilla frutescens (L.) and Their Contents in Cultivars and Germplasm

  • The influenza neuraminidase (NA, E.C. 3.2.1.18), an antiviral, has been the target of high pharmaceutical companies due to its essential role in viral replication cycle. Perilla frutescens (P. frutescens) is used in traditional Chinese medicine for various diseases, such as cold due to wind-cold, headache and cough. In this context, four major polyphenolic compounds including rosmarinic acid-3-O-glucoside (1), rosmarinic acid (2), luteolin (3), and apigenin (4) isolated from P. frutescens were evaluated for their inhibitory effect on recombinant virus H1N1 neuraminidase (rvH1N1 NA). Among the test compounds, rosmarinic acid and luteolin inhibited the rvH1N1 NA with an $IC_{50}$ of 46.7 and $8.4{\mu}M$, respectively. The inhibition kinetics analyzed by the Dixon plots indicated that rosmarinic acid and luteolin were noncompetitive inhibitors and that the inhibition constant, $K_I$, was established as 43.9 and $14.3{\mu}M$, respectively. In addition, 578 genetically diverse accessions and 39 cultivars of P. frutescens were analyzed using HPLC to characterize the diversity of polyphenolic composition and concentration. The individual and total compositions exhibited significant difference (P < 0.05), especially rosmarinic acid which was detected as the predominant metabolite in all accessions (58.8%) and cultivars (62.8%). Yeupsil and Sangback cultivars exhibited the highest rosmarinic acid ($3,393.5{\mu}g/g$) and luteolin ($383.3{\mu}g/g$) content respectively. YCPL177-2 with the high concentration ($889.8{\mu}g/g$) of luteolin may be used as a genetic resource for breeding elite cultivars.

Nematode-Trapping Fungi Showed Different Predacity among Nematode Species (선충 종류별 4종 포식성곰팡이의 포식력 차이)

  • Nematode-trapping fungi develop trap and consume nematodes are an important part of the subsoil ecosystem and they share a special predator-prey relationship. Four nematode-trapping species, there with adhesive network, Arthrobotrys oligospora, A. sinensis, A. thaumasia and one with constricting ring, Drechslerella brochopaga were collected from soils in Korea and tested their predacity against 12 different nematode species. They were three feeding groups, plant-parasitic (Meloidogyne incognita and Pratylenchus penetrans), fungivorous (Aphelenchus avenae), bacteriovorous (Betlerius sp. and Diplogasteritus sp. in diplogasterid, Panagrolaimus labiatus, P. multidentatus in panagrolaimid, Mesorhabditis irregularis, Pelodera strongyloides and Rhabditis sp., in rhabditid, and Acrobeloides sp. in cephalobid). Results showed that nematode-trapping fungi successfully captured most of nematodes in Petri dish in the group of plant-parasitic nematodes and rhabditids, moderately and variably in other nematodes in 15 days. But it didn't captured A. avenae and Acrobeloides sp. both belongs to c-p group 2. Numbers of Acrobeloides sp. and A. avenae even increased during the test period. The results of this study indicated that nematode-trapping fungi may have specificity among nematode species.

Association analysis of polymorphisms of G protein-coupled receptor 54 gene exons with reproductive traits in Jiaxing Black sows

  • Objective: The aim of this study was to detect single nucleotide polymorphisms (SNP) of G protein-coupled receptor 54 (GPR54) gene and explore association of this candidate gene with reproductive traits in Jiaxing Black sows. Methods: Six pairs of primers of the gene were designed to amplify all exons thus sequences of which were detected by means of direct sequencing and then SNP loci were scanned. The effects of SNPs on total number of piglets born (TNB), number of piglets born alive (NBA), number of still born piglets (NSB), and litter weight at birth (LWB) of Jiaxing Black sows were analyzed. Results: Three SNP loci, including T3739C, C3878T and T6789C, were identified via comparison of sequencing and two genotypes (AB, BB) at each SNP site were observed. T3739C resulted in the change of amino acid ($Leu{\rightarrow}Pro$) in corresponding protein, and C3878T resulted in synonymous mutation ($Ile{\rightarrow}Ile$). Statistical results demonstrated that allele B was the preponderant allele at the three SNP loci and Genotype BB was the preponderant genotype. Meanwhile, Chi-Square test of these three SNPs indicated that all mutation sites fitted in Hardy-Weinberg equilibrium (p>0.05). For GPR54-T3739C locus, Jiaxing Black sows with genotype BB had 1.23 TNB and 1.28 NBA (p<0.01) that were more than those with genotype AB, respectively. Jiaxing Black sows that had the first two parities with genotype BB had additional 2.23 TNB, 2.27 NBA (p<0.01), and 1.94 LWB (p<0.05) compared to those with genotype AB, respectively. However, for other two loci, no significant difference was found between TNB, NBA, NSB, and LWB, and different genotypes of Jiaxing Black sows. Conclusion: In conclusion, the polymorphisms of GPR54-T3739C locus were significantly associated to TNB, NBA, and LWB and could be used as a potential genetic marker to improve reproductive function of Jiaxing black sows.

Breeding of a new cultivar of White Flammulina velutipes, 'Seolhan' (흰색 팽이 신품종 '설한' 육성)

  • Flammulina velutipes is the representative edible mushroom in Korea. It makes up around 18% of the total mushroom production and accounts for about 36% of the total mushroom export. Despite being one of the most industrially important mushroom in Korea, the distribution rate of domestic cultivar was low (~31.6%) in 2020. Therefore, we need a new domestic cultivar with high quality as the main export mushroom. We developed a new white domestic cultivar, 'Seolhan', bred by mating two monokaryotic strains isolated from Baekseung and Woori-1ho. The optimum mycelial growth temperature was 25℃ and Seolhan had higher mycelial growth than the control at 20 and 25℃. In bottle cultivation, the optimum temperature of incubation was from 15℃ to 17℃ and the growth temperature of Seolhan was 7-8℃, which was slightly higher than that of the control. The yield of Seolan was 226.5±8.0 g, an increase of 18.4% compared to the control. The period of primodia formation also was a couple of days ahead. In morphological traits, the pileus of Seolhan was smaller than that of the control, the stipe was thicker, and the color brighter. As the results of replacement culture and genetic diversity test, it was confirmed that Seolhan was genetically different from other white varieties.

Detection of the Carbapenem Resistance Gene in Gram-negative Rod Bacteria Isolated from Clinical Specimens (임상검체에서 분리된 그람음성막대균으로부터 카바페넴 내성 유전자 검출)

  • Carbapenem-resistant Enterobacteriaceae (CRE) poses an increasing public health threat and has limited treatment options with high associated mortality. Genotypes of carbapenemase that threaten public health (blaKPC, blaNDM, blaIMP, and blaVIM) and blaOXA-48-like genes were detected by phenotypic and molecular diagnosis, and related gene distribution patterns were investigated. Phenotypic testing using the modified Hodge test confirmed positivity in all 41 strains examined, and carbapenemase inhibitory testing using meropenem+phenyl boronic acid or meropenem+EDTA confirmed positivity in 18 and 8 strains, respectively. Polymerase chain reaction revealed the presence of amplification products in 28 strains of blaKPC, 25 strains of blaNDM, 5 strains of blaIMP, 1 strain of blaVIM, and 13 blaOXA-48-like strains. In addition, 7 strains of blaKPC+blaNDM, 1 strain of blaKPC+blaIMP, 1 strain of blaNDM+blaOXA-48-like, 1 strain of blaNDM+blaVIM, 4 strains of blaKPC+blaNDM+blaIMP, and 4 strains of blaKPC+blaNDM+blaOXA-48-like were identified. Melting curve analysis using real-time PCR was wholly consistent with PCR results. The study shows genetic identification of highly specific CRE by real-time PCR could be used to provide early diagnoses and infection control, improve surveillance, and prevent the transmission of CRE.

Dietary effect of energy levels on growth performance and carcass characteristics of White Pekin duck over 21 days

  • This experiment was conducted to determine the maximum dietary energy levels on growth performance and carcass characteristics of White Pekin duck. the Six dietary treatments were formulated based on their apparent metabolizable energy (AME) concentrations from 2,700 to 3,200 kcal/kg with a 100 kcal/kg gap to evaluate the accurate dietary AME requirement to address current knowledge and further issues for fulfilling the genetic potential of meat-type white Pekin ducklings. A total of 432 one-day-old male White Pekin ducklings were randomly allocated into one of six dietary treatments with six replicates (12 birds per pen). The diets were formulated as corn-soybean meal-based diets to meet or exceed the Nutrient Requirement of Poultry specification for meat-type ducks. Growth performance indices (i.e. average daily gain [ADG], average daily feed intake, feed conversion ratio) were measured weekly. Medium body weight (BW) ducklings from each pen were sacrificed to analyze the carcass traits and abdominal fat content on day 21. Obtained data were analyzed to estimate significant effect using the one-way ANOVA of IBM SPSS Statistics (Version, 25). If the p-value of the results were significant, differences in means among treatments were separated by Tukey's post hoc test. Significant differences were then analyzed with a linear and quadratic broken model to estimate the accurate concentration of AME. Ducklings fed higher dietary AME diets increased (p < 0.05) BW, ADG. Ducklings fed higher AME than 2,900 kcal/kg diets increased abdominal fat accumulation and leg meat portion. The estimated requirement by linear plateau method showed from 3,000.00 kcal/kg to 3,173.03 kcal/kg whereas the requirement by quadratic plateau method indicated from 3,100.00 kcal/kg to 3,306.26 kcal/kg. Collectively, estimated dietary requirements exhibit diverse results based on the measured traits and analysis methods. All the estimated requirements in this experiment present higher than previous research, the maximum requirement for the next diet formulation should be selected by the purpose of the diet.

Characterization of broad bean wilt virus 2 isolated from Perilla frutescens in Korea (국내 잎들깨에서 발생한 잠두위조바이러스2의 특성 구명)

  • Broad bean wilt virus 2 (BBWV2) is a species in the genus Fabavirus and family Secoviridae, which is transmitted by aphids and has a wide host range. The BBWV2 genome is composed of two single-stranded, positive-sense RNAs, RNA-1 and RNA-2. The representative symptoms of BBWV2 are mosaic, mottle, vein clearing, wilt, and stunting on leaves, and these symptoms cause economic damage to various crops. In 2019, Perilla fructescens leaves with mosaic and yellowing symptoms were found in Geumsan, South Korea. Reverse-transcription polymerase chain reaction (RT-PCR) was performed with specific primers for 10 reported viruses, including BBWV2, to identify the causal virus, and the results were positive for BBWV2. To characterize a BBWV2 isolate (BBWV2-GS-PF) from symptomatic P. fructescens, genetic analysis and pathogenicity tests were performed. The complete genomic sequences of RNA-1 and RNA-2 of BBWV2-GS-PF were phylogenetically distant to the previously reported BBWV2 isolates, with relatively low nucleotide sequence similarities of 76-80%. In the pathogenicity test, unlike most BBWV2 isolates with mild mosaic or mosaic symptoms in peppers, the BBWV2-GS-PF isolate showed typical ring spot symptoms. Considering these results, the BBWV2-GS-PF isolate from P. fructescens could be classified as a new strain of BBWV2.

Epistatic Interaction Analysis of Two Dull Genes, wx-mq and du1, Affecting Amylose Content Using Nearly Isogenic Lines in Rice

  • Glutinous rice is a key grain quality trait occupying an important part during rice processing in most rice growing areas. Amylose content (AC) of rice determine eating quality which is one of the major traits in rice breeding program. In this study, a gene pyramiding approach was used to introduce two dull genes, responsible for low amylose contents, for glutinous rice breeding using marker assisted selection (MAS). Two dull genes were located on chromosome 6 (wx-mq, AC: 12.7 %) and chromosome 10 (du1, AC: 10.3%), respectively. To test whether these two dull genes have an epistatic interaction, we developed an F2 population by crossing two nearly isogenic lines(NILs) harboring wx-mq and du1. Gene based marker and KASP marker were used to select NILs(NIL-nor, NIL-wxmq, NIL-du1, and NIL-wxmq/du1) from the F2 population. A two-way ANOVA revealed an epistatic interaction between the two genes in the F2 population. The mean of Amylose contents for NIL-nor, NIL-wxwq, NIL-(du1, and NIL-wxmq/du1 were 17.3%, 12.5%, 9.7%, and 7.2%, respectively. This interaction was confirmed by an analysis of NILs indicating that both genes are involved in the same genetic mechanism controlling amylose contents. This result will be useful for rice breeding related to amylose content.
