Acidovorax valerianellae had been reported a causal agent of bacterial black spot disease on corn salad in France, 2003 and on watermelon in Korea 2011. In this study, difference in host specificity between 2 groups, corn salad strains and watermelon strains, of Acidovorax valerianellae was recognized and compared. In the pathogenicity test, all 5 watermelon strains showed pathogenicity on the 6 Cucurbitaceae plants but not on corn salad, whereas 4 corn salad strains showed pathogenicity only on the corn salad. Utilization of Biolog substrates was different between watermelon strains and corn salad strains on 4 substrates, Malonic Acid, ${\alpha}-Hydroxybutyric$ Acid, ${\alpha}-Keto$ Butyric Acid, and Glycyl-L Glutamic Acid. The phylogenetic tree built with the 16S rDNA sequences showed that all of A. valerianellae stains was grouped into 1 clade separating from the other species of Acidovorax genus. Within A. valerianellae clade, watermelon strains and corn salad strains were separated into 2 sub-groups. REP-PCR analysis also separated the two groups. Host specificity, substrate utilization, and some genetic characteristics suggested that there are two pathogenic groups, watermelon group and corn salad group in A. valerianellae.
This study investigated genetic, mineralogical and spectral characteristics of oil shale and coal samples in Dundgobi area, Mongolia. Based the Rock/Eval and Total organic carbon (TOC) analysis, kerogen type, hydrogen quantity, thermal maturity and depositional environment were confirmed. Moreover, the mineral composition of oil shale and coal samples were analyzed by XRD and spectroscopy. The result of Rock Eval/TOC analysis revealed that the samples of Eedemt deposit are immature to mature source rocks with sufficient hydrocarbon potential, and the kerogen types were classified as Type I, Type II and Type III kerogen. On the other hand, the samples from Shine Us Khudag deposit were mature with good to very good hydrocarbon potential rocks where kengen types are defined as Type I, Type II/III and Type III kerogen. According to the carbon and sulfur contents, the depositional environment of the both sites were defined as a freshwater depositional environment. The XRD analysis revealed that the mineral composition of oil shale and coal samples were quartz, calcite, dolomite, illite, kaolinite, montmorillonite, anorthoclase, albite, microcline, orthoclase and analcime. The absorption features of oil shale samples were at 1412 nm and 1907 nm by clay minerals and water, 2206 nm by clay minerals of kaolinite and montmorillonite and 2306 nm by dolomite. It is considered that spectral characteristics on organic matter content test must be tested for oil shale exploration using remote sensing techniques.
RAPD test and the observation of morphological, cultural characteristics of fourteen selected entomogenous fungi were conducted to investigate the analysis of their internal relationships. Paecilomyces tenuipes showed snow flower form attached to numerous white conidiophores, produced globular and semi-egg types on the club types of phialides. Cordyceps militaris formed globosely conidiophores, dark yellow fruiting body on pupa. The phialide as on Acremonium-type in global conidiophores. Beauveria bassiana covered with conidia was not formed fruiting body and adhered conidia on conidiophore of zigzag type. The PDA and SDAY medium were confirmed as an optimum growth of them. P. tenuipes showed to velvet and plane types in several media whereas C. militaris was belong to centrally raised and floccose in the morphological type. In contrast, B. bassiana covered with conidia on velvet shape. The size of amplified products were analyzed by RAPD using URP primer and were from 100 bp to 2.0 kb with $10{\sim}14$ geuomic DNA. Total similarities of two groups were by dendrogram of UPGMA analysis. The homology of P. tenuipes groups was 94.8 to 100%. It also showed 70.1 to 96.6% in C. militaris group and B. bassiana was higher similarity than any other. The internal change of C. militaris, produced telemorph fruiting body, was higher seperated in species than P. tenuipes and B. bassiana in the RAPD.

We already reported four groups which contains some similar strains based on URP-PCR in the previous paper. The objective of this study was to confirm those strains by the amplified fragment length polymorphism (AFLP) and vegetative compatibility group (VCG). AFLP analysis showed no difference among these strains except ASI 2595 and 2183 in Weonhyeong group and ASI 2829 in Suhan group. They showed specific DNA bands only in the result of P + AG/M + AAG and P + GT/M + ATG primer combinations out of eight different combinations. The AFLP primers produced a total of 330 fragments between 80 and 1000 bp in length for 31 Pleurotus ostreatus strains. At a genetic similarity of 0.96, the UPGMA analysis separated the isolates into four distinct clusters. Each group was classified by similar strains. Confrontation test by vegetative compatibility groups (VCGs) also showed distinct line between strains from different groups, but no line between similar strains within the cluster. Our results indicate that most of similar strains was not distinctness. Thus, similar strains are considered to be very close on the genealogy of their parent or same strain with different name.

Squamous cell carcinoma is the most prevalent oral cancer, which is characterized by its low survival rate, high malignancy, mortality with facial defects, and poor prognosis. Exact cause and pathogenesis of the squamous cell carcinoma is still unknown. Various routes including smoking, radiation, and viral infections predispose its genesis, and recent studies revealed that genetic defects which fail to prevent cancer proliferation play a role. Generally, a cancer develops from the decreased rate of apoptosis which is an active and voluntary cell death, and from the altered cell cycles. Anticancer effect can be obtained by recovering the apoptotic process, and by suppressing the cell cycles. Among the apoptosis related factors, bcl-2, caspase-9, and VDAC (voltage-dependent anion channel)are produced in mitochondria of the cell. Cyclosporin-A is known to induce apoptosis through its activation with VDAC. This study was to reveal the anticancer effect of Cyclosporin A to the oral squamous cell carcinoma. The inverted microscope was used to find alterations in the tissue, and sensitivity test to the anticancer cells was performed with MTT (Tetrazolium-based colorimetric) assay. Following cell line culture of primary and metastastic oral squamous cell carcinoma, electrophoresis was performed with extracted total RNA. Finally, semi-quantitative study was carried out through RT-PCR (Reverse Transcription-Polymerase Chain Reaction). The results of this study are as follows: 1. The inverted microscopic observation revealed a poorly defined cytoplasm at $2000ng{\sim}3000ng/ml$, indistinct nucleus, and apoptosis. 2. The Growth of cancer cells was decreased at 1000ng/ml of cyclosporin-A. No cancer cell growth was observed at over 2000ng/ml concentration of cyclosporin-A, and at one week, growth of cancer cells was ceased. 3. The MTT assays were decreased as cyclosporin-A concentration was increased. This means that the activation of succinyl dehydrogenase in mitochondria was decreased following administration of cyclosporin A. 4. A result of RT-PCR showed that amount of mRNA of VDAC-2 was decreased half times at a cyclosporine-A concentration of 2000ng/ml. In bcl-2, amount of mRNA was significantly decreased 1/5 times at 2000ng/ml. caspase-9, however, showed slight increase compared to the control group. From the results obtained in this study, administration of cyclosporin-A to the cell lines of oral squamous cell carcinoma induced alterations in morphology and growth of the cells as its concentration increased. Since apoptosis related factors such as VDAS-2, bcl-2, and caspase-9 also showed distinct alterations on their mRNAs, further research on cyclosporin A as an anti-cancer agent will be feasible.
This study was conducted to investigate composition of understory vegetation and indicator species by altitude and slope azimuth in Mt. Gyebang designated as Protected Area for Forest Genetic Resource Conservation and National Park. Tracheophytes were 350 taxa; 80 families, 203 genera, 303 species, 38 varieties, 5 forma and 4 sub-species in research area. The species of greatest importance value were Tripterygium regelii (9.143%), Acer pseudosieboldianum (7.594%), Symplocos chinensis for. pilosa (6.347%) in the shrub layer and were Sasa borealis (8.653%), Isodon excisus (2.936%) and Carex siderosticta (2.897%). In the herb layer as a result of NMS analysis, the distribution range of the major species were found to be affected by the altitude (shrub layer: $R^2$ > 0.3, herb layer: $R^2$ > 0.6). The result of plexus diagram analysis showed that Acer pseudosieboldianum was associated with Magnolia sieboldii, Acer barbinerve, Euonymus oxyphyllus etc. in the shrub layer; Meehania urticifolia was associated with Aconitum jaluense, Veratrum oxysepalum, Prunus padus etc. in the herb layer. The significant indicator species were analyzed for 60 species by the altitude and investigated for 30 species in accordance with the slope azimuth. As a consequence of MRPP, interspecies composition along the altitude group was heterogeneous and the species composition according to the azimuth slope was extremely different between the NE and SW.

Geoni, a new variety of Auricularia polytricha, was bred in JARES(Jeollanamdo Agricultural Research and Extension Services) in 2012. Geoni was selected through the monospore hybridization with JNM21008 and JNM21014 in 2010. Based on a performance test conducted from 2011 to 2012, the Geoni was selected from a line showing an excellent light brown pileus and strong pest resistance. Geoni has a favorable chewiness, light brown and smooth pileus. In addition, Geoni was rich in dietary fiber. MCM(mushroom complete medium), Malt and PDA(Potato Dextrose Agar) mediums were suitable for cultivating the Geoni. The number of effective stipes was 39 ea/0.9 kg and minor axis and major axis of pileus were 6.9 cm and 8.7 cm respectively. The yield of Geoni was 291 g/0.9 kg in plastic bag. Geoni was required 40~54 days for culture at $20^{\circ}C$ and 24days for the primordia and growth period, which is longer than that of the control(Pung-un). Somatic incompatibility was formed between parental strains and Geoni. Analysis of the genetic diversity of the new variety "Geoni"revealed a different profile from the parental strains when RAPD(random Amplified Polymorphic DNA) primers were used.

The coat protein gene (AF296280) of the Korean isolate Potato leafroll virus (PLRV) was cloned and the open reading frame (627 bp) was transformed into potato (Solanum tuberosum cv. Superior). Out of seventeen individual transgenic lines, five lines were identified to confer resistance to PLRV through the five generation's selection program in the greenhouse as well as isolated trial field. Successful introduction and genetic stability of coat protein gene in the genome of potato were confirmed by polymerase chain reaction (PCR), Southern blot hybridization and northern blot hybridization. Some of the transgenic lines were highly resistant to PLRV but did not show any resistance to less homologous Potato virus Y (PVY). Our results suggest that the resistance to PLRV is due to homology dependent gene silencing by sense strand coat protein gene. In addition, the results of field test through five generations showed that there were no significant differences comparing to nontransgenic potatoes in the morphological aspect of shoot as well as tuber, Ho remarkable differences were also observed in the major agronomic characters and yields except for the resistance to PLRV.

A peculiar virus-like disease of tomato showing yellow mosaic and necrotic spots on leaves and necrosis on veins, petioles and stems was observed at the Tomato Experimental Station (TES), Buyeo, Chungcheongnamdo, Korea. The disease incidence at TES fields ranged from 21 to 35% infecting different tomato cultivars. For this reason, to identify the virus infecting tomato and to characterize the virus based on biology, serology, cytology and at molecular level. Here, leaf samples were randomly collected from different infected tomato cultivars at TES fields and greenhouses and tested by ELISA using Pepper mottle virus (PePMoV) and Tomato mosaic virus (ToMV) antisera. Infected saps were mechanically inoculated in different host plants to test for pathogenicity, symptomatology and host ranges. Infected tissues and ultrathin sections were examined by electron microscopy. Finally, putative coat protein and 3'-untranslated region (CP/3'-UTR) fragment was amplified and cloned for sequence determination and analyzed its genetic relationship to existing PepMoV and PVY sequences at the Genbank. Results showed 69% of the samples were positive with PepMoV, 13% with ToMV and 19 % were doubly infected with PepMoV and ToMV. Symptoms greatly varied from different host plants inoculated with tomato leaf sap infected with PepMoV alone and discussed in detailed in this paper. Electron microscopy from infected tissues showed filamentous particles of 720-750nm in length, a typical morphology and size of PepMoV. In addition, cylindrical inclusion bodies, pinwheels, scrolls and laminates with masses of fibrillar inclusions were also found in ultrathin sections. Alignment of the sequences of the CP/3'-UTR revealed >96% sequence identity with PepMoV and only <61% with PVY. Taken together, all these evidences presented clearly indicated that the causal agent infecting tomato at TES was PepMoV and we designated this PepMoV infecting tomato as Tom-sd2 strain in this study.
A multiple-trait stochastic computer simulation model was constructed to predict the breeding schemes and selection methods on Hanwoo(Korean cattle). The model could be used four kinds of selection criteria (random, phenotype and true or estimated breeding values). At the test run in various population size for 20 years, all estimated parameters of the each simulated populations were resulted similar to input parameters. The deviations between input and output values of parameter in the large population were smaller than in the small population. The simulated results obtained from ten small populations consisted with one sire and ten dams in each population for 500 years were as follows; Inbreeding coefficients of population were similar to theoretical estimating function. Mean values of each traits selected were randomly drifted by generation, but they were converged into a value when inbreeding coefficients came close to one. Additive genetic variances within each population were reduced by generation, and they were converged into zero when inbreeding coefficients came close to one. These results indicated that the simulated populations hold to statistical properties of input parameters.
